Estimating the contribution of bacteriophage to the dissemination of antibiotic resistance genes in pig feces النص الكامل

The transfer of antibiotic resistance genes (ARGs) in the environment is a threat to both human and animal health. However, the contribution of bacteriophages to the dissemination of resistance genes via transduction is rarely explored. In this study, we screened pig feces from three commercial farms in China for 32 clinically relevant ARG types to assess the presence of the ARG population in bacteria and bacteriophage and further to estimate the contribution of bacteriophages to the dissemination of antibiotic resistance. We found that bacteriophage DNA contained 35.5% of the target ARG types and sul1, blaTEM and ermB were found in 100% of the phage DNA samples. The most abundant genes in the bacterial population were ermB and fexA whereas ermB was the most abundant in bacteriophage. In contrast, floR was the least abundant ARG in both populations. Also, the ratio index of the abundance of ARGs in bacteriophage and bacteria was firstly used in this study as an estimator of bacteriophage ability to transmit ARGs. The ratio for qnrA was the greatest (about 10−1) and differed from the most abundant bacteriophage ARG ermB. In addition, fexA had the lowest ratio value (about 10−6) and not floR. These results illustrate that ARGs abundance and detection rates used alone probably be not suitable for comprehensively judging the contribution of bacteriophage to the dissemination of antibiotic resistance. A more suitable model is the application of three indices; occurrence rate, absolute abundance in bacteriophage and the ratio value as warning and monitoring tools for environmental ARG assessments in bacteriophages.

Microplastics and mercury are environmental pollutants of great concern. The main goal of the present study was to investigate the effects of these pollutants, both individually and in binary mixtures, on the swimming performance of juvenile European seabass, Dicentrarchus labrax. Microplastics alone, mercury alone and all the mixtures caused significant reduction of the swimming velocity and resistance time of fish. Moreover, changes in behavioural responses including lethargic and erratic swimming behaviour were observed. These results highlight that fish behavioural responses can be used as sensitive endpoint to establish the effects of contamination by microplastics and also emphasizes the need to assess the combined effects of microplastics and other environmental contaminants, with special attention to the effects on behavioural responses in fish and other aquatic species.

Recent studies have indicated that urban streets can be hotspots for emissions of methane (CH4) from leaky natural gas lines, particularly in cities with older natural gas distribution systems. The objective of the current study was to determine whether leaking sewer pipes could also be a source of street-level CH4 as well as nitrous oxide (N2O) in Cincinnati, Ohio, a city with a relatively new gas pipeline network. To do this, we measured the carbon (δ13C) and hydrogen (δ2H) stable isotopic composition of CH4 to distinguish between biogenic CH4 from sewer gas and thermogenic CH4 from leaking natural gas pipelines and measured CH4 and N2O flux rates and concentrations at sites from a previous study of street-level CH4 enhancements (77 out of 104 sites) as well as additional sites found through surveying sewer grates and utility manholes (27 out of 104 sites). The average isotopic signatures for δ13C-CH4 and δ2H-CH4 were −48.5‰ ± 6.0‰ and −302‰ ± 142‰. The measured flux rates ranged from 0.0 to 282.5 mg CH4 day−1 and 0.0–14.1 mg N2O day−1 (n = 43). The average CH4 and N2O concentrations measured in our study were 4.0 ± 7.6 ppm and 392 ± 158 ppb, respectively (n = 104). 72% of sites where fluxes were measured were a source of biogenic CH4. Overall, 47% of the sampled sites had biogenic CH4, while only 13% of our sites had solely thermogenic CH4. The other sites were either a source of both biogenic and thermogenic CH4 (13%), and a relatively large portion of sites had an unresolved source (29%). Overall, this survey of emissions across a large urban area indicates that production and emission of biogenic CH4 and N2O is considerable, although CH4 fluxes are lower than those reported for cities with leaky natural gas distribution systems.

Many of the world's large coastal cities discharge partially treated wastewater effluents containing various endocrine disrupting chemicals (EDCs) to coastal environments. Nonylphenols (NP) and bisphenol A (BPA) were found to be the most abundant EDCs in sewage effluents in Hong Kong. The environmental fate and ecological risk of these two EDCs remains largely unknown, particular for coastal systems with complex hydrodynamic flows. Based on a validated three-dimensional (3D) multiple-scale hydrodynamic model, a field-based study was conducted to track the two EDCs from potential sources to the only marine reserve in Hong Kong. The two compounds were detected in all seawater, suspended particle, and sediment samples, with higher aqueous concentrations in wet season than in dry season. High concentrations in sediments suggest sediment is a sink, posing an ecological risk to the benthos. The fate and transport of the two EDCs was predicted using a 3D near-field Lagrangian jet model seamlessly coupled with a 3D shallow water circulation model. The results suggested the NP and BPA in the marine reserve cannot be solely attributed to the nearby submarine sewage outfall, but likely concurrently contributed by other sources. This study calls for more effective measures of reducing the use and release of these EDCs, and research to investigate their impacts on the marine benthos.

