Unveiling resistome profiles in the sediments of an Antarctic volcanic island

The Deception Island, located in Maritime Antarctica, is a volcanic island with geothermal activity and one of the most visited by tourists. However, the extent of the anthropogenic impact remains largely unknown and the factors shaping the resistance/tolerance mechanisms in the microbiomes from Whalers Bay ecosystems have never been investigated. In this context, this study aimed to reveal the resistome profiles of Whalers Bay sediments and correlate them with environmental factors. Samples were collected at four sites during the summer 2014/2015 along a transect of 27.5 m in the Whalers Bay sediments. DNA isolated from sediment samples was sequenced using the Illumina HiSeq platform. Bioinformatic analyses allowed the assembly of contigs and scaffolds, prediction of ORFs, and taxonomic and functional annotation using NCBI RefSeq database and KEGG orthology, respectively. Microorganisms belonging to the genera Psychrobacter, Flavobacterium and Polaromonas were shown to dominate all sites, representing 60% of taxonomic annotation. Arsenic (As), copper (Cu) and iron (Fe) were the most abundant metal resistance/tolerance types found in the microbiomes. Beta-lactam was the most common class related to antibiotics resistance/tolerance, corroborating with previous environmental resistome studies. The acridine class was the most abundant amongst the biocide resistance/tolerances, related to antiseptic compounds. Results gathered in this study reveal a repertoire of resistance/tolerance classes to antibiotics and biocides unusually found in Antarctica. However, given the volcanic nature (heavy metals-rich region) of Deception Island soils, this putative impact must be viewed with caution.

Groundwater is an essential public and drinking water supply and its protection is a goal for global policies. Here, we investigated the presence and prevalence of antibiotic residues, antibiotic resistance genes (ARGs), mobile genetic elements (MGEs), and microbial contamination in groundwater environments at various distances from urban areas. Antibiotic concentrations ranged from below detection limit to 917 ng/L, being trimethoprim, macrolide, and sulfonamide the most abundant antibiotic classes. A total of eleven ARGs (aminoglycoside, β-lactam, chloramphenicol, Macrolide-Lincosamide-Streptogramin B - MLSB, sulfonamide, and tetracycline), one antiseptic resistance gene, and two MGEs were detected by qPCR with relative abundances ranging from 6.61 × 10−7 to 2.30 × 10−1 copies/16S rRNA gene copies. ARGs and MGEs were widespread in the investigated groundwater environments, with increased abundances not only in urban, but also in remote areas. Distinct bacterial community profiles were observed, with a higher prevalence of Betaproteobacteria and Bacteroidetes in the less-impacted areas, and that of Firmicutes in the contaminated groundwater. The combined characteristics of increased species diversity, distinct phylogenetic composition, and the possible presence of fecal and/or pathogenic bacteria could indicate different types of contamination. Significant correlations between ARGs, MGEs and specific taxa within the groundwater bacterial community were identified, revealing the potential hosts of resistance types. Although no universal marker gene could be determined, a co-selection of int1, qacEΔ1 and sulI genes, a proxy group for anthropogenic pollution, with the tetC, tetO, tetW resistance genes was identified. As the tet group was observed to follow the pattern of environmental contamination for the groundwater samples investigated in this study, our results strongly support the proposal of this group of genes as an environmental tracer of human impact. Overall, the present study investigated several emerging contaminants in groundwater habitats that may be included in monitoring programs to enable further regulatory and protection measures.

