Changes in viable bacterial counts and physicochemical parameters of water used during live transportation of Pangasius catfish (Pangasianodon hypophthalmus) in Bangladesh

Objective: The study was undertaken to assess the changes in viable bacterial counts and physicochemical parameters of water used during the live transportation of Pangasius catfish (Pangasianodon hypophthalmus). The correlations between the changing patterns of these parameters were also established. Materials and Methods: Water samples were collected every 2 h interval, plated onto agar plates for assesing viable bacterial counts. Physicochemical parameters, namely, water temperature, dissolved oxygen (DO), pH, and ammonia (NH3) concentration in the water were measured with a glass thermometer, DO test kit, pH test kit, and total NH3 measurement kit, respectively. Results: The viable bacterial counts increased significantly from 0 to 2 h in all the studied chan¬nels and remained almost similar up to the end of the supply channels. The water temperature was almost stable regardless of the supply channels and transportation period at around 30°C. The DO concentration and pH level decreased, and NH3 concentrations increased gradually in all the supply channels. The viable bacterial counts were inversely correlated to the DO levels and directly associated with the NH3 concentrations of the water used during the live transportation of Pangasius catfish. Conclusions: Gradual increase in viable bacterial counts and fluctuation in some vital physicochemical parameters with the duration of transportation indicated an unfavorable environment for the survival of Pangasius catfish. Objectives: The goal of this research was to figure out the effect of the local sorghum as silage on the performance of Bligon goats. Microbial protein synthesis, digestible nutrients, and average daily weight gain (ADWG) were measured to evaluate the goats' performance. Materials and Methods: The study was designed in a completely randomized design with a one-way pattern. Twelve female Bligon goats with 24.33 ± 2.83 kg (mean ± SEM) initial body weight were divided into three groups of total mixed ration (TMR) treatments. Group 1 received fresh Napier grass (FNG) as a control, group 2 received imported sorghum silage (ISS) of brown midrib resistance (BMR), and group 3 received local sorghum silage (LSS) of super-2. Analysis of variance was used to analyze the data on microbial protein synthesis and feed intake during the research. On the contrary, analysis of covariance was used to analyze ADWG with initial weight as a covariate. Results: Microbial protein synthesis, feed-intake, and ADWG of goats that received TMR based on silage of two varieties of sorghum, namely BMR (ISS) and super-2 (LSS), were lower (p < 0.05) than control. However, there was no significant difference between both TMR based on sorghum silages. ISS's feed conversion was better than LSS (p < 0.05), and FNG was the best. Sorghum silage as a basal ration in TMR had lower microbial protein synthesis but higher total digestible nutrient content than fresh forage, such as Napier grass. The sorghum varieties did not affect the microbial protein synthesis, digestible nutrients, and ADWG of Bligon goats. However, ISS treatment had higher feed efficiency than LSS. Conclusion: The local sorghum (super-2) silage can be used as ruminant feed as well as imported sorghum (BMR) offered as TMR. However, regardless of the cultivar, TMR based on sorghum silage cannot replace TMR based on fresh Napier grass. [J Adv Vet Anim Res 2022; 9(2.000): 175-183]

Objective: The research was conducted to determine β-carotene and antioxidant activities and screening of phytochemical substances of Moringa oleifera extraction using organic solution. Materials and Methods: 550 gm of M. oliefera leaf flour was macerated. This research was conducted by laboratory experiments using the maceration method. The extraction was performed using three kinds of solvents, which are n-hexane, ethyl acetate, and methanol; for 3 x 24 h, they were concentrated with a rotary evaporator. Then, the flavonoid, phenolic, β-carotene isolation, and antioxidant tests were conducted using the 2,2-diphenyl-1-picrylhydrazyl on each fraction (n-hexane, ethyl acetate, and methanol). Results: The results of weighing each concentrated extract from the maceration process of each fraction (n-hexane, ethyl acetate, and methanol) were 12.67, 35.67, and 49.29 gm, with the total phenolic content (1.4595 ± 0.361, 46.5489 ± 1.832, and 39.74574 ± 0.786) and total flavonoid content of each fraction (3.3056 ± 0.039, 58.6389 ± 2.051, and 48.9056 ± 0.0809), respectively. The antioxidant activity test on the crude extract from the ethyl acetate fractionation showed that the IC50 value was 30.309 mg/ml. The ethyl acetate fraction has a high total phenolic and flavonoid content. The results of the isolation of β-carotene from M. oleifera leaf flour were 0.4798 gm, or equivalent to 0.956% carotenoids. Conclusions: Based on the results of the research, M. oleifera leaves are identified to have a fairly high antioxidant activity, which is 30.309 mg/ml, resulting from the potential compounds in M. oleifera leaves that function as inhibitors of antioxidant activity, which are the groups of phenolic and flavonoid compounds. Objective: Brucellosis is an endemic zoonotic infection in Saudi Arabia, including Jazan Province, and epidemiological reports about this disease are limited. In this study, we aimed to investigate knowledge, attitudes, and