Intensity dependent disruptive effects of light at night on activation of the HPG axis of tree sparrows (Passer montanus)

Artificial light at night (ALAN) has become increasingly recognized as a disruptor of the reproductive endocrine process and behavior of wild birds. However, there is no evidence that ALAN directly disrupt the hypothalamus-pituitary-gonadal (HPG) axis, and no information on the effects of different ALAN intensities on birds. We experimentally tested whether ALAN affects reproductive endocrine activation in the HPG axis of birds, and whether this effect is related to the intensity of ALAN, in wild tree sparrows (Passer montanus). Forty-eight adult female birds were randomly assigned to four groups. They were first exposed to a short light photoperiod (8 h light and 16 h dark per day) for 20 days, then exposed to a long light photoperiod (16 h light and 8 h dark per day) to initiate the reproductive endocrine process. During these two kinds of photoperiod treatments, the four groups of birds were exposed to 0, 85, 150, and 300 lux light in the dark phase (night) respectively. The expression of the reproductive endocrine activation related TSH-β, Dio2 and GnRH-I gene was significantly higher in birds exposed to 85 lux light at night, and significantly lower in birds exposed to 150 and 300 lux, relative to the 0 lux control. The birds exposed to 85 lux had higher peak values of plasma LH and estradiol concentration and reached the peak earlier than birds exposed to 0, 150, or 300 lux did. The lower gene expression of birds exposed to 150 and 300 lux reduced their peak LH and estradiol values, but did not delay the timing of these peaks compared to the control group. These results reveal that low intensity ALAN accelerates the activation of the reproductive endocrine process in the HPG axis, whereas high intensity ALAN retards it.

Thermal desorption (TD) is a universal solvent-free pre-concentration technique. It is often used to pre-concentrate semi-volatile and volatile organic compounds in various sample types. Polycyclic aromatic hydrocarbons (PAHs) are widespread contaminants from incomplete combustion of organic matter and fossil fuel, which have carcinogenic effects on human health. Conventional methods for determining PAHs, represented by solvent extraction, are gradually being replaced by solvent-free methods, typically the TD technique, because of TD's many advantages, including time savings and environmentally friendly treatment. This work presents an extensive review of the universal methods used to determine PAHs in the atmosphere based on the TD technique. The methods currently used for collection and detection of both gas- and particle-phase PAHs in the air are critically reviewed. In addition, the operating parameters of the TD unit are summarized and discussed. The design shortcomings of existing studies and the problems that researchers should address are presented, and promising alternatives are suggested. This paper also discusses important parameters, such as reproducibility and limit of detection, that form a crucial part of quality assurance. Finally, the limitations and the future prospects of the TD technique for use in airborne PAH analyses are addressed. This is the first review of the latest developments of the TD technique for analysis of PAHs and their derivatives in the atmosphere.

Although alternative Flame Retardant (FR) chemicals are expected to be safer than the legacy FRs they replace, their risks to human health and the environment are often poorly characterized. This study used a small volume, fish embryo system to reveal potential mechanisms of action and diagnostic exposure patterns for TBPH (bis (2-ethylhexyl)-tetrabromophthalate), a component of several widely-used commercial products. Two different concentration of TBPH were applied to sensitive early life stages of an ecologically important test species, Fundulus heteroclitus (Atlantic killifish), with a well-annotated genome. Exposed fish embryos were sampled for transcriptomics or chemical analysis of parent compound and primary metabolite or observed for development and survival through larval stage. Global transcript profiling using RNA-seq was conducted (n = 16 per treatment) to provide a non-targeted and statistically robust approach to characterize TBPH gene expression patterns. Transcriptomic analysis revealed a dose-response in the expression of genes associated with a surprisingly limited number of biological pathways, but included the aryl hydrocarbon receptor signal transduction pathway, which is known to respond to several toxicologically-important chemical classes. A transcriptional fingerprint using Random Forests was developed that was able to perfectly discriminate exposed vs. non-exposed individuals in test sets. These results suggest that TBPH has a relatively low potential for developmental toxicity (at least in fishes), despite concerns related to its structural similarities to endocrine disrupting chemicals and that the early life stage Fundulus system may provide a convenient test system for exposure characterization. More broadly, this study advances the usefulness of a biological testing and analysis system utilizing non-targeted transcriptomics profiling and early developmental endpoints that complements current screening methods to characterize chemicals of ecological and human health concern.

