Effect of long-term equilibration on the toxicity of molybdenum to soil organisms

To determine if long-term equilibration may alleviate molybdenum toxicity, earthworms, enchytraeids, collembolans and four plant species were exposed to three soils freshly spiked with Na₂MoO₄.2H₂O and equilibrated for 6 or 11 months in the field with free drainage. Total Mo concentrations in soil decreased by leaching, most (up to 98%) in sandy soil and less (54–62%) in silty and clayey soils. Changes in residual Mo toxicity with time were inconclusive in sandy soil. In the other two soils, toxicity of residual total Mo was significantly reduced after 11 months equilibration with a median 5.5-fold increase in ED50s. Mo fixation in soil, i.e. the decrease of soil solution Mo concentrations at equivalent residual total soil Mo, was maximally a factor of 2.1 only. This experiment shows natural attenuation of molybdate ecotoxicity under field conditions is related to leaching of excess Mo and other ions as well as to slow ageing reactions.

Predicting the transfer of contaminants in soils is often hampered by lacking validation of mathematical models. Here, we applied Hydrus-2D software to three agricultural soils for simulating the 1900–2005 changes of zinc and lead concentration profiles derived from industrial atmospheric deposition, to validate the tested models with plausible assumptions on past metal inputs to reach the 2005 situation. The models were set with data from previous studies on the geochemical background, estimated temporal metal deposition, and the 2005 metal distributions. Different hypotheses of chemical reactions of metals with the soil solution were examined: 100% equilibrium or partial equilibrium, parameterized following kinetic chemical extractions. Finally, a two-site model with kinetic constant values adjusted at 1% of EDTA extraction parameters satisfactory predicted changes in metal concentration profiles for two arable soils. For a grassland soil however, this model showed limited applicability by ignoring the role of earthworm activity in metal incorporation.

This study aims to evaluate toxic effects of exposure to chlortetracycline (CTC) in soil on reproductive endpoints (juvenile counts and cocoon counts), biochemical responses, and genotoxic potentials of the earthworm Eisenia fetida. Results showed that juvenile counts and cocoon counts of the tested earthworms were reduced after exposure to CTC. The effective concentrations (EC₅₀ values) for juvenile and cocoon counts were 96.1 and 120.3 mg/kg, respectively. Treatment of earthworms with CTC significantly changed the activity of catalase (CAT), superoxide dismutase (SOD) and glutathione S-transferase (GST). An increase in malondialdehyde (MDA) indicated that CTC could cause cellular lipid peroxidation in the tested earthworms. The percentage of DNA in the tail of single-cell gel electrophoresis of coelomocytes as an indication of DNA damage increased after treatment with different doses of CTC, and a dose-dependent DNA damage of coelomocytes was found. In conclusion, CTC induces physiological responses and genotoxicity on earthworms.

We assessed dietary exposure of the little owl Athene noctua to trace metal contamination in a Dutch Rhine River floodplain area. Diet composition was calculated per month for three habitat types, based on the population densities of six prey types (earthworms, ground beetles and four small mammal species) combined with the little owl’s functional response to these prey types. Exposure levels showed a strong positive relationship with the dietary fraction of earthworms, but also depended on the dietary fraction of common voles, with higher common vole fractions resulting in decreasing exposure levels. Spatio-temporal changes in the availability of earthworms and common voles in particular resulted in considerable variation in exposure, with peaks in exposure exceeding a tentative toxicity threshold. These findings imply that wildlife exposure assessments based on a predefined, average diet composition may considerably underestimate local or intermittent peaks in exposure.

Short-term changes in levels of expression of nine stress response genes and oxidative damage of proteins were examined in Eisenia fetida exposed to polyvinylpyrrolidone (PVP) coated Ag nanoparticles (Ag-NP) and AgNO₃ in natural soils. The responses varied significantly among days with the highest number of significant changes occurring on day three. Similarity in gene expression patterns between Ag-NPs and AgNO₃ and significant relationships of expression of CAT and HSP70 with Ag soil concentration suggest similarity in toxicity mechanisms of Ag ions and NPs. Significant increases in the levels of protein carbonyls on day three of the exposure to both ions and Ag-NPs indicate that both treatments induced oxidative stress. Our results suggest that Ag ions drive short term toxicity of Ag-NPs in E. fetida. However, given that <15% of Ag in the NPs was oxidized in these soils, dissolution of Ag-NPs is likely to occur after or during their uptake.

The close relationship between soil organic matter and the bioavailability of POPs in soils suggests the possibility of using it for the extrapolation between different soils. The aim of this study was to prove that TOC content is not a single factor affecting the bioavailability of POPs and that TOC based extrapolation might be incorrect, especially when comparing natural and artificial soils. Three natural soils with increasing TOC and three artificial soils with TOC comparable to these natural soils were spiked with phenanthrene, pyrene, lindane, p,p′-DDT, and PCB 153 and studied after 0, 14, 28, and 56 days. At each sampling point, total soil concentration and bioaccumulation in earthworms Eisenia fetida were measured. The results showed different behavior and bioavailability of POPs in natural and artificial soils and apparent effects of aging on these differences. Hence, direct TOC based extrapolation between various soils seems to be limited.

Lead forms stable compounds with phosphate and the immobilized Pb becomes less available to soil biota. In this study, we tested the bioavailabilty of Pb using earthworms (Eisenia fetida) and plants after immobilization of Pb by a soluble P compound and an insoluble rock phosphate compound in the presence of phosphate-solubilizing bacteria (Enterobacter sp.). Rock phosphate in the presence of phosphate-solubilizing bacteria and a soluble P compound enhanced Pb immobilization as measured by NH4NO3-extractable Pb concentration, thereby reduced its bioavailability as evaluated by earthworm Pb loading and sunflower (Helianthus annuus) Pb uptake under greenhouse conditions. However, soluble P treatment increased the concentration of Pb in soil solution thereby inhibited the root elongation of mustard (Brassica hirta) seedlings. Sunflower plants in the Pb-spiked soil without P amendments showed symptoms of necrosis and stunting because of Pb toxicity. Both soluble and insoluble P treatments significantly increased shoot and root weight and decreased Pb concentration in shoot by more than 50% compared to the control. However, high Pb concentration in soil solution was found in soluble P treatment, which can be attributed to dissolved organic carbon–Pb complex formation, thereby increasing Pb mobility. The inoculation of phosphate-solubilizing bacteria can facilitate phytostabilization of Pb-contaminated site.

In order to enhance the removal of heavy metals such as Ni, Cu, Zn and Cd from wastewater, different cow dung/sewage sludge ratios were tested to assess the effect of these metals on the adaptability of Eisenia fetida earthworms to the treatment process carried out in a typical plant located in Tamaulipas, Mexico. Two experimental water treatment setups were proposed. The first set of experiments was planned to determine the adequate sewage sludge/cow dung ratio(s), whereas the second arrangement was designed to evaluate the growth performance and fecundity of the earthworms under high heavy metal concentrations. To achieve the objectives, the experiments were conducted for 90 days under controlled environmental conditions. Maximum worm biomass and growth rates were attained in samples containing 25 wt.% of sewage sludge. Weight and mortality of worms were significantly affected by the high levels of heavy metals, making difficult the metal accumulation in the worm tissues.
