Cohorts of yearlings were sampled over a period of 6 years in a retrospective serological survey to establish the annual prevalence of serotype specific antibody to equine encephalosis virus on Thoroughbred stud farms distributed within defined geographical regions of South Africa. Seasonal seroprevalence varied between 3.6% and 34.7%, revealing both single and multiple serotype infections in an individual yearling. During the course of this study serotypes 1 and 6 were most frequently and extensively identified while the remaining serotypes 2, 3, 4, 5 and 7 were all identified as sporadic and localized in fections affecting only individual horses. This study of the seasonal prevalence of equine encephalosis virus has a corollary and serves as a useful model in the seasonal incidence of the serotypes of African horse sickness and bluetongue in regions where the respective diseases are endemic.

This work reports the first molecular analysis study of chicken anaemia virus (CAV) in backyard chickens in Africa using molecular cloning and sequence analysis to characterize CAV strains obtained from commercial chickens and Nigerian backyard chickens. Partial VP1 gene sequences were determined for three CAVs from commercial chickens and for six CAV variants present in samples from a backyard chicken. Multiple alignment analysis revealed that the 6 % and 4 % nucleotide diversity obtained respectively for the commercial and backyard chicken strains translated to only 2 % amino acid diversity for each breed. Overall, the amino acid composition of Nigerian CAVs was found to be highly conserved. Since the partial VP1 gene sequence of two backyard chicken cloned CAV strains (NGR/Cl-8 and NGR/Cl-9) were almost identical and evolutionarily closely related to the commercial chicken strains NGR-1, and NGR-4 and NGR-5, respectively, we concluded that CAV infections had crossed the farm boundary.

Serum samples from 237 randomized rabbits from the five ecological zones of Nigeria, i.e. Northwest (NW), Northeast (NE), Southeast (SE), Southwest (SW) and Northcentral (NC), were evaluated for the presence of antibodies to Encephalitozoon cuniculi by the indirect immunofluorescent antibodies test. A titre of 10 or more was taken as positive. Thirty-nine (16.5 %) of the 237 samples were positive with 11, 10, 8, 6 and 4 seropositive rabbits occurring in the NW, NE, SE, SW and NC zones of Nigeria, respectively. Age, sex, live mass and access to grass as a feed supplement were not statistically (P > 0.05) associated with seropositivity, but cage type (single-versus multi-rabbit type), contact with free-range rats and previous illness were strongly (P < 0.05) associated with it. The practice of selling unscreened and untreated 5 to 10-week-old weaners to prospective buyers as foundation stock, use of multi-rabbit communal cages, occasional release of rabbits in runs and contact with free-range house rats should be discouraged. Regular prophylactic and curative treatments, occasional serological screening to remove carriers, and the practice of a high level of hygiene in rabbit colonies are effective control measures.

Previous studies have shown that crude extracts from Pavetta harborii as well as dried plant material have cardiotoxic effects on rats and sheep that can lead to heart failure. The active component has since been isolated and identified. This substance has been named pavetamine. The aim of this study was to determine whether pavetamine has cardiotoxic effects similar to those seen in previous reports, when administered to rats intraperitoneally. Sprague Dawley rats received two doses, initially 4 mg / kg and then 3 mg / kg pavetamine respectively and were monitored for 35 days before cardiodynamic parameters were measured by inserting a fluid-filled catheter into the left ventricle via the right carotid artery. These values were compared to those of control rats that had received only saline. Pavetamine significantly reduced systolic function and body mass in the treated rats, which indicates that it has the potential to induce heart failure in this animal model.

