Phylogenetic analysis of HPAI H5N1 virus from duck swab specimens in Indonesia النص الكامل

Objective: A phylogenetic study was carried out on the avian influenza virus (AIV) isolated from a disease outbreak in Sidenreng Rappang Regency, South Sulawesi, Indonesia, in 2018. Material and Methods: Oropharyngeal swabs and organ samples were obtained from ducks that showed clinical symptoms: torticollis, fascial edema, neurological disorders, the corneas appear cloudy, and death occurs less than 1 day after symptoms appear. In this study, isolate A/duck/ Sidenreng Rappang/07180110-11/2018 from duck was sequenced and characterized. Results: It was found that each gene segment of the virus has the highest nucleotide homology to the Indonesian highly pathogenic avian influenza (HPAI) H5N1 clade 2.3.2.1c. Multiple alignments of the sample Hemagglutinin (HA) gene with the avian influenza references virus showed that the pattern of amino acid arrangement in the cleavage site PQRERRRK-RGLF is the characteristic of the HPAI virus. In addition, the HA gene contained Q222 (glutamine) and G224 (glycine), signify¬ing a high affinity to avian receptor binding specificity (SA α2,3 Gal). Furthermore, there was no genetic reassortment of this virus based on the phylogenetic analysis of HA, NA, PB1, PB2, PA, NP, M, and NS genes. Conclusion: The HPAI H5N1 clade 2.3.2.1c virus was identified in duck farms in South Sulawesi, Indonesia. [J Adv Vet Anim Res 2021; 8(2.000): 346-354]

Objective: Chicken infectious anemia virus (CIAV) is an economically important emerging infec¬tion of poultry as it causes immunosuppression and reduces egg production. Although it is world¬wide distributed and first reported (single case) in Bangladesh in 2002, no epidemiological and serological investigations have been conducted. The current study aimed to conduct a serologi¬cal investigation on the prevalence of CIAV infection in broiler breeder and layer farms in some selected areas of Bangladesh. Materials and Methods: A total number of 460 sera samples were randomly collected from unvac¬cinated broiler breeder and layer flocks, of which 276 were from 11 broiler breeder farms and 184 from 12 layer farms. The sera samples were subjected to a commercially available enzyme-linked immunosorbent assay kit to observe antibodies induced by CIAV. Results: Results demonstrated that the overall prevalence of CIAV was 83.6% among a total of 460 samples. In broiler breeder birds, the prevalence was 89.9%, whereas it was 78.3% in layer birds. A higher number of female birds was found to be seropositive than male birds. However, chickens of all age groups were found to be susceptible to the virus. Conclusions: These results indicate the presence of CIAV in Bangladesh, which may be the sequel of naturally occurring either vertical or horizontal infection in all bird flocks tested without clinical symptoms of the disease. A further epidemiological investigation will be required, followed by molecular isolation and characterization of the virus for suitable vaccine candidate selection and/ or preparation. Objective: Present research aims to isolate, identify, and determine the virulence of the Streptococcus agalactiae (group B Streptococcus; GBS), isolated from popped eye disease affected Tilapia and Vietnamese Koi (V. Koi) fishes. Materials and Methods: A total of 330 fish samples were collected, of which Tilapia (n = 180) and V. Koi (n = 150), were collected from 35 affected ponds of four selected districts of Bangladesh. Isolation of the bacterium was done using different culture media (Nutrient broth, Plate count agar, Tryptic Soy Agar, and Blood agar), and identification by using various biochemical tests (con¬ventional and using API 20 Strep kit) and polymerase chain reaction (PCR) using primers against 16S rRNA gene of S. agalactiae. Antibiotic susceptibility of the bacteria was performed using seven different antibiotics disc (Tetracycline, Oxytetracycline, Chlortetracycline, Streptomycin, Ciprofloxacin, Gentamicin, and Neomycin). Virulence of the isolated S. agalactiae was determined by infecting healthy Tilapia and V. Koi fishes through experimental infection. Results: Isolated bacteria were found Gram-positive paired and chained cocci, β-hemolytic and non-motile. Findings of biochemical and serological tests indicate that the isolated bacterium belongs to Group B Streptococcus of Lancefield classification. PCR result also confirmed that the bacteria were S. agalactiae. The bacterial isolates possessed resistance property against all the seven antibiotics used in this study. The isolated GBS was found highly virulent and showed 80%90% mortality for Tilapia and V. Koi fishes in experimental infection within 16 days of post-infection. Conclusion: From the findings of this study, it may be concluded that isolated GBS from the Tilapia and V. Koi fishes were highly virulent and possessed multidrug-resistance properties. Objective: The study was carried out for molecular characterization and antibiotic resistance anal¬ysis of Escherichia coli isolated from different turkey farms in Dinajpur, Bangladesh. Materials and methods: A total of 45 samples comprising feces (n = 23) and cloacal swabs (n = 22) were collected randomly from turkeys. The samples were subjected to isolation and identification of E. coli by cultural and biochemical characteristics, followed by polymerase chain reaction and sequencing. An antibiogram of the isolated