Emission of antibiotics into riverine environments affects aquatic ecosystem functions and leads to the development of antibiotic resistance. Here, the profiles of forty-four antibiotics and eighteen antibiotic resistance genes (ARGs) were analyzed in two large rivers of the Pearl River System. In addition, the risks of ecotoxicity and resistance selection posed by the antibiotics were estimated. As compared to the reservoirs, the river sections close to the urban and livestock areas contained more antibiotics and ARGs. Seasonal variations of antibiotics (higher in the dry season) and relative ARGs (normalized by 16S rRNA gene, higher in the wet season) were found in the water, but not in the sediment. Sulfonamide resistance genes were the most prevalent ARGs in both river water and sediment. Antibiotic concentration was correlated with ARG abundance in the water, indicating that antibiotics play a critical role in ARG spread. In addition, oxytetracycline was the most abundant antibiotic with concentrations up to 2030 ng/L in the water and 2100 ng/g in the sediment respectively, and posed the highest risks for resistance selection. Oxytetracycline, tetracycline and sulfamethoxazole were expected to be more ecotoxicologically harmful to aquatic organisms, while ofloxacin, enrofloxacin, norfloxacin, chlortetracycline, oxytetracycline and tetracycline posed ecotoxicological risks in the sediment. The Nanliujiang river with intensive livestock activities was contaminated by antibiotics and ARGs and faced high ecotoxicological and resistance selection risks. Collectively, these findings reflect the impacts of anthropogenic activities on the spread of antibiotic resistance in large river basins.

Current swine industry practice is to house animals in confinement facilities which capture and store feces and urine as slurry in pits below the production area. Additives and disinfectants may be introduced into the manure pits. This study was conducted to measure the effects of additives and disinfectants on temporal changes in swine slurry characteristics. Slurry from a commercial swine production facility in southeast Nebraska, USA was collected and transferred to 57 L reactors located within a greenhouse. Selected additives and disinfectants were added to the reactors and physical properties, chemical characteristics, and antibiotic concentrations were monitored for 40 days. Concentrations of dry matter (DM), total nitrogen (TN), phosphorus pentoxide (P₂O₅), calcium (Ca), magnesium (Mg), zinc (Zn), iron (Fe), manganese (Mn), and copper (Cu) were significantly greater than the Control in each of the reactors containing additives. The reactors in which the additives MOC-7, More Than Manure®, Sludge Away, and Sulfi-Doxx were introduced had significantly greater values of chemical oxygen demand (COD), total volatile solids (TVS), total suspended solids (TSS), total solids (TS), dry matter (DM), TN, P₂O₅, Ca, Mg, Zn, Fe, Mn, Cu and chlortetracycline than the other additive treatments. Concentrations of TVS and TSS were significantly lower in the reactors containing Clorox® and Virkon™ than the other disinfectant treatments. The total dissolved solids (TDS) concentration of 26,500 mg L⁻¹ and pH value of 7.27 obtained for the reactors containing Tek-Trol were significantly greater than measurements obtained for the other treatments. Concentrations of chlortetracycline and tiamulin of 8840 and 28.8 ng g⁻¹, respectively, were significantly lower for the treatments containing Tek-Trol. The sodium (Na) concentration of 1070 mg L⁻¹ measured in the reactors containing Clorox® was significantly greater than values for the other disinfectant treatments. The introduction of selected additives and disinfectants may influence certain physical properties, chemical characteristics, and antibiotic concentrations of swine slurry.

The potential impact of six antibiotics (chlortetracycline, tetracycline and tylosin; sulfamethoxazole, sulfamethazine and trimethoprim) on plant growth and soil quality was studied by using seed germination test on filter paper and plant growth test in soil, soil respiration and phosphatase activity tests. The phytotoxic effects varied between the antibiotics and between plant species (sweet oat, rice and cucumber). Rice was most sensitive to sulfamethoxazole with the EC10 value of 0.1 mg/L. The antibiotics tested inhibited soil phosphatase activity during the 22 days' incubation. Significant effects on soil respiration were found for the two sulfonamides (sulfamethoxazole and sulfamethazine) and trimethoprim, whereas little effects were observed for the two tetracyclines and tylosin. The effective concentrations (EC10 values) for soil respiration in the first 2 days were 7 mg/kg for sulfamethoxazole, 13 mg/kg for sulfamethazine and 20 mg/kg for trimethoprim. Antibiotic residues in manure and soils may affect soil microbial and enzyme activities. Terrestrial ecotoxicological effects of antibiotics are related to their sorption and degradation behavior in soil.

