Livestock manure is a reservoir for antibiotic resistance genes (ARGs), and aerobic composting is used widely for recycling animal manure. This study investigated the effects of adding nano-zerovalent iron (nZVI) at 0, 100, and 1000 mg/kg on the fates of ARGs and mobile genetic elements (MGEs) during swine manure composting. Under nZVI at 100 mg/kg, the relative abundances of sul1, sul2, dfrA7, ermF, and ermX decreased by 33.26–99.31% after composting, and the relative abundances of intI2 and Tn916/1545 decreased by 95.59% and 97.65%, respectively. Most of the ARGs and MGEs co-occurred and they had strong correlations with each other. The bacterial community structure was significantly separated by the composting periods, and they clustered together under different treatments in the same phase. Network analysis showed that Solibacillus, Clostridium_sensu_stricto_1, Terrisporobacter, Romboutsia, Turicibacter, Lactobacillus, Planococcus, Dietzia, and Corynebacterium_1 were common potential hosts of ARGs and MGEs. Redundancy analysis suggested that MGEs had key effects on the variations in the relative abundances of ARGs. Adding 100 mg/kg nZVI could reduce the environmental risk of ARGs by decreasing the abundances of MGEs.

Fish-associated antibiotic resistance genes (ARGs) have attracted increasing attention due to their potential risks to human beings via the food chain. However, data are scarce regarding the antibiotic resistance in fish themselves. Herein, the antibiotic resistance genes (ARGs) were assessed in the gut of four major Chinese freshwater carp (i.e., silver carp, grass carp, bighead carp, and crucian carp) from food retail markets. Results show that the abundances of target ARGs (e.g., tetA, tetO, tetQ, tetW, sulI, sulII, and blaTEM₋₁) and class 1 integrase (intI1) were in the range 9.4 × 10⁻⁶ - 1.6 × 10⁻¹ and 6.7 × 10⁻⁵ - 5.2 × 10⁻² gene copies per 16S rRNA gene, respectively. The sulI, sulII, and tetQ strongly correlated with silver and mercury resistance genes (e.g., silE and merR). The microbial taxa of fish gut could be partly separated among retail markets based on the PCA analysis. About 15.0% of the OTUs in fish gut were shared and 74.5% of the shared OTUs were identified as Acidobacteria, Bacteroidetes, Chloroflexi, Cyanobacteria, Firmicutes, and Proteobacteria. These phyla may constitute the core microbiota in the guts of the four Chinese freshwater carp. The possible ARG hosts were revealed based on the network analysis, and the presence of pathogen-associated resistant genera in fish gut highlights the need to fully understand their potential human health risks.

Vermicomposting is a green technology used in the recycling of sewage sludge using the joint action of earthworms and microorganisms. Although tetracycline is present in abundance in sewage sludge, little attention has been given to its influence on vermicomposts. This study investigated the effects of different tetracycline concentrations (0, 100, 500 and 1000 mg/kg) on the decomposition of organic matter, microbial community and antibiotic resistance genes (ARGs) during vermicomposting of spiked sludge. The results showed that 100 mg/kg tetracycline could stimulate earthworms’ growth, accompanied by the highest humification and decomposition rates of organic matter in the sludge. The abundance of active microbial cells and diversity decreased with the increase in tetracycline concentrations. The member of Bacteroidetes dominated in the tetracycline spiked treatments, especially in the higher concentration treatments. Compared to its counterparts, the addition of tetracycline significantly increased the abundances of ARGs (tetC, tetM, tetX, tetG and tetW) and Class 1 integron (int-1) by 4.7–186.9 folds and 4.25 folds, respectively. The genera of Bacillus and Mycobacterium were the possible bacterial pathogen hosts of ARGs enriched in tetracycline added group. This study suggests that higher concentration of tetracycline residual can modify microbial communities and increase the dissemination risk of ARGs for final sludge vermicompost.