E. coli isolates was carried out by following the Kirby Bauer disk diffusion method. Results: Out of the 45 samples, 28 (62.21%) were positive for E. coli, of which 16 (35.55%) fecal samples were positive and 12 (26.66%) cloacal swabs were positive. The antibiotic sensitivity analysis revealed that all the E. coli isolates were 100% sensitive to levofloxacin, norfloxacin, neo¬mycin, gentamicin, and nitrofurantoin. On the other hand, all the isolates were 100% resistant to amoxicillin, azithromycin, erythromycin, tetracycline, bacitracin, cephalexin, nalidixic acid, van¬comycin, methicilin, piperacillin, pefloxacin, novobiocin, cefepime, trimethoprim, netilmicin, and aztreonam. Objectives: Probiotics are well documented for their health benefits by developing a balanced intestinal microbiota and boosting immunity. The present study was conducted to determine the effects of a probiotic preparation EnzimsporinTM (consisting of spore-forming bacteria Bacillus subtilis B-2998D, B-3057D, and Bacillus licheniformis B-2999D) on the biochemical, hematological, immunological parameters, intestinal microbiota, and growth dynamics of sheep and lambs. Materials and methods: Enzimsporin was fed to lambs and sheep at different doses to determine the bacterias probiotic effects. Sheep were divided into three groups (six each), which received 0, 1, and 3 gm of Enzimsporin/per head/day, respectively, and two groups of lambs (10 each), who received 0 gm and 1 gm of Enzimsporin/per head/day for 30 days in addition to their regular ration. On day 30, blood samples were collected, followed by the determination of biochemical, hematological, and natural resistance indicators. Fecal samples were examined to determine the intestinal microflora, and animals were weighed daily to determine their growth dynamics. Results: Supplementation of probiotics (EnzimsporinTM) improved the lambs body weight gain by 18.8%. Analysis of the clinical parameters showed improvements in the levels of total protein, globulins, and urea by 5.3%, 10.8%, and 6.2%, respectively, in the blood of probiotic-supplemented lambs. Similarly, an increment in the total protein, albumins, and globulins was observed in the sheep with EnzimsporinTM supplementation. The decrease in bilirubin and cholesterol levels in the blood and increased bactericidal and phagocytic index in the sheep and lambs with probiotic supplementation indicated a positive influence of EnzimsporinTM on the liver function and natural resistance. Furthermore, an increase in Lactobacillus and Bifidobacterium and a decrease in the Escherichia coli, Enterococcus, and Yeast in the fecal contents of experimental sheep and lambs indicated the potentiality of EnzimsporinTM on maintaining good gut health. Conclusion: Spore-forming bacteria B. subtilis B-2998D, B-3057D, and B. licheniformis B-2999D can be used in feeding sheep and lambs of 2 months of age to increase body weight gain, improve intestinal microbiota, strengthen the immune system, and maintain normal metabolic processes. Objective: The article aimed to study the relationship between the physico-chemical qualities of pig meat and their pre-slaughter weights. Materials and Methods: In this study, 60 pigs were reared for fattening under the same condi¬tions of keeping and feeding, slaughtered, and used to sample the longest back muscle meat with pre-slaughter weights of 110 and 130 kg. The samples were evaluated according to generally accepted methods for assessing the physico-chemical qualities in pig meat. Results: Samples of meat from animals slaughtered at 130 kg had higher values for marbling by 2.0 points or 5.24% (p < 0.01), active acidity pH in ham muscles by 0.20 pH or 3.57% (p < 0.01), and the longest muscle of the back by 0.10 pH or 1.82% (p < 0.001). The pre-slaughter weight factor substantially affected the pH of ham muscles at 10.35% and on the marbling of meat in the longest back muscle at 13.31%. Pigs slaughtered at 110 kg had a greater increase in the color intensity of the meat and an increase in its water holding capacity. At a pre-slaughter weight of 130 kg, increasing the marbling and decreasing the softness of the flesh demonstrated a modest, adverse relationship. Objective: Ivermectin (IVM) could be used effectively to treat bovine cutaneous papillomatosis, a widespread viral skin disease that causes major economic losses in cattle. This study aimed to evaluate the regression of bovine cutaneous papillomas induced by IVM by estimating oxidative stress markers, besides clinicopathological and hematological findings. Materials and Methods: Twenty naturally infected animals with cutaneous papillomatosis were chosen randomly and diagnosed clinically and histopathologically. All the infected animals were divided into groups: Group I (n = 10), which received no treatment and was considered the con¬trol group. In Group II (n = 10), the animals were subcutaneously injected at 0.2 mg/kg of IVM 2 weeks apart during the 90-day experimental period. Papilloma regression was tracked clinically, papilloma biopsies were taken for histopathological analysis, and blood samples were taken for hematological and oxidative parameter testing. Results: From the 15th to 45th day after receiving IVM, papillomas began to fade. Necrotic areas, ulcerations, and lymphocytic infiltration were found in the histopathological studies, besides a decrease in papilloma epidermal proliferation. total erythrocytes count, packed