Seasonal patterns of the bacterioplankton community composition in a lake threatened by a pesticide disposal site النص الكامل

Background aim and scope The objective of the study was to determine the effects of ca. 35 years of pesticide contamination (pesticide dump--PD) of Lake Szeląg Wielki (located in the north-eastern Poland) on changes in the microbial communities of aquatic ecosystems. In the years 2008-2009, analyses were carried out for seasonal changes in the quantity and composition of bacterioplankton in the lake examined, which is of high significance to the tourism and fishing industries and is located in the vicinity of an area subjected to reclamation after a pesticide dump. Methods Bacterioplankton composition was assayed by fluorescence in situ hybridisation technique for the contribution of major groups of the Bacteria domain: ά-, β- and γ-Proteobacteria, Cytophaga-Flavobacterium and Actinobacteria as well as bacteria capable of degrading pesticides in an aquatic environment--Pseudomonas spp. Seasonal patterns of the total number of bacteria were determined by direct counting of 4′,6-diamidino-2-phenylindole (DAPI)-stained cells. Results The percentage of the detected Eubacteria (EUB 338 probe) relative to all the DAPI-stained bacteria in Lake Szeląg Wielki ranged from 46% to 63%. Bacteria capable of degrading pesticides in an aquatic environment--Pseudomonas spp.--were identified with a highly specific probe PEA 998. The highest mean values of this parameter reached 5.1%. In the spring, Pseudomonas spp. bacteria accounted for up to 80% of all Gamma-Proteobacteria microbes. Conclusion The study showed that the qualitative and quantitative changes in the bacterioplankton of the lake can be characterised by tendencies which are typical of a eutrophic water reservoir. However, a higher contribution of microorganisms capable of degrading sparingly degradable, toxic compounds and pesticides was determined in bacterioplankton from the PD-contaminated lake, as compared to microbial communities of a lake not contaminated with pesticides.

Introduction Balya and its associated villages which is a town of the Balikesir region of Turkey have very rich zinc, lead, and manganese mines. These mines have been operating since the thirteenth century and now there is heavy metal contamination in both the soil and natural waters in these areas. Materials and methods Soils were collected from Sarı su, Enverpaşa, and Hastanetepe which are in Balya town and Kadıköy, Kaşıkcı, Müstecap, Patlak, Çakallar, and Bengiler which are the villages near Balya and the mine areas. Nine trace analytes (As, Ba, Cd, Cr, Cu, Mn, Ni, Pb, and Zn) were determined using an acid extraction procedure as well as from PBET in vitro gastro-intestinal experiments using ICP-OES. Results and discussion The results showed that high As, Ba, Pb, Zn, and Cd concentrations were found in these soils. The amounts ingested by pica behavior of children at the rate of 10 g day⁻¹ are calculated using the results of in vitro intestinal bio-accessibility experiments. Conclusion The results showed that the amount of As, Pb, Ba, and Cd levels ingested by pica behavior are substantially higher than tolerable daily intake values in most of the soils. When normal ingestion is taken into account, the tolerable daily limits are only exceeded for one element (Pb) and even then, only at two sites.

BACKGROUND, AIM, AND SCOPE: Topographical and meteorological factors, such as monsoons, can drastically affect ambient dioxin levels. In order to better understand these phenomena, four sampling lines (comprising 16 sampling sites) were selected in accordance with the topographical characteristics and the transport paths of the Taichung Basin. MATERIALS AND METHODS: The ambient air samples were simultaneously collected according to US EPA TO-9A. Seventeen polychlorinated dibenzo-p-dioxins and dibenzofurans (PCDD/Fs) were analyzed by a high-resolution gas chromatograph/high-resolution mass spectrometer. RESULTS: Field measurements indicated that, during monsoons, PCDD/Fs concentrations varied from 128 to 4,230 fg m−3. Two locations, the pass between two tablelands and the valley in front of a mountain, had the highest PCDD/Fs concentrations; those concentrations were about two to five times greater than the average concentration. There were 189 analyses of 11 kinds of PCDD/Fs emission sources used to develop emission fingerprints for a CMB8.2 simulation. Chemical mass balance (CMB8.2) model simulation with PCDD/Fs and surface wind field measurements were performed. Results of CMB8.2 modeling indicated that the major source of most sites was secondary copper smelting. CONCLUSIONS: The CMB8.2 model indicated the effects of hills and river valleys on pollutant transport. Results indicated that secondary copper smelting must be controlled to reduce the ambient concentrations of PCDD/Fs in the Taichung Basin.

