Pharmacokinetics of combination antiparasitic drug preparation for dogs and cats in the form of spot-on solution النص الكامل

Objective: The object of the study was to examine the major pharmacokinetic parameters after a single application of a complex drug preparation for veterinary use based on fipronil, praziquantel, moxidectin, and pyriproxyfen in cats and dogs. Materials and Methods: For dogs, the drug preparation was administered spot-on solution in the following dosage of active pharmaceutical substances: fipronil 27.0 mg/kg body weight (bwt), praziquantel 10.8 mg/kg bwt, moxidectin 6.75 mg/kg bwt, and pyriproxyfen 5.4 mg/kg bwt; for cats, the dosage was the following: fipronil 43.2 mg/kg bwt, praziquantel 17.28 mg/kg bwt, mox-idectin 4.32 mg/kg bwt, and pyriproxyfen 8.64 mg/kg bwt. The blood samples were taken from dogs and cats. The principle of the method for determining praziquantel, trans-4-hydroxyprazi-quantel, pyriproxyfen, and fipronil in serum samples was chromatographed in a high-pressure liquid chromatograph with detection by means of a mass-spectrometric detector. The moxidectin content of the blood was detected by high-performance liquid chromatography. Results: The drug preparation active substances: praziquantel, fipronil, and moxidectin are absorbed into the blood of dogs and cats. The penetration of praziquantel into the systemic circulation and further into organs and tissues was proved. After topical administration, moxidectin is absorbed and distributed systemically and is slowly removed from the plasma, which manifests itself in detectable concentrations of moxidectin in the blood for 1 month. Conclusion: The present results of pharmacokinetic investigations may promote to the determination of effective therapy strategy and prophylaxis of parasitic diseases in dogs and cats. Objective: The purpose of our research was to evaluate the effect of mechanochemical technology on the efficacy of supramolecular complex of fenbendazole (SMCF) with polyvinylpyrrolidone (PVP) polymer against some helminthosis of animals. Materials and Methods: The SMCF samples with PVP were synthesized using a solid-state mechanochemical technology in activators of impact-abrading type and their physicochemical properties were analyzed. The efficacy of SMCF was studied on the laboratory model of Hymenolepis nana and Trichinella spiralis infection of mice and helminthosis of sheep. Results: In the trials conducted on laboratory models, the supramolecular complex showed 93.94% and 98.56 % efficacy at the dose of 1 mg/kg of body weight (b/w), while the substance of fenbendazole showed 7.97% and 8.33% efficacy at the same dose. A high efficacy (>94%) of the SMCF was revealed at the dose of 2.0 mg/kg of b/w at oral administration against nematodes in naturally infected sheep by the results of the fecal examination, while the substance of fenbendazole was active at the dose of 5.0 mg/kg at single oral administration. Moreover, the SMCF demonstrated 97.37% efficacy at the dose of 2 mg/kg against Moniezia spp. infection of sheep. Physicochemical studies confirmed the increase in solubility of the complex, reducing of particle sizes, amorphization of fenbendazole substance, and incorporating it with micelles of PVP. Conclusion: According to the results, supramolecular complex of fenbendazole with PVP was more active than the basic substance of fenbendazole and its anthelmintic properties were expanded. Objective: Anopheles (Cellia) epiroticus Linton & Harbach, a coastal mosquito (also called a brack¬ish mosquito), is a secondary vector species of malaria distributed throughout eastern and south¬ern regions of Thailand. This research aimed to investigate the differences of wing size and shape of this female Aonpheles species in Samut Songkhram Province, Thailand occurring over time between 2015 and 2017. Materials and Methods: Coordinates of 13 landmarks were selected and digitized. Centroid size (CS) was used to estimate wing size. Shape variables were used to estimate wing shape and were calculated from the Generalized Procrustes Analysis following principal components of the par¬tial warp. The statistically significant differences of the average wing size based on CS and wing shape based on Mahalanobis distances in each year were estimated using the non-parametric permutation testing with 1,000 cycles after Bonferroni correction with a significance level of 0.05 (p < 0.05). Results: The A. epiroticus population in year 2016 had the highest average (3.61 mm), and the population in year 2017 had the lowest (3.47 mm). In this study, there was no difference in the size of wing between A. epiroticus population in the years 2015 and 2016 (p > 0.05). The A. epiroticus population in year 2017 was significantly smaller than the population in the years 2015 and 2016 (p < 0.05). All pairwise comparisons of wing shape Mahalanobis distances were significantly different in year 2017 compared with 2015 and 2016 (p < 0.01). Conclusion: These results indicate differences of wings occur over time that affect the morpho¬logical variability of A. epiroticus. The differences in weather conditions in each year affect the adaptive and morphological changes of mosquitoes in coastal areas. [J Adv Vet Anim Res 2019; 6(2.000): 208-214]

