The Use of Astacus leptodactylus (Eschscholtz, 1823) as a Test Species for Toxicity Evaluation of Municipal Wastewater Treatment Plant Effluents

This study evaluated the responses of biochemical biomarkers in Astacus leptodactylus exposed to treated municipal effluents discharged into Keban Dam Lake, Elazığ Elazig, Turkey. A. leptodatyclus were exposed to treated municipal effluents and Catalase (CAT), Superoxide dismutase (SOD) activity and lipid peroxidation (TBARS), glutathione (GSH) levels were measured as oxidative stress biomarkers. SOD activity was increased after exposing to treated municipal effluents for 24th and 96th h. CAT activities were decreased from 25.29 to 14.12 nmol/min/ml compared to control in the group exposed to treated municipal effluents for 24 h but it increased after 96 h exposure. GSH levels were decreased from 9.08 to 3.77 µM compared to control, but MDA levels were increased both at 24 th h and 96 th h after exposure to treated municipal effluents. CAT, SOD activities and MDA and GSH levels in the hepatopancreas of A. leptodactylus are sensitive and suitable biochemical biomarkers for evaluating the toxicity of the treated municipal effluent complex mixtures. Treated municipal effluents exposure was found to cause sub-lethal responses in A. leptodactylus suggesting oxidative stress.

Penconazole is one of the most widely used fungicides all over the world, and since it spreads to large environments, its toxic effects on non-target organisms are of great concern. The toxic effects of penconazole on crayfish (Astacus leptodactylus), which is a bioindicator in freshwater ecosystems and consumed economically, are not known. Therefore, in this study, the purpose was to contribute to the literature on the potential harmful effects of penconazole on a non-target species, Astacus leptodactylus. For this aim, the acute toxicity (96 h) of penconazole was examined. The 96-h LC₅₀ value of penconazole was detected as 18.7 mg L⁻¹. Four concentrations of penconazole (18.7 mg L⁻¹, 9.35 mg L⁻¹, 4.68 mg L⁻¹, 2.34 mg L⁻¹) were applied to crayfish for 96 h. The results showed that penconazole had destructive effects on esterase mechanisms by inhibiting acetylcholinesterase (AChE) and carboxylesterase (CaE) activities. Significant increases were observed in all antioxidant parameters (superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione reductase (GR), glutathione S-transferase (GST), reduced glutathione (GSH), malondialdehyde (MDA)) in all doses except the lowest concentration (2.34 mg L⁻¹). All adenosine triphosphatase (ATPase) activities (Na⁺/K⁺-ATPase, Mg²⁺-ATPase, Ca²⁺-ATPase, total ATPase) had significant dose-related inhibition in both gill and muscle tissues. In summary, our findings show that acute penconazole administration to crayfish causes significant toxic effects on esterase, antioxidative parameters, and metabolic enzymes.

Bisphenol A (BPA), an endocrine-disrupting chemical (EDC), has strong potential for daily exposure to humans and animals due to its persistence and widespread in the environment, so its effects directly concern public health. Although invertebrates represent important components of aquatic ecosystems and are at significant risk of exposure, there is little information about the biological effects of EDCs in these organisms. Astacus leptodactylus used in this study is one of the most consumed and exported freshwater species in Europe. In this study, the 96-h effect of BPA on A. leptodactylus was examined using various biomarkers. The LC₅₀ value of BPA was determined as 96.45 mg L⁻¹. After 96 h of exposure to BPA, there were increases in superoxide dismutase (SOD), glutathione peroxidase (GPx), glutathione S-transferase (GST), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and lactate dehydrogenase (LDH) activities and levels of malondialdehyde (MDA), and total oxidant status context (TOSC), and there were decreases in the activity of glutathione reductase (GR), carboxylesterase (CaE), acetylcholinesterase (AChE), Na⁺/K⁺ ATPase, Mg²⁺ ATPase, Ca²⁺ ATPase, and total ATPase and the total antioxidant context (TAC). From the results of this study, it can be concluded that BPA has significant toxic effects on A. leptodactylus based on the selected biochemical parameters of antioxidant, cholinergic, detoxification, and metabolic systems in crayfish even at low doses. Thus, it can be said that BPA can seriously threaten the aquatic ecosystem and public health.