Parabens are added into foodstuffs, pharmaceuticals and personal care products (PCPs) as additives extensively due to their excellent antiseptic and antibacterial effects. In the past decade, parabens have raised great concerns on their potential harm to humans. Existing studies have suggested positive correlations between PCP application and urinary paraben concentrations in females, but little is known about paraben exposure levels and health risks arising from PCP use. In this study, 150 PCP samples covering eleven categories were collected from South China and measured for the concentrations of five parabens, including methyl paraben (MeP), ethyl paraben (EtP), propyl paraben (PrP), butyl paraben (BuP) and benzyl paraben (BeP). Parabens were widely detected in PCPs, with a detection frequency of 100%, 99.3%, 80.0%, 74.0% and 13.3%, for MeP, EtP, PrP, BuP and BeP, respectively. The median concentration of Σ₅parabens was 126 μg/g with a range of 6.38–424 μg/g across all PCP samples. The contents of MeP, EtP and PrP measured in leave-on PCPs were obviously higher than those in the rinse-off ones (p < 0.05). MeP and PrP were the main paraben analogues, together accounting for 93.6% of Σ₅parabens in all PCPs. The daily intakes of parabens through dermal absorption by Chinese adult females estimated by measurements obtained in the present study were 0.15 and 83.2 μg/kg-bw/day on basis of the application of rinse-off and leave-on PCPs, respectively. Among the eleven categories, sunscreen, body lotion and mask constituted the main exposure sources of parabens to females. The hazard quotients of parabens were far less than 1, indicating no considerable health risk for Chinese adult females.

Numerous studies have confirmed the damage caused by excessive exposure to ultraviolet-A rays. Malignant melanoma and skin cancer are two of the most serious health consequences. Thus, the UV-A protectant is intended to protect the skin, especially the two primary layers of skin (epidermis that represents the interface between the body and its surroundings and dermis). Spider silk is the most powerful natural fibre due to its regeneration, biocompatibility, antimicrobial, wound healing, antiseptic, and blood clotting properties. This work targeted to determine the protective effect of spidroin extract against UV-A radiation damage. Earthworms Aporrectodea caliginosa were collected from Assiut University’s farm. Each set of ten earthworms was separated into six groups and placed in a plastic container. Webs of spiders collected from trees and old houses. Spidroin was extracted and utilised in this work to determine the potential effects of topical application on UV-A protection. The experiment is divided into two sections: (1) UV-A exposure and (2) the use of spidroin extract to protect the earthworms from ultraviolet radiation. Two control groups (1،2) of worms were not received UV-A exposure, and four groups (3,4,5,6) were exposed to UVR-A. In contrast, groups (5,6) were received spidroin extract before exposure to UV-A. Each group from the groups (3,4,5,6) was exposed for three consecutive days (¼ hour/day, ½ hour/day, and 1 h/day), using a UV-Lamp with a wavelength of 366 nm. The histopathological changes revealed that after 1⁄4 h of UV exposure, the cuticle was swollen with a slightly detached epithelium. The cuticle was down after 1⁄2 h of exposure, and the epidermis was totally damaged and necrosed. After 1 h, the exposure showed destruction of the epidermis in the circular muscle with a loss of muscle filament integrity, varying size, and altered nucleus form, along with mild disintegration of longitudinal muscle. Spidroin extract is critical for earthworm protection against UV-A radiation damage and able to regeneration. For the first time, morphological and histological analysis was established to detect the Spidroin extract evaluated for topical application on earthworms. Earthworms can be considered as a robust human skin model prior to UV-A exposure. It induces a complete protective effect against UV-A radiation damage in earthworms.

This study focused on the presence of three biocidal products specific to healthcare facilities, i.e. chlorhexidine digluconate (CHD), bis(aminopropyl)laurylamine (BAPLA), and didecyldimethylammonium chloride (DDAC), in a hospital sewage system. Five sampling campaigns were conducted in 2016 and 2017 throughout the entire Poitiers University Hospital sewage system. DDAC concentrations ranging from 933 ± 119 to 3250 ± 482 μg/L were detected in 24-h composite samples, while lower concentrations (both within the same range) were detected for the two other compounds (i.e. 25 ± 5 to 97 ± 39 μg/L for CHD and 18 ± 3 to 142 ± 16 μg/L for BAPLA). Based on these findings, a mass balance was determined for these discharged compounds to compare the quantities detected in discharges to the amounts used for healthcare in the hospital. Hence, 60–90% of the quantities of DDAC used were found to be present at the hospital sewage outfall. Higher percentages of CHD (100–242%) were noted because of the high presumably quantities used for antiseptic applications, which were not considered in mass balance calculation. Finally, only 10–30% of BAPLA quantities used were detected at the site outfall. Analysis of the results for the different sampling points revealed the nature of the emission sources. For surface applications of DDAC and BAPLA, management of hospital linen is thus a major source of discharged biocidal products, probably following the washing of biocide-soaked textiles used for hospital facility maintenance. Moreover, discharge of biocidal products from a healthcare establishment depends especially on biocide handling practices in the emitting establishment. For BAPLA, compliance with hospital recommended dosages and practices whereby operators are required to prepare tailored quantities of detergents and disinfectants for each specific task could largely explain the limited release of this compound.