practices regarding brucellosis among the general population of Jazan Province, southwestern Saudi Arabia. Materials and Methods: This is a cross-sectional study conducted using a pretested questionnaire that assessed the level of knowledge, attitudes, and practices toward Brucellosis. The calculated sample size was 384, and 1,055 participants were included representing various genders, ages, and levels of education. Data were collected between March and April 2022 and analyzed using descriptive, chi-square, and t-test analyses. Results: Only 50% of the included participants have heard about Brucellosis. Among those who had heard about Brucellosis, 70% had a good knowledge about Brucellosis and that was significantly associated with male gender, being healthcare workers (HCWs), and having a higher level of education. Participants who directly involved in animal care represented 9%, and attitudes and practices regarding brucellosis were varied from average to unsatisfactory. This variation was significantly affected by the level of knowledge. Conclusion: About 50% of the general population of Jazan Province had never heard about Brucellosis, in a region found prevalent for this disease. Good knowledge was recorded in 70% of those who were aware of Brucellosis, which was significantly associated with the male sex, job as being HCW, and having a higher level of education. These results necessitate public awareness campaign activities to improve knowledge and practices, especially among women, the younger generation, and individuals with limited educational backgrounds. Objective: As one of the most valuable genetic resources of Ongole beef cattle globally, the Sumba Ongole (SO) cattle population is being studied in this investigation of myostatin (MSTN) gene polymorphism and its association with growth traits. Materials and Methods: Blood samples from 161 SO cattle were collected and analyzed. Deoxyribonucleic acid (DNA) was isolated. The DNA was electrophoresed and extracted, and finally, the annealing temperature was optimized, followed by amplification and sequencing. Next, we used a Basic local alignment search tool to assess the sequencing data. Results: The analysis revealed 22 single nucleotide polymorphisms (SNPs) in the MSTN gene in this region that showed genetic variation. Two SNPs, c.424 G > A, and c.467 G > C, were found to be significantly associated with SO cattle phenotypes of wither height, heart girth, and hip height (p < 0.05) but not with body weight or body length (p > 0.05). Conclusion: As a result of our findings, the MSTN gene polymorphism and its correlation with growth traits in SO cattle may be employed as a candidate marker in SO cattle and other beef cattle breeds. Objective: This study describes the newest deoxyribonucleic acid (DNA) barcoding Asian redtail catfish (Hemibagrus nemurus) in the Progo River, Magelang, Central Java, Indonesia. Materials and Methods: Ten fish were caught in the Progo River, Magelang, Central Java, Indonesia. The polymerase chain reaction was the molecular diagnosis to detect the sequences of DNA of Cytochrome Oxidase I compared to National Center for Biotechnology Information data (GenBank). Results: The results showed that the percent identity was not 100% with H. nemurus data from other locations (GenBank), including Indonesia. The closest percent identity was H. nemurus from Java Island (Accession ID: MK312566.1) with 97.6% similarity. The genetic mutation that hap¬pened might be due to environmental change (pollution) in the Progo River recently. Conclusions: This study showed a genetic mutation in H. nemurus from Progo River may be caused by environmental change. Low pollution exposure levels may not be detrimental (lethal) to fish. However, it can affect fish fertility, which leads to population degradation (gene variation). Attention must be increased for fish survival in the future. Objectives: This study aimed to develop a computerized deep learning (DL) technique to identify bacterial genera more precisely in minimum time than the usual, traditional, and commonly used techniques like cultural, staining, and morphological characteristics. Materials and Methods: A convolutional neural network as a part of machine learning (ML) for bacterial genera identification methods was developed using python programming language and the Keras API with TensorFlow ML or DL framework to discriminate bacterial genera, e.g., Streptococcus, Staphylococcus, Escherichia, Salmonella, and Corynebacterium. A total of 200 dig¬ital microscopic cell images comprising 40 of each of the genera mentioned above were used in this study. Results: The developed technique could identify and distinguish microscopic images of Streptococcus, Staphylococcus, Escherichia, Salmonella, and Corynebacterium with the highest accuracy of 92.20% for Staphylococcus and the lowest of 77.40% for Salmonella. Among the five epochs, the accuracy rate of bacterial genera identification of Staphylococcus was graded 1, and Streptococcus, Escherichia, Corynebacterium, and Salmonella as 2, 3, 4, and 5, respectively. Conclusion: The experimental results suggest using the DL method to predict bacterial genera included in this study. However, further improvement with more bacterial genera, especially of similar morphology, is necessary to make the technique widely used for bacterial genera identification. Objective: The study was conducted to determine the effect of inoculants of different types and doses on the nutrient quality and in vitro digestibility