The application of the fungicide thiabendazole (TBZ) in fruit packaging plants (FPP) results in the production of effluents which are often disposed in adjacent field sites. These require remediation to prevent further environmental dispersal of TBZ. We assessed the bioaugmentation potential of a newly isolated TBZ-degrading bacterial consortium in a naturally contaminated soil (NCS) exhibiting a natural gradient of TBZ levels (12000, 400, 250 and 12 mg kg⁻¹). The effect of aging on bioaugmentation efficacy was comparatively tested in a soil with similar physicochemical properties and soil microbiota, which was artificially, contaminated with the same TBZ levels (ACS). The impact of bioaugmentation and TBZ on the bacterial diversity in the NCS was explored via amplicon sequencing. Bioaugmentation effectively removed TBZ from both soils at levels up to 400 mg kg⁻¹ but failed at the highest contamination level (12000 mg kg⁻¹). Dissipation of TBZ in bioaugmented samples showed a concentration-dependent pattern, while aging of TBZ had a slight effect on bioaugmentation efficiency. Bioaugmentation had no impact on the soil bacterial diversity, in contrast to TBZ contamination. Soils from the hotspots of TBZ contamination (12000 mg kg⁻¹) showed a drastically lower α-diversity driven by the dominance of β- and γ-proteobacteria at the expense of all other bacterial phyla, especially Actinobacteria. Overall, bioaugmentation with specialized microbial inocula could be an effective solution for the recovery of disposal sites contaminated with persistent chemicals like TBZ.

Evaluating potential adverse effects of complex chemical mixtures in the environment is challenging. One way to address that challenge is through more integrated analysis of chemical monitoring and biological effects data. In the present study, water samples from five locations near two municipal wastewater treatment plants in the St. Croix River basin, on the border of MN and WI, USA, were analyzed for 127 organic contaminants. Known chemical-gene interactions were used to develop site-specific knowledge assembly models (KAMs) and formulate hypotheses concerning possible biological effects associated with chemicals detected in water samples from each location. Additionally, hepatic gene expression data were collected for fathead minnows (Pimephales promelas) exposed in situ, for 12 d, at each location. Expression data from oligonucleotide microarrays were analyzed to identify functional annotation terms enriched among the differentially-expressed probes. The general nature of many of the terms made hypothesis formulation on the basis of the transcriptome-level response alone difficult. However, integrated analysis of the transcriptome data in the context of the site-specific KAMs allowed for evaluation of the likelihood of specific chemicals contributing to observed biological responses. Thirteen chemicals (atrazine, carbamazepine, metformin, thiabendazole, diazepam, cholesterol, p-cresol, phenytoin, omeprazole, ethyromycin, 17β-estradiol, cimetidine, and estrone), for which there was statistically significant concordance between occurrence at a site and expected biological response as represented in the KAM, were identified. While not definitive, the approach provides a line of evidence for evaluating potential cause-effect relationships between components of a complex mixture of contaminants and biological effects data, which can inform subsequent monitoring and investigation.

Imazalil (IMZ) is an imidazole fungicide commonly used by fruit-packaging plants (FPPs) to control fungal infections during storage. Its application leads to the production of pesticide-contaminated wastewaters, which, according to the European Commission, need to be treated on site. Considering the lack of efficient treatment methods, biodepuration systems inoculated with tailored-made inocula specialized on the removal of such persistent fungicides appear as an appropriate solution. However, nothing is known about the biodegradation of IMZ. We aimed to isolate and characterize microorganisms able to degrade the recalcitrant fungicide IMZ and eventually to test their removal efficiency under near practical bioengineering conditions. Enrichment cultures from a soil receiving regular discharges of effluents from a FPP, led to the isolation of a Cladosporium herbarum strain, which showed no pathogenicity on fruits, a trait essential for its biotechnological exploitation in FPPs. The fungus was able to degrade up to 100 mg L⁻¹ of IMZ. However, its degrading capacity and growth was reduced at increasing IMZ concentrations in a dose-dependent manner, suggesting the involvement of a detoxification rather than an energy-gain mechanism in the dissipation of IMZ. The isolate could tolerate and gradually degrade the fungicides fludioxonil (FLD) and thiabendazole (TBZ), also used in FPPs and expected to coincide alongside IMZ in FPP effluents. The capacity of the isolate to remove IMZ in a practical context was evaluated in a benchtop immobilized-cell bioreactor fed with artificial IMZ-contaminated wastewater (200 mg L⁻¹). The fungal strain established in the reactor, completely dominated the fungal community and effectively removed >96% of IMZ. The bioreactor also supported a diverse bacterial community composed of Sphingomonadales, Burkholderiales and Pseudomonadales. Our study reports the isolation of the first IMZ-degrading microorganism with high efficiency to remove IMZ from agro-industrial effluents under bioengineering conditions.

