Human inflammatory response of endotoxin affected by particulate matter-bound transition metals النص الكامل

Bacterial endotoxins are a component of particulate matter (PM) with anticipated health implications, yet we know little about how host reception of endotoxin through toll-like receptor 4 (TLR4) is affected by its association with other PM components. Subsequently, we investigated the relationship between endotoxin concentration (recombinant Factor C (rFC) assay) and host recognition (HEK Blue-TLR4 NF-kB reporter cell line based assay) in various compositions of urban PM, including road traffic, industrial and urban green land use classes. While the assays did not correlate strongly between each other, the TLR4 reporter cell line was found to be better correlated to the IL-8 response of PM. Furthermore, the ability of the quantified endotoxin (rFC assay) to stimulate the TLR4/MD-2 complex was significantly affected by the urban land use class, where traffic locations were found to be significantly higher in bioactive endotoxin than the industrial and green locations. We subsequently turned our attention to PM composition and characterized the samples based on transition metal content (through ICP-MS). The effect of nickel and cobalt – previously reported to activate the hTLR4/MD-2 complex – was found to be negligible in comparison to that of iron. Here, the addition of iron as a factor significantly improved the regression model between the two endotoxin assays, explaining 77% of the variation of the TLR4 stimulation and excluding the significant effect of land use class. Moreover, the effect of iron proved to be more than a correlation, since dosing LPS with Fe²⁺ led to an increase up to 64% in TLR4 stimulation, while Fe²⁺ without LPS was unable to stimulate a response. This study shows that endotoxin quantification assays (such as the rFC assay) may not always correspond to human biological recognition of endotoxin in urban PM, while its toxicity can be synergistically influenced by the associated PM composition.

The Keban Reservoir, which is the second man-made waterbody in Turkey, has the biggest rainbow trout production in the country. In this study, the impacts of rainbow trout farms on water and sediment chemistry were investigated. Water and sediment samples were taken at distances of 0, 10, 25, 50 and 100 m from the edge of the cages at the three fish farms, and at the respective reference stations. Samples were also taken at 0 m stations and reference stations in the late August when there were no fish in the cages. Physico-chemical variables and trace metals were analysed in all samples. Due to likely high dilution rates and recycling processes in the water column of the reservoir, little changes in the water quality parameters associated with wastes of the fish farms were noticed. When compared with those in the sediment samples at the stations near the edge of cages, the lower concentrations of total phosphorus (TP), total nitrogen (TN), organic matter (OM), total carbon (TC), sulfide (S2−), arsenic (As), cobalt (Co), copper (Cu), manganese (Mn) and zinc (Zn), and higher values of redox potential (Eh) were found at the reference stations. According to organic enrichment classification based on S2− and Eh values, sediments of the three fish farms in the period when there were fish in the cages fell into the oxic category, whereas sediments in the August (no fish farming activity) fell into the normal category. Also, it was found in the August that most of sediment quality parameters at the 0 m stations had close values to those at the reference stations. These results revealed that a three-month period when there were no fish in the cages allows for sediments to return to reference station conditions.

To date, there is not sufficient knowledge to fully understand the occurrence, transport and fate of residual uranium (U) from uranium mill tailings (UMT). Herein this study investigated different U release behaviors from natural UMT (without grinding) under four simulated acid rain (pH = 2.0–5.0) compared with controlled scenario (pH = 6.0) for 25 weeks. The results showed that the most notable U release was observed from UMTₚH₂.₀, followed by UMTₚH₃.₀ whereas a nonlinear relationship between pH and U release was observed from UMTₚH₄.₀–₆.₀. The divergence of U release behaviors was attributed to the presence of minerals such as calcite and clinochlore. Autunite, a secondary mineral formed after leaching, might regulate U release in UMTₚH₃.₀–₆.₀. Fick theory model revealed the shift of U release mechanism from surface dissolution to diffusion transport for UMTₚH₂.₀, UMTₚH₃.₀ and UMTₚH₅.₀ at varied stage, whereas UMTₚH₄.₀ and UMTₚH₆.₀ displayed univocal dissolution and diffusion mechanism, respectively. This study highlights the necessity of performing long-term leaching tests to detect the “shift event” of leaching kinetics and to better understand the mechanism of U release influenced by mineralogy of the natural UMT under simulated acid rain conditions, which is conducive to developing UMT management strategies to minimize the risk of U release and exposure.

