Clinical pharmacology of tramadol and tapentadol, and their therapeutic efficacy in different models of acute and chronic pain in dogs and cats

Opioids are considered the gold standard to manage acute or chronic or mild to severe pain. Tramadol is a widely prescribed analgesic drug for dogs and cats; it has a synthetic partial agonism on μ-opioid receptors and inhibits the reuptake of norepinephrine and serotonin. However, the biotransformation and resultant metabolites differ between species and depend on cytochrome P450 interactions. Dogs mainly produce the inactive N-desmethyl tramadol metabolite, whereas cats exhibit an improved antinociceptive effect owing to rapid active O-desmethyltramadol metabolite production and a longer elimination half-life. Tapentadol, a novel opioid with dual action on μ-receptors and noradrenaline reuptake inhibitory activity, is a promising option in dogs, as it is less reliant on metabolic activation and is unaffected by cytochrome polymorphisms. Although scientific evidence on the analgesic activity of tapentadol in both species remains limited, experimental studies indicate potential benefits in animals. This review summarizes and compares the pharmacology, pharmacokinetics, and therapeutic efficacy of tramadol and tapentadol in dogs and cats with different pain conditions. According to the available data, tramadol seems a more suitable therapeutic option for cats and should preferably be used as a component of multimodal analgesia in both species, particularly dogs. Tapentadol might possess a superior analgesic profile in small animals, but additional studies are required to comprehensively evaluate the activity of this opioid to manage pain in dogs and cats. Objectives: Demodicosis is a vital skin problem in dogs. The present study has determined the prevalence and associated risk factors of demodicosis in dogs and the response to treatment. Materials and Methods: A total of 100 skin scrapings were collected from dogs having dermatolog¬ical lesions brought to the Teaching and Training Pet Hospital and Research Center of Chattogram Veterinary and Animal Sciences University for treatment purpose. The collected scrapings were dissolved in 10% potassium hydroxide to detect mites through microscopic examination. Various risk factors like breed, age, sex, hair type, health status, and management system (indoor and outdoor) were analyzed using the logistic regression model. Positive cases were treated with oral ivermectin (Scabo®; at 0.6 mg/kg/day) along with amitraz 12.5% (Ridd®) diluted to 0.05% for rub¬bing on the body after bathing with chlorhexidine shampoo (PetHex®). Clavulanate amoxicillin (Moxaclav®) and omega-3 fatty acids (OMG-3®) were also suggested to prevent secondary bacte¬rial infection and to maintain skin and hair coat integrity. The recovery rate was observed every 24 weeks of treatment upto 1113 weeks. Results: The overall prevalence of demodicosis was recorded as 27%. Hair type, health status, and management system were significantly (p < 0.05) associated with the occurrence of demodicosis in dogs. Following treatment, the first negative skin scraping for mite was noticed at 810 weeks of treatment, and in all cases, clinical signs completely disappeared at 8090 days of treatment. Conclusion: A good percentage of the dogs having dermatological lesions was determined as demodicosis. Long-term oral ivermectin and topical amitraz, an oral antibiotic, and nutritional therapy are effective against canine demodicosis. Keeping long-haired dogs with good body con¬dition and indoor management is highly suggestive of preventing and controlling the disease. [J Adv Vet Anim Res 2021; 8(1.000): 116-122]

Objective: The objective of the study was to assess the effect of A,D3,E (I/M) and oregano oil extract 15% on some clinicopathological parameters during lamb bacterial enteritis treatment. Materials and Methods: Sixty Barki lambs, 20 apparently healthy (control group) and, 40 suf¬fered from bacterial enteritis [enteric group (EG)], were subdivided into four treated groups (TGs): antibiotic group (AG), antibiotic + A,D3,E group (A + A,D3,E), antibiotic + oregano oil (AOG), and oregano group (OG). Fecal swabs were collected from EG then aseptically cultured, isolated, phe¬notypically identified, genotypically confirmed, and sequenced by PCR 16srRNA. Paper disk dif¬fusion test was used for estimation of oregano oil extract 15% antibacterial activity. After blood sample aspiration from all animals, they were clinicopathologically and statistically analyzed. Results: Escherichia coli, followed by Salmonella species and then Klebsiella species, was the main causative agents of lamb diarrhea and were susceptible to oregano oil extract 15%. A + A,D3,E and AOG showed significant (p < 0.05) enhancement of some clinicopathological parameters more than AG or OG. Matrix metalloproteinases (MMP-2 and MMP-9) and total antioxidant capacity (TAC), yielded area under the curve, sensitivity, negative predictive value as 1, 100% and 100% respectively, were determined in both EG and TGs. Conclusion: Oregano oil extract 15% has good antibacterial properties against enteric bacteria in vitro and in vivo. The combination between antibiotic and antioxidant vitamins or oregano plant extract of 15% has a good impact on some clinicopathological alterations in lamb bacterial enteri¬tis treatment. Objective: The feasibility of fermented litter as an alternative feed material