Evaluation and development of diagnostic tools for rapid detection of Riemerella anatipestifer and Pasteurella multocida in ducks

Objectives: Ducks suffer a huge economic loss as a result of infections with Pasteurella multocida and Riemerella anatipestifer, which cause high morbidity and mortality. Because these patho¬gens induce similar clinical symptoms when coinfections occur, it is very difficult to differentiate between them based just on clinical signs. Hence, these major pathogens must be quickly and accurately detected. Materials and Methods: A total of 104 birds ranging from 2 days to 4 weeks old were collected from Egyptian farms, and the outcomes were compared statistically. Conventional cultural iden¬tification procedures and a direct multiplex polymerase chain reaction assay were utilized to recognize both pathogens in a single tube reaction simultaneously. Then, the obtained isolates were characterized phenotypically and genotypically. Results: Clinical signs appear at 2–4 weeks of age with respiratory distress (dyspnea), white fluid feces, and stunting. The scrutinized data demonstrated a significantly higher detection rate by PCR directly compared to classical culture procedures. Pasteurella multocida was detected only by PCR. The disc diffusion technique against ten antibiotics showed absolute susceptibilities to amik¬acin, doxycycline, and florfenicol. High levels of beta-lactam resistance were observed. Riemerella anatipestifer isolates were screened for pathogenicity and plasmid-borne blaTEM genes. All six isolates harbored five virulence genes: aspC, RA46, m28, pstS, and Nlp/P60. Moreover, blaTEM was identified into four isolates and deposited to GenBank with accession numbers OP347083, OP347084, OP347085, and OP347086. Conclusion: These results suggest advanced PCR assays can be applied to the field for rapid and valuable diagnosis of two significant pathogens and focus on the worth of ducks in the propaga¬tion of transferable antibiotic resistance genes into the environment. Objective: This study was conducted to analyze the matrilineal structure of Gallus sp. in Southeast Asia, especially in the D-loop region. Materials and Method: A total of 563 sequences from eight countries (Laos, Myanmar, Vietnam, Malaysia, Indonesia, Cambodia, the Philippines, and Thailand) in Southeast Asia are used in this study. Data collected from National Center for Biotechnology Information (NCBI) regarding the genus Gallus sp. in a Southeast Asian country. Data analysis was performed using MEGA 7.2 and DnaSP v6. Results: In the haplotype found in Gallus sp. in Southeast Asia, there are 89 haplotypes. Using a neighbor-joining (Nj) analysis, 89 haplotypes found three haplogroups for Gallus sp. in Southeast Asia. In Southeast Asia, the genetic diversity of the d-loop is exceptionally high, with a haplotype diversity value of 0.524 to 1. Conclusion: D-loop cannot be used as a specific marker for breeds or country-specifics. [J Adv Vet Anim Res 2023; 10(4.000): 817-819]

Objective: The aim of this study is to detect the prevalence and diagnosis of molecular characteristics of Cryptosporidium infection in handlers of domestic pigeons in Baghdad City. Traditional and molecular diagnostic methods were employed to detect and identify Cryptosporidium species. Materials and methods: Sixty stool samples were collected from the handlers of domestic pigeons, and various techniques, including direct smear, flotation concentration, staining methods, and DNA extraction coupled with nested polymerase chain reaction (PCR), were utilized. Results: The results obtained from traditional methods indicated an overall infection rate of 55% in handlers of domestic pigeons, while significant variations were noted between male and female handlers. Age group 21–40 years were found to have higher infection rates were found to have higher infection rates. The prevalence of Cryptosporidium also displayed temporal variability throughout the study period. Molecular analysis using nested PCR revealed higher infection rates of 86% in handlers of domestic pigeon samples. Genotyping and phylogenetic analysis identified the presence of Cryptosporidium parvam species in handlers of domestic pigeons, indicating zoonotic transmission potential. Objective: Protein and carbohydrates are substances needed by ruminants, especially sheep. Providing protein and carbohydrates must pay attention to their degradation. In addition, balancing nutrients to meet the nutritional needs of rumen microbes is very important because the unbalanced availability of rumen degradable protein (RDP) and readily available carbohydrate (RAC) at one time can cause suboptimal microbial protein synthesis efficiency. Materials and Methods: Completely randomized design with a nonfactorial pattern of five treatments with three replications. Treatment includes RDP and RAC ratios, namely R1 = 2.30, R2 = 2.00, R3 = 1.70, R4 = 1.50, and R5 = 1.30. Results: The results showed that the balance of RDP and RAC in sheep ration formulation in vitro had a very significant effect (p < 0.01) on NH3, microbial protein synthesis, total gas, total microbes, and organic matter digestibility (OMD) but had no significant effect (p > 0.05) on rumen pH and dry matter digestibility (DMD). Conclusion: The optimal balance of RDP and RAC in the formulation of sheep rations in vitro was obtained at a ratio of 2.30 with NH3 (mM) 8.47, rumen pH 5.97, microbial protein synthesis (mg/100 ml) 123, gas (ml/g of material) 145, total microbes (cells/ml) 2.012 × 106, (log CFU cells/ml) 6.3025, DMD 61.0%, and OMD 63.1%. Objective: This study investigated the effectiveness of different physical forms of feed containing Ipomoea aquatica waste and concentrate feed on the rumen characteristics, chewing activity, and performance of sheep. Materials and Methods: Twenty-four rams (19.87 ± 2.19 kg) were arranged in a completely randomized design. Rams were fed dried I. aquatica waste and concentrate feed provided separately (RCF) (conventional feeding system), and total mixed ration consisted of mash complete feed (MCF), and pelleted complete feed (PCF). The data were analyzed using a one-way analysis of variance. Results: The dry matter intake (DMI), average daily gain, and feed conversion ratio of rams fed different physical forms of feed containing I. aquatica waste in their diet were similar between the treatments, ranging from 4.08% to 4.29% of body weight, 120–180 gm, and 6.32–9.17, respectively. Different physical feeds did not affect microbial synthesis in the rumen. Methane emissions per unit of production were similar between the groups. The PCF sheep ate faster (0.24 min/gm DMI) than the MCF sheep (0.38 min/gm DMI), but similar to the RCF sheep (0.31 min/gm DMI). Conclusion: Dried I. aquatica waste was useful as an alternative lamb feed roughage during the dry season. The pelleted mixed ration was more efficient than mash in increasing DMI. The dried I. aquatica waste was environmentally friendly for mitigating enteric methane emissions by sheep. [J Adv Vet Anim Res 2023; 10(4.000): 677-684]

