Evaluation of systemic and mucosal immune responses in mice administered with recombinant Salmonella Typhimurium expressing IutA protein

Avian pathogenic Escherichia coli (APEC) are known to cause extraintestinal disease in poultry, leading to substantial losses in the industry. IutA, iron-regulated aerobactin receptor is firmly associated with APEC. To assess the potential of IutA to induce protective immune responses, attenuated Salmonella Typhimurium strain expressing IutA was constructed and administered orally to BALB/c mice. The IutA-specific immune responses were measured with sera, vaginal and fecal samples by an enzyme-linked immunosorbent assay. We found that the Salmonella-IutA vaccine induced significantly higher immune responses as compared to the control inoculated with the attenuated S. Typhimurium containing the plasmid only. The IutA-specific immune responses were increased by second immunization at third week after initial immunization, whereas triple immunization induced lower immune responses than those induced by the double immunization. The Salmonella-IutA vaccine induced a nature of immunity biased to the Th1- type, as judged by the ratio of IutA-specific IgG isotypes (IgG2a/IgG1). Overall, these results suggest that the Salmonella-IutA vaccine appear to be suitable candidate for a vaccine against APEC. Keywords :

Transgenic plants have been tested as an alternative host for the production and delivery of experimental oral vaccines. Here, we developed transgenic potatoes that express the major antigenic sites A and D of the glycoprotein S from transmissible gastroenteritis coronavirus (TGEV-S0.7) under three expression vector systems. The DNA integration and mRNA expression level of the TGEV-S0.7 gene were confirmed in transgenic plants by PCR and northern blot analysis. Antigen protein expression in transgenic potato was determined by western blot analysis. Enzyme-linked immunosorbent assay results revealed that based on a dilution series of Escherichia coli-derived antigen, the transgenic line P-2 had TGEV-S0.7 protein at levels that were 0.015% of total soluble proteins. We then examined the immunogenicity of potato-derived TGEV-S0.7 antigen in mice. Compared with the wild-type potato treated group and synthetic antigen treated group, mice treated with the potato-derived antigen showed significantly higher levels of immunoglobulin (Ig) G and IgA responses.

This study evaluated two different surgical techniques for the treatment of palmar annular ligament desmitis (PAL) in horses. The group 1 (G1) consisted of 11 Thoroughbred submitted to desmotomy, and G2 of 22 horses that underwent PAL desmectomy. There was no statistically significant difference between mean palmar annular ligament thickness in both groups evaluated (p = 0.800). In this study, the horses of G1 had 12 times more chance to return to physical activity, when compared to animals of G2 (p = 0.033; O.R. = 12.0; C.I.95% = 1,142-126,122). Desmectomy was more efficient in promoting the resolution of the injury and the return to sports activity.

The use of haematological techniques to assess fish health is generally accepted. The aim of the current study was to determine selected haematological changes that occur in Clarias gariepinus (Burchell, 1822). infected with trypanosomes. Blood films were prepared according to standard techniques to confirm trypanosome infections and whole blood was collected, the serum and plasma separated, and prepared for albumin and total protein concentration analysis. Plasma albumin levels were significantly higher in infected wild caught fish than in uninfected wild caught fish and uninfected breeding stock. Serum albumin levels were significantly lower in infected wild caught fish when compared to uninfected breeding stock. The total plasma and serum protein levels were within the normal range for C. gariepinus, that is, 3 g - 6 g/100 mL. The total plasma protein levels varied significantly between the three groups. However, the total serum protein levels were only significantly different between uninfected breeding stock and uninfected wild caught fish, as well as uninfected breeding stock and infected wild caught fish. The total protein levels were significantly higher in infected wild caught fish than in the other groups, a possible indication of an infection or inflammatory host response.

Objective-To characterize biomechanical differences in gait between dogs with and without an amputated thoracic limb. Animals-Client-owned dogs (16 thoracic-limb amputee and 24 quadruped [control] dogs). Procedures-Dogs were trotted across 3 in-series force platforms. Spatial kinematic and kinetic data were recorded for each limb during the stance phase. Results-Amputees had significant increases in stance duration and vertical impulse in all limbs, compared with values for control dogs. Weight distribution was significantly increased by 14% on the remaining thoracic limb and by a combined 17% on pelvic limbs in amputees. Braking ground reaction force (GRF) was significantly increased in the remaining thoracic limb and pelvic limb ipsilateral to the amputated limb. The ipsilateral pelvic limb had a significantly increased propulsive GRF. The carpus and ipsilateral hip and stifle joints had significantly greater flexion during the stance phase. The cervicothoracic vertebral region had a significantly increased overall range of motion (ROM) in both the sagittal and horizontal planes. The thoracolumbar vertebral region ROM increased significantly in the sagittal plane but decreased in the horizontal plane. The lumbosacral vertebral region had significantly greater flexion without a change in ROM. Conclusions and Clinical Relevance-Compared with results for quadruped dogs, the vertebral column, carpus, and ipsilateral hip and stifle joints had significant biomechanical changes after amputation of a thoracic limb. The ipsilateral pelvic limb assumed dual thoracic and pelvic limb roles because the gait of a thoracic limb amputee during trotting appeared to be a mixture of various gait patterns.