Lactose in normal saline was administered intravenously to a group of Zebu cattle infected with Trypanosoma vivax to determine the bloodplasma kinetics at onset of an experimental infection and its ability to protect tissues against damage as part of preliminary studies to determine its suitability for use in the treatment of trypanosomosis. Significant (P <0.01) higher lactose concentrations were observed in the T. vivax-intecled bulls at 30 min and 1h (P< 0.05) post-infectio (p.i.) and by 4 h p.i. the plasma lactose remained above the level prior to infusion, after which it fell slightly below the preinfusion level in the uninfected group. Calculated pharmacokinetic parameters revealed delayed excretion of lactose in the T. vivax-intected group soon after infection. The total body clearance (C/B )was significantly (P < 0.05) reduced. The biological half-life (t1/2), elimination rate constant (kel) and apparent volume of distribution (Vd) were relatively decreased (P > 0.05) as a result of the T. vivax infection. Retention of lactose in the plasma was attributed to decreased plasma clearance l.t is suggested that the presence of trypanosomes in circulation rather than organic lesions could have been responsible for the delay observed in the excretion of lactose.At 12 weeks p.i., when the experiment was terminated, the group infected and given lactose infusion (despiteh igherp arasitaemia) had no gross or histopathological lesions in the brain, spleen, lymphnodes, heart, kidneys, liver and testes. However, the group infected but not infused with lactose were emaciated, had pale mucosae, watery blood, general muscular atrophy, serous atrophy of coronary fat and other adiposet issue, hepatomegalys, plenomegalys, wollen and oedematous lymph nodes, all of which are suggestive of trypanosomosis. Histopathological lesions included arrowing of Bowman's space and hypercellularity of glomerular tufts in the kidneys with the mean glomerula truft nucleairn dices (GTNs) in the group significantly higher (P <0.01)than the mean GTNs of the lactoseinfused and control bulls. Degenerative changes occurred in the myocardium, spleen, testes and epididymides. The tesicular and epididymal lesions are indicative of male reproductive dysfunction.

This study investigated the withdrawal periods (WP) of two intramammary antibiotics Cloxamast LC (Intervet SA) and Spectrazol Milking Cow (Schering-Plough Animal Health) in dairy goats and compared them to those recommended for use in cattle. The WP for Cloxamast LC, measured by the Thermo Resistant Inhibitory Substances (TRIS) test, was 60 h in composite samples, 56 h in udder half samples, and the dye was visible for up to 56 h. The WP was significantly shorter than the 72 h recommended WP for use in cattle. It was however significantly longer when the 24 h safety margin (48 h) was subtracted from the recommended WP for cattle. For Spectrazol Milking Cow the antibiotics could be detected by the TRIS test for 61 h in composite samples and 59 h in udder half samples. This did not differ significantly from the recommended 60 h WP for cattle. However, it was significantly longer than that recommended for use in cattle without the 24 h safety margin. There was no significant difference in WP between infected and non-infected udder halves, while there was a weak positive correlation between WP and stage of lactation (R2 = 0.253). There was a moderate positive correlation (R2 = 0.583) between the TRIS test and the presence of dye in milk in udder half samples and between WP in both udder half and composite milk samples (R2 = 0.456). Weak to moderate positive correlations were present between milk yield and the WP in both udder half (R2 = 0.414) and composite (R2 = 0.262) milk samples. Significant differences (P < 0.001) were also observed between the milk yield of udder halves with and without palpable udder damage and between samples that tested TRIS positive and negative on both composite (P = 0.008) and udder half samples (P < 0.001). There was no significant difference between the milk yield of samples with or without dye. There was a significant difference in milk yield between infected and non-infected udder halves (P = 0.054) and a weak negative correlation between milk yield and stage of lactation (R2 = -0.379).

Pigeon paramyxovirus type 1 (PPMV-1), a variant of Newcastle disease virus that primarily affects doves and pigeons has been isolated in South Africa since the mid-1980s. Phylogenetic evidence indicates that pigeon paramyxovirus type 1 viruses were introduced in to South Africa on multiple occasions, based on the presence of two separate lineages, 4bi and 4bii, that have been circulating in Europe and the Far East since the early 1990s. During 2006, a PPMV-1 virus was isolated from an African ground hornbil(l Bucorvus leadbeateri) which becamea cutely infected with PPMV-1 and died, probably after scavenging off infected dove carcasses in the region, since a closely-related PPMV-1 strain was also isolated from doves collected nearby. The hornbill isolate had lCPl and MDT values characteristic of PPMV-1s trains. The threat of PPMV-1 to poultry production and biodiversity in southern Africa highlights the importance of monitoring the spread of this strain.