for ruminant livestock is measured by organoleptic quality, fiber profile, heavy metal contamination, and the presence of worm eggs. This study aimed to examine the influence of broiler chicken litter fermentation with different fermentation lengths on organoleptic quality, and contents of cuprum (Cu), lead (Pb), worm eggs, fiber fractions including hemicellulose, cellulose, neutral detergent fiber (NDF), acid detergent fiber (ADF), lignin, and fermented litter fiber profile through analysis of scanning electron microscope-energy dispersive X-ray (SEM-EDX). Materials and Methods: This study used a complete randomized design of a unidirectional pat¬tern with four treatments and four repeats with long fermentation treatments of 0, 3, 6, and 9 weeks. Result: The results showed a real influence (p ≤ 0.05) of fermentation length on organoleptic qual¬ity, NDF, ADF, lignin, hemicellulose, and fiber profile with SEM-EDX observations, with no presence of worm eggs and heavy metal content is still at a safe level for feed materials. Conclusion: This study concluded that the processing of broiler chicken litter with 6 weeks of fer¬mentation gave the best results on organoleptic observations, fiber profile, no presence of worm eggs, and heavy metal contamination that is safe for livestock. Objective: The current study was undertaken to evaluate the seasonal dependency and prevalence of gastrointestinal roundworms (nematodes) infecting domestic fowls (Gallus gallus domesticus) in Kashmir. Materials and Methods: From August 2017 through July 2019, the investigation was undertaken during each of the four seasons. We tested 400 guts obtained from varied places around the Kashmir valley for nematode infestation. The nematodes found within the digestive tract were collected and identified using a variety of identification keys under the microscope. Statistical Package for the Social Sciences was used to analyze the data (version 20). Chi-square (χ2) test was carried out to analyze the sample data. Results: 196 hosts were infected with various nematodes, indicating an overall prevalence of 49% (196/400). The findings revealed that the prevalence of Ascaridia galli was 32.97% (61/185) in the first year and 35.34% (76/215) in the second year. Heterakis gallinarum had a prevalence rate of 20.80% (38/185) in the first year and 24.18% (52/215) in the second year, whereas Capillaria spp. had a prevalence rate of 10.81% (20/185) in the first year and 12.55% (27/215) in the second year. The overall prevalence of A. galli was determined to be 34.25% in both years (August 2017July 2019), with a mean intensity of 4.86. Summer months had the highest parasitic load. Heterakis gallinarum had a prevalence rate of 22.5% and a mean intensity of 26.83. Summer was shown to have the most considerable parasitic burden. Capillaria spp. had an overall prevalence of 11.75% and a mean intensity of 4.59; autumn had the highest parasite load. The most abundant species was identified as A. galli. It was shown that there is a significant (p < 0.01) link between seasonal¬ity and helminth parasite prevalence. Conclusion: The studys findings indicate that these gastrointestinal nematodes are ubiquitous throughout the year, but are particularly abundant in the summer and fall seasons among domes¬tic poultry in the study area. This study on the prevalence of gastrointestinal nematodes in Gallus gallus domesticus demonstrates the seasonality of infection rates and also offers various methods and techniques for framing effective strategies for controlling these helminthes to maximize profit from backyard chicken farming. Objective: In this case report, we have investigated the infectious bronchitis (IB) virus (IBV) out¬break with the co-infection of Escherichia coli in 2833-day-old broiler chickens in Malaysia. Materials and Methods: A farmer complained that Cobb 500 chickens, raised in the open house, were having bloody diarrhea, open mouth breathing, non-uniform growth, and ruffled feathers. The mortality was about 100 birds (from about 7000 birds) per day. The sick birds were isolated and subjected to physical examination, postmortem, and histopathological analyses. Gross lesions were observed and recorded. The lung samples have proceeded with histopathological evalua¬tions. The lungs, kidneys, trachea, air sac, and heart samples were collected to isolate bacteria and fungi through a series of conventional cultural methods, followed by molecular confirmation of the IBV. Results: Postmortem examination revealed air sacculitis, hemorrhagic tracheitis, pulmonary con¬gestion, fibrin deposition in the liver and air sac, hemorrhagic enteritis, and renomegaly. The bacterial culture and biochemical tests revealed E. coli in the lungs, trachea, liver, intestine, and kidney samples. However, no fungus could be isolated from those samples. Histological evalua¬tion of lung samples demonstrated infiltration of inflammatory cells in the pulmonary tissues. Apart from this, reverse transcription-polymerase chain reaction confirmed the presence of avian coronavirus responsible for infectious bronchitis (IB). Conclusion: The chickens were diagnosed with IB concurrent with E.coli. The chickens exhibited typical nephropathogenic strain of IBV infection, causing high mortality. Many countries of the world have been combating the new variant of severe acute respiratory syndrome coronavirus 2. Black fungus is an opportunistic foe that may cause fatal