As a result of their ubiquitous presence in the environment perfluoroalkyl acids (PFAAs) have been demonstrated in numerous organisms worldwide, in particular in those of higher trophic levels. The fact that PFAAs have been detected in various human matrices, together with the resultant potential human health risks, knowledge of possible paths of entry and distribution in various ecosystems, is of great importance. In this context pooled samples of terrestrial ecosystems – roe deer liver (n = 141), earthworms (n = 44) as well as leaves from beech and poplar trees (n = 70) – from the year 1989–2015 were obtained from the German Environmental Specimen Bank and examined for the presence of 11 PFAAs for the first time. In addition to determining individual and total PFAA concentrations, temporal trends have been deduced in order to determine the effectiveness of regulatory measures. The highest total mean concentration of PFAAs (sum of the concentrations of the 11 analytes) were 9.9 μg/kg in the roe deer liver samples, followed by earthworm samples with a mean PFAA concentration of 3.5 μg/kg and leaves with a mean total concentration of 2.5 μg/kg. In regard to temporal trends there was a significant reduction of concentrations for perfluorooctane sulfonate, perfluorooctanoic acid, perfluorononanoic acid and perfluorodecanoic acid in roe deer liver from 2003 to 2015, an indication of the effectiveness of regulatory measures. The same is true for the perfluorooctane sulfonate concentrations in earthworms and for perfluorooctanoic acid concentrations in the leaves. In contrast, an increase in perfluorobutanoic acid concentrations was observed from 2003 to 2015 in all three matrices. In summary it must be noted that in spite of the discernible effectiveness of minimization strategies, PFAAs are still detectable in terrestrial matrices and concentrations of other PFAAs as perfluorobutanoic acid appear to be increasing.

The extent of per- and polyfluoroalkyl substances (PFAS) in groundwater surrounding legacy landfills is currently poorly constrained. Seventeen PFAS were analysed in groundwater surrounding legacy landfills in a major Australian urban re-development precinct. Sampling locations (n = 13) included sites installed directly in waste material and down-gradient from landfills, some of which exhibited evidence of leachate contamination including elevated concentrations of ammonia-N (≤106 mg/L), bicarbonate (≤1,740 mg/L) and dissolved methane (≤10.4 mg/L). Between one and fourteen PFAS were detected at all sites and PFOS, PFHxS, PFOA and PFBS were detected in all samples. The sum of detected PFAS (∑₁₄PFAS) varied from 26 ng/L at an ambient background site to 5,200 ng/L near a potential industrial point-source. PFHxS had the highest median concentration (34 ng/L; range: 2.6–280 ng/L) followed by PFOS (26 ng/L; range: 1.3–4,800 ng/L), PFHxA (19 ng/L; range: <LOQ – 46 ng/L) and PFOA (12 ng/L; range: 1.7–74 ng/L). Positive correlations between ∑₁₄PFAS, PFOA and other perfluoroalkyl carboxylic acids (PFCAs) (e.g. PFHxA) with typical leachate indicators including ammonia-N and bicarbonate were observed. In contrast, no such correlations were found with perfluoroalkyl sulfonic acids (PFSAs) (e.g., PFOS and PFHxS). In addition, a strong positive linear correlation (R² = 0.69) was found between the proportion of PFOA in the sum of detected perfluorinated alkylated acids (PFOA/∑PFAA) and ammonia-N concentrations in groundwater. This is consistent with previous research showing relatively high PFOA/∑PFAA in municipal landfill leachates, and more conservative behaviour (e.g. less sorption and reactivity) of PFCAs during subsurface transport compared to PFSAs. PFOA/∑PFAA in groundwater may therefore be a useful indicator of municipal landfill-derived PFAA. One site with significantly elevated PFOS and PFHxS concentrations (4,800 and 280 ng/L, respectively) appears to be affected by point-source industrial contamination, as landfill leachate indicators were absent.