Although abundant evidence has suggested that early-life antibiotic exposure was associated with adipogenesis later in life, limited data were available on the effect of intrauterine antibiotic exposure on infant growth and growth speed. Additionally, few studies have investigated the role of the neonatal gut microbiota in the above association. In this study, we examined the association between intrauterine cumulative antibiotic exposure and infant growth and explored the potential role of the neonatal gut microbiota in the association. 295 mother-child pairs from the Shanghai Maternal-Child Pairs Cohort (MCPC) study were included, and meconium samples and infant growth measurements were assessed. Z-scores of length-for-age, weight-for-age (weight-for-age), and body mass index (BMI)-for-age (BMI-for-age) were calculated. Eighteen common antibiotics were measured in meconium. Multivariable linear regression models were applied to test the interrelationships between antibiotic exposure, diversity indicators, and the relative abundance of selected bacterial taxa from phylum to genus levels from least absolute shrinkage and selection operator (LASSO) and infant growth indicators. The detection rates of the 18 antibiotics, except for chlortetracycline, penicillin, and chloramphenicol, were below 10 %. Penicillin was found to be positively associated with infant growth at birth and with growth speed from 2 to 6 months. The Pielou and Simpson indexes were negatively associated with meconium penicillin. Nominally significant associations between penicillin and the relative abundances of several bacterial taxa from the phyla Proteobacteria, Bacteroidetes, and Firmicutes were found. The Pielou and Simpson indexes were also found to be negatively associated with infant growth. Among taxa selected from LASSO regression, the relative abundances of the phyla Actinobacteria and Firmicutes and order Bifidobacteriales were found to be significantly associated with weight and BMI growth speeds from 2 to 6 months. In conclusion, intrauterine antibiotic exposure can affect infant growth. The neonatal gut microbiota might play a role in the abovementioned association.

The residual characteristics and the adsorption-desorption behaviors of azoxystrobin (AZO) as well as the soil ecological effects in the individual repeated treatments of AZO and its combination with chlorothalonil (CTL), chlortetracycline (CTC) and ciprofloxacin (CIP) were systematically studied in organic manure (OM)-amended soil under laboratory conditions. The presence of CTL, CTC, and CIP, both individually and combined, decreased the sorption affinity of AZO with the Freundlich adsorption and desorption coefficient decreasing by 0.3–24.2%, and CTC and CIP exhibited greater adverse effects than CTL. AZO dissipated slowly and the residues significantly accumulated during ten repeated treatments. The dissipation of AZO was inhibited to different degrees in the combined treatments. Biolog analysis revealed that the soil microbial functional diversity in the OM-soil + AZO and OM-soil + AZO + CTL treatments was higher than that in the OM-soil treatment during the former three repeated treatments, but which was inhibited during the latter seven repeated treatments. The soil microbial functional diversity in the OM-soil + AZO + CTC, OM-soil + AZO + CIP and OM-soil + AZO + CTL + CTC + CIP treatments was inhibited during the ten repeated treatments compared with OM-soil treatment. Metagenomic results showed that all repeated treatments significantly increased the relative abundance of Actinobacteria, but significantly decreased that of Proteobacteria and Firmicutes during the ten repeated treatments. Furthermore, the relative abundance of soil dominant bacterial genera Rhodococcus, Mycobacterium and Arthrobacter in all the repeated treatments significantly increased by 1.5–1283.9% compared with the OM-soil treatment. It is concluded that coexistence of CTL, CTC and CIP, both individually and combined, with AZO can inhibit the dissipation of AZO, reduce the adsorption affinity of AZO on soil, and alter the soil microbial community structure and functional diversity.

