Ovine pulmonary adenocarcinoma (OPA) is a transmissible lung cancer caused by Jaggsiekte sheep retrovirus (JSRV). It is difficult to identify animals infected with JSRV but are clinically healthy. The virus does not induce a specific antibody response and, although proviral DNA sequences of JSRV can be found in mononuclear blood cells, the detection is inconsistent. The aim of this study was to investigate the presence of JSRV in the bone marrow of infected sheep and develop a more consistent screening method. Immunohistochemical examination of bone marrow samples from 8 asymptomatic JSRV-infected sheep revealed the presence of positively labelled cells. However, JSRV could not be detected by a highly sensitive polymerase chain reaction (PCR) in bone marrow aspirates periodically collected from these animals. Results suggest that JSRV-infected cells may be present in the bone marrow of symptomless animals, but the number is below the detectable level for PCR. Therefore, this technique does not seem to be helpful for preclinical diagnosis of OPA.

Objective—To construct and optimize a fiducial marker suitable for both CT and MRI. Sample—Fiducial markers containing serial dilutions of iopamidol mixed with water. Procedures—IV tubing sets were infused with serial dilutions (0% to 100%; increments of 10%) of iopamidol. Tubing ends were sealed; additional seals were added to create an equilateral triangle. A reference point was created by placing a crimp in 1 side. Markers were fixed to a gelatin soft tissue–attenuating phantom and evaluated by use of CT and MRI. For CT, simple linear regression analysis was used to assess the relationship between the percentage of marker contrast medium and quantitative variables, including marker attenuation, attenuation changes in the phantom, and beam-hardening artifact length. A subjective grading scheme for artifact creation on CT images and marker visibility on MRI images was used. Measurements were obtained by investigators who were unaware of the contents of each marker. Results—Percentage of contrast medium in each marker was strongly correlated with marker attenuation (r2 = 0.96), artifact length (r2 = 0.765), and mean attenuation changes within the phantom (r2 = 0.826) for CT. Subjective CT scores indicated that concentrations of contrast medium > 50% resulted in excessive artifacts. Markers with concentrations of iopamidol > 50% had poor subjective MRI visibility scores. No artifacts were seen on MRI. Conclusions and Clinical Relevance—A marker containing a 10% solution of iodinated contrast medium mixed with water provided ideal contrast for both CT and MRI.

Objective-To validate use of stress MRI for evaluation of stifle joints of dogs with an intact or deficient cranial cruciate ligament (CrCL). Sample-10 cadaveric stifle joints from 10 dogs. Procedures-A custom-made limb-holding device and a pulley system linked to a paw plate were used to apply axial compression across the stifle joint and induce cranial tibial translation with the joint in various degrees of flexion. By use of sagittal proton density-weighted MRI, CrCL-intact and deficient stifle joints were evaluated under conditions of loading stress simulating the tibial compression test or the cranial drawer test. Medial and lateral femorotibial subluxation following CrCL transection measured under a simulated tibial compression test and a cranial drawer test were compared. Results-By use of tibial compression test MRI, the mean +/- SD cranial tibial translations in the medial and lateral compartments were 9.6 +/- 3.7 mm and 10 +/- 4.1 mm, respectively. By use of cranial drawer test MRI, the mean +/- SD cranial tibial translations in the medial and lateral compartments were 8.3 +/- 3.3 mm and 9.5 +/- 3.5 mm, respectively. No significant difference in femorotibial subluxation was found between stress MRI techniques. Femorotibial subluxation elicited by use of the cranial drawer test was greater in the lateral than in the medial compartment. Conclusions and Clinical Relevance-Both stress techniques induced stifle joint subluxation following CrCL transection that was measurable by use of MRI, suggesting that both methods may be further evaluated for clinical use.

Objective-To assess the feasibility and reproducibility of longitudinal tissue Doppler ultrasonographic imaging with regard to determination of velocity, strain, and strain rate (SR) of the left atrium (LA) and use those data to characterize LA synchrony (LAS) for a group of healthy dogs. Animals-15 healthy dogs. Procedures-For each dog, apical 4- and 2-chamber echocardiographic views were obtained. Peak velocity, strain, and SR and time to peak value during systole, early diastole, and late diastole were measured for each of the 4 LA walls. To characterize LAS, mean and SD maximal late diastolic time difference (LAD) among the 4 walls were calculated on the basis of time to peak for velocity, strain, and SR; for each, the 95% confidence interval (mean ± 2SD) was calculated. Within-day and between-day intraobserver variability was calculated. Results-For all dogs, tissue velocity and SR had peak positive values during systole and 2 negative peaks during early and late diastole. Atrial strain had a peak positive value during systole, positive values during early diastole, and a negative peak value during late diastole. Reproducibility was acceptable for most variables. Diastolic strain and SR had the highest variability, but times to peak values were always reproducible. For velocity, strain, and SR, the 95% confidence interval for the maximal LAD was < 50 milliseconds and that for the SD of the LAD was < 23 milliseconds. Conclusions and Clinical Relevance-Longitudinal tissue Doppler imaging of LA deformation was feasible in healthy dogs, and its application may be useful for understanding atrial pathophysiologic changes associated with various cardiac diseases in dogs.

