Unexpectedly high concentrations of 2,4-di-tert-butylphenol in human urine
2019
Liu, Runzeng | Mabury, Scott A.
Synthetic phenolic antioxidants (SPAs) have received increasing attention due to the reports of toxicity and environmental contamination. Nevertheless, limited information was available on human burdens of these SPAs, with the exception of 2,6-di-tert-butyl-4-methylphenol (BHT). In our study, BHT as well as six other SPAs were analyzed in human urine samples from United States donors. Three SPA congeners were detected in human urine: BHT, 2,4-di-tert-butylphenol (DBP), and 3-tert-butyl-4-hydroxyanisole (BHA). BHT, which is the congener received most concerns, was detected at low concentrations [geometric mean (GM): 0.06 ng/mL], whereas four of its metabolites were detected at relatively high concentrations (GM: 1.68 ng/mL). Surprisingly, DBP was detected at extremely high concentrations (GM: 18.3 ng/mL). The concentrations of DBP (GM: 25.8 ng/mL), BHT (0.853 ng/mL), and metabolites (GM: 10.5 ng/mL) increased significantly after the urine samples were hydrolyzed by β-glucuronidase (p < 0.01), indicating the prevalence of the conjugated forms of SPAs and their metabolites in human urine. DBP, which has previously received little attention, was the predominant congener, contributing 88.2% and 63.6% to total target concentrations in the urine samples before and after β-glucuronidase hydrolysis, respectively. Thus, previous studies have vastly underestimated the burdens of SPAs to humans. To our knowledge, this is the first study revealing the presence of DBP in human urine.

Although isomer-specific bioaccumulation of dechlorane plus (DP) has been addressed in many studies, it remains unclear which factors determine this process and whether biotransformation of DP occurs in organisms. Comparative experiments were conducted in both in vivo and in ovo incubation using hens and eggs to identify the dominant factors determining the bioaccumulation of DP. Hens and fertilized eggs were exposed to DP isomers (syn- and anti-DP) by feeding and spiking, respectively, to investigate absorption, elimination, and metabolism. No significant differences were found between absorption efficiencies of DP isomers in the adult hens. Following first-order kinetics, anti-DP exhibited a slightly longer half-life than syn-DP as well as an elevated anti-DP fraction in laid eggs, thereby suggesting selective enrichment of anti-DP in adult hens. However, chicken embryos metabolized approximately 12% and 28% of the absorbed syn- and anti-DP, respectively, thereby verifying that anti-DP was preferably metabolized. This result indicated that stereo-selective excretion of syn-DP, rather than preferred metabolism of anti-DP, played a more prominent role in isomer-specific bioaccumulation of DP in chickens. Further studies on metabolites of DP are crucial to understanding the fate of DP in organisms.

The wide adoption of combined heat and power (CHP) can not only improve energy efficiency, but also strengthens energy system resiliency. While CHP reduces overall emissions compared to generating the same amount of electricity and heat separately, its on-site nature also means that CHP facilities operate in populated areas, raising concerns over their near-source air quality impact. Evaluation of the near-source impact of distributed CHP is limited by emission data availability, especially in terms of particulate matter (PM). In this paper, we report on stack emission testing results of a community-scale CHP plant with two natural gas turbine units (15 MW each) from measurements conducted in both 2010 and 2015, and assess the near-source air quality impact using an integrated modeling framework using the stack test results, site-specific meteorological data and terrain profiles with buildings. The NOx removal efficiency by selective catalytic reduction (SCR) is estimated to be ∼83% according to the emission testing. The integrated framework employs AERMOD to screen air quality in a 2.7 km × 2.3 km domain from 2011 to 2015 to identify the highest ground-level concentrations (GLCs). Examining the corresponding meteorological conditions, we find that those high GLCs appeared during the stable atmospheric boundary layer with relative high wind speed. Next, the worse-case scenarios identified from the screening process are simulated using the detailed Unsteady Reynolds Averaged Navier-Stokes (URANS) model coupled with a chemistry solver. The results generally show low GLCs of primary PM₂.₅ for this case study. However, our analysis also suggests greater building downwash impacts with the presence of taller and denser urban structures. Therefore, the near-source impact of natural gas-fired CHP in large metropolitan areas is worthy of further investigation.