The ammonium (NH4+) pool in the water column of eutrophic lakes is dynamic and undergoes tightly coupled production and consumption processes because of the metabolism of bacterial and algal communities, particularly in summer. However, NH4+ recycling rates along nutrient gradients at river-lake transitional zones and the extent to which NH4+ regeneration can compensate for consumption have been poorly studied. In August (flood period) and November (normal period), 2016, NH4+ regeneration rates (REGs) and potential uptake rates (Upots) were measured in northwestern Lake Taihu and adjacent rivers. Results showed that the REGs ranged from 0.09 to 3.30 μmol N L−1 h−1 and the Upots ranged from 0.20 to 4.88 μmol N L−1 h−1, with higher recycling rates occurring at the river sites. Yet, the lake sites showed significantly higher water column NH4+ demand (WCAD) than that of the adjacent river sites during both seasons (p < 0.05), probably as a result of the low REGs and the lack of exogenous nitrogen (N) inputs. The flood period showed significantly higher REG and Upot values than those of the normal period (p < 0.05), probably controlled by higher water temperature and algal biomass. This study confirms that regenerated NH4+ was more important than the ambient NH4+ for sustaining cyanobacterial blooms in northwestern Lake Taihu and indicates that the river–lake transitional zones are key areas for N control in this hypereutrophic system.

Blueberry anthocyanin (BA) have strong health benefits as an active natural antioxidant and perfluorooctanoic acid (PFOA) can result in oxidative stress in animals. In our study, the protective effects of BA against stress induced by PFOA was investigated in the planarian Dugesia japonica using oxidative stress biomarkers, ATP contents, ATPase activity, DNA methylation and mRNA expression. PFOA exposure could resulted in malondialdehyde production. At the same time, treatment with BA decreased the production of malondialdehyde in BA-exposed and co-treatment planarians. PFOA caused activities increase in glutathione peroxidase (GPx), glutathione S-transferase (GST) and activities decrease in glutathione reductase (GR). PFOA exposure decreased the GSH and ATP contents. Additionally, it increased the GSSG contents and ATPase activity. BA administration increased the activities of GPx, GST and GR in BA and co-treatment planarians. Meanwhile BA maintained the contents of ATP, ATPase activity, GSH and GSSG by alleviating PFOA toxicity. Moreover, PFOA and BA increased the contents of 5-methylcytosine and decreased 5-hydroxymethylcytosine in all group. In addition, PFOA and BA treated planarians significantly altered the expression of genes associated with above biochemical parameters. The results showed that the mRNA expression of gpx, Djgst, gr, Djnak and dnmt1 were significantly elevated in all groups. Alterations in the mRNA expression levels indicated a stress response to PFOA exposure and anthocyanin protection. These alterations regulated biomarkers of oxidative stress, energy metabolism and DNA methylation levels in planarians. These results indicate that BA attenuated PFOA-induced oxidative stress, energy metabolism, DNA methylation and gene expression disorders.

Rapid urbanization, which is closely related to economic growth, human health, and micro-climate, has resulted in a considerable amount of anthropogenic heat emissions. The lack of estimation data on long-term anthropogenic heat emissions is a great concern in climate and urban flux research. This study estimated the annual average anthropogenic heat fluxes (AHFs) in Beijing–Tianjin–Hebei region in China between 1995 and 2015 on the basis of multisource remote sensing images and ancillary data. Anthropogenic heat emissions from different sources (e.g., industries, buildings, transportation, and human metabolism) were also estimated to analyze the composition of AHFs. The spatiotemporal dynamics of long-term AHFs with high spatial resolution (500 m) were estimated by using a refined AHF model and then analyzed using trend and standard deviation ellipse analyses. Results showed that values in the region increased significantly from 0.15 W· m−2 in 1995 to 1.46 W· m−2 in 2015. Heat emissions from industries, transportation, buildings, and human metabolism accounted for 64.1%, 17.0%, 15.5%, and 3.4% of the total anthropogenic heat emissions, respectively. Industrial energy consumption was the dominant contributor to the anthropogenic heat emissions in the region. During this period, industrial heat emissions presented an unstable variation but showed a growing trend overall. Heat emissions from buildings increased steadily. Spatial distribution was extended with an increasing tendency of the difference between the maximum and the minimum and was generally dominated by the northeast–southwest directional pattern. The spatiotemporal distribution patterns and trends of AHFs could provide vital support on management decision in city planning and environmental monitoring.