Whereas early life stages are usually considered as particularly sensitive to both organic and inorganic contaminants, field studies assessing contaminant bioaccumulation in these stages are scarce. Selenium (Se) is thought to counteract Hg toxic effects when it is found at Se:Hg molar ratios above 1. However, the variation of this ratio in key fish tissues of different early life stages is mostly unknown. The present study therefore aimed to assess Hg and Se content in gravid female tissues (gonads, muscle, liver, gut, and brain) and different life stages (egg masses, newly hatched larvae (NHL), larvae and juvenile) of Yellow Perch (YP) in a large fluvial lake (Lake Saint-Pierre, Québec, Canada). Se:Hg molar ratios were measured for each compartment in order to fill associated knowledge gaps. Total Hg (THg) and methylmercury (MeHg) concentration varied between tissue according to the following trend: Muscle > Liver > Gut > Brain > Gonads. During YP early life stages, MeHg values increased according to an ontogenetic pattern (mg/kg dw) (mean ± SEM): Egg masses (0.01 ± 0.002) < NHL (0.015 ± 0.001) < Larvae (0.14 ± 0.01) < Juveniles (0.18 ± 0.01). Se concentrations in different YP tissues showed the following trend (mg/kg dw) (mean ± SEM): Gut (3.6 ± 0.1) > Liver (2.5 ± 0.1) > Gonads (1.92 ± 0.06) > Brain (1.26 ± 0.03) > Muscle (1.23 ± 0.06). In YP early life stages, Se concentrations were highest in NHL (3.0 ± 0.2), and then decreased as follows: Egg masses (2.8 ± 0.1) > Larvae (1.37 ± 0.04) > Juveniles (0.93 ± 0.05). Se:Hg molar ratios varied considerably and were systematically above 1. This is the first study to simultaneously report Hg and Se bioaccumulation through fish life cycle.

This study investigated by the moss-bag approach the pattern of air dispersed elements in 12 coupled indoor/outdoor exposure sites, all located in urban and rural residential areas. The aims were to discriminate indoor vs. outdoor element composition in coupled exposure sites and find possible relation between moss elemental profile and specific characteristics of each exposure site.Elements were considered enriched when in 60% of the sites, post-exposure concentration exceeded pre-exposure concentration plus two folds the standard deviation. Of the 53 analyzed elements, 15 (As, B, Ca, Co, Cr, Cu, Mn, Mo, Ni, Sb, Se, Sn, Sr, V, Zn) were enriched in moss exposed outdoor, whereas a subset of 7 elements (As, B, Cr, Mo, Ni, Se, V) were enriched also in indoor moss samples. The cluster analysis of the sites based on all elements, clearly separated samples in two groups corresponding to mosses exposed indoor and outdoor, with the latter generally exceeding the first. Among outdoor sites, urban were most impacted than rural; whereas other factors (e.g., heating and cooking systems, building material, residence time and family life style) could affect element profile of indoor environments. Based on the indoor/outdoor ratio, As derived from outdoor and indoor sources, B, Mo and Se were enriched mostly in outdoor sites; Ni, Cr and V were specifically enriched in most indoor samples, supporting the presence of indoor emitting sources for these elements. A PCA of all indoor sites based on enriched elements and site characteristics showed that traffic affected indoor pollution in urban areas. The moss bag approach provided useful information for a global assessment of human exposure.