Objective: This study aims to evaluate the effect of Clostridium perfringens sialidase treatment on monolayer cell behavior using computational screening and an in vitro approach to demonstrate interaction between enzyme-based drugs and ligands in host cells. Materials and Methods: The in silico study was carried out by molecular docking analysis used to predict the interactions between atoms that occur, followed by genetic characterization of sialidase from a wild isolate. Sialidase, which has undergone further production and purification processes exposed to chicken embryonic fibroblast cell culture, and observations-based structural morphology of cells compared between treated cells and normal cells without treatment. Results: Based on an in silico study, C. perfringens sialidase has an excellent binding affinity with Neu5Acα (2.3) Gal ligand receptor with Gibbs energy value (ΔG)—7.35 kcal/mol and Ki value of 4.11 μM. Wild C. perfringens isolates in this study have 99.1%–100% similarity to the plc gene, NanH, and NanI genes, while NanJ shows 93.18% similarity compared to the reference isolate from GenBank. Sialidase at 750 and 150 mU may impact the viability, cell count, and cell behavior structure of fibroblast cells by significantly increasing the empty area and perimeter of chicken embryo fibroblast (CEF) cells, while at 30 mU sialidase shows no significant difference compared with mock control. Conclusion: Sialidase-derived C. perfringens has the capacity to compete with viral molecules for attachment to host sialic acid based on in silico analysis. Objective: Cadmium chloride (CdCl2) is an environmentally toxic pollutant that can cause reprotoxicity. Cyperus esculentus and Euterpe oleracea are potent antioxidant plants currently used to counteract the action of harmful pollutants. The present experiment was intended to evaluate and comp are the role of C. esculentus hydroethanolic extract (CHE) and E. oleracea in treating the reprotoxicity induced by CdCl2 in rats. Materials and Methods: Forty adult male rats (160–210 gm) were allocated into five groups equally. Control group: received 5 ml of normal saline (NS); the other treatment groups were injected with CdCl2 as a single dose for two weeks to induce testicular toxicity. After 14 days, the four groups were treated orally daily for two months as follows: The cadmium group (Cd) received NS, the third group (TC) was administered 800 mg/kg BW of CHE, the fourth group (TO) received 500 mg/kg BW of E. oleracea, and the fifth group (TCO) received CHE with E. oleracea. Results: The live sperm and motility, serum testosterone, follicle-stimulating hormone (FSH), testicular superoxide dismutase (SOD), catalase (CAT), steroidogenic acute regulatory protein (StAR), 17β-hydroxysteroid dehydrogenase, and 3β-hydroxysteroid dehydrogenase (3β-HSD) were significantly increased in the TCO, TC, and TO groups compared with the Cd group. Testicular nitric oxide and malondialdehyde were elevated significantly in the Cd group compared to the TC, TO, TCO, and control groups. The fold changes of Fshβ, Lhβ, and Gnrh genes were upregulated in the TCO group compared to the Cd and control groups. Conclusion: The combination of CHE with E. oleracea showed improvements in rat testicles affected by cadmium toxicity via upregulated reproductive gene expression and its antioxidant effects. Objective: The aim of the study was to use a meta-analysis to identify the correlation between linear body measurements, including body length (BL), wither height (WH), heart girth (HG), and body volume (BV), and body weight in beef cattle by breed, sex, and age as categories. Materials and Methods: These results can be used as a method for predicting beef cattle body weight. This study used systematic review and meta-analysis guidelines to create a checklist. The first stage was searching for papers relevant to the study objectives. The second stage was searching using the keywords beef cattle, body weight, body measurement, and correlation. The third stage was reviewing the title and abstract. The fourth stage was abstracting information from selected papers, and the last stage was tabulating data. Results: The results from this study were obtained, and 32 papers were eligible for the meta-analysis stage. The correlation between linear body measurement and body weight of beef cattle showed that HG (r = 0.88) and BV (r = 0.97) were significantly (p < 0.05) different compared to BL (r = 0.74) and WH (r = 0.72). The correlation between HG and body weight, and the categorization of cattle breeds showed significantly (p < 0.05) different results. The correlation between BV and body weight of cattle according to breed categories showed results that were not significantly (p > 0.05) different, while age was significantly (p < 0.05). Conclusion: In conclusion, to predict beef cattle body weight, it is necessary to use HG or BV, with breed, sex, and age of cattle as categories. [J Adv Vet Anim Res 2023; 10(4.000): 630-638]

