Seed germination and seedling establishment are the stages most sensitive to abiotic stress in the plant life cycle. We analyzed the effects of copper, zinc and nickel on seed germination and early seedling growth of native Spartina maritima and invasive S. densiflora from polluted and non-polluted estuaries. Germination percentages for either species were not affected by any metal at any tested concentration (up to 2000 μM). However, the increase in metal concentration had negative effects on S. densiflora seedlings. The primary effect was on radicle development, representing initial seedling emergence. Spartina densiflora seedlings emerging from seeds from Tinto Estuary, characterized by high bioavailable metal loads, showed higher tolerance to metals than those from less polluted Odiel and Piedras Estuaries. Comparing our results to metal concentrations in the field, we expect S. densiflora seedling development would be negatively impacted in the most metal-polluted areas in Odiel and Tinto Estuaries.

Mangroves are highly susceptible to oil exposure. Depending on the severity, oil exposure can result in initial defoliation and eventual recovery through to mass mortality and complete loss of habitat. Some aspects of the impact of oil on mangroves and their recovery are well studied, but the focus has been on short-term responses, and the understanding of the longer-term trajectory of mangrove recovery from oiling is very limited. Here, we combine field results from sampling in the two years following a significant oiling event, with analysis of canopy cover in aerial images from before the event to 26 years afterwards. Approximately 100 ha of a monospecific stand of Avicenna marina mangroves were oiled as a result of a spill from the Era tanker in Spencer Gulf in southern Australia in September 1992. While lightly oiled trees made a full recovery, trees in heavily oiled areas experienced mass defoliation and ultimately mortality within several months of the oiling event. An analysis of aerial images indicated that there was no recovery in heavily oiled areas for 10 years following the oiling event. Between 10 and 25 years, seedling establishment and growth saw canopy cover increase to 35% of pre-oiling cover within heavily oiled areas. Predictive modelling estimates that complete recovery of mangroves to pre-oiling cover will take 55 years (median prediction in 2047). Our findings indicate that although mangroves can recover following a heavy oiling event, the rate of recovery can be slow, with full recovery in the order of half a century, much longer than has previously been anticipated.

Past mining activities have left a legacy of abandoned mine tailing deposits whose metal contaminants poses serious risks to ecosystems and human health. While the development of a vegetated cover in mine tailings can help in mitigating these risks, the local factors limiting plant establishment in these sites are not well understood, restricting phytostabilization efforts. Here, we explore some of the barriers that limit seedling establishment of two species (Vachellia farnesiana and Prosopis velutina) in a mine tailing deposit located in Nacozari, Sonora, Mexico, and assess whether compost addition can help in overcoming these barriers in pot and field experiments. Our field observations found 20 times more carbon and at least 4 times more nitrogen concentration in areas under vegetated patches than in non-vegetated areas, while a previous study found no difference in metal concentrations and other physicochemical parameters. This suggests that organic matter and nutrients are a major limitation for plant establishment. In agreement with this, species failed to establish without compost addition in the field experiment. Compost addition also had a positive effect on biomass accumulation, pH and microbial activity, but increased the substrate soluble concentration of As, Cu, and Zn. Nonetheless, only Cu, K, and Mo in P. velutina accumulated in tissues at levels considered toxic for animal consumption. Our study documents that compost addition facilitated plant establishment for the phytostabilization of mine tailings and help to prevent the dispersion of most metal contaminants via animal consumption. We encourage the use of complementary strategies to minimize the risk of dispersion of metal contaminants.

Using association of plants, nanomaterials, and plant growth-promoting bacteria (PGPR) is a novel approach in remediation of heavy metal-contaminated soils. Co-application of nanoscale zerovalent iron (nZVI) and PGPR to promote phytoremediation of Sb-contaminated soil was investigated in this study. Seedlings of Trifolium repens were exposed to different regimes of nZVI (0, 150, 300, 500, and 1000 mg/kg) and the PGPR, separately and in combination, to investigate the effects on plant growth, Sb uptake, and accumulation and physiological response of the plant in contaminated soil. Co-application of nZVI and PGPR had positive effects on plant establishment and growth in contaminated soil. Greater accumulation of Sb in the shoots compared to the roots of T. repens was observed in all treatments. Using nZVI significantly increased accumulation capacity of T. repens for Sb with the greatest accumulation capacity of 3896.4 μg per pot gained in the “PGPR+500 mg/kg nZVI” treatment. Adverse impacts of using 1000 mg/kg nZVI were found on plant growth and phytoremediation performance. Significant beneficial effect of integrated use of nZVI and PGPR on plant photosynthesis was detected. Co-application of nZVI and PGPR could reduce the required amounts of nZVI for successful phytoremediation of metalloid polluted soils. Intelligent uses of plants in accompany with nanomaterials and PGPR have great application prospects in removal of antimony from soil.

Due to the accelerating use of manufactured nanomaterials, more research is needed to define their impact on plants. The present investigation aimed at evaluating the effect of different levels (0.0, 10, 25, 50, and 100 mg/L) of ZnO nanoparticles (NPs) on Vicia faba during seed germination and seedling establishment. Additionally, V. faba root meristems were used as a model to monitor the cytotoxic and genotoxic effects resulting from exposure to ZnO NPs. The influence of ZnO NPs on three isoenzyme systems, peroxidase, α, and β esterase, was also evaluated using native-PAGE. Our results showed that lower concentrations of ZnO NPs (especially 10 and 25 mg/L) enhanced seed germination and improved seedling growth, while higher concentrations (100 and 200 mg/L) resulted in phytotoxicity. Cytological investigations of ZnO NPs-treated V. faba root cells denoted the clastogenic and aneugenic nature of ZnO NPs. Differential increase in mitotic index and significant alterations in cell cycle were observed upon exposure to ZnO NPs. High concentrations of ZnO NPs markedly induced chromosomal aberration, micronuclei, and vacuolated nuclei formation. Chromosomal breakage, chromosomal bridges, ring chromosomes, laggard chromosomes, and stickiness were also observed at a higher rate. The PAGE analysis showed that ZnO NPs treatments altered the expression patterns of all studied enzyme systems. Collectively, results from this work will help to further understand the phytotoxic effects of nanomaterials.
