Through quorum sensing, Pseudomonas aeruginosa resists noble metal-based nanomaterials toxicity

Noble metal–based nanomaterials (NMNs), such as platinum nanoparticles (Pt@NPs) and palladium nanoparticles (Pd@NPs), are increasingly being used as antibacterial agents. However, little information is available on bacterial resistance to NMNs. In this study, owing to their oxidase-like and peroxidase-like properties, both Pt@NPs and Pd@NPs induce reactive oxygen species (ROS) and manifest antibacterial activities: 6.25 μg/mL of either Pt@NPs or Pd@NPs killed >50% of Staphylococcus aureus strain ATCC29213. However, Pseudomonas aeruginosa strain PAO1 completely resisted 12.5 μg/mL of Pt@NPs and 6.25 μg/mL of Pd@NPs. Compared to the non-NMN groups, these NMNs promoted 2–3-fold upregulation of the quorum sensing (QS) gene lasR in strain PAO1. In fact, the lasR gene upregulation induced a 1.5-fold reduction in ROS production and increased biofilm formation by 11% (Pt@NPs) and 27% (Pd@NPs) in strain PAO1. The ΔlasR mutants (lasR gene knock out in strain PAO1), became sensitive to NMNs. The survival rates of ΔlasR mutants at 12.5 μg/mL Pt@NPs and Pd@NPs treatments were only 77% and 58%, respectively. This is the first report indicating that bacteria can resist NMNs through QS. Based on these results, evaluation of the ecological risks of using NMNs as antibacterial agents is necessary.

Mining activities in the world's largest platinum mining area in South Africa have resulted in environmental contamination with Pt (e.g., the Hex River's vicinity). The present study compared a Pt mining area with a non-mining area along this river in terms of (1) metal concentrations in different grain size fractions from soils and aquatic sediments; (2) the toxicological potential of aquatic sediments based on the Consensus-Based Sediment Quality Guideline (CBSQG); and (3) the chronic toxicity of aqueous eluates from soils and sediments to Caenorhabditis elegans. Platinum concentrations were higher in the mining area than in the non-mining area. For most metals, the sediment silt and clay fraction contained the highest metal concentrations. Based on the CBSQG, most sampling sites exhibited a high toxicological potential, driven by Cr and Ni. Eluate toxicity testing revealed that C. elegans growth, fertility, and reproduction inhibition were not dependent on mining activities or the CBSQG predictions. Toxicity was instead likely due to Cd, Fe, Mn, Ni, Pt, and Pb. In conclusion, the investigated region is loaded with a high geogenic background resulting in high reproduction inhibition. The mining activities lead to additional environmental metal contamination (particularly Pt), contributing to environmental soil and sediment toxicity.
اظهر المزيد [+] اقل [-]Adsorption and fractionation of Pt, Pd and Rh onto inorganic microparticles and the effects of macromolecular organic compounds in seawater

Adsorption and fractionation of Pt, Pd and Rh (defined here as platinum group elements, PGEs) onto the representative inorganic microparticles, including Fe2O3, MnO2, CaCO3, SiO2, Al2O3 and kaolinite in seawater were investigated. The effects of macromolecular organic compounds (MOCs) as the representatives of organic matter, including humic acids (HA), bovine serum albumin (BSA) and carrageenan, on the adsorption were also studied considering that organic matter is ubiquitous in seawater and indispensable to marine biogeochemical cycles. In the absence of MOCs, the representative mineral particles Fe2O3 and MnO2 had the strongest interaction with PGEs. The adsorption of PGEs onto the representative biogenic particles SiO2 and CaCO3 and lithogenic particles Al2O3 and kaolinite was similar or weaker than onto the mineral particles. MOCs inhibited the interaction between PGEs and the particles except for Pt and Pd onto the biogenic particles in artificial seawater. This impediment may be closely related to the interaction between particles, MOCs and elements. The partition coefficient (log Kd) of Pt was similar (∼4.0) in the presence of MOCs, indicating that the complexation between Pt and MOCs was less important than hydrolysis or adsorption onto the acid oxide particle surface. Rh tended to fractionate onto the mineral and lithogenic particles in the presence of HA and carrageenan, while Pd was more likely to fractionate onto the biogenic particles. However, BSA enhanced the fractionation tendency of Pd onto the mineral particles. The results indicate that the adsorption behavior of Pd onto inorganic particles was significantly affected by the composition or the type of MOCs. Hence, the interaction between PGEs and inorganic particles may be greatly affected by the macromolecular organic matter in the ocean.

