Waterborne zinc bioaccumulation influences glucose metabolism in orange-spotted grouper embryos

Fish embryos, as an endogenous system, strictly regulate an energy metabolism that is particularly sensitive to environmental pressure. This study used orange-spotted grouper embryos and stable isotope ⁶⁷Zn to test the hypothesis that waterborne Zn exposure had a significant effect on energy metabolism in embryos. The fish embryos were exposed to a gradient level of waterborne ⁶⁷Zn, and then sampled to quantify ⁶⁷Zn bioaccumulation and mRNA expressions of key genes involved glucose metabolism. The results indicated that the bioaccumulated ⁶⁷Zn generally increased with increasing waterborne ⁶⁷Zn concentrations, while it tended to be saturated at waterborne ⁶⁷Zn > 0.7 mg L⁻¹. As we hypothesized, the expression of PK and PFK gene involved glycolysis pathway was significantly up-regulated under waterborne ⁶⁷Zn exposure >4 mg L⁻¹. Waterborne ⁶⁷Zn exposure >2 mg L⁻¹ significantly suppressed PCK and G6PC gene expression involved gluconeogenesis pathway, and also inhibited the AKT2, GSK-3beta and GLUT4 genes involved Akt signaling pathway. Our findings first characterized developmental stage-dependent Zn uptake and genotoxicity in fish embryos. We suggest fish embryos, as a small-scale modeling biosystem, have a large potential and wide applicability for determining cytotoxicity/genotoxicity of waterborne metal in aquatic ecosystem.

The present study aimed to use primary liver cell culture derived from the orange-spotted grouper, Epinephelus coioides, to assess the toxic effects of nonylphenol (NP) on the hepatocyte viability and the liver antioxidant system. E. coioides was selected due to its commercial importance. NP was used in this study because of its high potential of producing oxidative stress due to increased reactive oxygen species (ROS). A liver of E. coioides was digested with PBS containing 0.1% collagenase IV. The digested cells were moved to Leibovitz L-15 culture medium with 20% fetal bovine serum (FBS), 100IUmL−1 penicillin, 100μgmL−1 streptomycin. Aliquots of cell suspension were seeded as a monolayer into sterile 25cm2 tissue culture flasks and incubated at 30°C for 14days. The medium, containing non-attached cells, was removed after 24 to 48h and a new medium was added. The IC50 of 10−4molL−1 was determined for nonylphenol using MTT assay. Cells were then incubated with L-15 medium containing 10−5, 2×10−5, 3×10−5molL−1 of NP and samples were taken after 6, 12 and 24h of incubation for analysis of LPO, SOD, CAT, GPx, LDH, AST, ALT, and ALP. Based on the results, the lowest concentration of NP was not markedly cytotoxic to primary hepatocytes and the cell sensitivity to NP increased dose-dependently. The activities of SOD, CAT and GPx decreased significantly, while activities of LPO, LDH, AST, ALT and ALP, increased significantly in a dose-related pattern in NP-treated cells. In conclusion, this study revealed that NP could induce the oxidative stress in cultivated hepatocytes of E. coioides during a short-term exposure. NP toxicity is mainly due to the induction of the reactive oxygen species (ROS), which lead to cell membrane disruption, damage of cellular metabolism, and interference with cellular macromolecules.

The objective of this study was to assess the environmental conditions of a heavily polluted marine habitat using descriptors of fish parasites. Epinephelus coioides from Jakarta Bay as well as off Jakarta Bay was studied for metazoan parasites. Based on 70 fish and considering previous studies (230 fish), an environmental indicator system was designed. Including the recent study, a total of 51 parasite species have been recorded for E. coioides in Indonesian waters. Seven of them combined with five parasitological indices are useful descriptors for the environmental status of marine ecosystems. The results are visualized in a star graph. A significant different parasite infection between nine analyzed localities demonstrates the negative influence of the megacity Jakarta onto the coastal environment. We herewith complete a parasite based indicator system for Indonesian coastal waters, and suggest that it can be used in other marine habitats as well as for further epinephelids.

The major aim of this study was to investigate heavy metal content of edible fish in the PRD. Eleven species of fish (consisting of 711 individuals) [catfish (Clarias fuscus), tilapia (Oreochromis mossambicus), grass carp (Ctenopharyngodon idellus), bighead carp (Aristichthys nobilis), mandarin fish (Siniperca kneri), snakehead (Channa asiatiea), black bass (Micropterus salmoides), mangrove snapper (Lutjanus griseus), star snapper (Lutjanu stellatus), snubnose pompano (Trachinotus blochii) and orange-spotted grouper (Epinephelus coioides)] were collected for the analyses of heavy metals. Overall concentrations (mg/kg, ww) in the fish muscles were: As (0.03–1.53), Pb (0.03–8.62), Cd (0.02–0.06), Ni (0.44–9.75), Zn (15.7–29.5), Cr (0.22–0.65), Cu (0.79–2.26), Mn (0.82–6.91). Significant level of Pb were found in tilapia at all locations. It is recommended that heavy metal concentrations in different fish species must be determined on a regular basis in the future so as to reduce human health risks from acute and chronic food intoxication.