Conjunctivitis has hazardous effects on patients’ quality of life through influencing school performance, work productivity, and daily activities such as driving. However, limited evidence is available on the contributory role of air pollution on conjunctivitis, particularly in China. We obtained data of 81,351 conjunctivitis outpatients from the largest comprehensive hospitals of four cities, China, between Jan 1, 2013 and Dec 31, 2014. Data on air pollutants, including particulate matter ≤2.5 μm in diameter (PM₂.₅), particulate matter ≤10 μm in diameter (PM₁₀), nitrogen dioxide (NO₂), ozone (O₃) and sulphur dioxide (SO₂) were collected from China National Environmental Monitoring Centre. Conjunctivitis outpatient visits were linked with air pollution concentrations by the visiting dates. A time-stratified case-crossover design with conditional logistic regression model was used to examine the effect of short-term exposure to air pollution on conjunctivitis outpatient visits. We found that the associations between air pollutants (per 10 μg/m³ increase) and hospital outpatient visits for asthma were [odds ratios (ORs) and 95% confidence intervals]: 1.004(1.002–1.007) for PM₂.₅, 1.004 (1.002–1.005) for PM₁₀, 1.012(1.005–1.020) for NO₂, 1.006 (1.001–1.011) for SO₂, and 1.007 (1.003–1.010) for O₃, respectively at lag0 day. Outpatients aged 35–64 years showed significant associations with exposure to PM₂.₅ (1.005, 1.001–1.010), PM₁₀ (1.005, 1.002–1.008), NO₂ (1.014, 1.003–1.026), and O₃ (1.005, 1.000–1.011), while those aged 15–34 years showed significant associations with exposure to O₃ (1.010, 1.004–1.017).Short-term exposure to air pollution has impacts on outpatient visits for conjunctivitis in China. This study suggests that improving air quality could protect eye health.

Growth of textile industries led to production of enormous dye varieties. These textile dyes are largely used, chemically stable and easy to synthesize. But they are recalcitrant and persist as less biodegradable pollutants when discharged into waterbodies. Potential use of enzyme-linked bioremediation of textile dyes will control their toxicity in waterbodies. Bioinformatics and Molecular docking tool provides an insight into remediation mechanism by predicting susceptibility of dye degradation using oxidoreductive enzymes. In this study, six dyes, Reactive Red F3B, Remazol Red RGB, Joyfix Red RB, Joyfix Yellow MR, Remazol Blue RGB and Turquoise CL-5B of azo, anthraquinone and phthalocyanine molecular class were identified as potential targets for degradation by laccase and azoreductase of Aeromonas hydrophila in addition to Lysinibacillus sphaericus through in silico docking tool BioSolveIT-FlexX. Azoreductase breaks azo bonds by ping-pong mechanism whereas laccase decolorizes dyes by free radical mechanism which is not specific in nature. Results were analyzed based on parameters like stability, catalytic action and selectivity for enzyme-dye interactions. Amino acids of enzymes interacted with several dyes substantiating variations in active site for enzyme-ligand binding affinity. This suggests the role of enzymes in decolorizing an extensive variety of textile dyes, thereby, aiding in understanding the enzyme mechanisms in Bioremediation.