Due to the increasing presence of platinum (Pt) in the environment, the caveat arises to identify its toxic potential in species at risk of being exposed – especially those found in aquatic environments where pollutants tend to accumulate. Comprehensive characterisation of possible adverse effects following exposure of aquatic organisms to Pt remains elusive. To address this, Zebra mussels (Dreissena polymorpha) were exposed to a range of Pt(IV) concentrations (0.1, 1, 10, 100 and 1000 μg/L) for one and four days, respectively, after which bioaccumulation was quantified and compared to alterations in biomarker profiles relevant to metal toxicity i.e. glutathione-S-transferase (GST) and catalase (CAT) activity, lipid peroxidation and metallothionein (MT) induction. Despite pre-conditioning of the tanks, Pt recovery in the exposure media was found to be 36% (0.1 μg/L), 42% (1 μg/L), 47% (10 μg/L), 68% (100 μg/L) and 111% (1000 μg/L) due to biological and non-biological processes. Pt concentrations in dried mussel soft tissue increased with exposure concentrations and were 20–153 times higher compared to quantified Pt concentrations in the exposure media. CAT activity was significantly increased in the tissue of mussels exposed to 0.1–100 μg/L Pt after Day 1 while the lowest effect concentration (LOC) for this response on both Day 1 and Day 4 was 0.1 μg/L. The effect on the GST activity was less pronounced but demonstrated a similar trend. However, enhanced lipid peroxidation was measured in the tissue of mussels exposed to ≥0.1 μg/L on Day 4. Bioaccumulation of Pt was also associated with a concentration-dependent increase in Pt-MT. Although these effects occurred at Pt levels higher than those present in the environment, it indicates that Pt has the ability to cause aberrancies in metal-associated biomarker profiles.

The aim of this study was to evaluate the mobility of platinum (Pt) and palladium (Pd) emissions from automotive catalysts in soils and to contribute to the risk assessment of platinum group metals (PGMs) discharged from catalysts in the environment. To address this question, for the first time risk assessment code (RAC) was applied to consider the results from sequential extraction of different Pd and Pt species from soils. For this purpose, model soil samples were prepared spiking defined Pd or Pt species, respectively, at known concentrations. In order to mimic emitted species as well as possible transformation products of traffic-related Pd and Pt emissions in soils, coated and uncoated elemental nanoparticles (cPd/cPt NPs, Pd/Pt NPs) and ionic divalent metal species (Pd(II)/Pt(II)) were applied. All model samples were characterized in detail and the developed sequential extraction scheme was validated. RAC values ranged between 24 and 8% revealing medium to low risk. The order of mobility for the studied species was found to be Pt(II) > cPd NPs » Pd(II) > Pd NPs > Pt NPs > cPt NPs. Furthermore, migration of Pd species in gravity columns was studied confirming highest transport of cPd NPs.

Anticancer drugs are continuously released into hospital and urban wastewaters, where they, most commonly, undergo conventional treatment in wastewater treatment plants (WWTPs). Wastewaters contain complex mixtures of substances including parent compounds, their metabolites and transformation products (TPs). In this study, samples of hospital effluents and WWTP influents and effluents from Slovenia and Spain were analyzed for twenty-two selected anticancer drugs, their metabolites and transformation products. Acute and chronic toxicity tests were performed on the crustacean Ceriodaphnia dubia, genotoxicity was determined with Tradescantia and Allium cepa micronucleus (MN) assays and in vitro comet assay in zebrafish (Danio rerio) liver cell line (ZFL cells). Sixty of the two hundred-twenty determinations revealed detectable levels of anticancer drug residues. Among the targeted compounds, platinum based were most frequently detected (90%). Furthermore, erlotinib was detected in 80%, cyclophosphamide and tamoxifen in 70% and methotrexate in 60% of the samples. Seven of ten samples were toxic to C. dubia after acute exposure, whereas after chronic exposure all samples reduced reproduction of C. dubia at high sample dilutions. Allium cepa proved insensitive to the potential genotoxicity of the tested samples, while in Tradescantia increased MN frequencies were induced by a hospital effluent and WWTP influents. In ZFL comet assay all but one sample induced a significant increase of DNA strand breaks. Correlations of chemotherapeutics or their TPs were detected for all bioassays except for Allium cepa genotoxicity test, however for each test the highest correlations were found for different substances indicating differential sensitivities of the test organisms.

Biochar was prepared from rice straw and modified with 15% H₂O₂ and 1:1 HNO₃/H₂SO₄, respectively. The unmodified biochars and HCl treated biochars for carbonate removal were used as control. The biochars were added to the acid paddy soil collected from Langxi, Anhui Province, China at the rate of 30 g/kg. The paddy soil was flooded and then air-dried, and soil pH and Eh were measured in situ with pH electrode and platinum electrode during wet-dry alternation. Soil pH buffering capacity (pHBC) was determined by acid-base titration after the wet-dry treatment. Then, the simulated acidification experiments were carried out to study the changing trends of soil pH, base cations and exchangeable acidity. The results showed that soil pHBC was effectively increased and the resistance of the paddy soil to acidification was apparently enhanced with the incorporation of H₂O₂- and HNO₃/H₂SO₄-modified biochars. Surface functional groups on biochars were mainly responsible for enhanced soil resistance to acidification. During soil acidification, the protonation of organic anions generated by dissociation of these functional groups effectively retarded the decline of soil pH. The modification of HNO₃/H₂SO₄ led to greater increase in carboxyl functional groups on the biochars than H₂O₂ modification and thus HNO₃/H₂SO₄-modified biochars showed more enhancement in soil resistance to acidification than H₂O₂-modified biochars. After a wet-dry cycle, the pH of the paddy soil incorporated with HNO₃/H₂SO₄-modified biochar increased apparently. Consequently, the addition of HNO₃/H₂SO₄-modified biochar can be regarded as a new method to alleviate soil acidification. In short, the meaning of this paper is to provide a new method for the amelioration of acid paddy soils.

