Microplastics are ubiquitous in the environment, with high concentrations being detected now also in river corridors and sediments globally. Whilst there has been increasing field evidence of microplastics accumulation in the guts and tissues of freshwater and marine aquatic species, the uptake mechanisms of microplastics into freshwater food webs, and the physical and geological controls on pathway-specific exposures to microplastics, are not well understood. This knowledge gap is hampering the assessment of exposure risks, and potential ecotoxicological and public health impacts from microplastics. This review provides a comprehensive synthesis of key research challenges in analysing the environmental fate and transport of microplastics in freshwater ecosystems, including the identification of hydrological, sedimentological and particle property controls on microplastic accumulation in aquatic ecosystems. This mechanistic analysis outlines the dominant pathways for exposure to microplastics in freshwater ecosystems and identifies potentially critical uptake mechanisms and entry pathways for microplastics and associated contaminants into aquatic food webs as well as their risk to accumulate and biomagnify. We identify seven key research challenges that, if overcome, will permit the advancement beyond current conceptual limitations and provide the mechanistic process understanding required to assess microplastic exposure, uptake, hazard, and overall risk to aquatic systems and humans, and provide key insights into the priority impact pathways in freshwater ecosystems to support environmental management decision making.

The aim of our study was to evaluate the water quality of an urban stream in southeastern Brazil by analyzing epibenthic ciliates, and to investigate the existence of phylogenetic signal for saprobity in ciliates. However, before conducting this type of phylogenetic study, it is necessary to evaluate if the saprobic classification used frequently in the northern Hemisphere is suitable for neotropical ecosystems. Sediment samples were collected from five sampling stations: two in rural areas and three in urban areas. During the one-year study, with monthly collections, 39 ciliates species were found, of which 32 are included in the saprobic system. Physical, chemical and biological parameters of water confirm the spatial heterogeneity of the sampling stations, with a clear influence of organic pollution on the composition and structure of ciliates taxocenosis. The saprobic index and the saprobic valence index were used to evaluate the water quality of the sampling stations and demonstrated clear heterogeneity between the stations and high degree of pollution of the urban area. These sampling stations were dominated by ciliates indicators of polysaprobric environments. Since we were able to successfully use the saprobic index in a limnic ecosystem in Brazil, we applied the phylogenetic signal validation as a tool for saprobity prediction of the limnic ciliate species not yet analyzed. A phylogenetic tree containing only 18S-rDNA nominal sequences of freshwater ciliates was estimated and used to explore the existence of the phylogenetic signal, which showed that the sensitivity/tolerance of ciliates to organic pollution reflected evolutionary divergence. The results confirm the existence of phylogenetic signal for the saprobrity in Ciliophora. Also, our results suggest that evolutionary analysis is a potential method to predict lineages of ciliates not yet classified for saprobity.

Joo, Junghee | Kwon, Eilhann E. | Lee, Jechan
Many aspects of modern life of our civilization are associated with using electrical and electronic devices (EEE). Ever-increasing demand for high-performance EEE and accelerated technological development make the replacement of EEE become frequent. This leads to the generation of a tremendous amount of electronic waste (E-waste). Challenges of the management of E-waste have recently arisen out of a dearth of proper technologies to treat E-waste. Pyrolysis process can thermochemically treat waste materials that have a complicated nature and inhomogeneity. This article gives a systematic review as an effort to tackle the challenges in the context of achievements in pyrolysis process in E-waste management sector. Pyrolysis mechanism and types of pyrolysis processes and pyrolysis reactors are first discussed. Various pyrolysis technologies applied to the E-waste treatment are then summarized and compared to each other. Points to be considered for further research and pending challenges of E-waste pyrolysis are also discussed. The pyrolysis treatment of E-waste is not yet fully industrialized mostly because of high costs. However, there should be much room for further developing the E-waste pyrolysis; hence, its industrialization and commercialization is just a matter of time.