In this study, we examined the removal of arsenite (As(III)) and arsenate (As(V)) by perilla leaf-derived biochars produced at 300 and 700 °C (referred as BC300 and BC700) in aqueous environments. Results revealed that the Langmuir isotherm model provided the best fit for As(III) and As(V) sorption, with the sorption affinity following the order: BC700-As(III) > BC700-As(V) > BC300-As(III) > BC300-As(V) (QL = 3.85–11.01 mg g⁻¹). In general, As removal decreased (76–60%) with increasing pH from 7 to 10 except for the BC700-As(III) system, where notably higher As removal (88–90%) occurred at pH from 7 to 9. Surface functional moieties contributed to As sequestration by the biochars examined here. However, significantly higher surface area and aromaticity of BC700 favored a greater As removal compared to BC300, suggesting that surface complexation/precipitation dominated As removal by BC700. Arsenic K-edge X-ray absorption near edge structure (XANES) spectroscopy demonstrated that up to 64% of the added As(V) was reduced to As(III) in BC700- and BC300-As(V) sorption experiments, and in As(III) sorption experiments, partial oxidation of As(III) to As(V) occurred (37–39%). However, XANES spectroscopy was limited to precisely quantify As binding with sulfur species as As2S3-like phase. Both biochars efficiently removed As from natural As-contaminated groundwater (As: 23–190 μg L⁻¹; n = 12) despite in the presence of co-occurring anions (e.g., CO3²⁻, PO4³⁻, SO4²⁻) with the highest levels of As removal observed for BC700 (97–100%). Overall, this study highlights that perilla leaf biochars, notably BC700, possessed the greatest ability to remove As from solution and groundwater (drinking water). Significantly, the integrated spectroscopic techniques advanced our understanding to examine complex redox transformation of As(III)/As(V) with biochar, which are crucial to determine fate of As on biochar in aquatic environments.

The aim of this study was to compare the toxicity effects of carbon nanomaterials (CNMs), namely fullerene (C60), reduced graphene oxide (rGO) and multi-walled carbon nanotubes (MWCNTs), on a mini-ecosystem of rice grown in a loamy potted soil. We measured plant physiological and biochemical parameters and examined bacterial community composition in the CNMs-treated plant–soil system. After 30 days of exposure, all the three CNMs negatively affected the shoot height and root length of rice, significantly decreased root cortical cells diameter and resulted in shrinkage and deformation of cells, regardless of exposure doses (50 or 500 mg/kg). Additionally, at the high exposure dose of CNM, the concentrations of four phytohormones, including auxin, indoleacetic acid, brassinosteroid and gibberellin acid 4 in rice roots significantly increased as compared to the control. At the high exposure dose of MWCNTs and C60, activities of the antioxidant enzymes superoxide dismutase (SOD) and peroxidase (POD) in roots increased significantly. High-throughput sequencing showed that three typical CNMs had little effect on shifting the predominant soil bacterial species, but the presence of CNMs significantly altered the composition of the bacterial community. Our results indicate that different CNMs indeed resulted in environmental toxicity to rice and soil bacterial community in the rhizosphere and suggest that CNMs themselves and their incorporated products should be reasonably used to control their release/discharge into the environment to prevent their toxic effects on living organisms and the potential risks to food safety.

Maternal 1,1-dichloro-2,2-bis(p-chlorophenyl)ethylene (DDE) serum levels during pregnancy have been negatively linked to child neurodevelopment in contrast to intake of omega-3 and -6 (ω-3 and ω-6) fatty acids.To assess whether maternal dietary intake of ω-3 and ω-6 during pregnancy modifies the association between exposure to DDE and child neurodevelopment from age 42–60 months.Prospective cohort study with 142 mother–child pairs performed in Mexico. DDE serum levels were determined by electron capture gas chromatography. Dietary ω-3 and ω-6 intake was estimated by questionnaire. Child neurodevelopment was assessed by McCarthy Scales.Docosahexaenoic (DHA) fatty acid intake significantly modified the association between DDE and motor component: increased maternal DDE was associated with lower motor development in children whose mothers had lower DHA intake (βlog2DDE = −1.25; 95% CI: −2.62, 0.12), in contrast to the non-significant increase among children whose mothers had higher DHA intake (βlog2DDE-motor = 0.50; 95% CI: 0.55, 1.56). Likewise, arachidonic fatty acid (ARA) intake modified the association between DDE and memory component: increased maternal DDE was associated with a significantly larger reduction in the memory component in children whose mothers had lower ARA intake (βlog2DDE = −1.31; 95% CI: −2.29, −0.32) than children whose mothers had higher ARA intake (βlog2DDE-memory = 0.17; 95% CI: −0.78, 1.11).Dietary intake of DHA and ARA during pregnancy may protect against child neurodevelopment damage associated with prenatal maternal DDE levels.

