Mercury (Hg) is a persistent and widespread heavy metal with neurotoxic effects in wildlife. While bioaccumulation of Hg has historically been studied in aquatic food webs, terrestrial consumers can become contaminated with Hg when they feed on aquatic organisms (e.g., emergent aquatic insects, fish, and amphibians). However, the extent to which dietary connectivity to aquatic ecosystems can explain patterns of Hg bioaccumulation in terrestrial consumers has not been well studied. Bats (Order: Chiroptera) can serve as a model system for illuminating the trophic transfer of Hg given their high dietary diversity and foraging links to both aquatic and terrestrial food webs. Here we quantitatively characterize the dietary correlates of long-term exposure to Hg across a diverse local assemblage of bats in Belize and more globally across bat species from around the world with a comparative analysis of hair samples. Our data demonstrate considerable interspecific variation in hair total Hg concentrations in bats that span three orders of magnitude across species, ranging from 0.04 mg/kg in frugivorous bats (Artibeus spp.) to 145.27 mg/kg in the piscivorous Noctilio leporinus. Hg concentrations showed strong phylogenetic signal and were best explained by dietary connectivity of bat species to aquatic food webs. Our results highlight that phylogeny can be predictive of Hg concentrations through similarity in diet and how interspecific variation in feeding strategies influences chronic exposure to Hg and enables movement of contaminants from aquatic to terrestrial ecosystems.

Recently, concerns have been raised that residues of glyphosate-based herbicides may interfere with the homeostasis of the intestinal bacterial community and thereby affect the health of humans or animals. The biochemical pathway for aromatic amino acid synthesis (Shikimate pathway), which is specifically inhibited by glyphosate, is shared by plants and numerous bacterial species. Several in vitro studies have shown that various groups of intestinal bacteria may be differently affected by glyphosate. Here, we present results from an animal exposure trial combining deep 16S rRNA gene sequencing of the bacterial community with liquid chromatography mass spectrometry (LC-MS) based metabolic profiling of aromatic amino acids and their downstream metabolites. We found that glyphosate as well as the commercial formulation Glyfonova®450 PLUS administered at up to fifty times the established European Acceptable Daily Intake (ADI = 0.5 mg/kg body weight) had very limited effects on bacterial community composition in Sprague Dawley rats during a two-week exposure trial. The effect of glyphosate on prototrophic bacterial growth was highly dependent on the availability of aromatic amino acids, suggesting that the observed limited effect on bacterial composition was due to the presence of sufficient amounts of aromatic amino acids in the intestinal environment. A strong correlation was observed between intestinal concentrations of glyphosate and intestinal pH, which may partly be explained by an observed reduction in acetic acid produced by the gut bacteria. We conclude that sufficient intestinal levels of aromatic amino acids provided by the diet alleviates the need for bacterial synthesis of aromatic amino acids and thus prevents an antimicrobial effect of glyphosate in vivo. It is however possible that the situation is different in cases of human malnutrition or in production animals.

The gut microbiome is sensitive to diet and environmental exposures and is involved in the regulation of host metabolism. Additionally, gut inflammation is an independent risk factor for the development of metabolic diseases, specifically atherosclerosis and diabetes. Exposures to dioxin-like pollutants occur primarily via ingestion of contaminated foods and are linked to increased risk of developing cardiometabolic diseases. We aimed to elucidate the detrimental impacts of dioxin-like pollutant exposure on gut microbiota and host gut health and metabolism in a mouse model of cardiometabolic disease. We utilized 16S rRNA sequencing, metabolomics, and regression modeling to examine the impact of PCB 126 on the microbiome and host metabolism and gut health. 16S rRNA sequencing showed that gut microbiota populations shifted at the phylum and genus levels in ways that mimic observations seen in chronic inflammatory diseases. PCB 126 reduced cecum alpha diversity (0.60 fold change; p = 0.001) and significantly increased the Firmicutes to Bacteroidetes ratio (1.63 fold change; p = 0.044). Toxicant exposed mice exhibited quantifiable concentrations of PCB 126 in the colon, upregulation of Cyp1a1 gene expression, and increased markers of intestinal inflammation. Also, a significant correlation between circulating Glucagon-like peptide-1 (GLP-1) and Bifidobacterium was evident and dependent on toxicant exposure. PCB 126 exposure disrupted the gut microbiota and host metabolism and increased intestinal and systemic inflammation. These data imply that the deleterious effects of dioxin-like pollutants may be initiated in the gut, and the modulation of gut microbiota may be a sensitive marker of pollutant exposures.

