Tetracyclines uptake from irrigation water by vegetables: Accumulation and antimicrobial resistance risks

Wastewater irrigation may introduce antibiotic residues in the soil-plant systems. This study aimed to investigate the uptake of tetracyclines by spinach and collard greens and assess associated ecological and human health risks. Synthetic wastewater spiked with 1 ppm and 10 ppm of oxytetracycline, doxycycline, and tetracycline was used to grow vegetables in a greenhouse pot experiment. The uptake and accumulation of the tetracyclines were low and residual concentrations in the soil were negligible. All the tetracyclines were detected at concentrations ranging from 1.68 to 51.41 μg/g (spinach) and 1.94–30.95 μg/g (collard greens). The accumulation rate was in a dose-response scenario with a bioconcentration factor of 6.34 mL/kg (spinach) and 2.64 mL/kg (collard greens). Oxytetracycline had the highest accumulation in leaves, followed by doxycycline and tetracycline, and the residual concentrations followed the same order. The highest residual concentration was in soils receiving 10 ppm oxytetracycline. Residual concentrations in the soil were lower than accumulated levels and exerted negligible ecological risks. Tetracyclines accumulation in spinach significantly differed between the vegetables demonstrating a subspecies difference in uptake and accumulation. Ecological risk quotient (RQ) and human health risk quotient (HQ) were below thresholds that would exert toxicity and resistance selection impacts. Although RQs and HQs are low (<0.1), this study shows that the vegetables accumulate tetracyclines from irrigation water, posing plausible human health risks to allergic individuals. Similarly, the ecological risks cannot be ignored because the synergistic and antagonistic effects of sublethal concentrations can perturb ecosystem processes.

Microplastics (MPs) pollution in Tangxun Lake, the largest urban lake in China, was investigated. The average MPs pollution in sediment (1.81 ± 1.75 × 10⁴ items kg⁻¹) is at a high level, while the MPs in lakeshore water (917.77 ± 742.17 items m⁻³) is in the middle to low level compared with existing studies, which is related to the government's protection. Fragments and fibers are the most common shapes in sediment and water, respectively. MPs size <1 mm dominates in the sediment, while the MPs in water has a larger size. The distribution of MPs in the inner lake shows that pellets tend to “hidden” in sediments. Suspected MPs are randomly selected for polymer detection by Micro-Raman microscopy. Polypropylene (PP), polyethylene (PE) and polyethylene terephthalate (PET) are the most common polymer types in water, sediment and atmospheric deposition MPs samples. The input of wastewater, fishery and surrounding human activities are the main sources of MPs in sediment. Atmospheric deposition has a great impact on the distribution of MPs, while the contribution of surface runoff to lake MPs is not remarkable. In addition, MPs in sediments have exceeded the environmental carrying capacity. More attention should be focused on the sediment, where huge amounts of MPs are “hidden”.

The inherent oxidation potential (OP) of atmospheric particulate matter has been shown to be an important metric in assessing the biological activity of inhaled particulate matter and is associated with the composition of PM₂.₅. The current study examined the chemical composition of 388 personal PM₂.₅ samples collected from students and guards living in urban and suburban areas of Beijing, and assessed the ability to predict OP from the calculated metrics of carcinogenic risk, represented by ELCR (excess lifetime cancer risk), non-carcinogenic risk represented by HI (hazard index), and the composition and sources of the particulate matter using multiple linear regression methods. The correlations between calculated ELCR and HI and the measured OP were 0.37 and 0.7, respectively. HI was a better predictor of OP than ELCR. The prediction models based on pollutants (Model_1) and pollution sources (Model_2) were constructed by multiple linear regression method, and Pearson correlation coefficients between the predicted results of Model_1 and Model_2 with the measured volume normalized OP are 0.81 and 0.80, showing good prediction ability. Previous investigations in Europe and North America have developed location-specific relationships between the chemical composition of particulate matter and OP using regression methods. We also examined the ability of relationships between OP and composition, sources, developed in Europe and North America, to predict the OP of particulate matter in Beijing from the composition and sources determined in Beijing. The relationships developed in Europe and North America provided good predictive ability in Beijing and it suggests that these relationships can be used to predict OP from the chemical composition measured in other regions of the world.

