Relationship between thyroid hormone parameters and exposure to a mixture of organochlorine pesticides, mercury and nutrients in the cord blood of newborns

The fetus is prenatally exposed to a mixture of organochlorine pesticides (OCPs), mercury (Hg), docosahexaenoic acid (DHA), eicosapentaenoic acid (EPA) and selenium (Se) through maternal seafood consumption in real-life scenario. Prenatal exposure to these contaminants and nutrients has been suggested to affect thyroid hormone (TH) status in newborns, but the potential relationships between them are unclear and the joint effects of the mixture are seldom analyzed. The aim of the study is to investigate the associations of prenatal exposure to a mixture of OCPs, Hg, DHA, EPA and Se with TH parameters in newborns. 228 mother-infant pairs in Shanghai, China were included. We measured 20 OCPs, total Hg, DHA, EPA and Se in cord blood samples as exposure variables. The total thyroxine (TT4), free thyroxine (FT4), total triiodothyronine (TT3), free triiodothyronine (FT3), and thyroid-stimulating hormone (TSH) levels and the FT3/FT4 ratio in cord serum were determined as outcomes. Using linear regression models, generalized additive models and Bayesian kernel machine regression, we found dose-response relationships of the mixture component with outcomes: among the contaminants, p,p'-DDE was the most important positive predictor of TT3, while HCB was predominantly positively associated with FT3 and the FT3/FT4 ratio, indicating different mechanisms underlying these relationships; among the nutrients, EPA was first found to be positively related to the FT3/FT4 ratio. Additionally, we found suggestive evidence of interactions between p,p'-DDE and HCB on both TT3 and FT3, and EPA by HCB interactions for TT3, FT3 and FT3/FT4 ratio. However, the overall effects of the mixture on thyroid hormone parameters were not significant. Our result suggests that prenatal exposure to p,p’-DDE, HCB and EPA as part of a mixture might affect thyroid function of newborns in independent and interactive ways. The potential biological mechanisms merit further investigation.

Fish is an important source of nutritional omega-3 (n-3) polyunsaturated fatty acids, but it also readily accumulates toxic mercury (Hg) and microcystins (MC) in eutrophic aquatic systems. In China, farmed fish was widely consumed, and aquaculture has caused pervasive eutrophication of freshwater lakes, resulting in the increasing accumulation of MC in fish tissue. To assess the risk-benefit of consuming farmed fish, 205 fish samples of 10 primary species were collected from the eutrophic Wujiangdu (WJD) Reservoir, SW China. The contents of Hg, microcystin-RR (MC-RR), microcystin-LR (MC-LR), and polyunsaturated fatty acids (PUFA) in fish were analyzed. The results showed that THg and MeHg concentrations in all fish sampls were well below the safety limit (500 ng/g w.w) established by the Standardization Administration of China, with average values of 22.9 ± 22.8 and 6.0 ± 6.6 ng/g wet weight (w.w.), respectively. Average concentrations of MC-RR and MC-LR were 40 ± 80 and 50 ± 80 ng/g w.w., respectively. MC-RR and MC-LR concentrations in fish were significantly higher in silver carp and black carp than in perch and catfish (p < 0.05). In nutritional terms, average concentrations of n-3 PUFA and the eicosapentaenoic (EPA) + docosahexaenoic acids (DHA) of fish were 2.0 ± 2.5 and 1.4 ± 0.5 mg/g w.w., respectively. The risk-benefit assessment suggests that the n-3 PUFA benefits from consuming all farmed fish species in the WJD Reservoir outweigh the adverse effects of MeHg. However, except for perch, most fish species still pose a high MC-LR exposure risk that created a requirement for fish consumption advisories and monitoring. Consequently, more attention should be paid on the health risk of combined exposure to pollutants by aquatic product consumption.

Aquaculture can affect the polyunsaturated fatty acids (PUFA) and mercury (Hg) in fish by altering their diet. Here, planktivorous (silver carp and bighead carp), omnivorous and carnivorous fish with different dietary strategies were selected from two reservoirs, one with on-going aquaculture (WJD) and another without aquaculture (HF) in Southwest China. We compared the total mercury (THg), methylmercury (MeHg) contents and PUFA profiles of fish and their potential diets in these two reservoirs. THg and MeHg contents in omnivorous and carnivorous fish were lower from the WJD Reservoir, which is related to the lower THg and MeHg contents in the artificial fish food. THg and MeHg contents in silver carp from the WJD Reservoir were lower than those from the HF Reservoir, while they were similar in bighead carps from the two reservoirs. The Hg variation in planktivorous fish were inconsistent with that in plankton. THg contents in phyto- and zooplankton from the HF Reservoir were higher than those from the WJD Reservoir, yet their MeHg contents were similar. Artificial fish food which contained higher total PUFA eicosapentaenoic (EPA; 20:5n-3) and docosahexaenoic acid (DHA, 22:6n-3), significantly increased the total PUFA and EPA + DHA contents in carnivorous fish, but had less effect on that in omnivorous fish from the WJD Reservoir. Eutrophication caused by aquaculture reduced total PUFA and EPA + DHA contents of plankton in WJD, yet did not reduce those in planktivorous fish. The impacts of aquaculture on Hg and PUFA accumulated in fish were varied among different fish species, and the mechanism needs further exploration.