Being one of the most common wood in furniture and decoration, Fraxinus mandshurica Rupr. (F. mandshurica) is a frequently used material for wood products. However, the resulting odor problem has not yet been solved so far. Thus, this study focused on exploring odor-active compounds and investigating the effect of environmental conditions on emission and odor attributes. F. mandshurica was studied via TD–GC–MS/O combined with microchamber. Totally, 15 kinds of key odor-active compounds were detected from F. mandshurica. The main odorous components were found to be aromatics, aldehydes and ketones, esters, alcohols and acids. Six kinds of attributes played a key role in the overall odor character of F. mandshurica. Fragrant was the predominant odor impressions, followed by vinegar and antiseptic solution. The proportion of the odor-active compounds concentration to total concentration decreased with an increase in the temperature and relative humidity, while the ratio of air exchange rate to the loading factor had little effect on this. The total concentration of F. mandshurica increased as the temperature and relative humidity increased, but these decreased as the ratio of air exchange rate to the loading factor increased, while the total odor intensity reduced with the rise in the ratio of air exchange rate to the loading factor. Time was found to be the most effective factor for eliminating emissions. Considering the environmental conditions in this study, 40 °C, 60% relative humidity and a 1.0 m³·m⁻² h⁻¹ ratio of air exchange rate to loading factor are recommended to accelerate the emission of F. mandshurica. The optimum condition was found to be higher temperature, higher relative humidity and higher ratio of air exchange rate to loading factor.

The occurrence of silver (Ag) in urban effluents is partly associated with the increasing use of silver nanoparticles (Ag NPs) as an antiseptic agent in various consumer products. Distinction among Ag forms must be taken into account in the assessment of exposure and toxicological effects to aquatic organisms. Wastewater treatment processes effectively remove Ag particles and colloids (mostly > 95%), but this still leaves notable concentrations (in order of ng/L) escaping to effluent-receiving waters. Total suspended Ag concentrations in various studied effluents ranged from 0.1 to 6 ng/L. The purpose of this study was then to measure and characterize Ag NPs in urban effluents for their concentrations and size distribution using the single particle ICP-MS technique (SP-ICP-MS). Wastewater influents and effluents from various treatment plants—from aerated lagoons to advanced treatment technology—were collected for three sampling days. Our results showed the presence of Ag NP in all samples with concentrations reaching 0.5 ng/L on a mass basis. However, on a particle number basis, Ag NP concentrations (expressed in particle/mL) in the 20–34-nm fraction (up to 3400 particles/mL) were much more abundant (> 700%) than in the > 35-nm larger fraction. The proportion of Ag at the nanoscale (1–100 nm) represents less than 8% of the total suspended Ag for all effluent samples, regardless of their origins. A significant correlation (linear regression: r² > 0.7) was observed between Ag NP and total suspended Ag concentrations in investigated effluents. Because Ag nanotoxicity is size dependent, the determination of size distribution and exposure concentration on a particle number basis is urgently needed for risk assessment of this class of nanoparticles.