Fine particulate matter (PM₂.₅) has been associated with risk of oral and respiratory diseases. However, the biological mechanisms of adverse oral and respiratory health response to PM₂.₅ fluctuation have not been well characterized. This study aims to explore the relationships of PM₂.₅ with airway inflammation, salivary biomarkers and buccal mucosa microbiota. We performed a panel study among 40 college students involving 4 follow-ups from August to October 2021 in Hefei, Anhui Province, China. Health outcomes included fractional exhaled nitric oxide (FeNO), salivary biomarkers [C-reactive protein (CRP), cortisol, lysozyme and alpha-amylase] and buccal mucosa microbial diversity. Linear mixed-effect models were applied to explore the cumulative impacts of PM₂.₅ on health indicators. PM₂.₅ was positively correlated with FeNO, CRP, cortisol and alpha-amylase, while negatively with lysozyme. Per 10-μg/m³ increase in PM₂.₅ was linked to maximum increments in FeNO of 10.71% (95%CI: 2.01%, 19.41%) at lag 0–24 h, in CRP of 7.10% (95%CI: 5.39%, 8.81%) at lag 0–24 h, in cortisol of 1.25% (95%CI: 0.44%, 2.07%) at lag 0–48 h, and in alpha-amylase of 2.12% (95%CI: 0.53%, 3.71%) at lag 0–24 h, while associated with maximum decrement in lysozyme of 0.53% (95%CI: 0.12%, 0.95%) at lag 0–72 h. Increased PM₂.₅ was linked to reduction in the richness and evenness of buccal microbe and o_Bacillales and o_Bacteroidales were identified as differential microbes after PM₂.₅ inhalation. Bio-information analysis indicated that immunity system pathway was the most important enriched abundant process altered by PM₂.₅ exposure. In summary, short-term PM₂.₅ exposure may impair oral and respiratory health by inducing inflammatory and stress responses, weakening immune function and altering buccal mucosa microbial diversity.

Chlorinated paraffins (CPs) are industrial chemicals produced in large quantities. Short-chain CPs (SCCPs) were classified as persistent organic pollutants under the Stockholm Convention in 2017. Medium-chain CPs (MCCPs) became candidate persistent organic pollutants in 2021. CPs are now ubiquitously found in the environment. Honey bees can be exposed to CPs during foraging, and this exposure subsequently results in the contamination of honey and other bee products along with colony food production and storage. Here, SCCP and MCCP concentrations in honey collected from Chinese apiaries in 2015 and 2021 were determined. Total CP concentrations in honey from 2021 to 2015 were comparable, but the ratio of MCCPs/SCCPs was higher in 2021 than in 2015. SCCP and MCCP congener group profiles in all honey samples were similar and dominated by C₁₀–₁₁Cl₆–₇ and C₁₄Cl₆–₇, respectively. MCCP concentrations were also higher than SCCP concentrations in bees, pollen, and wax but not in bee bread, which were all collected in 2021. The order of average CP concentrations was determined as wax > bee > pollen > bee bread > honey. Poor relationships were found between SCCP concentrations in honey and other samples, but a relationship between MCCP concentrations in honey and other samples was observed. Migration tests of CPs in plastic bottles showed essentially no migration into honey during storage. The risks to humans from CPs in honey are low.

The emergence of resistance genes is a global phenomenon that poses a significant threat to both animals and humans. Lakes are important reservoirs of genes that confer resistant to antibiotics and metals. In this study, we investigated the distribution and diversity of antibiotic resistance genes (ARGs) and metal resistance genes (MRGs) in the sediment of Daihai Lake using high-throughput sequencing and metagenomic analysis. The results indicated that all sampling sites had similar bacterial community structures, with Proteobacteria, Actinobacteria, Firmicutes, and Bacteroidetes being the most abundant. A total of 16 ARG types containing 111 ARG subtypes were deposited in the sediment. Among the resistance genes to bacitracin, multidrug, macrolide-lincosamide-streptogramin (MLS), tetracycline, beta-lactam, and sulfonamide were the dominant ARG types, accounting for 89.9–94.3% of the total ARGs. Additionally, 15 MRG types consisting of 146 MRG subtypes were identified. In all samples, MRGs of the same type presented resistance to Pb, Ni, Hg, W, Zn, Ag, Cr, Fe, As, Cu, and multimetals. Overall, the distribution and diversity of antibiotic and metal resistance genes showed no significant differences in the samples. Plasmids (91.03–91.82%) were the most dominant mobile genetic elements in the sediments of Daihai Lake. Network analysis indicated that the target ARGs and MRGs were significantly positively correlated with the microorganisms. Potential hosts for various ARGs and MRGs include Proteobacteria, Euryarchaeota, Actinobacteria, Chloroflexi, and Bacteroidetes.