Maternal 1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (DDE) serum levels during pregnancy have been negatively linked to child neurodevelopment in contrast to intake of omega-3 and -6 (ω-3 and ω-6) fatty acids.To assess whether maternal dietary intake of ω-3 and ω-6 during pregnancy modifies the association between exposure to DDE and child neurodevelopment from age 42–60 months.Prospective cohort study with 142 mother–child pairs performed in Mexico. DDE serum levels were determined by electron capture gas chromatography. Dietary ω-3 and ω-6 intake was estimated by questionnaire. Child neurodevelopment was assessed by McCarthy Scales.Docosahexaenoic (DHA) fatty acid intake significantly modified the association between DDE and motor component: increased maternal DDE was associated with lower motor development in children whose mothers had lower DHA intake (βlog2DDE = −1.25; 95% CI: −2.62, 0.12), in contrast to the non-significant increase among children whose mothers had higher DHA intake (βlog2DDE-motor = 0.50; 95% CI: 0.55, 1.56). Likewise, arachidonic fatty acid (ARA) intake modified the association between DDE and memory component: increased maternal DDE was associated with a significantly larger reduction in the memory component in children whose mothers had lower ARA intake (βlog2DDE = −1.31; 95% CI: −2.29, −0.32) than children whose mothers had higher ARA intake (βlog2DDE-memory = 0.17; 95% CI: −0.78, 1.11).Dietary intake of DHA and ARA during pregnancy may protect against child neurodevelopment damage associated with prenatal maternal DDE levels.

This study evaluated the spatial variability of risks and benefits of consuming fish from humic and clear lakes. Mercury in fish is a potential risk for human health, but risk assessment may be confounded by selenium, which has been suggested to counterbalance mercury toxicity. In addition to the risks, fish are also rich in eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), which are known to be beneficial for cardiovascular health and brain cognitive function in humans.We found that the concentrations of EPA + DHA and mercury in European perch (Perca fluviatilis) vary spatially and are connected with lake water chemistry and catchment characteristics. The highest mercury concentrations and the lowest EPA + DHA concentrations were found in perch from humic lakes with high proportion of peatland (30–50%) in the catchment. In addition, the ratio of selenium to mercury in perch muscle was ≥1 suggesting that selenium may counterbalance mercury toxicity.The observed variation in mercury and EPA + DHA content in perch from different lakes indicate that the risks and benefits of fish consumption vary spatially, and are connected with lake water chemistry and catchment characteristics. In general, consumption of perch from humic lakes exposed humans to greater risks (higher concentrations of mercury), but provided less benefits (lower concentrations of EPA + DHA) than consumption of perch from clear lakes.

Mercury, omega-3 (docosahexaenoic acid, DHA and eicosapentaenoic acid, EPA) and macronutrients (fat and proteins) were quantified on a wet weight (ww) basis in 20 species of fish taken as bycatch in tropical tuna fisheries. Based on a hazard quotient taking into account mercury and omega-3 contents, a benefit-risk assessment for the consumption of these pelagic species was conducted for three people categories: young children, children and adults. All fish bycatch were found to be an excellent source of proteins (min‒max = 14.4–25.2 g/100 g fillet), had low omega-6/omega-3 ratios (<1, except for silky shark), and had mercury content below the safety limits defined by sanitary agencies. Silky shark and Istiophoridae had the highest mercury contents (min‒max = 0.029–0.317 ppm ww). Omega-3 contents were the lowest in silky shark (0.2 ± 0.2 mg/100 g fillet) and the highest in striped marlin (3.6 ± 3.2 g/100 g fillet). Billfishes (Istiophoridae, including striped marlin), minor tunas (Scombridae), and Carangidae had the highest omega-3 contents (min‒max = 0.68–7.28 g/100 g fillet). The highest hazard quotient values obtained for silky shark and great barracuda reflected a lower nutritional benefit (i.e., low omega-3 source) than risk (i.e., mercury exposure), making them not advisable for consumption. Eight species had low hazard quotients, and among them cottonmouth jack and flat needlefish were found of high health interest (high protein, moderate fat contents, and low omega-6/omega-3 ratio). A daily serving portion of 85–200 g (according to people category) can be recommended for these species. Batfish, and to a lower extent pompano dolphinfish and brassy chub, can also be consumed safely and would provide greater health benefits than risks. These results advocate for a better access of these species to local populations.