Osteoporosis or enhanced bone loss is one of the most commonly occurring bone conditions in the world, responsible for higher incidence of fractures leading to increased morbidity and mortality in adults. Bone loss is affected by various environmental factors including diet, age, drugs, toxins etc. Microcystins are toxins produced by cyanobacteria with microcystin-LR being the most abundantly found around the world effecting both human and animal health. The present study demonstrates that MC-LR treatment induces bone loss and impairs both trabecular and cortical bone microarchitecture along with decreasing the mineral density and heterogeneity of bones in mice. This effect of MC-LR was found due to its immunomodulatory effects on the host immune system, wherein MC-LR skews both T cell (CD4+ and CD8+ T cells) and B cell populations in various lymphoid tissues. MC-LR further was found to significantly enhance the levels of osteoclastogenic cytokines (IL-6, IL-17 and TNF-α) along with simultaneously decreasing the levels of anti-osteoclastogenic cytokines (IL-10 and IFN-γ). Taken together, our study for the first time establishes a direct link between MC-LR intake and enhanced bone loss thereby giving a strong impetus to the naïve field of “osteo-toxicology”, to delineate the effects of various toxins (including cyanotoxins) on bone health.

Studies of circulating levels in difference sex and age classes, and maternal transfer of bisphenol A, 4-tert-octylphenol and 4- nonylphenol in the Baltic grey seal were performed from 2014-2017. Blood was collected from long-term captive adult males, pregnant females and pups. Milk was collected from nursing females. The aim of this study was not only to determine the concentrations of phenol derivatives, i.e. bisphenol A (BPA), 4-tert-octylphenol (OP) and 4-nonylphenol (NP), but also to try to evaluate the transfer of these compounds to the next generation in the final stage of foetal life and in the first few weeks of life in juvenile marine mammals. The measurements were carried out using high performance liquid chromatography. The obtained data show that all phenol derivatives are present in the blood of males, females and pups (range <0.07–101 ng·cm⁻³) and in female milk (range <0.1–406.3 ng·cm⁻³). The main source of phenol derivatives in organisms is food exposure. Gender, age, or number of births were not observed to have a significant effect on changes in phenol derivative levels in seal blood within the breeding group. In the prenatal stage of life, a small amount of BPA and alkylphenols was passed on to the offspring through the placenta. In the blood of the offspring the concentration of these compounds exceeded the concentration in the mother's blood 1.5-fold. During nursing, females detoxified their systems. Level of phenol derivatives in the pups blood increased linearly with its increasing concentrations in the mother's milk. On the other hand, the seafood diet which started after the physiological fasting stage of the pup, stabilised the levels of phenol derivatives below 10 ng ∙ cm⁻³.

Microplastics exposure could be detrimental to marine organisms especially under high concentrations. However, few studies have considered the multiphasic nature of marine invertebrates' life history and investigated the impact of experiencing microplastics during early development on post-metamorphic stages (legacy effect). Many planktonic larvae can feed selectively and it is unclear whether such selectivity could modulate the impact of algal food-sized microplastic. In this two-stage experiment, veligers of Crepidula onyx were first exposed to additions of algae-sized micro-polystyrene (micro-PS) beads at different concentrations, including ones that were comparable their algal diet. These additions were then either halted or continued after settlement. At environmentally relevant concentration (ten 2-μm microplastic beads ml⁻¹), larval and juvenile C. onyx was not affected. At higher concentrations, these micro-PS fed larvae consumed a similar amount of algae compared to those in control but grew relatively slower than those in the control suggesting that ingestion and/or removal of microplastic was/were energetically costly. These larvae also settled earlier at a smaller size compared to the control, which could negatively affect post-settlement success. Juvenile C. onyx receiving continuous micro-PS addition had slower growth rates. Individuals only exposed to micro-PS during their larval stage continued to have slower growth rates than those in the control even if micro-PS had been absent in their surroundings for 65 days highlighting a legacy effect of microplastic exposure.

There is increasing evidence linking levels of trace elements (TEs) in adipose tissue with certain chronic conditions (e.g., diabetes or obesity). The objectives of this study were to assess concentrations of a selection of nine essential and possibly-essential TEs in adipose tissue samples from an adult cohort and to explore their socio-demographic, dietary, and lifestyle determinants. Adipose tissue samples were intraoperatively collected from 226 volunteers recruited in two public hospitals from Granada province. Trace elements (Co, Cr, Cu, Fe, Mn, Mo, Se, V, and Zn) were analyzed in adipose tissue by high-resolution inductively coupled plasma mass spectrometry (HR-ICP-MS). Data were collected on socio-demographic characteristics, lifestyle, diet, and health status by face-to-face interview. Predictors of TE concentrations were assessed by using multivariable linear and logistic regression. All TEs were detected in all samples with the exception of Se (53.50%). Iron, zinc, and copper showed the highest concentrations (42.60 mg/kg, 9.80 mg/kg, and 0.68 mg/kg, respectively). Diet was the main predictor of Cr, Fe, Mo, and Se concentrations. Body mass index was negatively associated with all TEs (β coefficients = −0.018 to −0.593, p = 0.001–0.090) except for Mn and V. Age showed a borderline-significant positive correlation with Cu (β = 0.004, p = 0.089). Residence in a rural or semi-rural area was associated with increased Co, Cr, Fe, Mo, Mn, V and Zn concentrations and with β coefficients ranging from 0.196 to 0.544 (p < 0.05). Furthermore, individuals with higher educational level showed increased Cr, Co, Fe and V concentrations (β coefficients = 0.276–0.368, p = 0.022–0.071). This is the first report on the distribution of these TEs in adipose tissue and on their determinants in a human cohort and might serve as an initial step in the elucidation of their clinical relevance.