Keeping in view the expanding environmental pollution and irrigation water deficit, a pot experiment was performed for the upland (Huyou2, Hanyou737) and paddy rice cultivars (Taigeng8; Yixiang2292), to study soil liming effects on methane (CH₄) and nitrous oxide (N₂O) emission, bioavailability and accumulation of Cd, Pb in upland and paddy rice. Upland rice reduced 90% of soil CH₄ emission as compared to paddy conditions. Soil CH₄ emission decreased by 45% and 39% with dolomite, and it reduced by 35% and 33% with lime treatment both in upland and paddy conditions, respectively. Soil N₂O emission decreased by 44% and 52% with dolomite, and with the lime application, it was reduced by 37% and 44% for both upland and paddy conditions respectively. Reduction in soil DTPA-extractable Cd was between 37-53% and 43–80% with dolomite and 16–37% and 24–72% Cd decreased with lime application in upland and paddy conditions respectively. Soil DTPA-extractable Pb reduced by 27–44% and 25–53% with dolomite and 16–40% and 11–42% with soil-applied lime in upland and paddy conditions, respectively. Cd accumulation in rice grain was decreased by 47–88% and 62–79% with dolomite and 31–86% and 45–52% reduction by lime application in upland and paddy rice respectively. Rice grain Pb reduced by 58–91% and 66–78% with dolomite application and 32–71% and 44–71% with lime in upland and paddy rice, respectively. Our results showed that soil liming significantly reduced soil N₂O and CH₄ emission and Cd, Pb accumulation in rice grain, but dolomite was more effective as compared to lime. Altogether, results of this study suggest that upland rice can be cultivated in CdPb polluted soils with least soil CH₄ emission. Cd and Pb toxicity, accumulation, and N₂O emission in upland rice can be minimized by soil liming of 3 g kg⁻¹ and optimizing the nutrients composition of the soil.

2019
Tongue, Andrew D.W. | Reynolds, S James | Fernie, Kim J. | Harrad, Stuart
Given factors such as their persistence and toxicity, legacy brominated flame retardants (BFRs) like polybrominated diphenyl ethers (PBDEs) and hexabromocyclododecane (HBCDD), are designated as persistent organic pollutants (POPs) and are subject to regulation. Waste streams likely represent a substantial reservoir of legacy BFRs given that they were once widely applied to goods which are increasingly likely to be obsolete. Waste streams are also increasingly likely to be a source of emerging flame retardants, in particular, novel BFRs (NBFRs), the halogenated norbornene flame retardant Dechlorane Plus (DDC-CO) and the brominated, chlorinated or non-halogenated organophosphate triester flame retardants (PFRs). Many bird populations rely on landfill and its surrounding land-use for inter alia the opportunities it provides for activities such as foraging and resting. However, studies on captive and wild (free-living) birds have demonstrated deleterious effects of several FRs. Globally, approximately 250 bird species, including many of conservation concern, are reported to use landfill and surrounding habitat (including wastewater treatment operations), thus putting birds potentially at risk of exposure to such chemicals. We synthesise and critically evaluate a total of 18 studies covering eight avian species published between 2008 and 2018 (inclusive) across four continents that report flame retardant (FR) burdens in birds utilising landfill. Several such studies found FRs at among the highest concentrations detected in wild biota to date. We recommend that ongoing research be focused on landfill-associated birds, given that landfill is an important source of FRs and other anthropogenic chemicals, and particularly at sites where species are of conservation concern. We suggest ways in which the comparative power of studies could be enhanced in the future, the reporting of a minimum common suite of key chemicals, and where feasible, standardisation of the tissue compartments (i.e., eggs) to be studied. We conclude by identifying future research directions.

