Household prallethrin-based insecticide toxicity on different C. elegans life stage: A possible sign of Huntington Disease

Household insecticide is largely used for insect and ectoparasite control, in city centers as well as in the countryside. The pyrethroids are the most used class of insecticide, these compounds in low doses have low toxicity for mammalians, in comparison to other compounds, with insecticide effects. The contact of these compounds in sublethal doses begins in early life and many cases, in intrauterine life. Considerable diseases still with undefined etiology, such as neurodegenerative conditions, and Huntington's Disease (HD) is one of them. HD is related to overexpression of Polyglutamine (PolyQ40), its aggregation, and non-solubilization, which leads to neural, behavioral, and cognitive damage. In our study, we evaluate the effect of two sublethal doses of a prallethrin-based insecticide (P-BI), in three different Caenorhabditis elegans life stages transgenerational, neonatal, and lifespan. We evaluated the Body bends and pharyngeal pumping rate, and social feeding as behavioral biomarkers. As well as acetylcholinesterase activity (AChE), PolyQ40 aggregation, antioxidant enzymes, and heat shock protein (HSP) expression. We observe that the toxic effect of P-BI is more pronounced on transgenerational and lifespan exposure. Both sublethal doses of P-BI decreased the AChE activity and retard the HSP expression as well as increased the PolyQ40 aggregates indicating a clear biomarker for possible effect in the progression of the HD, by the environmental contamination.

Atlantic coastal bays of the US are essential habitat for young of year bluefish (Pomatomus saltatrix). Their residence in these estuaries during critical life stages, high lipid content, and piscivory make bluefish an ideal bioindicator species for evaluating estuarine health. Individual whole fish from four estuaries impacted by Hurricane Sandy were collected in August 2013, analyzed for a suite of persistent organic pollutants (POPs) including polychlorinated biphenyls, polybrominated diphenyl ethers and organochlorine pesticides and evaluated using health metrics. Concentrations in whole bluefish differed by estuary; however, concentrations for many POPs decreased or were similar to those observed prior to the hurricane. Prevalence of the ectoparasitic gill isopod (Lironeca ovalis) varied by estuary and no relationships between contaminants and lesions were observed. Bluefish should be considered for monitoring programs and, if sampled frequently, could be an effective bioindicator of incremental and episodic changes in contaminants within aquatic food webs.

We examined parasite populations and communities in the Mediterranean sentinel fish species, Mullus barbatus, sampled at a small-scale PCB gradient at the shelf sediments off Catalonian coasts of the Western Mediterranean. A recurrent feature at both the population and community level was the differentiation of the samples along the increasing PCB levels simultaneously registered in the sediments. Both directly transmitted ectoparasites and endoparasites with complex life-cycles transmitted via food chains exhibited a decrease in abundance with the increase in PCB levels. Parasite numerical responses translated into significant differences in infracommunity structure with decreasing predictability associated with increasing PCB levels. The abundance of two species, the specialist Opecoeloides furcatus and the generalist nematode Hysterothylacium fabri, contributed substantially to the observed dissimilarity between infracommunity samples along the gradient. The observed parasite responses to moderate levels of pollution were simultaneously validated by both chemical monitoring and effect biomarkers.

Transaminase profile and hepatic histopathological traits in Piaractus mesopotamicus exposed to insecticide Diflubenzuron

Diflubenzuron (DFB) is a widely used insecticide to control ectoparasites in fish farming. Although therapeutic concentrations (i.e., 50 to 100 mg/L) are safe as they fail to induce mortality, they can promote tissue changes. In Brazil, Pacu (Piaractus mesopotamicus) is a native species used for commercial production, and it remains crucial to determine underlying mechanisms to mitigate the potential effects of pathogens on productivity. The aim of this study was to analyze the transaminase profile and histopathological changes in the liver of P. mesopotamicus exposed to a DFB bath. Hence, the fish were exposed to an immersion bath containing a 70 mg/L nominal concentration of Difluchem 240 SC® (24% (m/m) DFB) for 30 (n = 10), 60 (n = 10), and 120 min (n = 10), every 24 h for 3 days. Following exposure, plasma transaminases and liver histology were analyzed. In DFB-exposed fish, levels of aspartate transaminase (AST) and alanine transaminase (ALT) were elevated when compared with the control at 30 and 60 min. Furthermore, liver morphology was altered based on exposure times. Compared with controls, the degree of reversible damage (degree of tissue change (DTC)) demonstrated high scores for all exposure times, with no difference between individual groups. Irreversible changes were increased in the 60 and 120-min baths. These findings highlight the impact of the therapeutic DFB concentration (i.e., 70 mg/L), revealing that 60-min and 120-min bathing induces irreversible and progressive hepatic changes.