Objectives: The objective of this study was to evaluate the impact of the antimicrobials nisin and lysozyme to control the growth of spoilage bacteria of pasteurized milk during cold storage. Materials and Methods: Nisin, lysozyme, and a mixture of them were inoculated into freshly pasteurized milk at 500 IU/ml concentrations each. The acidity, sensory evaluation, and bacteri¬ological quality of the treated pasteurized milk samples were examined at zero time and every 3 days till the samples showed the signs of spoilage, that were checked every day. Results: Obtained results showed that there was a slight increase of the titratable acidity of the control and treated samples during refrigerated storage, but the acidity increase was significantly lower in samples containing lysosomes and/or nisin than the control samples. Nisin and lyso¬zyme at 500 IU/ml concentration possessed inhibitory effect on the total bacterial, aerobic spore-formers, and psychrotrophic bacterial counts and extended the shelf-life of the treated samples. The efficacy of nisin 500 IU/ml combined with lysozyme 500 U/ml was assessed and synergistic activity has been detected, that was expressed in the form of higher inhibitory effect and extend¬ing the shelf-life of the samples up to 15 days at cold storage. Moreover, the sensory evaluation showed that nisin and lysozyme does not affect the acceptability of the examined samples. Conclusion: The obtained data indicate that nisin and lysozyme have the potential to enhance the post-process bacteriological safety of pasteurized milk during the storage period and could aid in the elimination of post-process contamination and prolong its shelf-life. Objective: The aim of this study is to determine the effect of laser diode as an alternative treat¬ment on liver dysfunction (in vivo study) that is caused by carbofuran using male mice (Mus musculus) strain Balb/C. Materials and Methods: The samples were divided into three groups, namely, Group CL (con¬trol group, no treatment), Group C+L (only treated by carbofuran treatment), and Group C+L+ (treatment group, treated by carbofuran and laser-puncture) with five replications each. After being treated, each liver slice of samples was observed using microscope to get the histology result and then scored. Results: Carbofuran contamination can lead to inflammation of cells and necrosis. The histology results and the scoring test showed that the liver cells repair with the energy dose of laser diode at 0.5 and 1.0 Joule. Conclusion: The optimum energy dose in this study was 1.0 Joule which had the closest score of inflammatory cells and necrosis to normal liver cells. [J Adv Vet Anim Res 2019; 6(4.000): 499-505]

Objective: The study was carried out to develop a wet litter model with magnesium chloride to assess the effectiveness of a polyherbal formulation (PHF) on growth performance, litter and cecal moisture (LCM) level, cecal consistency (CC) score, and footpad lesions (FPLs) score in Ross 308 broiler chickens. Materials and Methods: 1,200 one-day-old chicks were assigned into five groups: normal control, negative control [NTC; treated with 1.7% magnesium chloride hexahydrate (MgCl2.6H2O)], and three treatment groups, T1, T2, and T3, where 750, 1,000, and 2,000 gm/ton of PHF, respectively, were supplemented. All the groups were fed a basal diet until day 7. However, the NTC and treat¬ment groups were fed a diet with MgCl2 from days 8 to 42. Results: The addition of MgCl2 for 35 days worsened the growth performance traits in broilers and induced wet litter problems (FPL, high LCM, and poor CC) in the NTC group. However, PHF (750, 1,000, and 2,000 gm/ton) ameliorated the negative effect of a diet with MgCl2 on growth perfor¬mance and wet litter problems, but a better response with respect to LCM and CC was observed in 2,000 gm/ton of PHF group, followed by that in 1,000 gm/ton of PHF group and 750 gm/ton of PHF group on day 42. Conclusion: The wet litter broiler model was developed through excessive feeding of MgCl2, which caused the performance parameters to worsen and the emergence of problems associated with the wet litter. Supplementation with PHF ameliorated these problems and, therefore, it can be used for the management of wet litter in poultry. [J Adv Vet Anim Res 2019; 6(4.000): 536-543]