Freshwater crayfish are bioindicators of environmental pollution, often used for the assessment of heavy metal (HM) presence in the tissues, a time-consuming and expensive task. In this study, we propose the use of the vibrational spectroscopy to detect in a fast, non-destructive and sensitive way the presence of HM in the cephalothorax exoskeleton of the freshwater crayfish. Incorporation of HM into the cephalothorax exoskeleton was investigated under controlled laboratory conditions. In particular, the cephalothorax exoskeleton of five crayfish species (Astacus leptodactylus, Procambarus clarkii, Austropotamobius pallipes, Faxonius limosus, and Pacifastacus leniusculus) was analyzed by attenuated total reflection–Fourier transformed infrared (ATR–FTIR) spectroscopy in the presence or absence of cadmium (Cd), chromium (Cr), lead (Pb), nickel (Ni), and zinc (Zn) up to 4 weeks at various concentrations (0.01, 0.1, 1, 10, ppm). The ATR–FTIR profile of the crayfish cephalothorax exoskeleton was compatible with the presence of amorphous calcium carbonate, chitin, and proteins. The incubation with the HM revealed two main modifications: the shift of the peak from 859 to 872 cm⁻¹ and the appearance of a peak at 712 cm⁻¹. Both are ascribable to the HM interaction with calcium carbonate. The absorbance of both peaks increased along with the time of incubation, and the HM concentration. We conclude that ATR–FTIR analysis can be a useful, quick, and cost-sensitive tool to detect HM presence in the crayfish cephalothorax exoskeleton. However, it has to be regarded as a non-specific analytical technique for assessing HM contamination, since it is unable to discriminate between different HM.

The discharge of textile effluents into the environment without appropriate treatment poses a serious threat for the aquatic organisms. The present study was undertaken to investigate biochemical response of crayfish Astacus leptodactylus exposed to textile wastewater (TW) treated by indigenous white rot fungus Coriolus versicolor. Glutathione S-transferase (GST), cytochrome P450 1A1 (CYP1A1), and acetylcholinesterase (AchE) levels in hepatopancreas and abdomen tissues of crayfish exposed to untreated, treated, and diluted rates (1/10) in both TW during 24 and 96 h were tested. Physiochemical parameters (electrical conductivity (EC), chemical oxygen demand (COD), pH, and total dissolved solid (TDS)) of TW were determined before and after treatment. Physiochemical parameters of TW decreased after treatment. The GST activity and AchE were generally increased, but CYP1A1 activity was decreased in hepatopancreas tissue of crayfish exposed to different kinds of untreated TW. After treatment by indigenous white rot fungus (C. versicolor), GST and CYP1A1 activities were returned to control values, while AchE activities were increasing further. In this study, only GST and CYP1A1 activities of A. leptodactylus confirmed the efficiency of TW treatment with C. versicolor.

The crayfish play an essential role in the biomonitoring and may reflect ambient water quality through the biochemical, behavioural and physiological reactions. To assess whether narrow-clawed crayfish Astacus leptodactylus can respond by heart rate changes to presence in water of such biocide as chloramine-T, adult males were exposed to its low (2 and 5 mg L⁻¹), moderate (10 mg L⁻¹, commonly used in industry and aquaculture) and exceeded (20 and 50 mg L⁻¹) concentrations. In addition, a physical stress test evaluated energy expenditure following the chemical trials. Three key reactions (cardiac initial, first-hour and daily prolonged exposure) were discussed with particular focus on crayfish initial reaction as the most meaningful in on-line water quality biomonitoring. After short-term exposure to both chloramine-T concentrations, crayfish were found to respond rapidly, within 2–5 min. According to heart rate changes, the 1-h exposure did not adversely affect crayfish at either concentration, as well as during daily exposure to 10 mg L⁻¹. As assessed by the heart rate, the 24-h exposure to 50 mg L⁻¹of chloramine-T was toxic for crayfish and led to substantial loss of energy that became apparent during subsequently conducted physical stress. The results supported a hypothesis that crayfish vital functions are connected with environment they inhabit closely enough to serve as biological monitors. Crayfish were tolerant to short-term chloramine-T exposure, while rapid crayfish reaction to an increased chemical level indicated their high sensitivity, an essential attribute of real-time environmental assessment.
