Rapid method demonstration project at four New Jersey marine beaches using real time quantitative Polymerase Chain Reaction (qPCR) النص الكامل

Real time quantitative Polymerase Chain Reaction (qPCR) was used at four marine bathing beaches in New Jersey as part of a demonstration project to evaluate the potential for use of qPCR as part of a routine beach monitoring program. Split sample analyses for Enterococcus spp. using membrane filtration (MF) and qPCR were performed for 11weeks during the summer of 2011 using swimming advisories based on qPCR results. Comparison of qPCR and MF results from split samples indicated that there was an 82% overall agreement rate between the two methods. Results from the qPCR tests were available by noon the same day of sample collection and swimming advisories were posted on a dedicated website. The qPCR method can be more labor intensive and requires a higher level of training to perform, however, qPCR was able to assess beach water quality in a timelier manner compared to conventional MF techniques.

We investigated the effects of elevated pCO2 in seawater both on the acute mortality and the reproductive properties of the benthic copepod Tigriopus japonicus and gastropod Babylonia japonica with the purpose of accumulating basic data for assessing potential environmental impacts of sub-sea geological storage of anthropogenic CO2 in Japan. Acute tests showed that nauplii of T. japonicus have a high tolerance to elevated pCO2 environments. Full life cycle tests on T. japonicus indicated NOEC=5800μatm and LOEC=37,000μatm. Adult B. japonica showed remarkable resistance to elevated pCO2 in the acute tests. Embryonic development of B. japonica showed a NOEC=1500μatm and LOEC=5400μatm. T. japonicus showed high resistance to elevated pCO2 throughout the life cycle and B. japonica are rather sensitive during the veliger stage when they started to form their shells.

The environmental impact of elevated carbon dioxide (CO2) levels has become of more interest in recent years. This, in relation to globally rising CO2 levels and related considerations of geological CO2 storage as a mitigating measure. In the present study effect data from literature were collected in order to conduct a marine ecological risk assessment of elevated CO2 levels, using a Species Sensitivity Distribution (SSD). It became evident that information currently available from the literature is mostly insufficient for such a quantitative approach. Most studies focus on effects of expected future CO2 levels, testing only one or two elevated concentrations. A full dose-response relationship, a uniform measure of exposure, and standardized test protocols are essential for conducting a proper quantitative risk assessment of elevated CO2 levels. Improvements are proposed to make future tests more valuable and usable for quantitative risk assessment.

Since 2005, five different ballast water management systems (BWMSs) based on chlorination treatment have been tested by Norwegian Institute for Water Research (NIVA) according to guidelines from the International Maritime Organization (IMO). 25% and >50% of all the tested discharge samples exhibited acute and chronic toxic effects on algae, respectively. In most cases this toxicity was plausibly caused by a high free residual oxidant (FRO) level (>0.08mgCl/l). Of the 22 disinfection by-products (DBPs) that were identified in treated water at discharge, four compounds were at times found at concentrations that may pose a risk to the local aquatic environment. However, there seemed to be no clear indication that the measured DBP concentrations contributed to the observed algal toxicity. The addition of methylcellulose instead of lignin in the test water to comply with IMO requirements seemed to limit the formation of DBP.

Boring isopods damage expanded polystyrene floats under docks and, in the process, expel copious numbers of microplastic particles. This paper describes the impacts of boring isopods in aquaculture facilities and docks, quantifies and discusses the implications of these microplastics, and tests if an alternate foam type prevents boring. Floats from aquaculture facilities and docks were heavily damaged by thousands of isopods and their burrows. Multiple sites in Asia, Australia, Panama, and the USA exhibited evidence of isopod damage. One isopod creates thousands of microplastic particles when excavating a burrow; colonies can expel millions of particles. Microplastics similar in size to these particles may facilitate the spread of non-native species or be ingested by organisms causing physical or toxicological harm. Extruded polystyrene inhibited boring, suggesting this foam may prevent damage in the field. These results reveal boring isopods cause widespread damage to docks and are a novel source of microplastic pollution.