Acute and long-term effects of a single, relative lyhigh oral dose (0.25a nd 0.30 mg/kg) of sodium monofluoroacetate (1080) on the survival and productivity of sheep were evaluated to establish a better understanding of 1080 poisoning and identify more specific changes diagnostic of toxicosis. In survivors, clinical signs of acute 1080 toxicosis such as salivation and lethar gywere generally very mild. Fasted animals were more prone to 1080 toxicity. In animals that died, more severe signs, including tachypnoea, dyspnoea, and tremors occurred for 15-20 min prior to death. 1080 concentrations were highest in the blood> heart> skeletal muscle> liver. 1080 could not be detected in any of these organs of the animals that survived. Serum citratec oncentratione were elevated for 4 days after dosing. No clinical or biochemical abnormalities were found in any animal after 4 days. Histopathological lesions were most marked in the heart and lung with inflammation, necrosis, and scattered foci of fibrous tissue in the myocardium, pulmonary oedema and inflammation of the lung. No adverse longterm effects on general health or reproductive performance were observed in any sheep that survived the first 4 days following exposure to 1080. The most reliable diagnostic in dicators of 1080 exposure in sheep were measurement of its residues in blood, skeletal muscle and ruminal contents, increased serum citratec oncentratione; l evated heart rate,and characteristic electrocardiograpchh anges(up to 4 days after exposure). Death from 1080 is most likely to occur within 96 h, and animals that survived this period appeared normal.

In a preliminary survey conducted in 2005, the species composition and seasonality of ticks infesting cattle in Central Equatoria State, Southern Sudan was determined. Three locations were selected (Gumbo, Khor Rumla and Nyaing) and surveyed every 3 months. Two cattle herds in each of the three locations were visited four times during the study period. Total body collections of ticks were made from each of five cattle (Nilotic Zebu breed) kept in six different herds. Four tick genera and ten species were identified. The tick species identified were Amblyomma lepidum, Amblyomma variegatum, Boophilus annulatus, Boophilus decoloratus, Hyalomma marginatum rufipes, Hyalomma truncatum, Rhipicephalus appendiculatus, Rhipicephalus evertsi evertsi, Rhipicephalus praetextatus and Rhipicephalus sanguineus group. The highest number of ticks was collected in October during the rainy season. A finding of great significance was that R. appendiculatus, vector of East Coast fever, has now firmly established itself throughout the year with possible implications for cattle production in Central Equatoria State.

The prevalence and geographic distribution of the fowl tampan, Argas walkerae Kaiser & Hoogstraal, 1969 was determined in the eastern region of the Eastern Cape Province, South Africa by inspecting two fowl houses in the vicinity of each of 72 randomly selected communal cattle dip-tanks. Tampans were collected from 102 (70.8 %) of the 144 fowl houses in the neighbourhood of 57 (79.2 %) of the 72 selected dip-tanks, and the localities of the collections were mapped. Argas walkerae was present in fowl houses from the warm coastal regions of the Indian Ocean in the south to the cold and mountainous Drakensberg in the north-east of the Province. Taking into account the probable sensitivity of the sampling method, it is estimated that A. walkerae is likely to be present in fowl houses belonging to between 74 and 84 % of communities making use of cattle dip-tanks in the eastern region of the Eastern Cape Province, and that when it is present, between 64 and 75 % of fowl houses will be infested. The geographic distribution of A. walkerae seemed to be more strongly associated with the presence of fowls and fowl houses containing raw or processed wood in their structure than with climate.
