Use of integrated biomarker response for studying the resistance strategy of the earthworm Metaphire californica in Cd-contaminated field soils in Hunan Province, South China النص الكامل

Research was conducted to study the response and detoxification mechanisms of earthworms collected from Cd-contaminated areas in Hunan Province, South China. Metaphire californica, the dominant earthworm species in fields, referred as earthworm-A and -B that collected from low- (0.81 mg kg⁻¹) and high-Cd soil (13.3 mg kg⁻¹), respectively, for exchanging incubation in laboratory. The results showed that earthworm-A gradually accumulated higher Cd when exposed in the high-Cd soil, whereas Cd concentration of earthworm-B decreased after being transferred to low-Cd soil (albeit BAFCd >20). The integrated biomarker response index was calculated with the biomarkers of antioxidant systems (e.g., superoxide dismutase (SOD), peroxidase (POD), glutathione (GSH), glutathione peroxidase (GPx), glutathione-S transferase (GST), and malondialdehyde (MDA)) and energy index (e.g., protein and glycogen) in M. californica. GSH, GPx, and GST contributed the most to the integrated biomarker response (IBR) in earthworm-A when exposed in high-Cd soil for 14 d. Earthworm-B responded with higher GST and GPx activities and decreased protein content in low-Cd soil. For 28 d, the response of earthworm-A was not evident in either low- or high-Cd soil, and the inductive effect of metal stress on earthworm-B tended to be stable, except for the higher MDA content (p < 0.05) when exposed in low-Cd soil. The IBR index of earthworm-B (2.93 and 3.40) in low- and high-Cd soil, respectively, was higher than that of earthworm-A (0.89 and 1.0). Overall, earthworm-A exhibited a detoxification process to resist high-Cd toxicity from low-to high-Cd soil. Earthworm-B exhibited a physiological resilience once its habitat had changed to a normal or low-Cd soil environment, possibly owing to the cost of its resistance adaptation to the historical highly contaminated soil in fields.

Cadmium (Cd) concentration was investigated in parent rocks, surrounding soil of black shales outcrop, stream water, stream sediments, paddy soil as well as rice plants. Leaching test and sequential extraction procedure were applied to evaluate Cd mobility and bioavailability in soil samples. This study aims to emphasize ecological risk of Cd induced by black shales by combining various natural medias in black shales area and control area. The black shales parent rocks have elevated Cd concentration and act as a source of Cd. The liberated Cd from black shales outcrop temporarily accumulated in the acidized surrounding soil and could arise potential adverse impacts on environment due to rainfall. Although high concentration of Cd was not detected in stream water, Cd concentrated stream sediment was a hidden toxin for surface water system. Cd in paddy soil was primarily from geogenic source and effected little by anthropogenic source. The concentration as well as mobility and bioavailability of Cd were high in paddy soil in black shales area, which lead to elevated Cd concentration in roots, shoots and grains of rice. As a result, residents in black shales area suffer increased non-carcinogenic risk of Cd via food chain.

Fructose was utilized as an additional co-substrate to systematically investigate the molecular mechanism of its boosting effect for the degradation of refractory dye reactive black 5 (RB5) by a natural bacterial flora DDMZ1. A decolorizing rate of 98% was measured for sample YE + FRU(200) (with 3 g/L fructose additionally to yeast extract medium, 10% (v/v) inoculation size of flora DDMZ1, 200 mg/L RB5) after 48 h. This result was 21% and 77%, respectively, higher than those of samples with only yeast extract or only fructose. Fructose was found to significantly stimulated both intracellular and extracellular azoreductase secretion causing enhanced activity. Metagenomic sequencing technology was used to analyze the functional potential of genes. A label-free quantitative proteomic approach further confirmed the encoding of functional proteins by the candidate genes. Subsequently, the molecular mechanism of RB5 degradation by candidate genes and functional proteins of the dominant species were proposed. This study provides important perspectives to the molecular mechanism of co-metabolic degradation of refractory pollutants by a natural bacterial flora.

Cadmium (Cd), which is a toxic non-essential heavy metal capable of entering plants and thus the food chain, constitutes a major environmental and health concern worldwide. An understanding of the tools used by plants to overcome Cd stress could lead to the production of food crops with lower Cd uptake capacity and of plants with greater Cd uptake potential for phytoremediation purposes in order to restore soil efficiency in self-sustaining ecosystems. The signalling molecule nitric oxide (NO), whose function remains unclear, has recently been involved in responses to Cd stress. Using different mutants, such as nia1nia2, nox1, argh1-1 and Atnoa1, which were altered in NO metabolism, we analysed various parameters related to reactive oxygen and nitrogen species (ROS/RNS) metabolism and seedling fitness following germination and growth under Cd treatment conditions for seven days. Seedling roots were the most affected, with an increase in ROS and RNS observed in wild type (WT) seedling roots, leading to increased oxidative damage and fitness loss. Mutants that showed lower NO levels in seedling roots under Cd stress were more resistant than WT seedlings due to the maintenance of antioxidant systems which protect against oxidative damage.