Benzimidazoles are anthelmintics frequently used in sheep farming due to the high susceptibility of these animals to parasitic diseases. Sheep excreta are often disposed onto soils as a fertilizer, and they may contain benzimidazole residues that can contaminate soil and water. This work aimed to assess the leaching behavior of benzimidazole drugs (albendazole, fenbendazole, and thiabendazole) and their metabolites in two Brazilian soils of different textural classifications (sandy and clay), as well as sheep excreta-amended soils, following the OECD 312 Guidelines. Ewes received a single oral dose of 10 mg kg⁻¹ b.w. of either albendazole or fenbendazole. The feces were collected at 24, 48, 72, 96, and 120 h post-dose, and the parent drugs and their metabolites extracted using the QuEChERS approach and quantified by UHPLC-MS/MS. For the leaching assays, a benzimidazole solution was directly applied onto the soil columns, or an amount of 5 g of the medicated sheep feces was distributed over the top of the soil columns. In soil samples, benzimidazoles were extracted by solid-liquid extraction and quantified by UHPLC-MS/MS. For the leaching studies, atrazine was used as a reference substance to determine the relative mobility factor of the analytes of interest. Benzimidazoles were considered slightly to moderately mobile in both soils tested, with a leaching distance of up to 25 cm in a 30-cm soil column. Approximately 3 to 6% of the benzimidazoles present in ewe feces were able to leach into the soil columns. This finding is of concern since benzimidazoles are persistent in soil and may pose a risk to soil biota and induce the development of resistant strains of parasites.

A monitoring study was carried out in two micro-catchments in the Reventazón basin, in Northern Cartago, Costa Rica; pesticide occurrence and water quality were analyzed. Twelve pesticides were detected, five insecticides (chlorpyrifos, carbofuran, cypermethrin, imidacloprid, and oxamyl), four fungicides (carbendazim, imazalil, metalaxyl, and thiabendazole), and three herbicides (diuron, linuron, and terbutryn); eight of them presented risk quotients RQ >1, which implies a high risk for the environment. The water quality evaluation included fourteen physicochemical and microbiological parameters, out of which thermotolerant coliforms, nitrate, and total phosphorus exceeded a selected threshold value in every sample. Five metals were also included in the evaluation, Pb was the most frequent, followed by few detections of Cd, Cu, and Cr. Four water quality indexes (WQIs) were applied, two of them, the CCME WQI, based on physicochemical parameters, and the BMWP-CR WQI, based on benthic macroinvertebrate recount adapted to Costa Rican species, categorized all the sampling points as “bad” and “very bad” quality. This work of monitoring is important in the Latin American region, where there is a lack of information for regulation improvement and management decisions. These results showed poor management of the micro-catchments in this agricultural rural area.

Benzimidazoles (BZ) are among the most used drugs to treat parasitic diseases in both human and veterinary medicine. In this study, solutions fortified with albendazole (ABZ), fenbendazole (FBZ), and thiabendazole (TBZ) were subjected to photoperoxidation (UV/H₂O₂). The hydroxyl radicals generated by the process removed up to 99% of ABZ, and FBZ, in the highest dosage of H₂O₂ (i.e., 1.125 mmol L⁻¹; 4.8 kJ L⁻¹). In contrast, 20% of initial TBZ concentration remained in the residual solution. In the first 5 min of reaction (i.e., up to 0.750 mmol L⁻¹ of H₂O₂), formation of the primary metabolites of ABZ—ricobendazole (RBZ), albendazole sulfone (ABZ-SO₂), and oxfendazole (OFZ)—was observed. However, these reaction products were converted after the reaction time was doubled. The residual ecotoxicity was investigated using the Raphidocelis subcapitata microalgae and the marine bacteria Vibrio fischeri. The results for both microorganisms evidence that the residual solutions are less harmful to these microorganisms. However, after 30 min of reaction, the treated solution still presents a toxic effect for V. fischeri, meaning that longer reaction times are required to achieve an innocuous effluent.