In the UK, Government policy requires marine aggregate extraction companies to leave the seabed in a similar physical condition after the cessation of dredging. This measure is intended to promote recovery, and the return of a similar faunal community to that which existed before dredging. Whilst the policy is sensible, and in line with the principles of sustainable development, the use of the word ‘similar’ is open to interpretation. There is, therefore, a need to set quantifiable limits for acceptable change in sediment composition. Using a case study site, it is shown how such limits could be defined by the range of sediment particle size composition naturally found in association with the faunal assemblages in the wider region. Whilst the approach offers a number of advantages over the present system, further testing would be required before it could be recommended for use in the regulatory context.

Unmanned aerial systems (UAS, aka drones) are being used to map macro-litter on the environment. Sixteen qualified researchers (operators), with different expertise and nationalities, were invited to identify, mark and categorize the litter items (manual image screening, MS) on three UAS images collected at two beaches.The coefficient of concordance (W) among operators varied between 0.5 and 0.7, depending on the litter parameter (type, material and colour) considered. Highest agreement was obtained for the type of items marked on the highest resolution image, among experts in litter surveys (W = 0.86), and within territorial subgroups (W = 0.85). Therefore, for a detailed categorization of litter on the environment, the MS should be performed by experienced and local operators, familiar with the most common type of litter present in the target area. This work provides insights for future operational improvements and optimizations of UAS-based images analysis to survey environmental pollution.

Most crude oils spread on open water to an average thickness as low as 0.1mm. The application of dispersants enhances the transport of oil as small droplets into the water column, and when combined with the turbulence of 1m waves will quickly entrain oil into the top 1m of the water column, where it rapidly dilutes to concentrations less than 100ppm. In less than 24h, the dispersed oil is expected to mix into the top 10m of the water column and be diluted to concentrations well below 10ppm, with dilution continuing as time proceeds. Over the multiple weeks that biodegradation takes place, dispersed oil concentrations are expected to be below 1ppm. Measurements from spills and wave basin studies support these calculations. Published laboratory studies focused on the quantification of contaminant biodegradation rates have used concentrations orders of magnitude greater than this, as it was necessary to ensure the concentrations of hydrocarbons and other chemicals were higher than the detection limits of chemical analysis. However, current analytical methods can quantify individual alkanes and PAHs (and their alkyl homologues) at ppb and ppm levels. To simulate marine biodegradation of dispersed oil at dilute concentrations commonly encountered in the field, laboratory studies should be conducted at similarly low hydrocarbon concentrations.

A baseline dataset from 2005 was used to identify the spatial distribution of macrofaunal assemblages across the eastern English Channel. The range of sediment composition found in association with each assemblage was used to define limits for acceptable change at ten licensed marine aggregate extraction areas. Sediment data acquired in 2010, 4years after the onset of dredging, were used to assess whether conditions remained within the acceptable limits. Despite the observed changes in sediment composition, the composition of sediments in and around nine extraction areas remained within pre-defined acceptable limits. At the tenth site, some of the observed changes within the licence area were judged to have gone beyond the acceptable limits. Implications of the changes are discussed, and appropriate management measures identified. The approach taken in this study offers a simple, objective and cost-effective method for assessing the significance of change, and could simplify the existing monitoring regime.

The concept of Ecological Potential was explored using the macrobenthic communities of the Mondego estuary (Portugal). Different scenarios (loss of a primary producer and intertidal habitat, and habitat modification due to hydromorphological changes and continued press perturbation) were tested to predict changes in the biology if hydromorphological pressures could be reversed, assuming that differences with and without the pressure could indicate the potential.Results showed noticeable changes in the system biology in each scenario. The approach followed, indicates that when data sets exist, differences in the measurement of ecological status with and without the hydromorphological change could be a way forward to determine the potential. In the particular case of the Mondego estuary, the South arm (physically unaltered water body) proved to be richer than the North (HMWB). For the Ecological Potential determination, the South arm could thus be used to derive and adjust future reference conditions for the North.