The potential risk to human health of antibiotics that pass through the food chain has become an important global issue, but there are few reports on the response of ginger (Zingiber officinale) to antibiotic pollution. In this study, we investigated the enrichment characteristics and biological response of ginger to sulfamethoxazole (SMZ) and ofloxacin (OFL) residues, which are common in the environment. Lower levels of SMZ, OFL and their combined duplex treatment (SMZ+OFL) promoted the growth of ginger, but the critical doses necessary to stimulate growth differed among treatments: 10 mg L⁻¹ SMZ, 1 mg L⁻¹ OFL and 1 mg L⁻¹ (SMZ+OFL) had the strongest stimulating effects. At higher dosages, the root growth and light energy utilization efficiency of ginger were impaired, and (SMZ+OFL) had the strongest inhibitory effect. Treatments with lower levels of antibiotics had no significant effect on reactive oxygen species and antioxidant enzyme activities. However, when SMZ, OFL and SMZ+OFL concentrations exceeded 10 mg L⁻¹, the contents of H₂O₂, O₂⁻ and MDA continued to increase, while the activities of SOD, POD, CAT first increased and then decreased, especially in SMZ+OFL. Ginger accumulated more SMZ and OFL in rhizomes and less in leaves, and accumulation increased significantly as antibiotic concentration increased. When SMZ concentration was 1 mg L⁻¹, the SMZ concentrations in rhizomes, roots, and leaves were 0.23, 0.15, and 0.05 mg kg⁻¹, respectively, and the residual SMZ in the rhizome was 2.3 times higher than the maximum residue limit. The abundance of the resistance genes sul1, sul2, qnrS, and intI1 increased with increasing antibiotic concentrations, and intI1 abundance was the highest. OFL induced higher levels of intI1 expression than did SMZ.

High levels of air pollutants in Vietnam, especially particulate matters including PM₂.₅, can be important risk factors for respiratory diseases among children of the country. However, few studies on the effects of ambient air pollution on human health have been conducted in Vietnam so far. The aim of this study is to examine the association between PM₂.₅ and hospital admission due to acute lower respiratory infection (ALRI) among children aged < 5 years old in Ho Chi Minh city, the largest city of Vietnam. Data relating PM₂.₅ and hospital admission were collected from February 2016–December 2017 and a time series regression analysis was performed to examine the relationship between PM₂.₅ and hospital admission including the delayed effect up to three days prior to the admission. We found that each 10 μg/m³ increase in PM₂.₅ was associated with an increase of 3.51 (95%CI: 0.96–6.12) risk of ALRI admission among children. According to the analysis, male children are more sensitive to exposure to PM₂.₅ than females, while children exposed to PM₂.₅ are more likely to be infected with acute bronchiolitis than with pneumonia. The study demonstrated that young children in HCMC are at increased risk of ALRI admissions due to the high level of PM₂.₅ concentration in the city's ambient air.

Environmental monitoring and surveillance studies of pharmaceuticals routinely examine occurrence of substances without current information on human consumption patterns. We selected 10 streams with diverse annual flows and differentially influenced by population densities to examine surface water occurrence and fish accumulation of select psychoactive medicines, for which consumption is increasing in the Czech Republic. We then tested whether passive sampling can provide a useful surrogate for exposure to these substances through grab sampling, body burdens of young of year fish, and tissue specific accumulation of these psychoactive contaminants. We identified a statistically significant (p < 0.05) relationship between ambient grab samples and passive samplers in these streams when psychoactive contaminants were commonly quantitated by targeted liquid chromatography with tandem mass spectrometry, though we did not observe relationships between passive samplers and tissue specific pharmaceutical accumulation. We further observed smaller lotic systems with elevated contamination when municipal effluent discharges from more highly populated cities contributed a greater extent of instream flows. These findings identify the importance of understanding age and species specific differences in fish uptake, internal disposition, metabolism and elimination of psychoactive drugs across surface water quality gradients.