Rift Valley fever virus (RVFV) is an acute, zoonotic viral disease caused by a Phlebovirus, which belongs to the Bunyaviridae family. Among livestock, outbreaks of the disease are economically devastating. They are often characterised by large, sweeping abortion storms and have significant mortality in adult livestock. The aim of the current study was to investigate RVFV infection in the Kigoma region, which is nestled under the hills of the western arm of the Great Rift Valley on the edge of Lake Tanganyika, Tanzania. A region-wide serosurvey was conducted on non-vaccinated small ruminants (sheep and goats, n = 411). Sera samples were tested for the presence of anti-RVFV antibodies and viral antigen, using commercial enzyme-linked immunosorbent assay and reverse transcriptase polymerase chain reaction, respectively. The overall past infections were detected in 22 of the 411 animals, 5.4% (Confidence Interval (CI) 95% = 3.5% – 8.1%). The Kigoma rural area recorded the higher seroprevalence of 12.0% (CI 95% = 7.3% – 18.3%; p < 0.0001), followed by Kibondo at 2.3% (CI 95% = 0.5% – 6.5%; p > 0.05) and the Kasulu district at 0.8% (CI 95% = 0.0% – 4.2%; p > 0.05). The prevalence was 12.5% and 4.7% for sheep and goats, respectively. Reverse transcriptase polymerase chain reaction results indicated that only eight samples were found to be positive (n = 63). This study has confirmed, for the first time, the presence of the RVFV in the Kigoma region four years after the 2007 epizootic in Tanzania. The study further suggests that the virus activity exists during the inter-epizootic period, even in regions with no history of RVFV.

Haemoparasite infections are among the most economically important cattle diseases in sub-Saharan Africa. The present study investigated the occurrence of haemoparasites in 295 indigenous cattle from five villages (Mswakini, Lake Manyara, Naitolia, Makuyuni and Nanja) of the Monduli district, a wildlife-domestic animal-human interface area in northern Tanzania. The data showed that the overall occurrence of haemoparasites in the sampled cattle was 12.5% (95% CI: 8.7% – 16.3%), involving single and mixed infections with Theileria parva, Anaplasma marginale, Babesia bovis, Trypanosoma vivax and Trypanosoma brucei. The highest haemoparasite occurrence was recorded in Lake Manyara (18.3%; 95% CI: 8.5% – 28.1%), and the lowest was recorded in Nanja (6.5%; 95% CI: 0.4% – 12.6%). This preliminary study, furthermore, provided evidence of the possible arthropod vectors (ticks and tsetse flies) that may be involved in the transmission of haemoparasites to cattle in the Monduli district. It is envisaged that this survey will stimulate more studies to determine the prevalence of haemoparasites in livestock by using more sensitive molecular techniques.

Phylogeography data are of paramount importance in studying the molecular epidemiology dynamics of foot-and-mouth disease virus (FMDV). In this study, epithelial samples and oesophageal-pharyngeal fluids were collected from 361 convalescent animals (cattle and buffaloes) in the field throughout Tanzania between 2009 and 2013. The single plex real-time RT-PCR (qRT-PCR) assay for rapid and accurate diagnosis of FMDV employing the Callahan 3DF-2, 3DF-R primers and Callahan 3DP-1 probe were used. Preparation of the samples was performed according to the OIE manual, with a Kenya O serotype obtained from the attenuated vaccine serving as a positive control and samples collected from healthy animals serving as true negatives. The results indicated that 53.49% of samples (n = 176) were positive for FMDV genome by qRT-PCR, with Ct values ranging from 14 to 32. In addition, molecular typing of the FMDV genome positive samples using serotype specific primers revealed the existence of several serotypes: serotype South Africa Territory 1 (SAT1) (34.25%, n = 60), serotype A (68.92%, n = 98), serotype O (59.20%, n = 98) and SAT2 (54.54%, n = 96). The virus protein 1 sequences analysis for 35 samples was performed and the collective results indicated: 54.28% serotype O, 25.71% serotype A, 14.28% serotype SAT1 and 2.85% serotype SAT2. Therefore in this study, both the phylogenetic trees and spatial distribution of serotypes elucidated the phylodynamics of multiple FMDV field strains in Tanzania and neighbouring countries.

Ijara district in Kenya was one of the hotspots of Rift Valley fever (RVF) during the 2006/2007 outbreak, which led to human and animal deaths causing major economic losses. The main constraint for the control and prevention of RVF is inadequate knowledge of the risk factors for its occurrence and maintenance. This study was aimed at understanding the perceived risk factors and risk pathways of RVF in cattle in Ijara to enable the development of improved community-based disease surveillance, prediction, control and prevention. A cross-sectional study was carried out from September 2012 to June 2013. Thirty-one key informant interviews were conducted with relevant stakeholders to determine the local pastoralists’ understanding of risk factors and risk pathways of RVF in cattle in Ijara district. All the key informants perceived the presence of high numbers of mosquitoes and large numbers of cattle to be the most important risk factors contributing to the occurrence of RVF in cattle in Ijara. Key informants classified high rainfall as the most important (12/31) to an important (19/31) risk factor. The main risk pathways were infected mosquitoes that bite cattle whilst grazing and at watering points as well as close contact between domestic animals and wildlife. The likelihood of contamination of the environment as a result of poor handling of carcasses and aborted foetuses during RVF outbreaks was not considered an important pathway. There is therefore a need to conduct regular participatory community awareness sessions on handling of animal carcasses in terms of preparedness, prevention and control of any possible RVF epizootics. Additionally, monitoring of environmental conditions to detect enhanced rainfall and flooding should be prioritised for preparedness.
