Objective: To evaluate the use of mesenchymal stem cells (MSCs) in the attenuation of canine atopic dermatitis (AD). Materials and methods: Sixteen dogs were selected and divided into three groups, mild, moder¬ate, and severe, according to the Canine Atopic Dermatitis Extent and Severity Index (CADESI-4). They were evaluated for 82 days. The protocol recommended in this experiment was to inject 2 × 106/kg bodyweight of MSCs in all groups by the intravenous route with intervals of applications of 21 days. The degree of pruritus was evaluated by examining the visual analog scale, the CADESI-4, the histopathology of the skin, hematological and biochemical parameters, the pyogenic effect of MSCs, and the thickness of the epidermis. Results: There was a significant difference in the reduction of epidermal thickness in the moder¬ate and severe groups. Hematological, biochemical, and body temperature parameters remained within normal limits for the species with no side effects Conclusion: MSCs attenuated the clinical signs of AD. Objective: This study aims for molecular identification of naturally growing Bacillus cereus strain from a unique source, able to survive, and alleviate heavy metals from the nature. Materials and Methods: Pure isolate from Murrah buffalo milk was prepared in B. cereus selec¬tive Polymyxin pyruvate egg-yolk mannitolbromothymol blue agar (PEMBA) medium through a cascade of contamination free subcultures. The morphological and biochemical tests were done prior to 16S rRNA gene sequencing for strain identification and further physiological tests. The test strain was inoculated in both solid and suspension culture medium supplemented individually with Cd, Cu, Ag, and Zn to reveal the qualitative and quantitative heavy metal tolerance proper¬ties, respectively. Finally, the data collected from the in vitro assessment was statistically analyzed Results: Molecular analysis revealed that the test strain was B. cereus BF2, which was motile, catalase positive and Gram positive rod. B. cereus BF2 was found significant at 0.3% bile salt toler¬ance [two-way analysis of variance (ANOVA)p value is < 0.0001] where, t-test p value is < 0.0002 between Control Group (CG) and TGR-1; p < 0.037 between TGR-1 and 2; p < 0.0014 between CG and TGR-2. Similarly, B. cereus BF2 was significant in pH tolerant up to 8.0 with p < 0.0115 (in scale p < 0.05). The heavy metal tolerance test revealed that the test metals could not stop the growth of B. cereus BF2 even after 24 h of incubation but partially suppressed the growth kinetics for let¬ting into stationary phase. Among the four heavy metals, Cd and Zn showed partial antagonism to the growth of B. cereus BF2. The survivability was highly significant in the medium supplemented with Zn (p < 0.0001) and Ag (p < 0.018). Objective: This experiment was conducted to assess the effects of the housing system on the welfare and growth performance of the Muscovy duck. Materials and methods: A total number of 48 Muscovy duck aged 3-week old were divided ran¬domly into two groups (24 duck on each): deep litter system and cage system. Each group was subdivided into three replicates (8 birds on each) were identified with wing rings. Results: The study showed that feeding and drinking behaviors significantly increased (p < 0.05) in duck reared at the cage system. While there was no noticeable effect on lying and feather peck¬ing, duck raised in the deep litter had better growth performance with a substantial difference, which achieve a higher feed conversion rate with lower feed efficiency. Physical conditions were significantly better at cage management. The level of stress indicating hormones indicating free Thyroxin was increased with the cage. Therefore, the litter system improves duck welfare and their performance. Conclusion: It is concluded that the duck managed under the litter system has more opportunities that facilitate the expression of more natural behavior, and thus improve their health and perfor¬mance, as a result of improved feed conversion rate and feed efficiency. Objective: Granulosa cells (GCs) play a very important role in reproductive physiology due to their effect on developmental and functional changes. However, there are differing views regard¬ing the mechanism by which hormones stimulate GCs. Therefore, our study aims to determine whether GCs, in the absence of initial stimulation (17β-estradiol), select specific types of MMPs that reconstitute cells by stimulation of major hormones [follicle-stimulating hormone (FSH) or/ and luteinizing hormone (LH)]. Materials and methods: Early GCs were extracted from immature follicles of the porcine ovary to analyze the MMPs levels. Using early GCs in pigs, the cell development rate was evaluated by add¬ing 17β-estradiol, FSH, LH, or FSH + LH, respectively, to the DMEM containing 10% FBS. Real-time PCR, zymography, enzyme-linked immunosorbent assay, western blot, and immunofluorescence analysis were also performed to determine