Microplastics (MPs) are an emerging concern and potential risk to marine and terrestrial environments. Surface soils are reported to act as a sink. However, MP vertical mobility in the subsurface remains uncertain due to a lack of scientific data. This study focused on MP penetration in sand soil column experiments. Here we report the mobility of five different MPs, which consisted of polyethylene (PE) and polypropylene (PP) particles of various sizes and densities. We observed that the smallest sized PE MPs (21 μm) had the greatest movement potential. Moreover, it was found that when these MPs were subjected to greater numbers of wet-dry cycles, the penetration depth significantly increased, with an apparent linear relationship between depth and wet-dry cycle number (r2 = 0.817). In comparison, increasing the volume of infiltration liquid or the surface MP concentration had only negligible or weak effects on migration depth (r2 = 0.169 and 0.312, respectively). Based on the observed wet-dry cycle trend, we forecast 100-year penetration depths using weather data for 347 cities across China. The average penetration depth was calculated as 5.24 m (95% CI = 2.78–7.70 m), with Beijing Municipality and Hebei, Henan and Hubei provinces being the most vulnerable to MP vertical dispersion. Our results suggest that soils may not only represent a sink for MPs, but also a feasible entryway to subsurface receptors, such as subterranean fauna or aquifers. Finally, research gaps are identified and suggested research directions are put forward to garner a better understanding MP vertical migration in soil.

Perfluoroalkyl substances (PFASs) are known to be toxic, bioaccumulative, and persistent. However, exposure routes and toxic effects to humans are still widely unknown. Our objectives were to evaluate potential correlations between concentrations of PFASs in maternal plasma and infant cord blood with home characteristics and developmental effects, including wheezing. The concentrations of 17 PFASs were measured in plasma from prenatal women (n = 414), postnatal women (n = 247), and cord blood (n = 50) from a subset of participants in a population-based birth cohort in Winnipeg, Manitoba, Canada, using online solid phase extraction (SPE) with liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Multiple linear regression and principal component analysis (PCA) were used to evaluate possible associations with PFAS concentrations. Surveys were used to collect information regarding maternal characteristics (e.g. age, parity, duration of breastfeeding), infant characteristics (e.g. birth weight, birth length, head circumference, gestational age, and incidence of recurrent wheezing), and home characteristics (e.g. home age,carpet coverage in the most used room, presence of new furniture, or recent home renovations). PFASs in plasma were associated with maternal characteristics but not home characteristics or early childhood wheezing. PFASs were not associated with developmental effects, with the exception that perfluoroundecanoic acid (PFUA) was negatively associated with birth weight. Further studies investigating the potential influences of PFUA on birth weight are warranted.

Aerosol black carbon (BC) is the second strongest contributor to global warming, after CO₂, and it is linked to many adverse health effects. A sampling campaign of 15 months was carried out in León (Spain) in order to evaluate the scavenging of BC with an ensemble aethalometer-disdrometer. The aethalometer provides the concentration of equivalent black carbon (eBC), and the disdrometer, the raindrop size distribution. A total of seventy-five rain events were studied and in 73% of them there was an effective (eBCᵢₙᵢₜᵢₐₗ > eBCfᵢₙₐₗ) scavenging, with a mean decrease of 48 ± 37% in long rain events (>8 h) and 39 ± 38% in short rain events. The scavenging of BC is strongly related to its source. Thus, the scavenging coefficient (SC) mean value of the BC from fossil fuel (eBCff) for short and long rain events was 5.1 10⁻⁵ and 1.3 10⁻⁵ s⁻¹, respectively. For the BC from biomass burning (eBCbb), the SC values were 1.6 10⁻⁴ and 2.8 10⁻⁵ s⁻¹ in short and long events, respectively. There was a significant positive correlation between the SC and the number of drops with diameters between 0.375 and 2.5 mm. Rain scavenging of eBC was analyzed depending on the air mass origin obtaining an effective scavenging for air masses from Atlantic, Arctic and Africa. A linear model (R² = 0.72) was built to estimate the ΔeBC values with variables from an aethalometer, a disdrometer and a weather station: eBC concentration before rain, swept volume and precipitation accumulated. A Kolmogorov-Smirnov statistical test confirmed the goodness of fit of the model to the measured data.