Complexity of anthropogenic influences on coastal ecosystems necessitates use of an integrated assessment strategy for effective interpretation and subsequent management. In this study a multiple lines of evidence (LOE) approach for sediment assessment, that combined use of chemistry, toxicity, and benthic community structure in the sediment quality triad was used to assess spatiotemporal changes and potential risks of persistent toxic substances (PTSs) in sediments of Masan Bay highlighting “long-term changes” between 1998 and 2014. Specific target objectives encompassed sedimentary PTSs (PAHs, alkylphenols (APs), and styrene oligomers), potential aryl hydrocarbon receptor (AhR; H4IIE-luc assay)- and estrogen receptor (ER; MVLN assay)-mediated activities, and finally several ecological quality (EcoQ) indices of benthic community structure. Concentrations of target PTSs in Masan Bay sediments were generally less by half in 2014 compared to those measured in 1998. Second, AhR-mediated potencies in sediments also decreased during this time interval, whereas ER-mediated potencies increased (+3790%), indicating that there has been substantial ongoing, input of ER agonists over the past 16 years. Potency balance analysis revealed that only 3% and 22% of the AhR- and ER-mediated potencies could be explained by identified known chemicals, such as PAHs and APs, respectively. This result indicated that non-targeted AhR and ER agonists had a considerable presence in the sediments over time. Third, EcoQ indices tended to reflect PTSs contamination in the region. Finally, ratio-to-mean values obtained from the aforementioned three LOEs indicated that quality of sediments from the outer region of the bay had recovery more during the period of 16-years than did the inner region. Overall, the results showed that even with the progress supported by recent efforts from the Korean governmental pollution control, PTSs remain a threat to local ecosystem, especially in the inner region of Masan Bay.

Mercury (Hg) is a pervasive contaminant reaching Antarctic environments through atmospheric transport and deposition. Seabirds as meso to top predators can accumulate high quantities of Hg through diet. Reproduction is one of the most sensitive endpoints of Hg toxicity in marine birds. Yet, few studies have explored Hg exposure and effects in Antarctic seabirds, where increasing environmental perturbations challenge animal populations. This study focuses on the Antarctic petrel Thalassoica antarctica from Svarthamaren, Antarctica, where the world's largest breeding population is thought to be in decline. Hg and the stable isotopes of carbon (δ13C, proxy of feeding habitat) and nitrogen (δ15N, trophic position/diet) were measured in red blood cells from 266 individuals over two breeding years (2012–13, 2013–14). Our aims were to 1) quantify the influence of individual traits (size and sex) and feeding ecology (foraging location, δ13C and δ15N values) on Hg exposure, and 2) test the relationship between Hg concentrations with body condition and breeding output (hatching success and chick survival). Hg concentrations in Antarctic petrels (mean ± SD, 0.84 ± 0.25, min-max, 0.42–2.71 μg g−1 dw) were relatively low when compared to other Antarctic seabirds. Hg concentrations increased significantly with δ15N values, indicating that individuals with a higher trophic level (i.e. feeding more on fish) had higher Hg exposure. By contrast, Hg exposure was not driven by feeding habitat (inferred from both foraging location and δ13C values), suggesting that Hg transfer to predators in Antarctic waters is relatively homogeneous over a large geographical scale. Hg concentrations were not related to body condition, hatching date and short-term breeding output. At present, Hg exposure is likely not of concern for this population. Nevertheless, further studies on other fitness parameters and long-term breeding output are warranted because Hg can have long-term population-level effects without consequences on current breeding success.

Congenital heart defects (CHDs) are a major cause of death in infancy and childhood. Major risk factors for most CHDs, particularly those resulting from the combination of environmental exposures with social determinants and behaviors, are still unknown. This study evaluated the main effect of maternal environmental tobacco smoke (ETS), and its interaction with social-demographics and environmental factors on CHDs in China. A population-based, matched case-control study of 9452 live-born infants and stillborn fetuses was conducted using the Guangdong Registry of Congenital Heart Disease data (2004–2014). The CHDs were evaluated by obstetrician, pediatrician, or cardiologist, and confirmed by cardia tomography/catheterization. Controls were randomly chosen from singleton newborns without any malformation, born in the same hospital as the cases and 1:1 matched by infant sex, time of conception, and parental residence (same city and town to ensure sufficient geographical distribution for analyses). Face-to-face interviews were conducted to collect information on demographics, behavior patterns, maternal disease/medication, and environmental exposures. Conditional logistic regression was used to estimate odds ratios and 95% confidence intervals of ETS exposure on CHDs while controlling for all risk factors. Interactive effects were evaluated using a multivariate delta method for maternal demographics, behavior, and environmental exposures on the ETS-CHD relationship. Mothers exposed to ETS during the first trimester of pregnancy were more likely to have infants with CHD than mothers who did not (aOR = 1.44, 95% CI 1.25–1.66). We also observed a significant dose-response relationship when mothers were exposed to ETS and an increasing number of risk factors and CHDs. There were greater than additive interactions for maternal ETS and migrant status, low household income and paternal alcohol consumption on CHDs. Maternal low education also modified the ETS-CHD association on the multiplicative scale. These findings may help to identify high-risk populations for CHD, providing an opportunity for targeted preventive interventions.