the MMPs activation in the GCs. Results: Our results confirm that FSH or LH stimulation regulates cell development and intracel¬lular MMPs. In particular, FSH activity kept the MMP-2 and MMP-9 expressions constant in GCs. Conversely, LH activity initially led to rapid increases in the MMP-9 expression, which 96 h later was similar to the MMP-2 expression. Simultaneous utilization of FSH + LH maintained a steady MMP-9 expression and the development of GCs increased. Additionally, when FSH and LH were processed simultaneously, the number of cells increased without changes in cell size, while the cell size changed when LH alone was used. Conclusion: Therefore, the results of this study confirm that even without the initial stimulation of GCs, physiological changes occur according to hormonal changes in the environment, and there is variability in the expression of MMPs. Captive birds in zoological settings often harbor parasites, but little information is available about the potential for free-ranging avifauna to act as a source of infection. This review summarizes the gastrointestinal parasites found in zoo birds globally and in seven common free-ranging avian spe¬cies [mallard (Anas platyrhynchos), Eurasian blackbird (Turdus merula), common starling (Sturnus vulgaris), Eurasian jackdaw (Corvus monedula), house sparrow (Passer domesticus), European robin (Erithacus rubecula), and rock dove (Columba livia)] to identify the overlap and discuss the potential for cross-species transmission. Over 70 references were assessed, and papers spanned over 90 years from 1925 to 2019. A total of 60 studies from 1987 to 2019 met the eligibility cri¬teria. All examined free-ranging avifauna harbored parasite species that were also reported in zoo birds, except for the European jackdaw. Parasites reported in captive and free-ranging birds include nematodes (Capillaria caudinflata, Dispharynx nasuta, Ornithostrongylus quadriradiatus, Strongyloides avium, Syngamus trachea, and Tetrameres fissispina), cestodes (Dicranotaenia coronula, Diorchis stefanskii, Fimbriaria fasciolaris, and Raillietina cesticillus, Sobolevicanthus gracilis), trematode (Echinostoma revolutum), and protozoa (Cryptosporidium baileyi). Although no study effectively proved cross-transmission either experimentally or by genetic analysis, these parasites demonstrate low host specificity and a high potential for parasite sharing. There is potential for parasite sharing whenever determinants such as host specificity, life cycle, and hus¬bandry are favorable. More research should be carried out to describe parasites in both captive and free-ranging birds in zoological settings and the likelihood of cross-infection. Objective: The objective of this study was to investigate the intensity of phage infection caused by Phage SAvB14, which was isolated from dairy farms, depending on the initial number of Staphylococcus aureus cells in the medium. Material and methods: To evaluate the impact of the viable bacteria S. aureus var. bovis on the intensity of phage infection caused by Phage SAvB14, 1 mg of phagolysate (phage titer 105 CFU/ ml) was introduced in 9 ml of nutrient broth with an appropriate amount of daily culture of S. aureus var. bovis under study. The number of viable staphylococci was determined by total viable count/ml. Results: In this experiment, we found that the intensity of phages lytic activity was dependent on the number of sensitive bacterial cells in the volume of the culture medium. Effective phage ther¬apy requires a high concentration of phages in the medium (inflammation foci) for rapid contact of the virus with bacteria. Conclusion: When developing a phage drug to treat subclinical mastitis, it is necessary to increase the phage titer in the drug or its dosage compared to the clinical form, as there is a lower proba¬bility of phage contact with a susceptible microbial cell. Besides, at a high concentration of bacte¬ria, there is a gradual decrease in nutrients in the medium, resulting in phages going back to the condition of lysogeny. Objective: The trial was aimed at assessing the effect of phytogenic feed additive (PFA), a natural adaptogen, on growth performance, serum neopterin level, and cutaneous basophil hypersensi¬tivity (CBH) response in heat-induced stress model of broilers. Materials and Methods: One-day-old Ross 308 chicks (N = 360) were randomly distributed among normal control (NOR), heat-stress control (HSC), and PFA treatment (HSC plus PFA at 200 gm/ton of feed) group. HSC and PFA groups were subjected to heat stress (HS) (32°C36°C) from 9:00 a.m. to 5:00 p.m. for 35 days. The impact of HS on growth performance, serum neopterin level, and CBH response was assessed. Results: High ambient temperature worsened the performance traits [bodyweight (p < 0.05) and feed conversion ratio] and significantly lowered the serum neopterin level and CBH response in the HSC group when compared to the NOR group. However, supplementation of PFA at 200 gm/ ton of feed to birds mitigated the detrimental effects of HS. Conclusion: PFA at 200 gm/ton demonstrated the immunomodulatory effect through the resto¬ration of serum neopterin level, CBH response, and growth performance traits in heat-stressed broiler chickens. Thus, PFA can be used as a natural adaptogen to increase the stress resistance and mitigate the negative consequences of various stressors in broiler chickens. Objective: The objectives of this study were to determine the biofilm-forming capability and anti¬microbial susceptibility of Escherichia coli recovered from bovine endometritis samples. Materials and Methods: A total of 120 uterine specimens were collected from cows suffering from endometritis for bacteriological examination. Antimicrobial susceptibility testing was carried out for all isolated E. coli by using the disc diffusion method. The isolates were phenotypically studied for biofilm-forming ability by cultivation on yeast extract -casamino acids Congo red agar (CRA). Some randomly selected isolates were chosen for the molecular identification of some virulence and resistance genes. Results: A total of 58(48.3%) E. coli isolates could be isolated from the 120 samples. Antimicrobial susceptibility testing exhibited that 91.4%, 79.3%, 79.3%, 74.1%, and 58.6% of the isolates were sensitive to gentamicin, amoxicillin-clavulanic acid, ciprofloxacin, cephalexin, and sulfamethox¬azole-trimethoprim, respectively. On the other hand, 91.4% and 70.7% isolates were resistant to cefotaxime and doxycycline, respectively. Cultivation on CRA revealed that 46.6% of isolates were biofilm producers. The molecular detection of resistance and virulence genes declared that all iso¬lates harbored blaTEM, sul1, tetA, qnrS, blaCTX-M, and fimH with a percentage of 100%, papC (40%), and hlyA (10%). FimH was the most prevalent biofilm-associated gene. Conclusion: The present study highlights the high prevalence of multi-drug- resistant E. coli asso¬ciated with bovine endometritis. Objectives: The current research aimed at investigating growth performances and meat yield characteristics of commercial cockerels supplemented with antibiotics and probiotics to the diet. Materials and methods: A total of 1,350 commercial cockerels (ISA Brown) were indiscriminately distributed to 3 treatment groups, each having three replications of 150-day-old chicks based on completely randomized design. The basal diet was treated as the control, while the experimental groups receiving Enrofloxacin 1 gm/kg and Protexin 1 gm/kg feed were considered as antibiotic and probiotic groups, respectively. Bodyweight, feed intake (FI), feed conversion ratio (FCR), and other relevant characteristics were recorded weekly until 49 days of trial. In the end, similar number (10) of birds from each replicate group were slaughtered to determine the carcass characteristics. Results: Significantly, better results were found in the overall growth performances of the cock¬erels in the probiotic-fed treatment group. Highly significant differences were also found in live bodyweight, weight gain, daily gain, FI, FCR, survivability, dressing percentage, abdominal fat, breast meat, drumstick, and thigh weight in the probiotic-fed treatment group compared to the others. Conclusion: The results of the present study indicate that the inclusion of dietary probiotics has a superior performance to antibiotics and may have the potentiality to be used as an alternative growth enhancer in the diet of cockerels. Objective: This study was carried out to find out the immunolocalization of Interleukin 6 (IL-6) and Interleukin 10 (IL-10) in the testicular tissue of testicular dysfunction rat treated with secretome from human umbilical stem cells. Materials and methods: Rats were induced with cisplatin for testicular dysfunction condition. After that, the rats were grouped into two categories and were treated with secretome at 0.2 and 0.5 ml/kg BW once every week for 4 weeks. One week later, after the secretome treatment, the rats were sacrificed for histological evaluation using the immunohistochemical method. The preparation slides were examined using a light microscope and were analyzed descriptively and quantitatively. Results: There were no IL-6 and IL-10 immunoreactivities seen in the testicular tissue after cispla¬tin induction. However, the immunoreactivities of IL-6 and IL-10 were detected after secretome treatment, with both dosages of 0.2 and 0.5 ml/kg BW. These immunoreactivities were detected in the spermatogonia, spermatid/luminal tissue of seminiferous tubule, spermatogenic cells, and Leydig cells. In the cell calculation, the numbers of IL-6 immunoreactive cells were higher at the lower secretome dosage. In contrast, the numbers of IL-10 immunoreactive cells were higher at the higher secretome dosage. Conclusion: IL-6 and IL-10 can be localized in the testicular tissue of testicular dysfunction rat after secretome treatment. [J Adv Vet Anim Res 2020; 7(3.000): 514-520]