Employing biochar for environmental remediation has been widely practiced. Nonetheless, the reduction mechanisms of hexavalent chromium (Cr(VI)) in the presence of biochar have not been fully elucidated (i.e., direct or indirect reduction of Cr(VI) by biochar). In particular, the effect of light on Cr(VI) reduction by biochar was rarely reported. Thus, to clarify the reduction mechanisms of Cr(VI) by biochar at the fundamental level, this study laid great emphasis on the photo-induced reduction of Cr(VI) in the application of biochar. Biochar releases dissolved organic matter (DOM), the DOM can extract Fe(III) from soil by complexation, and the complexes can be photo-reacted under the light. In these respects, Fe(II) formed by the photo-induced reaction of DOM-Fe(III) was particularly evaluated in this study. To evaluate that, three biomass samples (rice straw, granular sludge from an up-flow anaerobic sludge blanket, and spent coffee ground) were torrefied to biochar. To circumvent the adsorption of Cr(VI) onto biochar, biochar extractives (served as a source for DOM) and Fe(III) solution were tested with/without UV light to prove Fe(II) formation. This study experimentally proved that the more Fe(II) under the UV radiation was formed in the co-existence with biochar extractives and Fe(III). All experimental data from three biochar samples were indeed very similar. Cr(VI) reduction by Fe(II) from GB, RB, and CB reached up to 96, 79, and 100%, respectively. The different reduction efficiency signified that the low molecular weight of organic acids, such as oxalate, were more sensitive to the UV light, thereby resulting in the enhanced Fe(II) formation. Such Fe(II) formation subsequently led to the high reduction efficiency of Cr(VI).

Hu, Xue Lei | Niu, Jiao Jiao | Meng, Qi | Chai, Yuet Hung | Chu, Ka Hou | Chan, King Ming
Juvenile hormone analogue (JHA) insecticides are endocrine disrupters that interfere with hormonal action in insects by mimicking their juvenile hormones (JH). As the structure and functions of methyl farnesoate in crustaceans are similar to those of JH in insects, exogenous JHA insecticides could have adverse effects on the development and reproduction of crustaceans. This study examined the toxic effects of two JHA insecticides, fenoxycarb and methoprene, on a freshwater shrimp model of cherry shrimp, Neocaridina davidi. Both insecticides had detrimental effects on cherry shrimp, but fenoxycarb was more toxic than methoprene. Chronic exposure to these insecticides reduced the shrimp's body length and molting frequency. Based on transcriptome annotations for N. davidi, we identified important gene homologues that were active in both insect JH biosynthetic and degradative pathways as well as JH and ecdysteroid signaling pathways. Chronic treatments with JHAs had significant effects on these genes in N. davidi. Our transcriptomic analysis showed that genes involved in the pathways related to cuticle development, serine protease activity, and carbohydrate, peptide and lipid metabolic processes were differentially expressed in shrimp exposed to JHAs. These results demonstrate the toxicity of fenoxycarb and methoprene to freshwater crustaceans and indicate the need to monitor the use of JHA insecticides.

Microplastics (MPs), are tiny plastic fragments from 1 μm to 5 mm generally found in the aquatic environment which can be easily ingested by organisms and may cause chronic physical but also toxicological effects. Toxicological assays on fish cell lines are commonly used as an alternative tool to provide fast and reliable assessment of the toxic and ecotoxic properties of chemicals or mixtures. Rainbow trout liver cell line (RTLW-1) was used to evaluate the toxicity of pollutants sorbed to MPs sampled in sandy beaches from different islands around the world during the first Race for Water Odyssey in 2015. The collected MPs were analyzed for polymer composition and associated persistent organic pollutants: polycyclic aromatic hydrocarbons (PAHs), polychlorobiphenyls (PCBs) and dichlorodiphenyltrichloroethane (DDT). In addition, DMSO-extracts from virgin MPs, MPs artificially coated with B[a]P and environmental MPs were analyzed with different bioassays: MTT reduction assay (MTT), ethoxyresorufin-O-deethylase (EROD) assay and comet assay. Microplastics from sand beaches were dominated by polyethylene, followed by polypropylene fragments with variable proportions. Organic pollutants found on plastic from beach sampling was PAHs (2–71 ng g⁻¹). Samples from Bermuda (Somerset Long Bay) and Hawaii (Makapu'u) showed the highest concentration of PAHs and DDT respectively. No toxicity was observed for virgin microplastics. No cytotoxicity was observed on cells exposed to MP extract. However, EROD activity was induced and differently modulated depending on the MPs locations suggesting presence of different pollutants or additives in extract. DNA damage was observed after exposure to four microplastics samples on the six tested. Modification of EROD activity level and DNA damage rate highlight MPs extract toxicity on fish cell line.