infection in immunocompromised and steroid-treated coronavirus disease 2019 (COVID-19) patients. The COVID-19 associated mucormycosis (CAM) is now a serious concern throughout the world, includ-ing many Asian countries. Therefore, along with early and accurate diagnostic facilities, special care, and prompt, but coordinated approach are recommended to combat the CAM in patients. Objective: The purpose of this study was to determine the effects of the ethanolic extract of avocado seed flour on the estrous cycle characteristics, the concentrations of reproductive hormones [luteinizing hormone (LH) and estradiol], and the activities of some tissues (ovarian and uterine) that are markers of oxidative stress in female guinea pigs (Cavia porcellus). Materials and Methods: Twenty-four female cavies with normal estrous cycles and equivalent body weights (464.25 and 71.88 gm) were randomly assigned to four groups, each with six females. The control group received 1 ml of distilled water orally, whereas the EE100, EE200, and EE400 groups received 100, 200, and 400 mg/kg bw of ethanolic extract of Persea americana seed, respectively. Following that, three consecutive estrous cycles were observed using vaginal smears. After the trial, the females were slaughtered, and blood and organs were gathered for study. Results: The mean duration of the estrus phase is considerably (p < 0.05) longer in group EE100 animals than in control animals. LH concentrations were substantially (p < 0.05) higher in females in group EE200 than in controls. Total cholesterol levels typically dropped in females in the extract groups, but only significantly (p < 0.05) in those in group EE100 compared to the control group. Serum estradiol levels increased considerably (p < 0.05) in treated females compared to controls. Catalase activity rose considerably (p < 0.05) in the ovaries of group EE400 when compared to the control. Superoxide dismutase activity rose considerably (p < 0.05) in the uterus of female cavies given the extract compared to the control. Conclusion: Avocado seed ethanolic extract prolongs the estrus phase, increases estradiol and LH levels, and protects the uterus from oxidative stress in female cavies. Objective: The work aimed to determine the state of lipid peroxidation products and the activity of the antioxidant system in cats with aseptic inflammation and purulent-inflammatory processes of varying severity. Materials and Methods: The intensity of the process of lipid peroxidation- antioxidant system processes in blood plasma was evaluated using commercial kits. The level of diene conjugates (DC), the content of malondialdehyde (MDA), the level of medium-weight molecules were deter¬mined from lipid peroxidation indices. The state of antioxidant protection was assessed by indi¬cators of superoxide dismutase (SOD) activity, catalase (CT), ceruloplasmin (CP) concentration, glutathione peroxidase activity, glutathione reductase (GLR), and total antioxidant activity (AOA). Results: With aseptic inflammation in the blood of cats, a slow activation of peroxidation reactions occurred. The DC indicator increased by 1.4 times, the SOD level rose by 1.38 times, the amount of GLR by 1.04 times, and the activation of total AOA by 9.06. In sepsis, the values of DC, MDA, and medium-weight molecules increased by 4.4, 8.4, and 8.8 times, respectively. With abscesses in the blood of cats, an increase in CT, CP, and GLR is observed against a background of a decrease in glutathione peroxidase. With purulent wounds, the total AOA of plasma increases by 2.5 times; with abscesses, the total AOA increases by 1.9 times, and with sepsis, it decreases by 1.5 times. Conclusion: With surgical infections in cats, deep biochemical changes or irreversible biochemical changes (in sepsis) occur that indicate a significantly enhanced catabolic orientation of metabolic processes and the accumulation of toxic metabolites, which leads to damage and a decrease in tissue regenerative capacity. Objective: Sertoli cells (SCs) are important sustentacular cells in the seminiferous tubules of the testis. Isolation and identification of SCs are the premise for studying their functions. Since New Zealand rabbit is a stable strain which is widely used for biomedical research and animal farming, this study aimed to develop a simple and effective protocol for SC isolation in New Zealand rabbits. Materials and Methods: The SCs of three 30-day-old New Zealand rabbits were isolated by incu¬bation with enzymatic digestion I (Dulbeccos modified Eagle medium supplemented with 1 mg/ ml collagenase IV and 50 μg/ml DNase I) and digestion II (digestion I + 1 mg/ml hyaluronidase + 1 mg/ml trypsin), as well as differential plating. The cells were enriched and identified by using immunocytochemical staining and reverse transcription polymerase chain reaction analysis. Results: Homogeneous cells were obtained. They presented the typical large cell body and an irregular pyramidal shape after differential plating and passaging. These cells expressed mRNA of the SC marker sex-determining region Y-box 9 (SOX9) instead of the Leydig cell marker StAR. Immunocytochemically, they are positive of SOX9, GATA binding protein 4, and androgen-binding protein. Conclusion: The SCs were enriched from the testicular tissues of prepubertal New Zealand rabbits by a simple and effective protocol, which provides a basis for further theoretical researches and practical applications.