Objective—To evaluate the effects of perioperative oral administration of carprofen and meloxicam on concentrations of 3 acute-phase proteins in dogs undergoing elective ovariohysterectomy (OVH). Animals—18 healthy adult anestrous female dogs undergoing elective OVH. Procedures—Dogs were allocated to 3 groups (6 dogs/group). A placebo treatment, carprofen (2.0 mg/kg), or meloxicam (0.2 mg/kg) was orally administered to the dogs of the respective groups. The initial doses were administered 30 minutes before premedication prior to OVH; additional doses were administered once daily for 4 days after surgery. Blood samples were collected 45 minutes before premedication and 4, 8, 12, 24, 36, 48, 72, 96, and 120 hours after the end of OVH; samples were used for measurement of total WBC and neutrophil counts and concentrations of C-reactive protein (CRP), ceruloplasmin, and fibrinogen. Results—Values did not differ significantly among groups for WBC and neutrophil counts, serum concentrations of CRP and ceruloplasmin, and plasma concentrations of fibrinogen. Concentrations of all inflammatory markers, except serum ceruloplasmin, increased significantly following OVH, but in a similar manner for each group. No significant changes were detected in serum ceruloplasmin concentrations over time. Conclusions and Clinical Relevance—Perioperative administration of both carprofen and meloxicam did not significantly affect the concentrations of CRP, ceruloplasmin, and fibrinogen in dogs undergoing OVH. Thus, use of carprofen or meloxicam should not affect clinical interpretation of results for these 3 acute-phase proteins.

The rabbit corpus luteum becomes an estradiol-dependent tissue by day 6 of gestation, and adequate estrogen is critical to avoid pregnancy failure. The aim of this study was to investigate the effect of parity (primiparous or multiparous), day of mating (11 or 21 d postpartum), and suckling status (suckling or nonsuckling) on various reproductive traits in hybrid rabbit does (n = 96). Ovarian structures on day 6 after coitus were evaluated by means of ultrasonography. Blood samples were collected that day, and the serum was analyzed for estradiol-17β by radioimmunoassay (RIA). Parity and suckling had significant effects on mating rate (P < 0.01 and P < 0.05, respectively). More does accepted the male on day 11 than on day 21 (P < 0.05). Ovulation frequency was significantly affected by parity (P < 0.05), day of mating (P < 0.01), and suckling (P < 0.01). Fewer ovarian large follicles and lower estradiol-17β levels were detected in suckling compared with nonsuckling rabbits (P < 0.01). Since estrogen concentrations are commonly used to assess follicular growth and steroidogenic capacity, the lower hormonal levels in the suckling rabbits may reveal poorer ovarian activity, which could result in reduced reproductive efficiency. Our observations confirm the existence of a partial antagonism between lactation and reproduction in rabbits. Further research is needed to elucidate these phenomena, including when artificial insemination is done. Ultrasonography could represent a noninvasive and reliable method for studying several reproductive functions and dysfunctions in rabbits.

This study was done to evaluate the hypoglycemic potentials of ginger (zingiber officinale ) in controlling hyperglycemia ,hypoinsulinemia and their effect on total cholesterol in cadmium-induced pancreotoxicity in male rabbits Twenty four of adult male rabbits were divided randomly into four equal groups. Group 1 was given distilled water orally and considered as control group, group 2 received 10mg/kg B.w of cadmium chloride, group 3 received 100mg/ kg B.w of ginger extract plus 10mg/kg B.w of cadmium chloride and group 4 received 200mg/ kg B.w of ginger extract plus 10mg/kg B.w of cadmium chloride. All treatments were given a daily dose for 35 days. The blood sample was taken from these animal groups after 35 days of treatment for biochemical analysis to estimate : insulin hormone, blood glucose ,total cholesterol, Aspartate amino transferase (AST) , Alanine aminotranferase(ALT) and Alkaline phosphatase (ALP). The results showed that cadmium chloride administration resulted in a decrease in the level of insulin concentration associated with an increase in glucose and total cholesterol concentration in rabbits .The enzyme activities of serum ALT, AST and ALP were also significantly increased . While , the groups of animal that received ginger showed significant decrease in serum glucose , cholesterol , AST, ALT and ALP. The level of insulin was also increased in these groups of animal.

Effect of age and sex were measured during and before sexual maturation of golden local quail males and females. Maels erythrocytes count showed higher value compared with that of adult females . Haemoglobin and haematocrit of adult males were significantly higher than those of adult females. However total leukocyte count and H/L ratio of the adult female were higher than those of males. Diabetes mellitus (DM) a disease which is characterized by hyperglycemia, lipoprotein abnormalities and oxidative stress. This study was undertaken to investigate the beneficial effect of vitamin C and Zinc on some biochemical parameter when induced diabetic in rabbits by alloxan. Male rabbits groups divided into four groups: Group I (Negative control) received normal saline orally, alloxan diabetic rabbits : Group II (Positive control) received normal saline orally , while Group III were orally administered 50 mg/kg body weight of vitamin C and group IV were orally administered 100 mg/kg body weight Zinc for 15 days. The results showed that vitamin C administration and zinc significantly (P< 0.05) reduced the serum glucose, total cholesterol, triglyceride, ALT and AST concentration, while there were significant changes (p< 0.05) was observed on the serum levels of total protein, albumin, globulin and albumin/ globulin ratio in diabetic group treated with vitamin C, Zinc respectively.