BACKGROUND, AIM, AND SCOPE: Fishery wastewater treatment can be compromised due to seasonal production. The use of sequencing batch reactors is not completely successful, despite flexibility being one of the principal advantages. Most research on activated sludge is performed using synthetic wastewater to ensure a stable and constant feed. The current work compared biomass morphology and settling ability using image analysis of synthetic and real fishery wastewaters, with and without NaCl addition. RESULTS: The final effluent presented higher turbidity for both wastewaters after NaCl addition, and lower SVI values. For synthetic wastewater, NaCl addition led to the total aggregates' area (TA) increase from 1.46 to 2.09 mm2/μL, alongside the growth of intermediate aggregates into larger aggregates. The addition of NaCl to the fishery wastewater led to a decrease of the TA from 4.43 to 1.72 mm2/μL. The biomass composition decreased in larger and intermediate structures, opposite to the smaller aggregates' area percentage increase. DISCUSSION: NaCl addition to synthetic wastewater incited flocculation increasing sludge settling ability. A slight aggregate disruption was responsible for a turbidity increase. A strong deflocculation was identified in fishery wastewater with NaCl from the decrease of intermediate and large aggregates. This contrasted with pinpoint flocs release, which increased the turbidity levels. CONCLUSIONS, RECOMMENDATIONS, AND PERSPECTIVES: It could be established that synthetic wastewater biomass flocculation and fishery wastewater biomass deflocculation, observed during 0.5% NaCl experiments, were related to sludge settling and effluent turbidity changes. Furthermore, the biomass changes obtained with synthetic wastewater cannot be extrapolated to fishery wastewater.

Introduction Inorganic ion concentrations in event-based wet-only precipitation samples collected during the south-west (SW) monsoon at an urban location in Western India, Ahmedabad between July 2000 and September 2002 were measured by Rastogi and Sarin (2007). Methods For the first time at a location in India, an advanced factor analysis model was retrospectively applied to the measured concentrations of ions (Rastogi and Sarin 2007) in precipitation for source apportionment. Positive matrix factorization resolved five factors, including crustal material, sea salt, nitrate/sulfate-rich factor, ammonium-rich factor, and free acidity. Results and discussion Amongst the model-resolved factors, crustal material was the highest contributor to the total dissolved solids (TDS) accounting for 44.1% on average. Potential source contribution function (PSCF) analysis identified source locations along the eastern coast of Somalia, Yemen, Oman, and the United Arab Emirates for this factor. Sea salt was the second highest contributor accounting for 29.8%. The potential source regions of this factor were also identified in the Arabian Sea and the southern Indian Ocean along the coast of Africa, and the Arabian Gulf. This study also examined the spatial relationships between the source locations of chemical species in precipitation and in ambient aerosol (resolved in an earlier study). Conclusions Crustal material was the highest contributor to TDS at the study location. Spatial relationships between aerosol and precipitation factor source regions suggested that below-cloud scavenging of aerosol particles was a likely contributor to the chemical species apportioned to various precipitation factors. Additionally, source types of chemical species in precipitation resolved in this study were qualitatively compared with those identified at other locations in India. The comparison showed that soil was an important contributor to the dissolved mass of chemical species in precipitation at all locations in India.

INTRODUCTION: In this study, we provide strategies for detecting and quantifying the structural isomers of polyfluorinated di- and tri-alkyl surfactants (PFAS) by mass spectrometry (MS). We specifically investigate polyfluorinated dialkylated phosphate ester surfactants (x:2/y:2 diPAPS, (F(CF2) x CH2CH2O-P(O)(O)−-OCH2CH2(CF2) y F)) and their thioether analogues (x:2/y:2 S-diPAPS, F(CF2) x CH2CH2SCH2-C[CH2O)2P(O)(O)−]-CH2SCH2CH2(CF2) y F), which are used for industrial applications, such as oil- and water-repellent coatings on paper and board. DiPAPS have been found in human blood and are metabolised to the persistent perfluoroalkyl carboxylic acids (PFCA) in rats. MATERIALS AND METHODS: A microwave popcorn bag extract was analysed by ultrahigh-pressure liquid chromatography coupled to a negative electrospray ionisation-quadrupole time-of-flight MS. RESULTS AND DISCUSSION: The extract contained S-diPAPS, diPAPS and trialkylated (triPAPS) impurities. TriPAPS were also present in industrial and synthetic diPAPS standards, and were verified with an 8:2/8:2/8:2 triPAPS standard. The eight elemental compositions (m/z’s) of diPAPS in the extract represent 19 precursor ion structures, and the six S-diPAPS m/z’s represent at least 13 structures. The diPAPS had [M-H]− precursor ions of m/z 789, 889,…1,489 and the S-diPAPS of m/z 921, 1,021,…1,421, corresponding to fluorinated chains from C6–18. Each m/z appeared as one to three chromatographic peaks of structural isomers, where, e.g. m/z 1,189 was present as 10:2/10:2, 8:2/12:2 and 6:2/14:2 diPAPS. The isomers formed different products ions, thus only half of the m/z 1,189 diPAPS concentration was measured with one precursor ion > product ion transition. CONCLUSION: In general, knowledge about structural isomers of poly-alkylated PFAS is needed for the estimation of types and amounts of perfluorinated degradation products, such as PFCA from diPAPS.

