Cyanobacterial harmful algal blooms (CyanoHABs) have been found to transmit from N₂ fixer-dominated to non-N₂ fixer-dominated in many freshwater environments when the supply of N decreases. To elucidate the mechanisms underlying such “counter-intuitive” CyanoHAB species succession, metatranscriptomes (biotic data) and water quality-related variables (abiotic data) were analyzed weekly during a bloom season in Harsha Lake, a multipurpose lake that serves as a drinking water source and recreational ground. Our results showed that CyanoHABs in Harsha Lake started with N₂-fixing Anabaena in June (ANA stage) when N was high, and transitioned to non-N₂-fixing Microcystis- and Planktothrix-dominated in July (MIC-PLA stage) when N became limited (low TN/TP). Meanwhile, the concentrations of cyanotoxins, i.e., microcystins were significantly higher in the MIC-PLA stage. Water quality results revealed that N species (i.e., TN, TN/TP) and water temperature were significantly correlated with cyanobacterial biomass. Expression levels of several C- and N-processing-related cyanobacterial genes were highly predictive of the biomass of their species. More importantly, the biomasses of Microcystis and Planktothrix were also significantly associated with expressions of microbial genes (mostly from heterotrophic bacteria) related to processing organic substrates (alkaline phosphatase, peptidase, carbohydrate-active enzymes) and cyanophage genes. Collectively, our results suggest that besides environmental conditions and inherent traits of specific cyanobacterial species, the development and succession of CyanoHABs are regulated by co-occurring microorganisms. Specifically, the co-occurring microorganisms can alleviate the nutrient limitation of cyanobacteria by remineralizing organic compounds.

Sanguinarine has strong inhibitory effects against the cyanobacterium Microcystis aeruginosa. However, previous studies were mainly limited to laboratory tests. The efficacy of sanguinarine for mitigation of cyanobacterial blooms under field conditions, and its effects on aquatic microbial community structure remain unknown. To elucidate these issues, we carried out in situ cyanobacterial bloom mitigation tests. Our results showed that sanguinarine decreased population densities of the harmful cyanobacteria Microcystis and Anabaena. The inhibitory effects of sanguinarine on these cyanobacteria lasted 17 days, after which the harmful cyanobacteria recovered and again became the dominant species. Concentrations of microcystins in the sanguinarine treatments were lower than those of the untreated control except during the early stage of the field test. The results of community DNA pyrosequencing showed that sanguinarine decreased the relative abundance of the prokaryotic microorganisms Cyanobacteria, Actinobacteria, Planctomycetes and eukaryotic microorganisms of Cryptophyta, but increased the abundance of the prokaryotic phylum Proteobacteria and eukaryotic microorganisms within Ciliophora and Choanozoa. The shifting of prokaryotic microbial community in water column was directly related to the toxicity of sanguinarine, whereas eukaryotic microbial community structure was influenced by factors other than direct toxicity. Harmful cyanobacteria mitigation efficacy and microbial ecological effects of sanguinarine presented in this study will inform the broad application of sanguinarine in cyanobacteria mitigation.

Phycoremediation technologies significantly contribute to solving serious problems induced by heavy metals accumulation in the aquatic systems. Here we studied the mechanisms underlying Al stress tolerance in two diazotrophic cyanobacterial species, to identify suitable species for Al phycoremediation. Al uptake as well as the physiological and biochemical responses of Anabaena laxa and Nostoc muscorum to 7 days Al exposure at two different concentrations i.e., mild (100 μM) and high dose (200 μM), were investigated. Our results revealed that A. laxa accumulated more Al, and it could acclimatize to long-term exposure of Al stress. Al induced a dose-dependent decrease in photosynthesis and its related parameters e.g., chlorophyll content (Chl a), phosphoenolpyruvate carboxylase (PEPC) and Ribulose‒1,5‒bisphosphate carboxylase/oxygenase (RuBisCo) activities. The affect was less pronounced in A. laxa than N. muscorum. Moreover, Al stress significantly increased cellular membrane damage as indicated by induced H₂O₂, lipid peroxidation, protein oxidation, and NADPH oxidase activity. However, these increases were lower in A. laxa compared to N. muscorum. To mitigate the impact of Al stress, A. laxa induced its antioxidant defense system by increasing polyphenols, flavonoids, tocopherols and glutathione levels as well as peroxidase (POX), catalase (CAT), glutathione reductase (GR) and glutathione peroxidase (GPX) enzymes activities. On the other hand, the antioxidant increases in N. muscorum were only limited to ascorbate (ASC) cycle. Overall, high biosorption/uptake capacity and efficient antioxidant defense system of A. laxa recommend its feasibility in the treatment of Al contaminated waters/soils.

