Investigation of the effects of dichlorvos poisoning on AMPK signaling pathway in chicken brain tissues

Dichlorvos is a common crop insecticide widely used by people which causes extensive and serious environmental pollution. However, it has been shown that organophosphorus poisoning causes energy metabolism and neural disorders. The overall purpose of this study was to investigate the damage to brain tissue and the changes in AMPK signaling pathway-related gene expression after dichlorvos poisoning in chickens. White-feathered broiler chickens, as the research subjects of this experiment, were divided into three groups: control group, low-dose group (77.5% dichlorvos at 1.13 mg/kg dose) and high-dose group (77.5% dichlorvos at 10.2 mg/kg dose). Clinical symptoms were observed after modeling, and an integrative analysis was conducted using HE staining microscopy, immune-histochemical microscopy, electron microscopy and PCR arrays. The results showed that the high-dose group had more obvious dyspnea, salivation, convulsion and other neurological phenomena. Pathological sections showed that nuclear disintegration of neurons was most obvious in the low-dose group, and apoptosis of brain cells was most obvious in the high-dose group, and the mitochondrial structure was destroyed in the two poisoned group, i.e. low-dose group and high-dose group. PCR arrays showed that AMPK signaling pathway was inhibited and the expressions of genes involved in energy metabolism (ACACA and PRKAA1) were significantly changed. Furthermore, genes associated with protein synthesis (EIF4EBP1) were significantly upregulated. FASN and HMGCR expressions were significantly increased. There were significant changes in the expressions of cell cycle-related genes (STK11, TP53 and FOXO3). Organophosphate poisoning can cause a lot of nuclear disintegration of brain neurons, increases cell apoptosis, disrupts the energy metabolism of mitochondrial structure, and inhibits the AMPK signaling pathway. These results provide a certain idea and basis for studying the mechanism of AMPK signaling after organophosphorus poisoning and provide a research basis for the prevention and treatment of organophosphorus poisoning.

Baseline emission values of greenhouse gases were not well established for commercial poultry barns in cold regions, including Canada, due to a lack of well-designed field studies. Emission factors of carbon dioxide (CO₂), methane (CH₄), and nitrous oxide (N₂O), were acquired for a commercial broiler barn and cage-layer barn in the Canadian Prairies climate. Between March 2015 and February 2016, monthly measurements throughout the year for the layer barn and over 6 flocks for the broiler barn, and diurnal measurements in the mild, warm, and cold seasons for both barns were conducted, respectively. The ventilation rate was estimated based on a CO₂ mass balance method; thus CO₂ emissions were quantified by the CIGR (2002) models. The CH₄ and N₂O emissions present at low levels from global perspective for both barns; the cold climate proved to be a major reason for the lower CH₄ emission from the layer barn. Considerable seasonal effect was observed only for N₂O emissions from the broiler barn, and for CH₄ and N₂O emissions from the layer barn, both with higher emissions in the mild and warm seasons than in the cold season. The big diurnal variations of CO₂ emissions for the layer barn demonstrated the uncertainty of the seasonal results by snapshot measurements and correction factors (from −20.9% to −22.5%) were obtained. Besides, the difference of CH₄ and N₂O concentrations and emissions as well as CO₂ concentrations between best-case (the first day after manure removal) and worst-case conditions (the last day before manure removal) was not obvious for the layer barn. Additionally, changes of temperature and ventilation rate were likely to have more impact on N₂O emission for the broiler barn and more impact on CH₄ emission for the layer barn than on the other two gas emissions, both with positive correlations.

Particulate matter (PM) released from the processes of livestock production has a negative impact on the health of animals and workers. Herein, the concentration, major chemical components, morphology and microbiological compositions of particulate matter 2.5 (PM2.5, particles with aerodynamic diameter less than 2.5 μm) in a broiler breeding house were investigated. The results showed that the PM2.5 distribution in the chicken house was affected by the illumination, draught fans, chicken frame structure and activity of the chickens in the broiler breeding house. Component analysis showed that organic carbon (OC) accounted for the largest proportion, and followed by element carbon (EC), SO42−, NO3−, NH4+, Na+, K+ and Ca2+. Ultrastructural observations revealed that the shape of PM2.5 had a round, rectangular, chain-like and irregular shape. The concentration of endotoxin was approximately 0.3 EU/m3. Microbiological analysis showed that at the genus level, the pathogenic bacteria included Staphylococcus, Corynebacterium, Enterococcus, Parabacteroides, Escherichia and Megamonas. The abundant harmful fungi were Aspergillus, Scopulariopsis, Wallemia, and Fusarium. Through redundancy analysis (RDA) analysis, we determined that OC, EC, Na+, K+, and NH4+ had strong correlations with Brachybacterium, Brevibacterium, Corynebacterium, Escherichia, Scopulariopsis and Microascus. SO42− was closely related to Scopulariopsis and Salinicoccus. Salinicoccus was also strongly correlated with NO3−. Our results indicated that feed, faeces, and outside soot are contributed to the increase in PM2.5 concentration in the chicken house, while the sources of the dominant bacterial and fungi might be feed, faeces, suspended outside soil and cereal crops.