Polychlorinated biphenyls (PCBs), one type of lipophilic pollutant, are ubiquitous in daily life. PCBs exposure has been implicated in the alterations of gut microbial community which is profoundly associated with diverse metabolic disorders, including obesity. High-fat diet (H) is a dietary pattern characterized by a high percentage of fat. According to the theory that similarities can be easily solvable in each other, PCBs and H exposures are inevitably and objectively coexistent in a real living environment, prompting great concerns about their individual and combined effects on hosts. However, the effects of PCBs-H interactions on gut microbiota and obesity are still incompletely understood. In the present study, the effects of PCBs and/or H on the gut microbiota alteration and obesity risk in mice were examined and the interactions between PCBs and H were investigated. Obtained results showed that PCBs and/or H exposure induced prominent variations in the gut microbiota composition and diversity. Exposure to PCBs also resulted in higher body fat percentage, greater size of abdominal subcutaneous adipocytes and increased expression of proinflammatory cytokines including TNF-α, iNOS and IL-6. Such PCBs-induced changes could be further enhanced upon the co-exposure of H, implying that obese individuals may be vulnerable to PCBs exposure. Taken together, the present study is helpful for a better understanding of the gut microbiota variation influenced by PCBs and/or H exposure, and furthermore, provides a novel insight into the mechanism of PCBs-H interactions on host adiposity.

Ecological communities are increasingly exposed to natural and anthropogenic stressors. While the effects of individual stressors have been broadly investigated, there is growing evidence that multiple stressors are frequently encountered underscoring the need to examine interactive effects. Pesticides and infectious diseases are two common stressors that regularly occur together in nature. Given the documented lethal and sublethal effects of each stressor on individuals, there is the potential for interactive effects that alter disease outcomes and pesticide toxicity. Using larval wood frogs (Lithobates sylvaticus), we examined the reciprocal interaction between insecticides (carbaryl and thiamethoxam) and the viral pathogen ranavirus by testing whether: (1) prior ranavirus infection influences pesticide toxicity and (2) sublethal pesticide exposure increases susceptibility to and transmission of ranavirus. We found that prior infection with ranavirus increased pesticide toxicity; median lethal concentration (LC50) estimates were reduced by 72 and 55% for carbaryl and thiamethoxam, respectively. Importantly, LC50 estimates were reduced to concentrations found in natural systems. This is the first demonstration that an infection can alter pesticide toxicity. We also found that prior pesticide exposure exacerbated disease-induced mortality by increasing mortality rates, but effects on infection prevalence and transmission of the pathogen were minimal. Collectively, our results underscore the importance of incorporating complexity (i.e. order and timing of exposures) into research examining the interactions between natural and anthropogenic stressors. Given the environmental heterogeneity present in nature, such research will provide a more comprehensive understanding of how stressors affect wildlife.

To reveal the degradation capacity of bacteria in PAH polluted soil and rhizosphere we combined bacterial extradiol ring-cleavage dioxygenase and 16S rRNA analysis in Betula pubescens rhizoremediation. Characterisation of the functional bacterial community by RFLP revealed novel environmental dioxygenases, and their putative hosts were studied by 16S rRNA amplification. Plant rhizosphere and PAH amendment effects were detected by the RFLP/T-RFLP analysis. Functional species richness increased in the birch rhizosphere and PAH amendment impacted the compositional diversity of the dioxygenases and the structural 16S rRNA community. A shift from an Acidobacteria and Verrucomicrobia dominated to an Alpha- and Betaproteobacteria dominated community structure was detected in polluted soil. Clone sequence analysis indicated catabolic significance of Burkholderia in PAH polluted soil. These results advance our understanding of rhizoremediation and unveil the extent of uncharacterized functional bacteria to benefit bioremediation by facilitating the development of the molecular tool box to monitor bacterial populations in biodegradation.

Freshwater mussels are one of the most threatened taxonomic groups in the world, and many species are on the brink of local or global extinction. Human activities have altered mussel living conditions in a plethora of ways. One of the most destructive human-induced impacts on running waters is the catastrophic spill of harmful substances, which results in massive die-offs. Even though Finland is regarded as the world’s top country in terms of environmental regulation quality, riverine systems are not safe. In 2014, River Kokemäenjoki in western Finland experienced the worst NiSO4 spill in the country’s history, visibly affecting the mussel community – including protected Unio crassus – along the river. Because freshwater mussel toxicology is grossly understudied (particularly in Europe), any pollution –linked die-offs offer valuable opportunities to study the issue in natural environment. Here, we report the mussel investigations from 2014 and a follow-up study conducted in 2017 in order to assess the variation in species sensitivity on nickel pollution. In total, 104 sites were sampled, and over 20 000 mussels were identified and counted. Our results indicate that the most impacted species (i.e. that which experienced the highest spill-induced mortality) was Anodonta anatina (62%), followed by Unio pictorum (32%), U. crassus (24%) and Unio tumidus (9%). The underlying reason for the sensitivity of A. anatina is not resolved, hence more research is urgently needed. The low mortality among most of the species in 2017 highlights the temporal nature of the pollution impact and the recovery potential of the mussel community. However, the case is more complex with U. crassus population, which may be experiencing delayed impacts of the spill. Because nickel is one of the most commonly produced industrial metals in the world (hence the pollution incident risk is high) and River Kokemäenjoki hosts mussel community typical for European rivers, our results may benefit many researchers and stakeholders dealing with riverine environments.