INTRODUCTION: This study aimed to analyze antioxidant responses and oxidative damage induced by two inorganic forms of arsenic (As; AsIII and AsV) in an estuarine polychaete species, Laeonereis acuta (Nereididae). The capacity of arsenic biotransformation was also evaluated through the methylation process considering the activity of a key enzyme involved in the metabolization process. MATERIALS AND METHODS: Worms were exposed to 50 μg (AsIII or AsV)/l during 2 or 7 days, plus a control group. Endpoints analyzed included concentration of reactive oxygen species (ROS), activities of antioxidant enzymes such as glutathione reductase (GR), total glutathione-S-transferase (GST), and omega isoform (GST Ω), glucose-6-phosphate deshydrogenase (G6PDH), levels of the antioxidant glutathione (GSH), and lipid peroxides concentration (TBARS). RESULTS AND DISCUSSION: Results showed: (1) GR inhibition after 2-day exposure to both As forms (p < 0.05); (2) GST Ω inhibition after 7-day exposure to AsIII paralleled by an increase in total GST activity (p < 0.05); (3) augmented G6PDH activity after 7-day exposure to both As forms (p < 0.05); (4) no differences in terms of ROS and TBARS; and (5) inhibition of GST Ω activity in AsIII exposed worms, which was concomitant with a lowering of mono- and dymethylated arsenic species. CONCLUSION: These results confirm the reactivity of some biochemical variables of L. acuta to As and indicates its importance as a sentinel species in estuarine regions with presence of arsenic.

INTRODUCTION: This study investigated the effect of copper on chemical oxygen demand (COD) removal efficiency and on the properties (mainly settling and dewatering) and the composition of extracellular polymeric substances (EPS) when 20 mg/L Cu(II) was continuously dosed to a sequencing batch reactor (SBR) inoculated with activated sludge. MATERIALS AND METHODS: The results showed that the continuous addition of 20 mg/L Cu(II) seriously inhibited the removal of sodium benzoate (provided as a model organic pollutant) by activated sludge in a SBR. RESULTS AND DISCUSSION: After 40 days of acclimation, the removal efficiency presented a slight but unsteady recovery and the settling and dewatering properties improved, indicating that sludge bulking had been inhibited. Additionally, the proportion of loosely bound EPS in the total EPS increased with time and the relative composition of the total organics was polysaccharide > humic substances > protein > DNA. CONCLUSION: The effects of copper on the composition of EPS and the settling and dewatering properties of bulking activated sludge were also discussed for the first time in this paper.

INTRODUCTION: The influence of sintering temperature on the physico-mechanical characteristics (such as water absorption, apparent porosity, bulk density, weight loss on ignition, firing shrinkage, and compressive strength), leachability, and microstructure of shale brick containing oil well-derived drilling waste (DW) was investigated. METHODS: The experiments were conducted at a temperature ranging from 950°C to 1,050°C with 30% DW addition. RESULTS: The results indicate that increasing the sintering temperature decreases the water absorption and apparent porosity and increases the shrinkage, density, and compressive strength of sintered specimens. Moreover, the physico-mechanical properties of samples sintered at 1,050°C meet the requirements of the MU20 according to GB/T 5101-2003 (in China). The heavy metal concentrations of the leachate are much lower than the current regulatory limits according to GB16889-2008. CONCLUSION: The results from XRD and SEM show that increasing sintering temperature results in an increase of the high temperature liquid phase, which may have a significant effect on the densification process of the samples.

PURPOSE: The purpose this research is to investigate the interaction of Cr(VI) species, present as Cr₂O₇ ²⁻, at ambient temperature with brick clay pre-fired at different temperatures. METHODS: A multi-technique approach was used for this investigation. Experiments such as surface titrations, Langmuir and Freundlich adsorption isotherms, mass-firing temperature investigation, scanning electron microscopy, Fourier transform infrared spectra, X-ray fluorescence spectra, and X-ray diffraction were conducted in this investigation. RESULTS: Fired brick clay, which bears a negative charge according to surface titration measurements, shows affinity towards Cr(VI) species despite the negative charge of the source of Cr(VI). The Cr(VI)—brick clay heterogeneous system, which shows the strongest interaction with brick clay fired at 200°C, obeys both the Langmuir and the Freundlich adsorption isotherms with high regression coefficients. Investigation on surface charge, constituents of brick clay, acid treatment of clay particles, and the effect of firing temperature suggests that the reduction of Cr(VI) to Cr(III) by reducing agents present in brick clay makes a significant contribution for adsorption of chromium species followed by subsequent removal. Scanning electron microscopic images support the adsorption of chromium species, and further, many metal ions are released as a result of Cr(VI)—brick clay interaction according to X-ray fluorescence studies. CONCLUSION: It is concluded that fired brick clay shows strong adsorption capacity on Cr(VI), having the maximum interaction with brick clay fired at 200°C. It is proposed that this methodology be extended for treatment of effluents containing Cr(VI) species.