Understanding the patterns of genetic diversity of fish species is essential for marine conservation and management. This is particularly important in the Arabian Gulf where marine life is subject to extreme environmental conditions that could impact genetic diversity. Here we assess genetic diversity of the most commercially important fish in the United Arab Emirates; groupers (Epinephelus spp.). Sequencing of 973bp mitochondrial DNA from 140 tissue samples collected in four main fish markets revealed 58 haplotypes clustered within three groups. Data analysis revealed the presence of three distinct Epinephelus species being marketed as one species (hammour): Epinephelus coioides, Epinephelus areolatus and Epinephelus bleekeri. We report species-specific genetic markers and demonstrate that all three species exhibit relatively low levels of genetic variation, reflecting the effect of overfishing and environmental pressures. In light of the genetic evidence presented here, conservation and management of groupers in the UAE warrant the implementation of species-specific measures.

The occurrence of endocrine disrupting chemicals (EDCs) has been determined in two widely consumed fish species from Persian Gulf i.e., Epinephelus coioides and Platycephalus indicus by applying a validated analytical for the simultaneous detection of fourteen EDCs. The concentrations of all detected EDCs were greater in the liver than in the muscle (except for bisphenol A in P. indicus), suggesting a prolonged exposure of the fishes to these pollutants in the Persian Gulf. Specifically, the results showed that di (2-ethylhexyl) phthalate (DEHP) was the compound detected most frequently and at the highest concentration in both species. DEHP levels in ranged from 6.68 to 297.48 μg g-dw⁻¹ and from 13.32 to 350.52 μg g-dw⁻¹, in muscle and in liver, respectively. A risk assessment study was conducted, and demonstrated that consuming two fish based- meals per week may result in a moderate risk especially for vulnerable population groups.

Our previous study in 2011 reported the detection of BPA and PFAAs in 20 species of marine and freshwater fishes. With an emerging evidence to suggest the metabolic-disrupting effects of BPA/PFAAs in animals, the present study was aimed to provide a time-trend analysis to determine the current concentrations of PFAAs and BPA in 20 commercially available Hong Kong species of fishes. Since the manufacture and use of BPA is being prohibited in most nations, the introduction of BPA alternatives has recently been incorporated in the markets. Therefore, the concentrations of BPB, BPF and BPS were determined. In the present study, all freshwater and seawater fish samples showed quantified concentrations [>Limit of Quantification (LOQ<0.5ng/g)] of BPA. BPF was detected in some marine (yellow seafin, bigeye, goldspotted rabbitfish, snubnose pompano, tongue sole, Bleeker's grouper and orange-spotted grouper) and freshwater fishes (mud carp, crucian carp, tilapia, catfish, mandarin fish, grass carp, grey mullet and spotted snakehead). Two of the compounds, BPS and BPB could only be identified in the marine fishes (snubnose pompano, yellow seafin). In PFAA analysis, PFOA, PFDA, PFOS, PFUdA and PFDoA were found in most of the marine and freshwater fishes. PFOS and PFOA were shown to be the two predominant PFAAs in fishes. On the basis of the measured concentrations of bisphenols, BPs (BPA, BPB, BPF, BPS) and PFAAs, the average daily intake for BPs (20.5–31.5ng/kgb.w./day) and PFAAs (1.17–1.83ng/kgb.w./day) were calculated and found to be lower than values of tolerable daily intake (TDI) established in Europe. However, as compared with our previous study in 2011, the present study revealed an approximate 10-fold increase in the concentrations of BPA in the fish samples. Although the hazard ratio of consuming fishes for BPA and PFAA exposure is expected to remain low, possible additive metabolic-disrupting effect of BPA and its analogues as well PFAAs should be taken into consideration for human health risk assessment.

The status of Kuwait’s fisheries landings and relative abundance for major species was reviewed using research data from Kuwait Institute for Scientific Research and landing data from the Kuwait’s Central Statistical Bureau. Landing data showed significant decreases for major commercial species such as zobaidy (Pampus argenteus), suboor (Tenualosa ilisha), hamoor (Epinephelus coioides), newaiby (Otolithes ruber) and hamra (Lutjanus malabaricus) while abundance data for the shrimp Penaeus semisulcatus showed significant reduction in the recent years mainly because of overfishing. The catch-rate data showed continuous decline for major species such as zobaidy, newaiby and hamoor, which indicate that stock abundances of these species are low. The reduction in stock abundance in context with changes in habitat quality, particularly the effects of reduced discharge of the Shatt Al-Arab, is discussed.

The aim of the study is to evaluate the impact of mariculture on phthalate esters speciation and distribution in sediments and cultured fish in the Hong Kong regions and near mainland China. Concentrations of ∑phthalate esters in mariculture surface sediments (0 to 5 cm) ranged from 0.20 to 54.3 mg/kg dw (mean 10.3 mg/kg dw), with the highest recorded at M2 (20.4 mg/kg dw). Concentrations of phthalate esters were not significantly (p > 0.05) enriched in surface and sediment cores at mariculture sites relative to the reference sediments, 1 to 2 km away in areas without mariculture activities. Among different congeners, only butyl benzyl phthalate (BBP) concentrations demonstrated a significant correlation (R² = 0.40, p < 0.05) with TOC values of sediments. The median concentrations of di-2-ethylhexyl phthalate (DEHP) and di-n-butyl phthalate (DBP) in the sediments were 1.57 and 6.96 times higher than the environmental risk levels (ERL), which may pose environmental risks. Results of health risk assessments revealed that the cultured fish (snubnose pompano, orange-spotted grouper, and red snapper) were safe for consumption, in terms of phthalate esters. This is the first study to assess the differences of phthalate esters contamination between mariculture and natural coastal sediments.