Although the poultry production sector plays a key role in sustaining the majority of animal protein demand in Egypt, the deleterious effects of widespread antibiotic resistance on health and environment are currently not well recognized. Litter and dropping samples from broiler and layer poultry farms as well as, tilapia samples from the Nile River and aquaculture farms were collected from Upper Egypt. Samples were extracted and examined for tetracycline residues [tetracycline (TC), chlortetracycline (CTC), oxytetracycline (OTC) and doxycycline (DC)] using HPLC. In addition, tetracycline resistance genes [tet (M), tet (W), tet (Q) and tet (G)] were screened from pooled intestinal contents collected from twelve broiler farms in Upper Egypt. The antibiotic resistance genes results revealed that tet (W) was confirmed to be expressed in all intestinal samples. In contrast, tet (Q) and tet (M) were detected only in 42% and 17% of the samples, respectively. CTC and OTC were the antimicrobial compounds with the highest concentrations in poultry litter and droppings, with concentrations of 6.05 and 2.47 μg g−1 (CTC) and 5.9 and 1.33 μg g−1 (OTC), respectively. However, the concentrations of DC were significantly higher than those of the other compounds in both aquaculture and Nile River tilapia. The tetracycline residue levels in aquaculture tilapia were significantly higher than those in Nile River tilapia. The hazard quotients (HQs) exceeded 1 for OTC, CTC and DC, which highlights the great risk of using broiler litter to fertilize agricultural land. Moreover, the presence of DC and CTC indicates that consumption of aquaculture tilapia poses a considerable health risk. Therefore, poultry litter or droppings containing tetracycline residues and tet resistance determinants used for aquaculture or as farmland fertilizers could be major sources of antibiotic resistance in fish, humans and environment.

The current experiment was conducted to evaluate the effects of feeding dried sweet orange peel (SOP) and lemon grass leaves (LGL) as feed additives on broiler growth performance, serum metabolites, and antioxidant status. A total of 192-day-old (Ross 308) broiler chickens were distributed randomly into 4 dietary treatments with 4 replicates per each treatment. The dietary treatments included a control diet without any feed additive (T1), a diet containing 0.8 % SOP (T2), a diet containing 0.8 % LGL (T3), and a diet containing combination of 0.4 % SOP + 0.4 % LGL (T4) was fed during the growth period from 22 to 42 days. Feed intake (FI), body weight gain (BWG), feed conversion ratio (FCR), carcass traits, serum components, and antioxidant status were measured. At the end of the experimental period, the results indicated that supplementation of SOP and LGL alone or in combination did not significantly (P > 0.05) affect BWG, FI, FCR, and carcass characteristics in broiler chickens. Serum total protein was increased significantly (P < 0.05) in T3 and T4 compared to the other treatments. Also, serum globulin increased significantly (P < 0.05) in the treated groups. Serum glucose, low density lipoprotein, triglyceride, and very low density lipoprotein decreased significantly (P < 0.05) in the treatment groups, while cholesterol and high-density lipoprotein decreased in T2 compared to the other groups. Significantly (P < 0.05) higher total antioxidant status was observed in T2 compared to the other treatments. In conclusion, these results indicate that SOP, LGL, and their combination may positively modify some serum components and the antioxidant status without any beneficial effect on growth performance and carcass traits in broiler chickens.

Tibial dyschondroplasia (TD) is most the common tibiotarsal bone disease in rapidly growing birds throughout the world. There is accumulating evidence that COX-2 abnormal expression in tibia plays an important role in TD progression. So, the regulation of COX-2 is an ever more appealing target for therapeutic intervention in TD. Astragaloside IV has an indispensable role in maintaining COX-2 expression in many diseases. So, we designed this study to use Astragaloside IV (AST-IV) against TD-affected chickens. A total of 180 Arbor Acres chickens were randomly divided in the control group, TD group, and Astr (AST-IV-treated chickens) group. During the experiment, mortality, feed conversion ratio, physiological changes, biochemical criterion, liver antioxidant enzymes, and gene expression of COX-2 were examined in all the chicken groups at various days. The results showed that AST-IV administration restored the growth performance and tibia lesions and decreased the mortality as compared with TD chickens. The biochemical criterion (ALP, AST, and ALT) of serum and liver antioxidant enzymes (SOD, GSH-Px, MDA, and T-AOC) improved after the administration of AST-IV. The COX-2 gene was upregulated significantly (P < 0.05) in TD chickens. Whereas, AST-IV treatment downregulated both gene and protein expression of COX-2 significantly (P < 0.05) in TD-affected chickens. AST-IV recovered tibial dyschondroplasia chickens by increasing the growth performance, ameliorating tibial cartilage damage, and decreasing COX-2 expression. In conclusion, AST-IV can be used to prevent thiram-induced TD in chickens.