The fate of veterinary antibiotics (VAs) in soil environment is determined by the hydrophilic performance and solubility of VAs and the type of soil. In this study, sulfadiazine (SDZ) and chlortetracycline (CTC) were selected as target pollutants, and a batch sorption method was used to find out the single and sorption competitive behavior and mechanism of the target pollutants on loess soil. Kinetic studies showed the apparent sorption equilibrium was reached 0–6 h for CTC and 0–12 h for SDZ. The sorption kinetics of VAs on loess soil were fitted well with a pseudo-second order kinetic model. Sorption thermodynamic data indicated the isotherm sorption of both SDZ and CTC on loess soil was fitted well with Freundlich isothermal (R², 0.960–0.975) and linear models (R², 0.908–0.976). The sorption affinity of CTC (Kd, 290–1620 L/kg for CTC) was much greater than that of SDZ (Kd, 0.6–4.9 L/kg for SDZ). The results also suggest that SDZ may be easily mobilized or leached from loess soil at neutral and alkaline pH, while CTC may be easily mobilized or leached at neutral pH. The sorption of each single target pollutant on the outer layer complex decreased with increasing ionic strength. Higher initial concentrations resulted in greater sorption capacity of target pollutants on loess soil increased. The sorption capacities of CTC and SDZ in the mixed system were lower than the sorption capacity of each single system, showing a competitive sorption behavior of CTC and SDZ during the sorption process. Overall, CTC showed the highest sorption potential in loess soil, whereas SDZ showed a high leaching risk in loess soil. These findings contribute to understanding the fate of different VAs in loess in the natural environment.

Tetracyclines (TCs) are frequently detected in agricultural soils worldwide, causing a potential threat to crops and human health. In this study, diffusive gradients in thin films technique (DGT) was used to measure the distribution and exchange rates of three TCs (tetracycline (TC), oxytetracycline (OTC) and chlortetracycline (CTC)) between the solid phase and solution in five farmland soils. The relationship between the accumulated masses with time suggested that TCs consumption in soil solution by DGT would induce the supply from the soil solid phase. The distribution coefficient for the labile antibiotics (Kdl), response time (Tc) and desorption/adsorption rates (kb and kf) between dissolved and sorbed TCs were derived from the dynamic model of DIFS (DGT induced fluxes in soils). The Kdl showed similar sizes of labile solid phase pools for TC and OTC while larger pool sizes were observed for CTC in the soils. Although the concentrations of CTC were lowest in soil solution, the potential hazard caused by continuous release from soil particles could not be ignored. The long response time (>30 min in most cases) suggested that the resupply of TCs from soil solids was limited by their desorption rates (1.26-121 × 10−6 s−1). The soils in finer texture, with higher clay and silt contents (<50 μm) showed a greater potential for TCs release.

Antibiotic and heavy metal pollution of aquatic environments are issues of serious concern, and the macrophyte Myriophyllum aquaticum may provide a viable solution for the removal of these contaminants. However, the toxic effects of coexisting tetracyclines (TCs) and Cu(II) on this plant species are currently unclear. In the present study, we constructed wetland microcosms planted with M. aquaticum and spiked these with three TCs (tetracycline, oxytetracycline, and chlortetracycline) and Cu(II) at concentrations ranging from 100 to 10,000 μg/L to investigate how Cu(II) influences the growth and tolerance responses of plants to TCs. After 12 weeks, we found that TCs had accumulated in the plants, and that plant growth and characteristics were significantly affected by the levels of both TCs and Cu(II). While low Cu(II) levels had a synergistic effect on the accumulation of TCs, high levels were observed to reduce accumulation. However, low levels of TCs and Cu(II) had a hormesis effect on plant growth, with plant biomass and leaf chlorophyll content decreasing and the malondialdehyde content and activities of antioxidant enzymes gradually increasing with an increase in TC dosage. The coexistence of low levels of Cu(II) was, however, found to alleviate these adverse effects. Principal component analysis revealed a close relationship among plant biomass, chlorophyll content, malondialdehyde content, and antioxidant enzyme activities. Considering that the Cu/TC ratio was shown to markedly affect M. aquaticum growth, the respective proportions of these pollutants should be taken into consideration in the future design of constructed wetlands.