A diet fortified with 2,2′, 4,4′-tetrabromodiphenyl ether (BDE-47: 0, 10, 100, and 1000 ng/g) was dosed to 4–7-day-old post-hatch medaka fish for 40 days to evaluate the effects on the swimming activity of fish using a miniaturized swimming flume. Chlorpyrifos (CF)-exposed fish were selected as the positive control to assess the validity and sensitivity of the behavioral findings. After 20 and 40 days of exposure, the locomotor activity was analyzed for 6 min in a flume section (arena). The CF positive control for each time point were fish exposed to 50 ng CF/ml for 48 h. Swimming patterns, presented as two-dimensional heat maps of fish movement and positioning, were obtained by geostatistical analyses. The heat maps of the control groups at time point 20 revealed visually comparable swimming patterns to those of the BDE-47-treated groups. For the comparative fish positioning analysis, both the arenas were divided into 15 proportional areas. No statistical differences were found between residence times in the areas from the control groups and those from the BDE-47-treated groups. At time point 40, the heat map overall patterns of the control groups differed visually from that of the 100-ng BDE-47/g-treated group, but a comparative analysis of the residence times in the corresponding 15 areas did not reveal consistent differences. The relative distances traveled by the control and treated groups at time points 20 and 40 were also comparable. The heat maps of CF-treated fish at both time points showed contrasting swim patterns with respect to those of the controls. These differential patterns were statistically supported with differences in the residence times for different areas. The relative distances traveled by the CF-treated fish were also significantly shorter. These results confirm the validity of the experimental design and indicate that a dietary BDE-47 exposure does not affect forced swimming in medaka at growing stages.

This study evaluated the effect of growth of different tissue compartments on the bioaccumulation of mercury (Hg) and polychlorinated biphenyls (PCBs) in Silver Carp (Hypophthalmichthys molitrix) and Bighead Carp (Hypophthalmichthys nobilis) from the Three Gorges Reservoir (TGR), China. A non-steady state bioenergetics/toxicokinetic model was developed to simulate PCB and Hg concentrations in these two species and compared with field data. Simulations using constant whole body growth rate and constant tissue to whole body weight ratios were contrasted against simulations adopting age specific whole body and tissue/age specific growth rates for their goodness of fit to field data. The simulations using age/tissue specific growth rates demonstrated better fit to field data for PCBs compared to the constant growth rate models (22% improved R²), while both models explained similar variation in Hg concentration data. Both species demonstrated higher growth rates of lipids (on a daily basis) relative to whole body and protein contributing to higher growth dilution of PCBs compared to Hg. Although stable isotope data indicated some degree of diet and/or habitat shift, simulations assuming a constant diet concentration explained between 36 and 40% of the variation in fish concentrations for both contaminants and fish species. This study demonstrates that differences in the bioaccumulation rate of PCBs and Hg by Asian carp can be partially explained by differences in the growth rates of key tissue storage compartments associated with each contaminant. These differences in chemical-specific growth dilution subsequently contribute to differences in chemical retention and bioaccumulation patterns of Hg and PCBs by fish.

Mining in the Athabasca Oil Sands Region (AOSR) has contributed extensively to increased exposure of wildlife to naturally occurring polycyclic aromatic compounds (PACs), yet little is known about the toxicity of PACs to wildlife in this region. We identified reproductive and developmental changes in tree swallows (Tachycineta bicolor) breeding in close proximity to mining-related activities in the AOSR, and determined these changes in relation to the birds' exposure and accumulation of 41 PACs (parent-, alkylated-PAHs), dibenzothiophenes (DBTs; previously published), diet (carbon (δ13C), nitrogen (δ15N)), volatile organic compounds, and weather variables. Tree swallow pairs (N = 43) were compared among mining-related (OS1, OS2) and reference (REF1, REF2) sites. At OS2, clutch initiation was slightly advanced (2012) but reproductive success (65%) was much lower than at the other sites (≥ 79%). Fledgling production by each pair was influenced by the timing of clutch initiation (years combined); in a highly inclement brood rearing period (2013), additional influences included the nestlings' exposure to ΣDBTs, accumulation of C1-naphthalene, the trophic position of the prey in their diet (δ15N), and record-breaking rainfall. Nestlings at OS2 were significantly lighter at day (d) 9 and d14, and in poorer body condition (d9). Nestling body mass was influenced by multiple stressors that varied by site: mass of younger nestlings (d9) was related to dietary source (δ13C; e.g., wetlands, terrestrial fields), exposure and/or accumulation of C1-phenanthrenes, C2-fluorenes, Σalkyl-PAHs and ΣDBTs, while for older nestlings (d14), body mass was related to sex, hatch date and/or rainfall during brood rearing. The swallows' exposure and accumulation of parent-PACs, alkyl-PACs and DBTs, the timing of hatching, their diet and exposure to highly inclement rains, contributed to their reproductive and developmental changes.