This study evaluated the effects of Bacillus subtilis, Saccharomyces boulardii, oregano, and calcium montmorillonite on the physical growth, intestinal histomorphology, and blood metabolites in Salmonella-challenged birds during the finisher phase. In this study, a total of 600 chicks (Ross 308) were randomly distributed into the following dietary treatments: basal diet with no treatment; infected with Salmonella; T1, infected + avilamycin; T2, infected + Bacillus subtilis; T3, Saccharomyces boulardii; T4, infected + oregano; T5, infected + calcium montmorillonite. Our results indicated that feed consumption, body weight gain, total body weight, and feed conversion ratio increased significantly (P < 0.01) in T1 and T2. Villus width increased significantly (P < 0.01) in T1 while the total area was significantly (P < 0.01) higher in T1 and T2 among the treatment groups. Blood protein was significantly (P < 0.01) high in T3 and T4; however, the glucose concentration was significantly (P < 0.01) high in T2, T3, and T4. The treatments increased significantly (P< 0.01) in the treatment groups compared to the negative control. Aspirate aminotransferase (AST) was significantly (P < 0.05) low in T3 compared to the positive control. In conclusion, the results indicated that supplementation of Bacillus subtilis and calcium montmorillonite improved the production performance compared to other feed additives in broiler chicks infected with Salmonella during the finisher phase.

Limited data are available on ammonia (NH₃), nitrous oxide (N₂O), carbon dioxide (CO₂) and methane (CH₄) emissions from poultry housing in Mediterranean countries. The aim of the present study was to assess the NH₃, N₂O, CO₂ and CH₄ emission rates from commercial breeding hen and broiler houses under Mediterranean climate conditions. Research was conducted at one commercial breeding hen house and in two commercial broiler houses located in central Portugal. The environmental conditions, gas concentrations and ventilation rates were measured in the cold (8.0 ± 2.1 °C) and hot (20.7 ± 1.9 °C) season for the breeding hen house, whereas for the two broiler houses, measurements were made during one fattening cycle in the fall (17.3 ± 1.7 °C) season. Results showed that the annual average emission rates for breeding hen and broiler houses were 0.52 ± 0.27 and 0.06 ± 0.01 for NH₃, 0.030 ± 0.042 and 0.006 ± 0.001 for N₂O, 169.6 ± 56.2 and 58.0 ± 15.1 for CO₂ and 0.092 ± 0.131 and 0.0113 ± 0.0002 g day⁻¹ bird⁻¹ for CH₄, respectively. The N₂O emission rates observed in breeding hen houses may have been overestimated, being higher than previously reported for Mediterranean countries.

Chromium is used in daily life and has a wide range of functions. It plays an important role in protein synthesis and carbohydrate and lipid metabolism. Chromium is found in trivalent Cr(III) and hexavalent Cr(VI) form; Cr(III) is relatively stable and intimately participates with many phenomena of metabolisms. Whereas, Cr(VI) is toxic, which results in growth inhibition and leading to changes in components of antioxidant systems as well as secondary metabolites. However, the molecular mechanism that is involved in Cr (VI)-induced hepatotoxicity is still unclear. For this purpose, 40 chickens were randomly assigned into two groups: the normal group (feeding the basic diet and clear water), the chromium group (16%LD₅₀, 74.24 mg/kg/day K₂Cr₂O₇). The samples were subjected to pathological examination and UHPLC-QE-MS non-target metabolomics method for metabolomics analysis of broiler liver using principal component analysis (PCA) and partial least squares discriminant analysis (OPLS-DA). The central venous cells of the broiler liver in the chromium poisoning group showed turbidity and flaky necrosis, nuclear condensation, nuclear rupture, and even nuclear dissolution. The differential metabolite analysis between the chromium poisoning and the control group showed that 32 differential metabolites were upregulated and 15 were downregulated in positive ion mode. Whereas,17 differential metabolites were downregulated, and 35 were downregulated in negative ion mode (P ≤ 0.05). The potential marker substances are oleic acidamide, farnesylacetone, betaine, taurine, choline, and galactinol. Additionally, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways showed that the lipid metabolism, carbohydrate metabolism, nucleotide metabolism, amino acid metabolism, energy metabolism, membrane transport, digestive system, and nervous system were the most important metabolic pathways in the liver. This study provides a theoretical basis for the future understanding of the pathogenesis of chromium poisoning and a new insight of the subsequent molecular mechanism of chromium hepatotoxicity.