Formaldehyde (FA) is an aldehyde used in antiseptics and adhesives. The World Health Organization (WHO) and other institutes have linked FA to sick building syndrome and allergic diseases. Recent studies have reported that cadavers embalmed using formalin and ethanol-based preservative solutions release FA vapor during dissection and that FA vapor may adversely affect students and lecturers in gross anatomy laboratories. However, few details have been reported correlating dissection stage with increased FA vapor release. In this study, we evaluated the vapor level of FA released in each dissection stage. Six cadavers for which consent was given for use in anatomy research and education were examined in this study. Using an active sampling method, FA vapor was collected above the thoracoabdominal region of each dissected cadaver. FA was eluted from each sampler using acetonitrile and analyzed by high-performance liquid chromatography. Our data show that FA levels significantly increase after skin incision and that the vapor level of FA released differs between male and female cadavers. We also found that subcutaneous adipose tissues of the thoracoabdominal-region release FA vapor and that female cadavers release significantly higher levels of FA per kilogram of subcutaneous adipose tissue than do male cadavers. Based on these data, we propose the methods be developed to prevent exposure to FA vapors released from cadavers.
اظهر المزيد [+] اقل [-]Fate of para-toluenesulfonamide (p-TSA) in groundwater under anoxic conditions: modelling results from a field site in Berlin (Germany)

This article reports on a field modelling study to investigate the processes controlling the plume evolution of para-toluenesulfonamide (p-TSA) in anoxic groundwater in Berlin, Germany. The organic contaminant p-TSA originates from the industrial production process of plasticisers, pesticides, antiseptics and drugs and is of general environmental concern for urban water management. Previous laboratory studies revealed that p-TSA is degradable under oxic conditions, whereas it appears to behave conservatively in the absence of oxygen (O₂). p-TSA is ubiquitous in the aquatic environment of Berlin and present in high concentrations (up to 38 μg L⁻¹) in an anoxic aquifer downgradient of a former sewage farm, where groundwater is partly used for drinking water production. To obtain refined knowledge of p-TSA transport and degradation in an aquifer at field scale, measurements of p-TSA were carried out at 11 locations (at different depths) between 2005 and 2010. Comparison of chloride (Cl⁻) and p-TSA field data showed that p-TSA has been retarded in the same manner as Cl⁻. To verify the transport behaviour under field conditions, a two-dimensional transport model was setup, applying the dual-domain mass transfer approach in the model sector corresponding to an area of high aquifer heterogeneity. The distribution of Cl⁻and p-TSA concentrations from the site was reproduced well, confirming that both compounds behave conservatively and are subjected to retardation due to back diffusion from water stagnant zones. Predictive simulations showed that without any remediation measures, the groundwater quality near the drinking water well galleries will be affected by high p-TSA loads for about a hundred years.
اظهر المزيد [+] اقل [-]The treatment of wastewater containing pharmaceuticals in microcosm constructed wetlands: the occurrence of integrons (int1–2) and associated resistance genes (sul1–3, qacEΔ1)

The aim of this study was to analyze the occurrence of sulfonamide resistance genes (sul1–3) and other genetic elements as antiseptic resistance gene (qacEΔ1) and class 1 and class 2 integrons (int1–2) in the upper layer of substrate and in the effluent of microcosm constructed wetlands (CWs) treating artificial wastewater containing diclofenac and sulfamethoxazole (SMX), which is a sulfonamide antibiotic. The bacteria in the substrate and in the effluents were equipped with the sul1–2, int1, and qacEΔ1 resistance determinants, which were introduced into the CW system during inoculation with activated sludge and with the soil attached to the rhizosphere of potted seedlings of Phalaris arundinacea ‘Picta’ roots (int1). By comparing the occurrence of the resistance determinants in the upper substrate layer and the effluent, it can be stated that they neither were lost nor emerged along the flow path. The implications of the presence of antibiotic resistance genes in the effluent may entail a risk of antibiotic resistance being spread in the receiving environment. Additionally, transformation products of SMX were determined. According to the obtained results, four (potential) SMX transformation products were identified. Two major metabolites of SMX, 2,3,5-trihydroxy-SMX and 3,5-dihydroxy-SMX, indicated that SMX may be partly oxidized during the treatment. The remaining two SMX transformation products (hydroxy-glutathionyl-SMX and glutathionyl-SMX) are conjugates with glutathione, which suggests the ability of CW bacterial community to degrade SMX and resist antimicrobial stress.