Frequently detected residuals of antibiotics in crops has drawn increasing attention from research community and the general public. This study was conducted under the controlled environmental conditions to investigate the uptake, translocation and distribution of three different veterinary antibiotics (VAs) in plants of Zea mays L. (maize, the third largest crop in the world, especially in China) and the associated mechanisms. The distribution color-maps of mixed-VAs showed that the highest RCF (root concentration factors) values of chlortetracycline (CTC) and sulfamethoxazole (SMZ) were found in the 0.5–2.0 mm zone (cell division zone), while the highest RCF value of sulfathiazole (ST) was in the 6.0–8.0 mm zone (elongation zone) of root tips (0.5–10.0 mm) after 120 h of exposure to VAs. The translocation factor (TF) of CTC was greater than 1.0, but the TFs of SMZ and ST were less than 1.0 under addition of single antibiotic. However, the TFs of three VAs were all greater than 1.0 at the end of exposure under addition of mixed-VAs. The dissipation of antibiotics by maize was also demonstrated by harvesting all plant parts in an enclosed system. The possible mechanisms for uptake and translocation of VAs in maize were investigated by adding multiple respiration inhibitors into the culture solution. The RCFs of VAs were suppressed heavily by salicylhydroxamic acid (SHAM) and sodium azide (NaN3), which indicates that the uptake of VAs was an active process. The results of TFs and stem concentration factors (SCFs) of CTC and SMZ in HgCl2 treatments revealed that the translocation of VAs was associated with the aquaporin activity in maize. The findings from this study will have significant implications for the management of crop food contamination by VAs and for the development of phytoremediation technology for antibiotics in the environment.

Manure treatment technologies are rapidly developing to minimize eutrophication of surrounding environments and potentially decrease the introduction of antibiotics and antibiotic resistant genes (ARGs) into the environment. While laboratory and pilot-scale manure treatment systems boast promising results, antibiotic and ARG removals in full-scale systems receiving continuous manure input have not been evaluated. The effect of treatment on ARGs is similarly lacking. This study examines the occurrence and transformation of sulfonamides, tetracyclines, tetracycline degradation products, and related ARGs throughout a full-scale advanced anaerobic digester (AAD) receiving continuous manure and antibiotic input. Manure samples were collected throughout the AAD system to evaluate baseline antibiotic and ARG input (raw manure), the effect of hygenization (post-pasteurized manure) and anaerobic digestion (post-digestion manure) on antibiotic and ARG levels. Antibiotics were analyzed by liquid chromatography-tandem mass spectrometry (LC-MS/MS), and the ARGs tet(O), tet(W), sul1 and sul2 were analyzed by quantitative polymerase chain reaction (Q-PCR). Significant reductions in the concentrations of chlortetracycline, oxytetracycline, tetracycline and their degradation products were observed in manure liquids following treatment (p < 0.001), concomitant to significant increases in manure solids (p < 0.001). These results suggest sorption is the major removal route for tetracyclines during AAD. Significant decreases in the epimer-to-total residue ratios for chlortetracycline and tetracycline in manure solids further indicate degradation is desorption-limited. Moreover, sul1 and sul2 copies decreased significantly (p < 0.001) following AAD in the absence of sulfonamide antibiotics, while tetracyclines-resistant genes remained unchanged. A cross-sectional study of dairy farms utilizing natural aeration and liquid-solid separation treatments was additionally performed to compare levels of antibiotics and ARGs found in AAD with the levels in common manure management systems. The concentration of antibiotics in raw manure varied greatly between farms while minimal differences in ARGs were observed. However, significant (p < 0.01) differences in the levels of antibiotics and ARGs (except tet(W)) were observed in the effluents from the three different manure management systems.