Taihu Lake is one of the largest freshwater lakes in China and serves as an important source for drinking water. This lake is suffering from eutrophication, cyanobacterial blooms and fecal pollution, and the inflow Tiaoxi River is one of the main contributors. The goal here was to characterize the bacterial community structure of Tiaoxi River water by next-generation sequencing (NGS), paying attention to bacteria that are either fecal-associated or pathogenic, and to examine the relationship between environmental parameters and bacterial community structure. Water samples collected from 15 locations in three seasons, and fecal samples collected from different hosts and wastewater samples were used for bacterial community analysis. The phyla Proteobacteria, Actinobacteria, Bacteroidetes, and Cyanobacteria were predominant in most of the water samples tested. In fecal samples, Bacteroidetes, Firmicutes, and Proteobacteria were abundant, while wastewater samples were dominated by Proteobacteria, Bacteroidetes, Acidobacteria, and Chloroflexi. The cluster analysis and principal coordinate analysis indicated that bacterial community structure was significantly different between water, fecal and sewage samples. Shared OTUs between water samples and chicken, pig, and human fecal samples ranged from 4.5 to 9.8% indicating the presence of avian, pig and human fecal contamination in Tiaoxi River. At genus level, five bacterial genera of fecal origin and sequences of seven potential pathogens were detected in many locations and their presence was correlated well with the land use pattern. The sequencing data revealed that Faecalibacterium could be a potential target for human-associated microbial source-tracking qPCR assays. Our results suggest that pH, conductivity, and temperature were the main environmental factors in shaping the bacterial community based on redundancy analysis. Overall, NGS is a valuable tool for preliminary investigation of environmental samples to identify the potential human health risk, providing specific information about fecal and potentially pathogenic bacteria that can be followed up by specific methods.

Activated sludge (AS) has been regarded as the main driver in the removal of organic pollutants such as pesticides due to a high diversity and abundance of microorganisms. However, little is known about the biodegradation genes (BDGs) and pesticide degradation genes (PDGs) harbored in the AS from wastewater treatment plants (WWTPs). In this study, we explored the bacterial communities and BDGs/PDGs in the AS from five WWTPs affiliated with pesticide factories across four consecutive seasons based on high-throughput sequencing. The AS in pesticide WWTPs exhibited unique bacterial taxa at the genus level. Furthermore, a total of 17 BDGs and 68 PDGs were explored with a corresponding average relative abundance of 0.002–0.046% and 2.078–7.143% in each AS sample, respectively, and some BDGs/PDGs clusters were also identified in the AS. The bacterial communities and BDGs/PDGs were season-dependent, and the total variations of 50.4% and 76.8% were jointly explained by environmental variables (pesticide types, wastewater characteristics, and temperature). In addition, network analysis and distribution patterns suggested that the potential hosts of BDGs/PDGs were Thauera, Stenotrophomonas, Mycobacterium, Hyphomicrobium, Allochromatium, Ralstonia, and Dechloromonas. Our findings demonstrated the linkages of bacterial communities and BDGs/PDGs in the AS, and depended on the seasons and the pesticide wastewater characteristics.

Urban and peri-urban forests provide a multitude of Ecosystem Services to the citizens. While the capacity of removing carbon dioxide and gaseous compounds from the atmosphere has been tested, their capacity to sequestrate particles (PM) has been poorly investigated. Mediterranean forest ecosystems are often located nearby or inside large urban areas. This is the case of the city of Rome, Italy, which hosts several urban parks and is surrounded by forested areas. In particular, the Presidential Estate of Castelporziano is a 6000 ha forested area located between the Tyrrhenian coast and the city (25 km downtown of Rome). Under the hypothesis that forests can ameliorate air quality thanks to particle deposition, we measured fluxes of PM1, 2.5 and 10 with fast optical sensors and eddy covariance technique. We found that PM1 is mainly deposited during the central hours of the day, while negligible fluxes were observed for PM 2.5 and 10. A Hybrid Single–Particle Lagrangian Integrated Trajectory model (HYSPLIT v4) simulated PM emission from traffic areas in the city of Rome and showed that a significant portion of PM is removed by vegetation in the days when the plume trajectory meets the urban forest.