Traffic-related air pollutants are major contributors to deteriorating urban air quality and pose a serious threat to pedestrians. From both a scientific and a regulatory standpoint, it is important and challenging to understand the contributions of local and non-local sources to accurately apportion specific sources such as traffic emissions contribution to on-road and near-road microenvironment air quality. In this study, we deployed mobile sensors on-board buses to monitor NO, NO₂, CO and PM₂.₅ along ten important routes in Hong Kong. The measurements include two seasons: April 2017 and July 2017. Two types of baseline extraction methods were evaluated and applied to separate local and background concentrations. The results show NO and NO₂ are locally dominated air pollutants in spring, constituting 72%–84% and 58%–71%, respectively, with large inter-road variation. PM₂.₅ and CO largely arise from background sources, which contribute 55%–65% and 73%–79% respectively. PM₂.₅ displays a homogeneous spatial pattern, and the contributions show seasonal change, decreasing during summer. Regional transport pollution is the primary contributor during high pollution episodes. Isolated vehicle plumes show highly skewed concentration distributions. There are characteristic polluted segments on routes and they are most evident at rush hours. The most polluted road segments (top 10%) cluster at tunnel entrances and congested points. Some of these polluted locations were observed in Hong Kong's Low Emission Zones and suggest limitations to the existing control strategies, which only address larger buses. Our work gives new insights in the importance of regional cooperation to improve background air pollution combined with local control strategies to improve roadside air quality in Hong Kong.

Long-term exposure to air pollutants, especially particulates, in adulthood is related to cardiovascular diseases and vascular markers of atherosclerosis. However, whether vascular changes in children is related to exposure to air pollutants remains unknown. This study examined whether childhood exposure to air pollutants was related to a marker of cardiovascular risk, carotid intima–media thickness (CIMT) in children aged 11–12 years old. Longitudinal Study of Australian Children (LSAC) recruited parents and their children born in 2003–4. Among the participants, CheckPoint examination was conducted when the children were 11–12 years old. Ultrasound of the right carotid artery was performed using standardized protocols. Average and maximum far-wall CIMT, carotid artery distensibility, and elasticity were quantified using semiautomated software. Annual and life-time exposure to air pollutants was estimated using satellite-based land-use regression by residential postcodes. A total of 1063 children (50.4% girls) with CIMT data, serum cholesterol, and modeled estimates of NO₂ and PM₂.₅ exposure for the period 2003 to 2015 were included. The average and maximum CIMT, carotid distensibility, and elasticity were 497 μm (standard deviation, SD 58), 580 μm (SD 44), 17.4% (SD 3.2), and 0.48%/mmHg (SD 0.09), respectively. The life-time average concentrations of PM₂.₅ and NO₂ were 6.4 μg/m³ (SD 1.4) and 6.4 ppb (SD 2.4), respectively. Both average and maximum CIMT were significantly associated with average ambient PM₂.₅ concentration (average CIMT: +5.5 μm per μg/m³, 95% confidence interval, CI 2.4 to 8.5, and maximum CIMT: +4.9 μm per μg/m³, CI 2.3 to 7.6), estimated using linear regression, adjusting for potential confounders. CIMT was not significantly related to NO₂ exposure. Carotid artery diameter, distensibility, and elasticity were not significantly associated with air pollutants. We conclude that life-time exposure to low levels of PM₂.₅ in children might have measurable adverse impacts on vascular structure by age 11–12 years.

This research explored the occurrence, epigenetic toxic profiling and main toxic pollutants of POPs in surface water of Dongjiang River, southern China. The concentrations of selected POPs including polycyclic aromatic hydrocarbons (PAHs), endocrine disrupting chemicals (EDCs), phthalate esters (PAEs) and polybrominated diphenyl ethers (PBDEs) of surface water from 18 sites were investigated. ∑₁₆PAHs and ∑₄EDCs were at a moderate level, while ∑₆PAEs and ∑₆PBDEs had low pollution levels. PAHs, EDCs and PAEs showed higher concentrations in dry season than those in wet season, and the loading of selected POPs in tributaries was higher than those in mainstream due to intensive manufactures and lower runoff volume. Moreover, activities of DNA methyltransferase (DNMT)1, histone deacetylase (HDAC2, HDAC8) were confirmed to be sensitive indicators for epigenetic toxicity. The DNMT1-mediated epigenetic equivalency toxicity of organic extracts in Dongjiang River were more serious than those of HDAC2 and HDAC8. Correlation analysis shown binding affinity between POPs and DNMT1, HDAC2 and HDAC8 could be regarded as toxic equivalency factors. Risk assessment suggested that 4-nonylphenol and bisphenol A were the largest contributors to epigenetic risk. This study is the first attempt to quantify epigenetic toxicity and epigenetic risk evaluation of river water.