Despite many efforts for pollution abatement in aquatic ecosystems, there are still some cases of high accumulation of industrial pollutants due to past activities. In Flix reservoir (Ebro River, Spain), there are around 200,000-360,000 tons of industrial pollutants with a high concentration of heavy metals and organochlorides due to the activity of an organochlorine industry during more than half a century. This exceptional amount of pollutants provides a good opportunity (and need) to analyse their effects on fish populations under natural conditions, which is rarely available to ecotoxicologists. We compared the reproductive traits and prevalence of diseases and parasites at this impacted area with a neighbouring upstream reservoir unaffected by the pollution (reference sites) and also to downstream sites. Deformity, eroded fin, lesion and tumour (DELT) anomalies and ectoparasites were clearly more frequent at the impacted area for several fish species (common carp, roach and pumpkinseed). A significant negative impact of Flix reservoir on condition (eviscerated and liver weights, adjusted for fish size with analysis of covariance) and reproductive traits (gonadal weight and number of mature eggs, adjusted for fish size) was also detected for several fish species. The responses to the pollutants were species-specific, and common carp (Cyprinus carpio) was the species with the clearest effects on fitness-related traits at the impacted area, despite also being among the most tolerant to pollution.

Transgenic Bt insect-resistant plants are highly resistant to Lepidoptera stockpile pest Indian meal moth, Plodia interpunctella Hübner (Lepidoptera: Pyralidae), a storage pest. Habrobracon hebetor (Say) (Hymenoptera: Braconidae), which is an ectoparasitic wasp of Indian meal moth, may be exposed to the Bt protein through the food chain. In the current study, high dose of Cry1C protein was injected into the hemolymph of P. interpunctella by microinjection, and the hemolymph was used as the carrier to deliver Bt protein to the H. hebetor. Using this method, we developed a new Tier-1 risk assessment system for ectoparasitoid, successfully avoided “host/prey quality-mediated effect,” and improve the accuracy of safety evaluation. Results showed that injected Cry1C was stable and bioactive in the hemolymph of P. interpunctella parasitized by H. hebetor, and high dose of Cry1C has no negative impacts on egg hatching rate, developmental duration from egg to adult, survival egg to adult, pupa weight, adults weight (male and female), adult longevity and reproduction, and activity of stress-related enzymes of H. hebetor. However, the hemolymph of P. interpunctella injected into Galanthus nivalis L. agglutinin (the positive control) had significant negative impact on these biological parameters of H. hebetor. The results indicate that H. hebetor are not sensitive to Cry1C protein at the tested concentration and there were no detrimental effects of Cry1C protein on any biological parameters tested in the present study. More importantly, we constructed a new efficient and simple system for the biosafety assessment on the larvae of ectoparasitoid of target pest.

The aquatic environment represents the final repository for many human-generated pollutants associated with anthropogenic activities. The quality of natural freshwater systems is easily disrupted by the introduction of pollutants from urban, industrial and agricultural processes. To assess the extent of chemical perturbation and associated environmental degradation, physico-chemical parameters have been monitored in conjunction with biota in numerous biological monitoring protocols. Most studies incorporating organisms into such approaches have focussed on fish and macroinvertebrates. More recently, interest in the ecology of parasites in relation to environmental monitoring has indicated that these organisms are sensitive towards the quality of the macroenvironment. Variable responses towards exposure to pollution have been identified at the population and component community level of a number of parasites. Furthermore, such responses have been found to differ with the type of pollutant and the lifestyle of the parasite. Generally, endoparasite infection levels have been shown to become elevated in relation to poorer water quality conditions, while ectoparasites are more sensitive, and exposure to contaminated environments resulted in a decline in ectoparasite infections. Furthermore, endoparasites have been found to be suitable accumulation indicators for monitoring levels of several trace elements and metals in the environment. The ability of these organisms to accumulate metals has further been observed to be of benefit to the host, resulting in decreased somatic metal levels in infected hosts. These trends have similarly been found for host–parasite models in African freshwater environments, but such analyses are comparatively sparse compared to other countries. Recently, studies on diplozoids from two freshwater systems have indicated that exposure to poorer water quality resulted in decreased infections. In the Vaal River, the poor water quality resulted in the extinction of the parasite from a site below the Vaal River Barrage. Laboratory exposures have further indicated that oncomiracidia of Paradiplozoon ichthyoxanthon are sensitive to exposure to dissolved aluminium. Overall, parasites from African freshwater and marine ecosystems have merit as effect and accumulation indicators; however, more research is required to detail the effects of exposure on sensitive biological processes within these organisms.