of fermented rice bran. Materials and Methods: The study was designed using a completely randomized design with a 3 × 3-factorial pattern. The first factor was the type of inoculum, consisting of Saccharomyces cerevi¬siae (SC), Effective Microorganism-4, and Saus Burger Pakan (SBP). The second factor is inoculum doses, which are as follows: levels 2%, 4%, and 6%. The variables measured included chemical composition, fiber fraction content, dry matter digestibility and organic matter digestibility. Results: The results showed that the type of inoculation treatment and the doses of inoculation did not affect the dry matter (DM) content of fermented bran, and the organic matter content of fermented bran was only affected by the inoculation dose factor (p < 0.05). The highest crude protein and Extract Ether (EE) were obtained in the SBP inoculants, which increased linearly with increasing inoculation doses (p < 0.05). While a significant decrease (p < 0.05) occurred in crude fiber content. The cellulose, hemicellulose, lignin, acid detergent fiber (ADF), and neutral deter¬gent fiber (NDF) fractions were significantly lower in the SBP treatment as the dose increased. The SBP inoculant type produced the highest DMD (p < 0.05) but showed a response that was not different from the SC inoculant treatment for OMD. Increasing inoculation doses of 2%, 4%, and 6% linearly increased the DMD and OMD of fermented bran (p < 0.05). Overall, inoculant application on fermented bran showed an interaction effect except for the components of DM, EE, ADF, NDF, and DMD of fermented bran. Conclusions: It was concluded that the SBP at 6% and their combination resulted in the best chemical quality and digestibility of rice bran. [J Adv Vet Anim Res 2022; 9(4.000): 625-633]

Objectives: This research aimed to isolate, identify, and characterize a new strain of Bacillus cereus through different molecular biology approaches so that it could be further studied for therapeutic purposes against selective enteric pathogens. Materials and Methods: Pure isolates of B. cereus were prepared from buffalo yogurt samples in REMBA medium. Initially, the morphological, physiological, and biochemical properties were studied accordingly. Following the tests, the molecular identification for the strain identification was conducted through plasmid DNA extraction, PCR, agarose gel electrophoresis, and 16S rRNA sequencing up to 1.37 kb. Afterward, the antibiotic sensitivity [Epsilometer test (E-Test)] and anti¬fungal activity were tested considering different concentrations. Being classified from the aforementioned tests, a comprehensive antimicrobial activity test was conducted using the cell-free-su¬pernatant (CFS) of the test strain against selective enteric pathogens in humans in vitro. Besides, the different clusters of genes were identified and characterized for understanding the presump¬tive bacteriocins present in the CFS of the strain in silico, where molecular string properties were calculated. Finally, the evolutionary relationship among diversified bacteriocins synthesized by different Bacillus strains was studied to predict the CFS-containing bacteriocins of the new strain. Results: Purified isolates of B. cereus were Gram-positive rods and showed significant tolerance (p < 0.0001) to different concentrations of pH, phenol, bile salt, and NaCl. 16S rRNA revealed the strain as LOCK 1002, which was strongly sensitive to all the antibiotics used and resistant to selec¬tive antifungal agents. The CFS of B. cereus LOCK 1002 was found to be a very promising antago¬nist to all the enteric pathogens used in the culture condition. Two gene clusters were predicted to be interconnected and responsible for different presumptive bacteriocins. Conclusion: The newly identified LOCK 1002 can be a very potent strain of B. cereus in use as an antimicrobial agent for having different bacteriocin coding gene clusters. Objectives: This study was designed to examine the effects of various concentrations of tris (hydroxymethyl) aminomethane (tris) and egg yolk on the quality of cryopreserved buck sperm. Materials and Methods: The collected semen samples were pooled, washed, and diluted into five different freezing extender groups, viz., extender I (tris 0% + egg yolk 0%), extender II (tris 1.41% + egg yolk 4%), extender III (tris 2.41% + egg yolk 8%), extender IV (tris 3.41% + egg yolk 16%), and extender V (tris 4.41% + egg yolk 24%). The sperm parameter of the five groups of extenders was evaluated after equilibration and cryopreservation. Results: The results showed that extenders II–V provided significantly higher semen progressive motility and total motility percentages than extender I after equilibration (p < 0.05). The higher percentages of semen progressive motility, total motility, viability, and plasma membrane integ¬rity (by both HOST under light microscopy and stain after HOST under light microscopy) were found in the sperm cryopreserved with extender IV than extender I, extender II, and extender III groups after thawing (p < 0.05). In addition, semen progressive motility, total motility, and viability were not further increased, or plasma membrane integrity (by both HOST tests) was decreased by the addition of tris and egg yolk (extender V) after cryopreservation (p < 0.05). Conclusion: In conclusion, our result indicates that the following washing, the supplementation of tris (3.41% + egg yolk 16%) on the freezing extender are suitable for improving the semen quality of buck after freezing and thawing.