Objective: Andaliman (Zanthoxylum acanthopodium) is a potent medicinal plant in Asia. This present study aimed to reveal the effectivity of Andaliman fruit extract in alleviating hyperglycemia, sensory and motoric balance disorders, histopathology of the cerebellum, and tissue oxidative stress in diabetic mice induced by alloxan. Materials and Methods: Diabetes induction was performed by intraperitoneally injecting alloxan monohydrate [200 mg/kg body weight (BW)]. Subsequently, the mice were treated daily with an ethanolic extract of Andaliman fruit (0, 150, 300, 450 mg/kg BW per oral) for 28 days, followed by measurements of blood glucose, paw sensitivity, motoric balance, histopathology of the cerebellum, and malondialdehyde (MDA) levels. Moreover, the phytochemical constituents of the extract were elucidated by liquid chromatography. Results: Higher doses of Andaliman fruit extract could significantly attenuate the elevation of random and fasting blood glucose (p < 0.05) and improve paw sensitivity responses (p < 0.05) and motoric balances (p < 0.05) in diabetic mice. Moreover, Andaliman fruit extract could significantly attenuate the degeneration of cerebellar Purkinje cells (p < 0.05) and suppress MDA levels in the blood (p < 0.05) while blunting the MDA in the brain tissue (p < 0.05). Phytochemical screening revealed 39 compounds in the Andaliman extract belonging to the groups of alkaloids (26 compounds), flavonoids (12 compounds), and terpenoids (1 compound). Conclusion: The ethanolic extract of Andaliman fruit is capable of ameliorating diabetic neuropathy, motor balance disorders, and Purkinje cell degeneration while also reducing oxidative stress in the peripheral system. Hence, Andaliman extract is a promising candidate for formulation as an herbal remedy against the detrimental outcomes of diabetes mellitus. [J Adv Vet Anim Res 2023; 10(4.000): 608-619]

Objective: This research aims to investigate the microbial diversity of Budu prepared from fresh and frozen fish from the Pariaman and Pasaman districts in West Sumatra Province, Indonesia, as well as provide basic information about Budu quality. Materials and Methods: To obtain the bacterial microbial composition, deoxyribonucleic acid extraction was carried out using amplicon-sequencing of the 16S-rRNA gene in the V3–V4 region from two types of Budu and carried out in duplicate. Results: Budu prepared with fresh (Pariaman) or frozen (Pasaman) fish was dominated by Firmicutes (78.455%–92.37%) and Proteobacteria (6.477%–7.23%) phyla. The total microbial species in Budu from Pariaman were higher (227 species) than in Pasaman (153 species). The bacterial species found are Lentibacillus kimchi (1.878%–2.21%), Staphylococcus cohnii (0.597%–0.70%), Peptostreptococcus russeli (0.00%–0.002%), Clostridium disporicum (0.073%–0.09%), Clostridium novyi (0.00%–0.01%), Nioella sediminis (0.00%–0.001%), and Shewanella baltica (0.00%–0.003%). Lentibacillus kimchi, S. cohnii, and C. disporicum are found in both Budu. Nioella sediminis and S. baltica are found in Budu Pariaman. Peptostreptococcus russeli and C. novyi were found in Budu Pasaman. Conclusion: Metagenomic analysis of Budu from different fish, Pariaman (fresh fish) and Pasaman (frozen fish) showed that the biodiversity of bacteria was barely different. Both Budu found lactic acid bacteria from the Enterococcaceae family, genus Vagococcus, and pathogenic bacteria, such as S. cohnii, P. russeli, C. disporicum, and S. baltica. The discovery of various species of pathogenic bacteria indicates that development is still needed in the Budu production process to improve Budu quality.