Black-spotted frogs and bullfrogs from an e-waste polluted area were collected and examined for legacy and emerging organophosphorus flame retardants (PFRs) and plasticizers. Total concentrations of PFRs and plasticizers were 0.62–15 ng/g wet weight (ww) and 316–4904 ng/g ww in muscles, 2.2–59 ng/g ww and 127–5757 ng/g ww in eggs and gonads, and 1.2–15 ng/g ww and 51–1510 ng/g ww in oviducts, respectively. For muscle tissues, concentrations of ∑PFRs, triethyl phosphate, tris-(2-chloroethyl) phosphate, and tris-(chloro-2-propyl) phosphate were significantly higher in the males than females (p < 0.05). However, for reproductive tissues, eggs exhibited higher levels of those contaminants than gonads (p < 0.05). No significant sex difference in levels of plasticizers was observed in muscles. In contrast, levels for (2-ethylhexyl) phthalate, di-n-butyl phthalate, and di-iso-butyl phthalate in gonads were significantly higher than those in eggs (p < 0.05). Significantly negative linear correlations between maternal transfer ratios and log KOW were found in female frogs. Paternal transfer potentials were first significantly and positively correlated to log KOW (<6) and then decreased afterward in the males. These results indicated that parental transfer was answer for the sex-specific accumulation of PFRs and plasticizers in frogs.

Acid mine drainage (AMD) is harmful to the environment and human health. Microorganisms-mineral interactions are responsible for AMD generation but can also remediate AMD contamination. Understanding the microbial response to AMD irrigation will reveal microbial survival strategies and provide approaches for AMD remediation. A terrace with sharp geochemical gradients caused by AMD flooding were selected to study the microbial response to changes in environmental parameters related to AMD contamination. AMD intrusion reduced soil microbial community diversity and further changed phylogenetic clustering patterns along the terrace gradient. We observed several genera seldom reported in AMD-related environments (i.e., Corynebacterium, Ochrobactrum, Natronomonas), suggesting flexible survival strategies such as nitrogen fixation, despite the poor nutritional environment. A co-occurrence network of heavily-contaminated fields was densely connected. The phyla Proteobacteria, Acidobacteria, Chloroflexi, and Euryarchaeota were all highly interconnected members, which may affect the formation of AMD. Detailed microbial response to different soil characterizations were highlighted by random forest model. Results revealed the top three parameters influencing the microbial diversity and interactions were pH, Fe(III), and sulfate. Various acidophilic Fe- and S-metabolizing bacteria were enriched in the lower fields, which were heavily contaminated by AMD, and more neutrophiles prevailed in the less-contaminated upper fields. Many indicator species in the lower fields were identified, including Desulfosporosinus, Thermogymnomonas, Corynebacterium, Shewanella, Acidiphilium, Ochrobactrum, Leptospirillum, and Allobaculum, representing acid-tolerant bacteria community in relevant environment. The detection of one known sulfate-reducing bacteria (i.e., Desulfosporosinus) suggested that biotic sulfate reduction may occur in acidic samples, which offers multiple advantages to AMD contamination treatment. Collectively, results suggested that the geochemical gradients substantially altered the soil microbiota and enriched the relevant microorganisms adapted to the different conditions. These findings provide mechanistic insights into the effects of contamination on the soil microbiota and establish a basis for in situ AMD bioremediation strategies.

In this study, total concentration and inhalation bioaccessibility (dissolution in simulated biological solution) of trace elements (TE) and rare earth elements (REE) were assessed in PM₁₀ from Canadian house dust samples with smoking (n = 25) and non-smoking (n = 25) status. Compared to the natural background concentrations in Canadian soils, median Zn, Pb, Cd and Cu concentrations in PM₁₀ were 10–23 fold higher, while median La, Ce and Pr concentrations were 1.6–2.4 fold higher. Mann-Whitney tests (α = 0.05) indicated no difference between the median TE concentrations based on the smoking status of the household; however, median REE concentrations were significantly higher in the PM₁₀ of smoking households. Additionally, Cd and Ni were positively correlated (Spearman r, p < 0.05) to La, Ce and Nd in smoking households, suggesting that tobacco combustion may have contributed REE in the PM₁₀ of these households. Median inhalation-ingestion bioaccessibility assay outcomes of arsenic (As) and lead (Pb) was higher in the non-smoking households when compared to smoking households (Mann Whitney test, α = 0.05), suggesting that tobacco combustion products may be associated with less soluble species of As and Pb. Although REE bioaccessibility was negligible in simulated lung epithelial fluid regardless of the smoking status of the household, bioaccessibility in the lung-gastric phase was 23.6–27.6% in the smoking household and 34.7–36.7% in the non-smoking households, indicating a significantly lower REE dissolution in PM₁₀ of smoking households. In contrast, between 17 and 21.9% bioaccessibility of REE was observed when artificial lysosomal fluid was used, where the outcome was not significantly affected by the smoking status. This study indicates that despite a higher median REE concentration in the PM₁₀ of smoking households, inhalation bioaccessibility may be significantly influenced by the mineralogy.