Platinum-Group Elements (PGEs, i.e. platinum; Pt, palladium; Pd and rhodium; Rh) are extensively employed in the production of automotive catalytic converters to catalyze and control harmful emissions from exhaust fumes. But catalytic converters wear out over time and the emission of PGEs along with the exhaust fumes are nowadays known to be the main reason of the presence of PGEs in urban environments. PGEs contents were studied on three gasoline 3-way catalytic convertors with low, medium and high kilometers. PGEs emission factors via exhaust gases from Euro 3, 4, 5 and 6 gasoline and diesel vehicles, were monitored using catalytic converters. Results show variable content for PGEs for the three converters, in the ranges of 6–511, 0.5–2507 and 0.1–312 mg kg⁻¹ for Pt, Pd and Rh respectively. PGEs contents in different catalyst supports show the replacement of Pt by Pd in more recent converters. Analysis of the exhaust gas shows that catalytic converters expel up to 36.5 ± 3.8 ng km⁻¹ of Pt, 8.9 ± 1.1 ng km⁻¹ of Pd and 14.1 ± 1.5 ng km⁻¹ of Rh. Higher emissions of PGEs have been observed by gasoline Euro 3 vehicle, possibly due to the older technology of motorization and of the catalytic converter in this vehicle. Euro 3 and 4 diesel vehicles seem to emit more PGEs during urban cycles. Emission of PGEs has been also observed during the cold start of the majority of vehicles which seems to be the result of incomplete combustion during the rise of temperature in the engine. Higher PGEs emissions were also observed during motorway cycles in newer (Euro 4 and 5) petrol and diesel vehicles, conceivably due to the greater combustion as the engine speeds up during this cycle.

Chemical and toxicological characterization of the water-soluble fraction of size-segregated urban particulate matter (PM) (<0.49, 0.49–0.97, 0.97–1.5, 1.5–3.0, 3.0–7.2 and >7.2 μm) was carried out at two urban sites, traffic and urban background, during the cold and the warm period. Chemical analysis of the water-soluble PM fraction included ionic species (NO3−, SO42−, Cl⁻, Na⁺, NH4⁺, K⁺, Mg²⁺, Ca²⁺), water-soluble organic carbon (WSOC), and trace elements (Al, As, Ba, Cd, Cr, Cu, Fe, Pb, Mn, Ni, Zn, Pt, Pd, Rh, Ru, Ir, Ca, and Mg). The dithiothreitol (DTT) assay was employed for the abiotic assessment of the oxidative PM activity. Cytotoxic responses were investigated in vitro by applying the mitochondrial dehydrogenase (MTT) and the lactate dehydrogenase (LDH) bioassays on human lung cells (MRC-5), while DNA damage was estimated by the single cell gel electrophoresis assay, known as Comet assay. The correlations between the observed bioactivity responses and the concentrations of water-soluble chemical PM constituents in the various size ranges were investigated. The results of the current study corroborate that short-term bioassays using lung human cells and abiotic assays, such as the DTT assay, could be relevant to complete the routine chemical analysis and to obtain a preliminary screening of the potential effects of PM-associated airborne pollutants on human health.

The increasing content of platinum group metal particles emitted into the environment by car traffic is gradually attracting the attention of the scientific community. However, the methods for the determination of platinum group metals in environmental matrices are either costly or suffer from low sensitivity. To facilitate the use of less sensitive, but significantly cheaper, devices, the preconcentration of platinum group metals is employed. For platinum, a multitude of preconcentration approaches have been published. On the contrary, the preconcentration approaches for palladium are still rare. In this work, the development, optimization, and testing of a new approach is described; it is based on a preconcentration of palladium on octadecyl modified silica gel together with the complexing agent dimethylglyoxime, and it is then analyzed with the high-resolution continuum-source atomic absorption spectrometry. For comparison, a newly developed sorbent, QuadraSil™ TA, with a high affinity for platinum group metals was also tested. The preconcentraiton approach was tested on the lichen Hypogymnia physodes, which served as a bioindicator of palladium emissions. The case study site was a mid-sized city in central Europe: Brno, Czech Republic. The dry “bag” monitoring technique was used to collect the palladium near roads with a large span of traffic density. The developed analytical approach confirmed an increasing concentration of palladium with increasing exposure time and intensity of the traffic. Consequently, a simple relationship between the amount of bioaccumulated palladium and traffic density was established.
