Comparative responses of cell growth and related extracellular polymeric substances in Tetraselmis sp. to nonylphenol, bisphenol A and 17α-ethinylestradiol

Estuarine ecosystems near mega-cities are sinks of anthropogenic endocrine disrupting chemicals (EDCs). As the most important primary producer, indigenous microalgae and their secreted extracellular polymeric substances (EPSs) might interact with EDCs and contribute to their fate and risk. Tetraselmis sp. is a representative model of estuarine microalga, for which EDC toxicity and its effects on EPS synthesis have rarely been studied. Through microalgal isolation, algal cell growth tests, EDC removal and the characterization of related EPS profiles, the present work intends to clarify the comparative responses of Tetraselmis sp. to nonylphenol (NP), bisphenol A (BPA) and 17α-ethinylestradiol (EE₂). The results showed that the half inhibitory concentration on cell growth was 0.190–0.313 mg/dm³ for NP, which was one order of magnitude lower than the comparable values for BPA and EE₂ at 2.072–3.254 mg/dm³. Regarding chlorophyll, NP induced its degradation, EE₂ led to its decreased production, and BPA had no obvious effect. Under EDC stress, only the concentrations of colloidal polysaccharides and proteins responded dose-dependently to EE₂. Except for the colloidal fraction in the EE₂ treatment group, the increase in neutral monosaccharides, especially glucose and galactose, was a common response to EDCs. Compared to the recalcitrant BPA, NP underwent abiotic degradation in alga-free water, and EE₂ could be biodegraded in water containing this microalga. The chemical-specific responses of cell growth, chlorophyll and related EPS profiles were driven by the different fates of EDCs, and the underlying mechanism was further discussed. The results obtained in the present work are of critical importance for understanding the fate and effects of different EDCs mediated by microalgae and their related EPSs.

Though the interaction between humic acid (HA) and heavy metals has been widely reported, the effects of HA on the toxicity of heavy metals to plants are still in debate. In this study, the regulation mechanisms of HA on Pb stress in tea plant (Camellia sinensis L.) was investigated through hydroponic experiments, and the experimental results were explained by using transmission electron microscope (TEM), scanning transmission X-ray microscopes (STXM) and isobaric tags for relative and absolute quantitation (iTRAQ) differential proteomics. Significant alleviation of Pb stress was found with HA coexistence. TEM results showed that HA greatly mitigated the damage of cells caused by Pb stress. Compared with sole Pb treatment, the addition of HA increased the contents of pectin and pectic acid in the cell wall by 10.5% and 30.5%, while arabinose (Ara) and galactose (Gal) decreased by 20.5% and 15.9%, respectively, which were beneficial for increasing Pb adsorption capacity of the cell wall and promoting cell elongation. Moreover, iTRAQ differential proteomics analysis proved that HA strengthened the antioxidant system, promoted the synthesis of cell wall, and stabilized protein and sulfur-containing substance metabolism in molecular level. Notably, the concentration of calcium (Ca) in the cell wall of HA coexistence treatment was 47.4% higher than Pb treatment. STXM results also indicated that the distribution of Ca in the cell wall was restored with the presence of HA. This might promote the formation of the egg-box model, thus alleviating Pb stress in cells. Our results reveal the regulation mechanisms of HA on Pb detoxification in plants and provide useful information for improving the safety of agricultural products.
اظهر المزيد [+] اقل [-]Toxicity assessment of metal oxide nano-pollutants on tomato (Solanum lycopersicon): A study on growth dynamics and plant cell death