Cadmium (Cd) is a toxic heavy metal that can be discharged into water environment through industrial activities, threatening the health of aquatic organisms and humans. MicroRNA (miRNA) plays an important role in the process of autophagy. The purpose of this experiment was to study the mechanism of Cd-induced autophagy in common carp hepatopancreas. We established a Cd poisoning model of common carp and explored ultrastructure, two oxidation indicators, three antioxidant indicators, miR-25-3p, two heat shock proteins (Hsps), and nine autophagy-related genes. The results confirmed that deleterious effect of Cd caused the injury of hepatopancreas and the appearance of hepatopancreas autophagic cells in common carp. At the same time, Cd exposure increased the contents of hydrogen peroxide (H₂O₂) and malonaldehyde (MDA), and decreased the activities of catalase (CAT), superoxide dismutase (SOD), and total antioxidative capacity (T-AOC), meaning that Cd caused oxidative stress via the imbalance between peroxide level and antioxidant capacity. Moreover, exposure to Cd increased mRNA expression of microtubule associated protein-1 light chain 3 beta (LC3-II), Dynein, Beclin 1, autophagy-related gene 5 (Atg5), and autophagy-related gene 12 (Atg12); and decreased mRNA expression of mechanistic target of rapamycin kinase (mTOR), indicating that excess Cd caused autophagy, and AMPK/mTOR/ULK1 signaling pathway took part in autophagy induced by Cd in common carp hepatopancreas. Furthermore, Cd down-regulated miR-25-3p and up-regulated its three target genes (AMPK, ULK1 as well as PTEN), suggesting that miR-25-3p mediated autophagy induced by Cd. In addition, we found that Hsps were activated via the up-regulation of Hsp70 and Hsp90. Moreover, oxidative stress mediated autophagy via Hsps in Cd-treated common carp hepatopancreas and Cd-induced autophagy was time dependent. In summary, miR-25-3p, oxidative stress, and Hsps participated in autophagy caused by Cd in common carp hepatopancreas. This study provided a new idea for the mechanism of Cd-induced autophagy in hepatopancreas.

2021
Morel, Elise | Cui, Lei | Zerges, William | Wilkinson, Kevin J.
In order to better understand the environmental risks of the rare earth elements (REEs), it is necessary to determine their fate and biological effects under environmentally relevant conditions (e.g. at low concentrations, REE mixtures). Here, the unicellular freshwater microalga, Chlamydomonas reinhardtii, was exposed for 2 h to one of three soluble REEs (Ce, Tm, Y) salts at 0.5 μM or to an equimolar mixture of these REEs. RNA sequencing revealed common biological effects among the REEs. Known functions of the differentially expressed genes support effects of REEs on protein processing in the endoplasmic reticulum, phosphate transport and the homeostasis of Fe and Ca. The only stress response detected was related to protein misfolding in the endoplasmic reticulum. When the REEs were applied as a mixture, antagonistic effects were overwhelmingly observed with transcriptomic results suggesting that the REEs were initially competing with each other for bio-uptake. Metal biouptake results were consistent with this interpretation. These results suggest that the approach of government agencies to regulate the REEs using biological effects data from single metal exposures may be a largely conservative approach.

Despite Australia's high reliance on coal for electricity generation, no study has addressed the extent to which mercury (Hg) deposition has increased since the commissioning of coal-fired power plants. We present stratigraphic data from lake sediments in the Hunter Valley (New South Wales) and Latrobe Valley (Victoria), where a significant proportion of Australia's electricity is generated via coal combustion. Mercury deposition in lake sediments increased in the 1970s with the commissioning of coal-fired power plants, by a factor of 2.9-times in sediments of Lake Glenbawn (Hunter Valley) and 14-times in Traralgon Reservoir (Latrobe Valley). Sediments deposited after the commissioning of power plants have distinct Hg isotope compositions, similar to those of combusted coals. Mercury emission, estimated using an atmospheric model (CALPUFF), was higher in the Latrobe Valley than in the Hunter Valley. This is a result of higher Hg concentrations in lignite coal, lax regulation and older pollution-control technologies adopted by coal-fired power plants in the Latrobe Valley. Near-source deposition of Hg in Australia is significantly higher than North America and Europe, where better emission controls (e.g. wet flue gas desulfurization) have been in effect for decades. The challenge for Australia in years to come will be to ratify the Minamata Convention and develop better regulation policies to reduce Hg emissions.