Animal studies have suggested that phthalate exposure alters the fatty acid composition of blood plasma. Therefore, we conducted an epidemiological study to examine whether urinary concentrations of phthalates are correlated with circulating fatty acids in the general US population. The 2003–2004 and 2011–2012 National Health and Nutrition Examination Survey were used in this study. Ten urinary phthalate metabolites and 23 fatty acids were measured. Fatty acid patterns were identified using principal component analysis (PCA) with an eigenvalue greater than 1. A two-step analysis was performed. We first performed multivariable linear regressions to evaluate whether urinary phthalate metabolites were related to the PCA-derived components of blood fatty acid levels. Then we performed multivariable linear regressions to investigate each of the fatty acids that were suggestively correlated with some of the phthalates in PCA. There were 994 participants (51.91% women). As for men, after adjustments for potential confounding factors, MECPP, MEHHP, and ∑DEHP were all positively correlated with gamma-linolenic, myristoleic, and myristic acids; both MEHHP and ∑DEHP were positively correlated with stearic acid; MMP was positively correlated with docosahexaenoic acid. As for women, MMP was negatively correlated with docosanoic, lignoceric, and arachidic acids; MBzP was negatively correlated with docosahexaenoic acid; both MEHP and MCPP were negatively correlated with docosatetraenoic acid; MEHP was negatively correlated with arachidonic acid, and MCPP was negatively correlated with docosapentaenoic-6 acid. Our findings support that phthalates may be correlated with circulating fatty acids.

Mercury (Hg) contamination testing was conducted on winter-caught male spiny dogfish (Squalus acanthias) in southern New England and results compared to available data on Hg concentrations for this species. A limited risk-reward assessment for EPA (eicosapentanoic acid) and DHA (docosahexanoic acid) lipid concentrations of spiny dogfish was completed in comparison with other commonly consumed marine fish. Mean Hg concentrations were 0.19ppm (±0.30) wet weight. In comparison, mean Hg concentrations in S. acanthias varied geographically ranging from 0.05ppm (Celtic Sea) to 2.07ppm (Crete, Mediterranean Sea). A risk-reward assessment for Hg and DHA+EPA placed S. acanthias in both “low-risk, high-reward” and “high-risk, high-reward” categories for consumption dependent on locations of the catch. Our results are limited and are not intended as consumption advisories but serve to illustrate the need for making more nuanced, geo-specific, consumption guidance for spiny dogfish that is inclusive of seafood traceability and nutritional benefits.

The current study was conducted to assess the beneficial effect of selenium (Se) on maneb-induced cardiotoxicity and fatty acid alterations in adult mice. Swiss albino male mice were assigned into four experimental groups. The first group consisted of negative controls. The second group represented the positive controls where mice received daily, via the diet, sodium selenite at a dose of 0.2 mg/kg. For the third group, mice were subjected to intraperitoneal injections of maneb (30 mg/kg BW). The fourth group (MB+Se) received daily the same dose of maneb as group 3 along with sodium selenite at the same dose as group 2. Mice exposure to maneb caused cardiotoxicity as indicated by an increase in malondialdehyde, hydrogen peroxide, and protein carbonyl levels, and an alteration of the antioxidant defense system (catalase, glutathione peroxidase, superoxide dismutase, glutathione, and vitamin C). Plasma lactate dehydrogenase activity and total cholesterol, triglyceride, and low-density lipoprotein cholesterol levels increased, while high-density lipoprotein cholesterol level decreased. Results showed also a decrease in the amount of n-3 PUFA, docosahexaenoic, docosapentaenoic, and eicosapentaenoic acids. However, an increase in the levels of MUFA, cis-vaccenic, and palmitoleic acids was observed. Co-administration of Se restored the parameters indicated above to near control values. The histopathological findings confirmed the biochemical results. Selenium could be a useful and efficient agent against maneb-induced cardiotoxicity.

Aquatic products contain both beneficial substances such as polyunsaturated fatty acids and harmful compounds such as methylmercury, and their combined health effects should be assessed at a common end point for a rational analysis. We used a risk–benefit model to assess net IQ point gain from consumption of Chinese mitten crabs (Eriocheir sinensis) from Jiangsu Province. Levels of methylmercury in the edible parts of the crabs ranged from 0.79 to 69.47 μg/kg (mean: 26.88 ± 15.09 μg/kg). Levels of methylmercury did not differ by sampling location. In aquaculture experiments, we simulated artificial pond farming using either iced trash fish or formulated feed. Crabs fed iced trash fish contained 28.72 ± 2.30 mg/g of eicosapentaenoic acid and docosahexaenoic acid and crabs fed formulated feed contained 19.64 ± 3.62 mg/g of eicosapentaenoic acid and docosahexaenoic acid. Model calculations estimated a gain of 7.52 IQ points from consumption of crabs fed formulated feed and 11.00 IQ points from consumption of crabs fed iced trash fish. They are all above the maximum theoretical IQ point gain value. Chinese mitten crab is a kind of high-quality food which is beneficial to the neural development of infants. Our findings may assist in improving crab farming methods and consumption patterns, accelerating the sustainable development of the Chinese mitten crab industry.