The present study for the first time demonstrated the interactions of metal oxide (MO) nano-pollutants (CuO and Al2O3-NPs) with tissues and cellular DNA of tomato plants grown in soil sand: silt: clay (667:190:143) and Hoagland-hydroponic system and assessed the hazardous effects of NPs on cell physiology and biochemistry. Results of SEM equipped with EDX revealed attachment of variably shaped CuO-NPs (18 nm) and Al2O3-NPs (21 nm) on roots, and internalization followed by translocation in plants by ICP-MS and TEM. Significant variations in foliage surface area, chlorophyll, proteins, LPO, and antioxidant enzymes were recorded. Roots and shoots accumulated 225.8 ± 8.9 and 70.5 ± 4 μgAl g−1 DW, whereas Cu accumulation was 341.6 ± 14.3 (roots) and 146.9 ± 8.1 μg g−1 DW (shoots) which was significant (p ≤ 0.0005) as compared to control. The total soluble protein content in roots, shoots, and leaves collected from Al2O3-NPs treated plants increased by 120, 80, and 132%, respectively while in CuO-NPs treatments, the increase was 68 (roots), 36 (shoots), and 86% (leaves) over control. The level of antioxidant enzymes in plant tissues was significantly (p ≤ 0.05) higher at 2000 μg ml−1 of MONPs over control. A dose-dependent increase in reactive oxygen species (ROS), biphasic change of lower and higher fluorescence in mitochondria due to dissipation of mitochondrial membrane potential (ΔΨm) and membrane defects using propidium iodide were observed. Comparatively, CuO-NPs induced higher toxicity than Al2O3-NPs. Perceptible changes in proteins (amide-I & II), cellulose, glucose, galactose and other carbohydrates were observed under FT-IR. The binding studies with TmDNA showed fluorescence quenching of EtBr-TmDNA and acridine orange-TmDNA complex only by CuO-NPs with -ΔG and +ΔH and +ΔS values. However, Al2O3-NPs induced lesser change in TmDNA conformation. Conclusively, the results are novel in better demonstrating the mechanistic basis of nano-phyto-toxicity and are important which could be used to develop strategies for safe disposal of Al2O3-NPs and CuO-NPs.

Graphene photocatalysis is a new method for harmful algae and water pollution control. However, microbial communities undergoing graphene photocatalysis treatment in freshwater lakes have been poorly studied. Here, using 16S rRNA and 18S rRNA gene high-throughput sequencing, the responses of microbial communities to graphene photocatalysis were analyzed in the eutrophic lake, Lake Xinyun, southwestern China. For microeukaryotes, we found that Arthropoda was dominant in summer, while its abundant level declined in spring under natural conditions. The evident reduction of Arthropods was observed after graphene photocatalysis treatment in summer and then reached a relatively stable level. For bacteria, Cyanobacteria decreased in summer due to the graphene photocatalysis-mediated inactivation. However, Cyanobacteria was higher in the treated group in spring with a genera group-shift. Functional analysis revealed that microeukaryotes showed higher potential for fatty acid oxidation and TCA cycle in the treated group in summer, but they were more abundant in control in spring. Pathways of starch and sucrose metabolism and galactose metabolism were more abundant in control in summer, while they were enriched in the treated group in spring for bacteria. This study offers insights into the effects of graphene photocatalysis on microbial communities and their functional potential in eutrophic lake.

Boron (B) has previously been shown to inhibit cadmium (Cd) uptake in wheat. Here, we investigated the physiological response of external B application (C for no B added, B for B added, B+Cd for B and Cd added, B/Cd for B 24 h pretreatment before Cd added, B and Cd were 46.2 μM and 5 μM, respectively) on wheat growth under Cd stress. The results showed that the wheat growth was significantly weaker under Cd treatment, while B application did not significantly improve the wheat growth under Cd stress. However, B application decreased Cd concentrations and malondialdehyde (MDA) concentrations of shoot and root. The key enzyme activities including superoxide dismutase (SOD) and peroxidase (POD) significantly increased under Cd treatments while decreased under B treatments. Further, a total of 198, 680 and 204 of the differential metabolites were isolated between B and C treatment, Cd and C treatment and B+Cd and Cd treatment, respectively. The metabolites with up-accumulation in B application (B+Cd) roots were mainly galactaric acid, citric acid, N6-galacturonyl-L-lysine, D-glucose, while the metabolites with down-accumulation were mainly threoninyl-tryptophan and C16 sphinganine. The differential metabolic pathways were mainly concentrated in linoleic acid metabolism, galactose metabolism, sphingolipid metabolism, glycolysis/gluconeogenesis, propanoate metabolism in diabetic complications between B+Cd treatment and B treatment. The results indicate that B alleviates Cd toxicity in winter wheat by inhibiting Cd uptake, increasing antioxidant enzyme activity and changing metabolites.