Controlling blooms of toxigenic phytoplankton, including cyanobacteria, is a high priority for managers of aquatic systems that are used for drinking water, recreation, and aquaculture production. Although a variety of treatment approaches exist, hydrogen peroxide (H2O2) has the potential to be an effective and ecofriendly algaecide given that this compound may select against cyanobacteria while not producing harmful residues. To broadly evaluate the effectiveness of H2O2 on toxigenic phytoplankton, we tested multiple concentrations of H2O2 on (1) four cyanobacterial cultures, including filamentous Anabaena, Cylindrospermopsis, and Planktothrix, and unicellular Microcystis, in a 5-day laboratory experiment and (2) a dense cyanobacterial bloom in a 7-day field experiment conducted in a nutrient-rich aquaculture pond. In the laboratory experiment, half-maximal effective concentrations (EC50) were similar for Anabaena, Cylindrospermopsis, and Planktothrix (average EC50 = 0.41 mg L−1) but were ∼10x lower than observed for Microcystis (EC50 = 5.06 mg L−1). Results from a field experiment in an aquaculture pond showed that ≥1.3 and ≥ 6.7 mg L−1 of H2O2 effectively eliminated Planktothrix and Microcystis, respectively. Moreover, 6.7 mg L−1 of H2O2 reduced microcystin and enhanced phytoplankton diversity, while causing relatively small negative effects on zooplankton abundance. In contrast, 20 mg L−1 of H2O2 showed the greatest negative effect on zooplankton. Our results demonstrate that H2O2 can be an effective, rapid algaecide for controlling toxigenic cyanobacteria when properly dosed.

Metalaxyl is a broad-spectrum chiral fungicide that used for the protection of plants, however extensive use of metalaxyl resulted in serious environmental problems. Thus, a study on the detoxification mechanism in algae/cyanobacteria and their ability for phycoremediation is highly recommended. Here, we investigated the physiological and biochemical responses of two cyanobacterial species; Anabaena laxa and Nostoc muscorum to R-metalaxyl toxicity as well as their ability as phycoremediators. Two different levels of R-metalaxyl, at mild (10 mg/L) and high dose (25 mg/L), were applied for one-week. We found that A. laxa absorbed and accumulated more intracellular R-metalaxyl compared to N. muscorum. R-metalaxyl, which triggered a dose-based reduction in cell growth, photosynthetic pigment content, and photosynthetic key enzymes’ activities i.e., phosphoenolpyruvate carboxylase (PEPC) and ribulose‒1,5‒bisphosphate carboxylase/oxygenase (RuBisCo). These decreases were significantly less pronounced in A. laxa. On the other hand, R-metalaxyl significantly induced oxidative damage markers, e.g., H₂O₂ levels, lipid peroxidation (MDA), protein oxidation and NADPH oxidase activity. However, these increases were also lower in A. laxa compared to N. muscorum. To alleviate R-metalaxyl toxicity, A. laxa induced the polyphenols, flavonoids, tocopherols and glutathione (GSH) levels as well as peroxidase (POX), glutathione peroxidase (GPX), glutathione reductase (GR) and glutathione-s-transferase (GST) enzyme activities. On the contrary, the significant induction of antioxidants in N. muscorum was restricted to ascorbate, catalase (CAT) and ascorbate peroxidase (APX), dehydroascorbate reductase (DHAR) enzyme activities. Although A. laxa accumulated more R-metalaxyl, it experienced less stress due to subsequent induction of antioxidants. Therefore, A. laxa may be a promising R-metalaxyl phycoremediator. Our results provided basic data for understanding the ecotoxicology of R-metalaxyl contamination in aquatic habitats and the toxicity indices among cyanobacteria.