Objective: The morphometric and meristic variations of Xenentodon cancila was studied based on the landmark-based truss network system to assess their phenotypic variations among four different freshwater stocks, viz. Boluhorpur baor, Jhenaidah (BBJ) (n = 29); Bhairab River, Jashore (BRJ) (n = 34); Arial Khan River, Madaripur (AKRM) (n = 28), and Bohnni baor, Gopalganj (BBG) (n = 25) in Bangladesh. Materials and methods: Seven meristic characters were counted by using a needle. Eight morphometrics and 28 truss measurements were measured by using tpsDigV.2.1 software. In meristic characters, KruskalWallis test was performed to determine any significant differences, whereas, in morphometrics and truss measurements, univariate statistics and discriminant function analy-ses were carried out by using SPSS 22 version. Results: Significant differences were observed in four meristic characters among seven meristic characters in the KruskalWallis test. In univariate statistics, only nine characters were observed significantly different among eight morphometrics and 28 truss measurements. The contribution of three discriminant function analyses (DFA), in which first DFA showed 49.2%, second DFA showed 33%, and third DFA showed 17.8% on behalf of both morphometric and truss measurements. In discriminant space, the four stocks were clearly separated. Two clusters were formed among four stocks, where BBG formed a single cluster, whereas BBJ and BRJ aggregately formed another cluster. Additionally, AKRM formed a sub-cluster with BBJ. Conclusion: The preliminary information generated from the current study would be beneficial for further genetic studies and in the assessment of ecological impacts on X. cancila stocks in Bangladesh. Objectives: Drug residues in poultry products could lead to the development of antibiotic-resistant bacteria as in any living animal and human alike. Extensive use of antibiotics in animals to promote growth rate, increase feed efficiency, and for prevention of intestinal infections has led to the development of resistant bacteria in the gastrointestinal tract. The present study was conducted to evaluate the effects of biological supplementation of probiotic, phytobiotic, and their combination over antibiotic on growth performance, microbial load, and hematological parameters in Broiler. Materials and methods: Sixty-five broiler chicken were divided into five groups (12 birds in each group), namely, group A (basal diet), group B (antibiotic, Renamycin 100®), group C (phytobiotic, Galibiotic®), group D (probiotic, Bio-Top®), and group E (combination, Galibiotic®+Galibiotic®) and five were sacrificed for baseline data on day 0. Results: Average final live weight gain was highest in group D (probiotic) than other groups. The feed conversion ratio was highest in group A and lowest in the probiotic group (group C). Blood samples were collected on 14th and 28th day for hematological studies. The mean hematology values regarding the total erythrocyte count, hemoglobin concentration, packed cell volume, and erythrocyte sedimentation rate differed significantly (p < 0.05) among groups. The pH of all the treatment groups was significantly decreased compared to the control group (p < 0.05) where group C was significantly (p < 0.05) lower than all other groups. Highest total viable cell count was observed in control (group A) and total coliform count in phytobiotic (group C) was significantly lower than in other treatment groups (p < 0.05). Conclusion: It may conclude that biological supplements have a significant positive impact on growth performance, hematological parameters, and gut microbial load in broiler chicken of which the probiotic showing the best effects. Supplementation of probiotic in feed could be one of the best candidates as an alternative to antibiotics as growth promoter for safe broiler production. [J Adv Vet Anim Res 2019; 6(3.000): 409-415]

Objective: The incidence of salmonellosis in humans and animals is still high due to the occur¬rence of virulence factors in Salmonella enterica which play a role in the process of infection in the host and the spread of disease and most of the S. enterica can infect humans and animals. The present study was aimed to identify Salmonella Enteritidis and detect virulence genes related to Salmonella pathogenicity islands (SPIs) and Salmonella plasmid virulence (Spv). Materials and Methods: A total of 27 S. Enteritidis archive isolates belonging to the National Veterinary Drug Assay Laboratory (NVDAL) were used in this study. The bacteria were collected in 2016 and 2017 from samples of the cloaca and fecal swabs from layer and broiler farms in five provinces of Java Island. Isolates were cultured in specific media, biochemical tests and Gram staining. Detection of S. Enteritidis and virulence genes was done by polymerase chain reaction (PCR) method. Results: Identification of serovar showed 100% (27/27) isolates were positive for the sdfI gene (304 bp). The result confirmed that all strains were S. Enteritidis. PCR based detection of virulence genes showed that 100% of isolates had virulence genes in SPI-1 to SPI-5, namely, invA, ssaQ, mgtC, spi4D, and pipA genes. All the isolates (27/27) were also positive to spvB gene-based PCR. Conclusion: All the isolates of S. Enteritidis in this study carry virulence genes related to SPI-1 to SPI-5 and plasmid virulence. The existence of virulent genes indicates that the S. Enteritidis strain examined in this study is highly virulent and poses a potential threat of worse disease outcome in humans and animals. Objective: The objective of this research work is to know the productive and reproductive perfor¬mances and problems of local, crossbred, Nilli, and Murrah buffalo cows in selected study areas in Bangladesh. Methodology: A total of 1,241 local, crossbred, Nilli, and Murrah buffalo cows were surveyed in the selected areas with a pre-set questionnaire. Among 1,241 buffalo cows, 112 buffalo cows were randomly selected at day 0 of the estrus cycle for studying ovarian features. Results: Results showed that the average age, body condition score, and body weight were sig¬nificantly (p < 0.05) different among the studied breeds. Milk production in Murrah and lactation length in Nilli cows were significantly (p < 0.05) higher than indigenous, crossbred, Nilli, and indig¬enous, crossbred, Murrah buffalo cows, respectively. Results also illustrated that sexual maturity, estrus cycle length, insemination time after the onset of estrus, and gestation length insignifi¬cantly (p > 0.05) varied among the surveyed breed. But, the fallout of the study denoted that estrus duration, first calving age, parity number, number of service per conception, calving inter¬val, and voluntary waiting period varied significantly (p < 0.05) in different breeds. Ovarian phys¬iological characteristics such as vaginal electrical resistance, average number of follicles in two ovaries, and largest follicular diameter, estrogen, and progesterone at day 0 of the estrus cycle of local, crossbred, Nilli, and Murrah buffalo cows showed insignificantly (p > 0.05) differences.