Aging is an ultimate reality that everyone has to face. D-galactose (D-gal) has been used extensively to develop aging model. Trace elements such as selenium (Se) have been used as a potential antioxidant for neuro-protection. The present work aims to develop therapeutic agents such as Se for the treatment of aging-induced neurological ailments such as anxiety, depression, and memory impairment. For this purpose, mice were treated with D-gal at a dose of 300 mg/ml/kg and various doses of Se (0.175 and 0.35mg/ml/kg) for 28 days. Behavioral tests were monitored after treatment days. After the behavioral assessment, mice were decapitated and their brains were collected. Hippocampi were removed from the brain for biochemical, neurochemical, and histopathological analysis. The present findings of behavioral analysis showed that D-gal-induced anxiety- and depression-like symptoms were inhibited by both doses of Se. D-gal-induced memory alteration was also prevented by repeated doses of Se (0.175 and 0.35mg/ml/kg). Biochemical analysis showed that D-gal-induced increase of oxidative stress and inflammatory markers and decrease of antioxidant enzymes and total protein contents in the hippocampus were prevented by Se administration. An increase in the activity of acetylcholinesterase was also diminished by Se. The neurochemical assessment showed that D-gal-induced increased serotonin metabolism and decreased acetylcholine levels in the hippocampus were restored by repeated treatment of Se. Histopathological estimations also exhibited; normalization of D-gal induced neurodegenerative changes. It is concluded that D-gal-induced dysfunction in mice hippocampus caused anxiety, depression, memory impairment, oxidative stress, neuro-inflammation, and histological alterations that were mitigated by Se via its antioxidant potential, anti-inflammatory property, and modulating capability of serotonergic and cholinergic functions.

Large quantities of waste biomass are generated annually worldwide by many industries and are vastly underutilized. However, these wastes contain sugars and other dissolved organic matter and therefore can be exploited to produce microbial biopolymers. In this study, four selected Halomonas strains, namely, Halomonas caseinilytica K1, Halomonas elongata K4, Halomonas smyrnensis S3, and Halomonas halophila S4, were investigated for the production of exopolysaccharides (EPS) using low-cost agro-industrial wastes as the sole carbon source: cheese whey, grape pomace, and glycerol. Interestingly, both yield and monosaccharide composition of EPS were affected by the carbon source. Glucose, mannose, galactose, and rhamnose were the predominant monomers, but their relative molar ratio was different. Similarly, the average molecular weight of the synthesized EPS was affected, ranging from 54.5 to 4480 kDa. The highest EPS concentration (446 mg/L) was obtained for H. caseinilytica K1 grown on cheese whey that produced an EPS composed mostly of galactose, rhamnose, glucose, and mannose, with lower contents of galacturonic acid, ribose, and arabinose and with a molecular weight of 54.5 kDa. Henceforth, the ability of Halomonas strains to use cost-effective substrates, especially cheese whey, is a promising approach for the production of EPS with distinct physicochemical properties suitable for various applications.

Natural polymeric flocculant shows effectiveness in wastewater treatment without increasing the environmental burden. The extracellular substance produced by Paenibacillus mucilaginosus WL412 was identified as an anionic polysaccharide composed of five types of monosaccharides, namely, D-mannose, D-glucuronic acid, D-glucose, D-galactose, and L-fucose with the molar ratio of 2.8:1.2:2.0:1.8:0.8. The purified polysaccharide, POS412, presented high efficiency in flocculating coal washing wastewater and kaolin suspension without the assistance of inorganic coagulants. Addition of POS412 resulted in the polymer bridging phenomenon in suspensions, which was observed by means of scanning electron microscopic imaging, size grading, and ζ-potential analyses. More importantly, POS412 exhibited satisfactory stability after storage in various conditions. The flocculation rate was more than 91% for coal washing wastewater when POS412 was stored for 264 h in the wide range of pH (3–11) and temperature (20–50 °C) before use. Results indicate that POS412 is a competent bioflocculant for wastewater treatment.