The imprudent agricultural practices are leading to an increasing load of pesticides in agricultural fields. Thus, there is a need to minimize the harmful effect of pesticides by adopting sustainable strategies. In the recent past decade, kinetin, a plant synthetic hormone, has been reported as a pesticide toxicity alleviator in higher plants. But its role in mitigating pesticide toxicity in cyanobacteria is still limited. Thus, in current study an attempt has been made to investigate the potential of kinetin in regulating cypermethrin, an insecticide, induced toxicity in Anabaena PCC 7120 and Nostoc muscorum ATCC 27893. Cypermethrin (Cyp₁; 2 μg ml⁻¹ and Cyp₂; 4 μg ml⁻¹) showed negative impact on growth, photosynthetic pigments, photosynthetic O₂-evolution and primary photochemistry of PS II (Phi_P₀, Psi_₀, Phi_E₀) resulting in decrease in performance index (PIABS). However, under similar conditions, increases in energy flux parameters (ABS/RC, TR₀/RC, ET₀/RC and DI₀/RC) were noticed. Cypermethrin at both the doses enhanced the level of oxidative stress biomarkers (SOR, H₂O₂, and MDA equivalent contents) despite of increased antioxidant enzymatic activity (SOD, POD, CAT and GST).Under similar condition, cypermethrin at tested doses caused substantial decrease in non-enzymatic antioxidant contents (proline, cysteine and NP-SH). Nevertheless, kinetin treatment attenuated cypermethrin induced oxidative stress by further up-regulating the activity of enzymatic antioxidants and by enhancing the contents of non-enzymatic antioxidants. Thus, with the application of kinetin improved photochemistry of PS II and growth yield of both the cyanobacteria were observed even in the presence of cypermethrin. Current results establish that cypermethrin induces toxicity on photosynthesis, photosynthetic pigments and growth, and this effect was more pronounced in Anabaena PCC 7120 than Nostoc muscorum ATCC 27893. Furthermore, the potential role of kinetin in mitigating the toxicity of cypermethrin in both the cyanobacteria provides an insight to be used in paddy fields for sustainable agricultural practices.

The monitoring of macroalgae is required by the Water Framework Directive (WFD) to achieve good ecological status for coastal waters and specific questions arise for tropical ecosystems belonging to the outermost European regions. To assess the suitability of macroalgae as a biological quality indicator for La Réunion reef flats (France), we performed multivariate analyses linking the abundance and composition of macroalgae to water physico-chemistry. Three hydrological groups of stations were identified according to dissolved inorganic nitrogen (DIN) concentrations and DIN/PO4 ratios. Some indicator species were found at the N-enriched stations (Bryopsis pennata, Caulerpa lamourouxii, Chaetomoropha vieillardii, Derbesia sp., Blennothrix lyngbyacea, Sphacelaria tribuloides), and others at the non-impacted stations (Anabaena sp1, Blennothrix glutinosa, Codium arabicum, Neomeris vanbosseae). Another key result was the significant increase in red algal cover at the most N-enriched station. Our findings are discussed in the context of the application of the WFD in the outermost French regions.