The acute and chronic toxicity of 3 common pesticides, namely, amitraz, chlorpyrifos and dimethoate, were tested in Apis mellifera and Apis cerana. Acute oral toxicity LC50 values were calculated after 24 h of exposure to contaminated syrup, and chronic toxicity was tested after 15 days of exposure to 2 sublethal concentrations of pesticides. The toxicity of the tested pesticides to A. mellifera and A. cerana decreased in the order of dimethoate > chlorpyrifos > amitraz. A. mellifera was slightly more sensitive to chlorpyrifos and dimethoate than A. cerana, while A. cerana was more sensitive to amitraz than A. mellifera. Chronic toxicity tests showed that 1.0 mg/L dimethoate reduced the survival of the two bee species and the food consumption of A. mellifera, while 1.0 mg/L amitraz and 1.0 mg/L chlorpyrifos did not affect the survival or food consumption of the two bee species. The treatment of syrup with amitraz at a concentration equal to 1/10th of the LC50 value did not affect the survival of or diet consumption by A. mellifera and A. cerana; however, chlorpyrifos and dimethoate at concentrations equal to 1/10th of their respective LC50 values affected the survival of A. cerana. Furthermore, intestinal bacterial communities were identified using high-throughput sequencing targeting the V3V4 regions of the 16S rDNA gene. All major honey bee intestinal bacterial phyla, including Proteobacteria (62.84%), Firmicutes (34.04%), and Bacteroidetes (2.02%), were detected. There was a significant difference in the microbiota species richness of the two species after 15 days; however, after 30 days, no significant differences were found in the species diversity and richness between A. cerana and A. mellifera exposed to 1.0 mg/L amitraz and 1.0 mg/L chlorpyrifos. Overall, our results confirm that acute toxicity values are valuable for evaluating the chronic toxicity of these pesticides to honey bees.

Reported here is the first evidence of plastic ingestion by freshwater fishes in the Amazon. Plastic bags, bottles, fishing gear, and other products are entering Amazonian water bodies and degrade into meso- and micro-plastic particles that may be ingested, either directly or indirectly via food chains, by fishes. Examination of stomach contents from 172 specimens of 16 serrasalmid species from lower Xingu River Basin revealed consumption of plastic particles by fishes in each of three trophic guilds (herbivores, omnivores, carnivores). Overall, about one quarter of specimens and 80% of species analyzed had ingested plastic particles ranging from 1 to 15 mm in length. Fourier transform infrared spectroscopy indicated 12 polymer types, including 27% identified as polyethylene, 13% polyvinyl chloride, 13% polyamide, 13% polypropylene, 7% poly(methyl methacrylate), 7% rayon, 7% polyethylene terephtalate, and 13% a blend of polyamide and polyethylene terephtalate. Dimensions of ingested plastic particles varied among trophic guilds, even though the frequency and mass of ingested particles were not significantly different among fishes with different feeding habits.

Microplastic pollution of intertidal mangrove ecosystems is receiving growing attention, and scientists suspect that the microplastic pollution of semi-enclosed seas is significantly different from that of other coastal types because of their unique geographical features. However, data on the distributions and characteristics of microplastics in the mangrove sediment of semi-enclosed seas are very limited. This study selected the Maowei Sea, a typical semi-enclosed sea, as its representative study site. The analysis revealed that the microplastic abundances in the river estuaries were much lower than those at the oceanic entrance zones, with values ranging from 520 ± 8 to 940 ± 17 items/kg. Polyethylene (PE)/polypropylene (PP)/polystyrene (PS), white/transparent, and <1 mm were the dominant type, colour, and size of the microplastics, respectively, in the observed mangrove sediments. Moreover, some other factors, including the rhizosphere/non-rhizosphere and the proportion of organic matter, codetermined the distribution and characteristics of microplastics. Specifically: (1) the percentage of colorful microplastics were higher in the rhizosphere due to the microbial activities and (2) positive linear relationships were found between the pore volume (PV) values of the free particulate organic matter (FPOM), occluded particulate organic matter (OPOM) (1.6–2.0 g/cm³ and >2.0 g/cm³), and the abundance of very small microplastics (<1 mm).

Hydrogeochemistry and isotope hydrology were carried out to investigate the spatial distribution of fluoride (F−) and the mechanisms responsible for its enrichment in the western region of the Ordos basin, northwestern China. Sixty-two groundwater samples from the unconfined aquifer and fifty-six from confined aquifer were collected during the pre-monsoon (June 2016). Over 77% of groundwater samples from the unconfined aquifer (F− concentration up to 13.30 mg/L) and approximately 66% from confined aquifer (with a maximum F− concentration of 3.90 mg/L) exhibit F− concentrations higher than the Chinese safe drinking limit (1.0 mg/L). High-F− groundwater presents a distinctive hydrochemical characteristic: a high pH value and HCO3− concentration with Ca-poor and Na-rich. Mineral dissolution (e.g., feldspar, calcite, dolomite, fluorite), cation exchange and evaporation in the aquifers predominate the formation of groundwater chemistry, which are also important for F− enrichment in groundwater. Mixing with unconfined groundwater is a significant mechanism resulting in the occurrence of high-F− groundwater in confined aquifer. These findings indicate that physicochemical processes play crucial roles in driving F− enrichment and that may be useful for studying F− occurrence in groundwater in arid and semi-arid areas.