Investigations of deleterious effects on non-target species, including earthworms, have been conducted for a number of pesticides, but there is a need for additional assessments of potential adverse effects. In the present study, the acute toxicity of eight pesticides to the earthworm Eisenia andrei was assessed and compared. The exposures were conducted using the filter paper contact toxicity method. Based on the 48-h LC₅₀ values, one pesticide was classified as supertoxic (combined fungicide containing difenoconazole and fludioxonil), four as extremely toxic (combined herbicide containing pethoxamide and terbuthylazine, combined fungicide containing fluopyram and tebuconazole, fungicide containing pyrimethanil, and combined fungicide containing thiram and carboxin), two as very toxic (combined fungicide containing flutriafol and thiabendazole, and herbicide containing fluroxypyr-meptyl), and one as moderately toxic (insecticide containing thiamethoxam). Additionally, effects of pesticides on the multixenobiotic resistance (MXR) activity were measured. Results showed that four pesticides caused significant effects with a recorded inhibition of the activity, which can consequently lead to a higher toxicity due to longer retention of the pesticides in the cells. Finally, for three chosen pesticides, gene expression of cat, sod, and gst was measured, and significant changes were observed. The obtained results show that earthworms could be significantly affected by pesticides commonly used in agriculture.

Thiabendazole (TBZ) is an ionizable anthelmintic agent that belongs to the class of benzimidazoles. It is widely used in veterinary medicine and as a fungicide in agriculture. Sorption and desorption are important processes influencing transport, transformation, and bioavailability of xenobiotic compounds in soils; data related to sorption capacity are therefore needed for environmental risk assessments. The aim of this work was to assess the sorption potential of TBZ in four Brazilians soils (sandy, sandy-clay, and clay soils), using batch equilibrium experiments at three pH ranges (2.3–3.0, 3.8–4.2, and 5.5–5.7). The Freundlich sorption coefficient (K F) ranged from 9.0 to 58 μg¹–¹/ⁿ (mL) ¹/ⁿ g⁻¹, with higher values generally observed at the lower pH ranges (2.3–3.0 and 3.8–4.2) and for clay soils. The highest organic carbon-normalized sorption coefficients (KOC) obtained at pH 3.8–5.7 (around the natural pH range of 4.1–5.0) for both clay soils and sandy-clay soil were 3255 and 2015 mL g⁻¹, respectively. The highest correlations K F vs SOM (r = 0.70) and K F vs clay content (r = 0.91) were observed at pH 3.8–4.2. Our results suggest that TBZ sorption/desorption is strongly pH dependent and that its mobility could be higher in the studied soils than previously reported in soils from temperate regions.

Many of South Africa’s freshwater impoundments are compromised by pesticide pollution, and the Roodeplaat Dam, near Pretoria, is no exception. This paper is, to the best of our knowledge, the first report of the herbicides Dacthal, metribuzin, simazine, tebuthiuron, terbuthylazine, and the fungicides azoxystrobin, carbendazim, epoxiconazole, metalaxyl (Ridomil), propiconazole, pyrimethanil and thiabendazole in a South African freshwater impoundment. This short note reports on the screening results of water and muscle tissue samples against a comprehensive library of pesticides, herbicides and fungicides in the polluted Roodeplaat Dam. Muscle samples of Oreochromis mossambicus screened positive for p,p'-DDE and p,p'-DDD and for DCPA (chlorthal-methyl). The muscle tissue of Clarias gariepinus screened positive for p,p'-DDE and p,p'-DDD, chlorpyrifos, trans-chlordane, DCPA and terbuthylazine. The presence of these pesticides, herbicides and fungicides in this impoundment is of great concern as there is substantial evidence of adverse health effects in fish exposed to these chemicals.

The simultaneous adsorption of both imazalil (IMZ) and thiabendazole (TBZ) fungicides in a Cu²⁺-exchanged Mt was studied in this work. Kinetic studies were used to determine the rate law which describes the adsorption of individual fungicides onto the adsorbent. Adsorption isotherm of individual and combined fungicides was done to evaluate synergic or antagonistic effects. The Mt-Cu material considerably improved TBZ and/or IMZ adsorption from aqueous suspensions with respect to raw Mt, leading to removal efficiencies higher than 99% after 10 min of contact time for TBZ and IMZ Cᵢ = 15 and 40 mg/L, respectively, when a solid dosage = 1 g/L was used. The adsorption sites involved were determined by a combination of X-ray diffraction (XRD) determinations and electron paramagnetic resonance (EPR), indicating that fungicides were bonded to Cu²⁺ cations, while the rate limiting step was the formation of coordination bonds. The adsorption mechanism proposed is that of ligand exchange between water and fungicide molecules in the metal coordination sphere. The single-crystal structure for the IMZ-Cu²⁺ complex indicated that four molecules were bounded to the copper centers, while two molecules of TBZ are bounded to copper explaining the higher IMZ uptake capacity for the Mt-Cu material.