Water quality assessment relies mostly on physico-chemical-based characterization; however, eutrophication and climate change advocate the abundance of toxic microcystins (MCs) producing cyanobacteria as emerging bio-indicator. In the present study, a spatial-temporal analysis was carried out at ten sampling sites of Prayagraj and Varanasi during June 2017 and March 2018 to determine the Ganga River water quality using physico-chemical parameters, cyanobacteria diversity, detection of MCs producing strains and MC-LR equivalence. Coliform bacteria, COD, NO₃-N, and phosphate are the significant contaminated parameters favoring the growth of putative MCs producing cyanobacteria. National Sanitation Foundation WQI (NSFWQI) indicates water quality, either bad or medium category at sampling points. The morphological analysis confirms the occurrence of diverse cyanobacterial genera such as Microcystis, Anabaena, Oscillatoria, and Phormidium. PCR amplification affirmed the presence of toxic microcystin (mcy) genes in uncultured cyanobacteria at all the sampling sites. The concentration of MC-LR equivalence in water samples by protein phosphatase 1 inhibition assay (PPIA) and high-performance liquid chromatography (HPLC) methods was observed in the range of 23.4–172 ng/L and 13.2–97.5 ng/L respectively which is lower than the harmful exposure limit by World Health Organization (WHO). Ganga isolate 1 was identified as Microcystis based on partial 16S rDNA sequence and its toxicity was confirmed due to presence of mcy genes and MCs production potential. These findings suggest the presence of MCs producers as new emerging parameter to monitor water quality index and identification up to species level will be valuable for restoration strategies of river Ganga.

The current study describes the mechanisms of zinc toxicity in the cyanobacterium Anabaena sphaerica after eight days treatment with 10 mg L⁻¹ ZnCl₂. The application of zinc not only showed elevated accumulation of the metal inside the cells but also exhibited devastating impacts on the cell numbers, morphology, and ultrastructure of A. sphaerica. The effects of zinc on the pigments contents, oxygen evolution rate, Fᵥ/Fₘ, electron transport rate, and carbohydrate content were also evaluated in A. sphaerica. Moreover, zinc adversely affected nutrient uptake and the cellular energy budget in the test cyanobacterium which in turn hampered heterocyst development and nitrogen fixation. Alongside, the cyanobacterium experienced zinc-mediated non-competitive inhibition of glutamine synthetase activity, curtailed synthesis of amino acids and proteins. Furthermore, drastically reduced total lipid and increased unsaturated lipid contents were also the prominent characteristics of zinc stressed A. sphaerica. Most importantly, zinc stress caused severe damages to the protein, lipid, and DNA by triggering hydrogen peroxide generation and accumulation of oxidized glutathione. Therefore, excess zinc is highly toxic to the cyanobacterium A. sphaerica, and the mechanisms of its toxicity followed a cascade of events including oxidative stress mediated geopardisation of growth and ultrastructure, metabolic derangements, and macromolecular damages.

The aim of this study was to evaluate the presence of microcystin-LR, microcystin-RR, microcystin-YR, and the neurotoxin anatoxin-a in water samples collected monthly during 1 year in San Roque reservoir (Córdoba, Argentina) to identify the environmental factors that could promote the presence of these cyanotoxins. The HPLC-UV and MS/MS analysis showed the presence of microcystin in most of the sampling times, even when Cyanobacteria were subdominant. Microcystin concentrations varied from not detectable levels to 119.0 μg L⁻¹. Thus, they frequently surpassed the guidelines suggested by WHO for drinking water (1 μg L⁻¹) and recreational exposure (20 μg L⁻¹). To the extent of our knowledge, this is the first report of anatoxin-a in freshwaters in South America. Anatoxin-a concentrations varied from not detectable levels to 6.6 ng L⁻¹, a thousand times below the provisional guideline adopted by New Zealand for drinking water. Microcystin showed significant correlation with Microcystis and Pseudoanabaena while anatoxin-a correlated with Oscillatoria and Anabaena counts. Linear discriminant analysis showed that higher pH levels and more variable chlorophyll-a concentrations were measured in San Roque reservoir when cyanotoxins were present. Lower inorganic nitrogen concentrations were observed in autumn, when the prevalence of Anabaena became significant in Cyanobacteria composition and highest anatoxin-a levels were measured. The observed dynamic of phytoplankton going together with the cyanotoxins occurrence could be explained by the hypothesis of cyanotoxins acting as allelopathic compounds. The microcystin levels measured plus the presence of anatoxin-a show the need of stronger management efforts to preserve human and wildlife health.