cell volume, total leucocytes count, and lymphocytes values were increased significantly, and a large decrease in glutathione peroxidase and glutathione reduced was identified as signs of IVM-induced oxidative stress. Objective: This research investigated the immunomodulatory potentials of two medicinally important wild epiphytic Bangladeshi orchids Cymbidium aloifolium and Papilionanthe teres using Swiss albino mice. Materials and Methods: Orchid extracts were prepared using a cold methanol extraction proce¬dure. To assess the immunomodulatory action, Swiss albino mice of either sex weighing 2535 gm were divided into five groups each with six animals. Sheep red blood cells (SRBC) of 0.5 × 109 cells/ml were used to immunize all mice on the 7th day, and a booster dose of the same quan¬tity of SRBC was given on the 11th day of the experiment. After 14 days of oral treatment with 100 and 200 mg/kg bw of orchid extract, the mice were sacrificed to collect serum and organs. Hematological assays, delayed type of hypersensitivity assays, phagocytic index (PI), and histo¬pathological investigations were used to assess in vivo immunomodulatory efficacy. Results: The body weight changes of the experimental animals were considerably greater at 100 mg/kg bw than at a higher dose (200 mg/kg bw). There was a substantial improvement of relative organ weights of the thymus and spleen at the low dose, but no effect on kidney weights was evi¬dent. The liver weight increased significantly (p < 0.05) at both doses. Total neutrophil, leukocyte, and lymphocyte counts, hemoglobin percentage, delayed hypersensitivity reaction, and PI were all significantly (p < 0.05) increased in mice receiving the lower dose. In contrast to the control group, the higher dose reduced immunological response, suggesting the negative influence of a higher dose of extracts on the immune reaction. Conclusions: The results demonstrate that orchid extracts can potentially modulate the innate immune system in the experimental animal. [J Adv Vet Anim Res 2021; 8(3.000): 479-488]

Objective: This study evaluated the different traits from day-old to age at first eggs of Fayoumi and White leghorn (WLH) F1 chickens and their reciprocal crossbreeds. Materials and Methods: In this study, 1,000 eggs were used to obtain experimental chicks. However, only 150 chicks were selected from each genotype for subsequent trial evaluation and reared on the same diet in a completely randomized design. The effects of genotype on each considered trait were analyzed by the statistical analysis system, and Duncans multiple range test made a significant difference. Results: Comparative evaluations made on all parameters illustrated significant differences (p < 0.05) throughout the study period. F1 acquired from WLH cocks and Fayoumi hens maximized weights almost at all ages during brooding, whereas pure line Fayoumi showed lower importance for the trait stages of growth. Offspring from the crossbreed of WLH cocks and Fayoumi hens had the highest weight gain than any other crossbreeds from 4 to 8 and 12 to 16 weeks of age. Pure line WLH and Fayoumi lay larger eggs and smaller eggs (41.67 and 34.00 g), respectively. The highest and smallest body weight at the age of first egg was 1,364.89 and 1,178.36 g in the main crossbreed and Fayoumi line. The main WLH cock and Fayoumi hens and reciprocal crossbreds advanced in age at first egg by 4 and 11 days, respectively. Conclusion: The leghorn hen and Fayoumi cock crosses could be used to develop potential egg-laying lines than their reciprocal crossbreeds. [J Adv Vet Anim Res 2021; 8(1.000): 1-6]

Objective: This study assessed the effect of cellulose sheets fortified with Natamycin-loaded algi¬nate nanoparticles on the growth of toxigenic Aspergillus flavus and aflatoxin production on the superficial layer of Egyptian Romy cheese after 12 weeks of maturation. Materials and Methods: Toxigenic A. flavus (GenBank accession No. MT645073) was inoculated into the outer surface of Egyptian Romy cheese (at 5 log CFU/gm) and wrapped with a cellulose sheet fortified with Natamycin-loaded alginate nanoparticles. Unwrapped control contaminated Romy wheels were made as well as non-contaminated wrapped cheese wheels for sensory eval¬uation. Romy cheese wheels were stored at a temperature similar to commercial methods for 12 weeks. Fungal counts were enumerated during this time, and enzyme-linked immune sorbent assay detected aflatoxin after the 4th week of maturation storage. Results: In cheese samples covered with cellulose sheets containing Natamycin-loaded alginate nanoparticles, the fungal count was reduced by 2 log approximately in contrast to control samples after the 2nd week of storage. However, within the 8th week of storage, the greatest significant reduction (p < 0.05) was seen where fungal growth was hindered entirely to the end of the rip¬ening period. The mean values for taste, color, flavor, and overall acceptability were 4, 4.7, 4.09, and 4.3, respectively. Furthermore, in the treated samples, the total aflatoxin concentration was decreased by 78.6% relative to the untreated control one. Conclusion: Using cellulose sheets fortified with Natamycin-loaded alginate nanoparticles in Egyptian Romy cheese wrapping could be an effective way of controlling A. flavus and subsequent aflatoxin production without influencing the typical taste, color, flavor, and overall appearance of traditional Romy cheese.