Ground-based Multi-Axis Differential Optical Absorption Spectroscopy (Max-DOAS) measurements of nitrogen dioxide (NO₂) were continuously obtained from January to November 2019 in northeastern China (NEC). Seasonal variations in the mean NO₂ vertical column densities (VCDs) were apparent, with a maximum of 2.9 × 10¹⁶ molecules cm⁻² in the winter due to enhanced NO₂ emissions from coal-fired winter heating, a longer photochemical lifetime and atmospheric transport. Daily maximum and minimum NO₂ VCDs were observed, independent of the season, at around 11:00 and 13:00 local time, respectively, and the most obvious increases and decreases occurred in the winter and autumn, respectively. The mean diurnal NO₂ VCDs at 11:00 increased to at 08:00 by 1.6, 5.8, and 6.7 × 10¹⁵ molecules cm⁻² in the summer, autumn and winter, respectively, due to increased NO₂ emissions, and then decreased by 2.8, 4.2, and 5.1 × 10¹⁵ molecules cm⁻² at 13:00 in the spring, summer, and autumn, respectively. This was due to strong solar radiation and increased planetary boundary layer height. There was no obvious weekend effect, and the NO₂ VCDs only decreased by about 10% on the weekends. We evaluated the contributions of emissions and transport in the different seasons to the NO₂ VCDs using a generalized additive model, where the contributions of local emissions to the total in the spring, summer, autumn, and winter were 89 ± 12%, 92 ± 11%, 86 ± 12%, and 72 ± 16%, respectively. The contribution of regional transport reached 26% in the winter, and this high contribution value was mainly correlated with the northeast wind, which was due to the transport channel of air pollutants along the Changbai Mountains in NEC. The NO₂/SO₂ ratio was used to identify NO₂ from industrial sources and vehicle exhaust. The contribution of industrial NO₂ VCD sources was >66.3 ± 16% in Shenyang due to the large amount of coal combustion from heavy industrial activity, which emitted large amounts of NO₂. Our results suggest that air quality management in Shenyang should consider reductions in local NO₂ emissions from industrial sources along with regional cooperative control.

Due to the high radiotoxicity in high concentrations, plutonium isotopes have drawn high attentions in the consideration of radiation risk, their sources, level, environmental behaviors, including deposition, retention and migration behaviors. However, such research in the Qinghai-Tibet Plateau is still missing, where is deemed as an environmental sensitive area. ²³⁹,²⁴⁰Pu in surface soil collected from the Qinghai-Tibet Plateau were determined for the first time in this work. The concentrations of ²³⁹,²⁴⁰Pu are in the range of 0.0176–1.95 Bq/kg, falling into the reported ranges in the background areas from the similar latitude belt. The ²⁴⁰Pu/²³⁹Pu atomic ratio range was measured to be 0.146–0.225, which is similar with the global fallout values. Both indicate that the global fallout is the major source of plutonium in this region, and the low plutonium level will not cause any radiation risk so far. Based on the statistical analysis of the possible parameters (organic content, moisture content, average annual precipitation, altitudes, topography and human activity), the large variations of ²³⁹,²⁴⁰Pu concentrations were mainly attributed to the retention process related factors including soil organic content and human activity disturbances. While, the deposition related factors including the average annual precipitation, altitudes, topography made insignificant influence on the spatial distribution of ²³⁹,²⁴⁰Pu concentrations due to the low ²³⁹,²⁴⁰Pu concentrations in atmosphere, less wet deposition amount and insignificant re-suspended amount. The highest ²³⁹,²⁴⁰Pu concentrations of 0.805–1.95 Bq/kg were mainly due to the good retention condition in the sampling sites with higher soil organic content and less human activity disturbances.

Considerable investigations have been carried out to address the relationship between ambient fine particulate matter (PM₂.₅) and blood pressure (BP) in patients with hypertension. However, few studies have explored the influence of PM₂.₅ and its constituents on Trimethylamine N-oxide (TMAO), an established risk factor for hypertension and cardiovascular disease (CVD), particularly in severely air-polluted areas. To explore the potential impact of PM₂.₅ constituents on BP, plasma hormones, and TMAO, a panel study was conducted to investigate changes in BP, plasma hormones, and TMAO in response to ambient air pollution exposure in stage 1 hypertensive young adults. Linear mixed effect models were used to estimate the cumulative effects of fine particulate matters (PM₂.₅) and its constituents on BP, plasma hormones and TMAO. We found that one interquartile range (IQR) (35 μg/m³) increase in 0–1 day moving-average PM₂.₅ concentrations was statistically significantly associated with elevated systolic blood pressure (SBP), diastolic blood pressure (DBP), and mean arterial pressure (MAP) with estimated values of 0.13 (95% confidence interval (CI): 0.03 to 0.23) mmHg, 0.18 (95% CI: 0.08 to 0.28) mmHg, and 0.17 (95% CI: 0.09 to 0.26) mmHg, respectively. Hormone disturbance in the renin-angiotensin-aldosterone system was also associated with PM₂.₅ exposure. Elevated TMAO levels with an IQR increase for 0–4, 0–5, 0–6 moving-average concentrations of PM₂.₅ were found, and the increased values ranged from 26.28 (95% CI: 2.92 to 49.64) to 60.78 (31.95–89.61) ng/ml. More importantly, the PM₂.₅-bound metal constituents, such as manganese (Mn), titanium (Ti), and selenium (Se) showed robust associations with elevated BP and plasma TMAO levels. This study demonstrates associations between PM₂.₅ metal constituents and increased BP, changes in plasma hormones and TMAO, in stage 1 hypertensive young adults. Source control, aiming to reduce the emission of PM₂.₅-bound metals should be implemented to reduce the risk of hypertension and CVD.
