Langone, M. | Petta, L. | Cellamare, C.M. | Ferraris, M. | Guzzinati, R. | Mattioli, D. | Sabia, G.
The occurrence of human pathogenic viruses in aquatic ecosystems and, in particular, in internal water bodies (i.e., river, lakes, groundwater, drinking water reservoirs, recreational water utilities, and wastewater), raises concerns regarding the related impacts on environment and human health, especially in relation to the possibility of human exposure and waterborne infections. This paper reviews the current state of knowledge regarding severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) presence and persistence in human excreta, wastewaters, sewage sludge as well as in natural water bodies, and the possible implications for water services in terms of fecal transmission, public health, and workers’ risk. Furthermore, the impacts related to the adopted containment and emergency management measures on household water consumptions are also discussed, together with the potential use of wastewater-based epidemiology (WBE) assessment as a monitoring and early warning tool, to be applied in case of infectious disease outbreaks. The knowledge and tools summarized in this paper provide a basic information reference, supporting decisions makers in the definition of suitable measures able to pursue an efficient water and wastewater management and a reduction of health risks. Furthermore, research questions are provided, in order to direct technical and public health communities towards a sustainable water service management in the event of a SARS-CoV-2 re-emergence, as well as a future deadly outbreak or pandemic.

Dioxybenzone is widely used in cosmetics and personal care products and frequently detected in multiple environmental media and human samples. However, the current understanding of the metabolic susceptibility of dioxybenzone and the potential endocrine disruption through its metabolites in mimicking human estrogens remains largely unclear. Here we investigated the in vitro metabolism of dioxybenzone, detected the residue of metabolites in rats, and determined the estrogenic disrupting effects of these metabolites toward estrogen receptor α (ERα). In vitro metabolism revealed two major metabolites from dioxybenzone, i.e., M1 through the demethylation of methoxy moiety and M2 through hydroxylation of aromatic carbon. M1 and M2 were both rapidly detected in rat plasma upon exposure to dioxybenzone, which were then distributed into organs of rats in the order of livers > kidneys > uteri > ovaries. The 100 ns molecular dynamics simulation revealed that M1 and M2 formed hydrogen bond to residue Leu387 and Glu353, respectively, on ERα ligand binding domain, leading to a reduced binding free energy. M1 and M2 also significantly induced estrogenic effect in comparison to dioxybenzone as validated by the recombinant ERα yeast two-hybrid assay and uterotrophic assay. Overall, our study revealed the potential of metabolic activation of dioxybenzone to induce estrogenic disrupting effects, suggesting the need for incorporating metabolic evaluation into the health risk assessment of benzophenones and their structurally similar analogs.

Plastic residues have become a serious environmental problem in areas where agricultural plastic film are used intensively. Although numerous of studies have been done to assess its impacts on soil quality and crop yields, the understanding of meso-plastic particles effects on plant is still limited. In this study, low density polyethylene (PE) and biodegradable plastic (Bio) mulch film were selected to study the effects of meso-plastic debris on soybean germination and plant growth with the accumulation levels of 0%, 0.1%, 0.5% and 1% in soil (w: w, size ranging 0.5–2 cm) by a pot experiment under field condition. Results showed that the germination viability of soybean seeds was reduced to 82.39%, 39.44% and 26.06% in the treatments with 0.1%, 0.5% and 1% added plastic debris compared to the control (CK), respectively, suggesting that plastic residues in soil inhibit the viability of soybean seed germination. The plastic debris had a significant negative effect on plant height and culm diameter during the entire growth stage of soybean. Similarly, the leaf area at harvest was reduced by 1.97%, 6.86% and 11.53% compared to the CK in the treatments with 0.1%, 0.5% and 1% plastic debris addition, respectively. In addition, the total plant biomass under plastic addition was reduced in both the flowering and harvesting stages, compared to the CK. For the different type of plastic residues, plant height, leaf area and root/shoot ratio at group PE were significantly lower than those of groups treated by Bio. In conclusion, PE debris had a greater negative effects on plant height, culm diameter, leaf area and root/shoot ratio while Bio debris mainly showed the adverse effects on germination viability and root biomass especially at the flowering stage. Therefore, further research is required to elaborate plastic particles’ effects on different stages of crops and soil quality.

This study evaluated the chronic toxicity (30 days) of different sizes of polyethylene terephthalate (PET) microplastics (60–3000 μm) provided alone or in combination with acid rain, on garden cress (Lepidium sativum). Both biometrical and physiological traits have been evaluated: i) percentage inhibition of seed germination, plant height, leaf number and fresh biomass production; ii) oxidative stress responses (hydrogen peroxide; ascorbic acid and glutathione production); iii) impairment in photosynthetic machinery in term of pigments production; iv) aminolevulinic acid and proline production. Results highlighted that different sizes of PET, alone or in combination with acid rain, are able to negatively affect both biometrical and physiological plant traits. In particular, the lower size of microplastics is able to negatively affect growth and development, as well as to trigger the oxidative burst. Regarding the pigments production, PET coupled with acid rain, induced a higher production of Chl-b, and an inhibition of aminolevulinic acid.

In modern agricultural management, the use of pesticides is indispensable. Due to their massive use worldwide, pesticides represent a latent risk to both humans and the environment. In the present study, 1056 frequently used pesticides were screened for oestrogen receptor (ER) agonistic activity by using in silico methods. We found that 72 and 47 pesticides potentially have ER agonistic activity by the machine learning methods random forest (RF) and deep neural network (DNN), respectively. Among endocrine-disrupting chemicals (EDCs), 14 have been reported as EDCs or ER agonists by previous studies. We selected 3 reported and 7 previously unreported pesticides from 76 potential ER agonists to further assess ERα agonistic activity. All 10 selected pesticides exhibited ERα agonistic activity in human cells or zebrafish. In the dual-luciferase reporter gene assays, six pesticides exhibited ERα agonistic activity. Additionally, nine pesticides could induce mRNA expression of the pS2 and NRF1 genes in MCF-7 cells, and seven pesticides could induce mRNA expression of the vtg1 and vtg2 genes in zebrafish. Importantly, the remaining 48 out of 76 potential ER agonists, none of which have previously been reported to have endocrine-disrupting effects or oestrogenic activity, should be of great concern. Our screening results can inform environmental protection goals and play an important role in environmental protection and early warnings to human health.

Wild organisms are increasingly exposed to multiple anthropogenic and natural stressors that can interact in complex ways and lead to unexpected effects. In aquatic ecosystems, contamination by trace metals has deleterious effects on fish health and commonly co-occurs with pathogens, which affect similar physiological and behavioral traits. However, the combined effects of metal contamination and parasitism are still poorly known. In addition, the sensitivity to multiple stressors could be highly variable among different fish populations depending on their evolutionary history, but this intraspecific variability is rarely taken into account in existing ecotoxicological studies. Here, we investigated i) the interactive effects of metal contamination (i.e., realistic mixture of Cd, Cu and Zn) and immune challenge mimicking a parasite attack on fish health across biological levels. In addition, we compared ii) the physiological and behavioral responses among five populations of gudgeon fish (Gobio occitaniae) having evolved along a gradient of metal contamination. Results show that single stressors exposure resulted in an increase of immune defenses and oxidative stress at the expense of body mass (contamination) or fish swimming activity (immune challenge). Multiple stressors had fewer interactive effects than expected, especially on physiological traits, but mainly resulted in antagonistic effects on fish swimming activity. Indeed, the immune challenge modified or inhibited the effects of contamination on fish behavior in most populations, suggesting that multiple stressors could reduce behavioral plasticity. Interestingly, the effects of stressors were highly variable among populations, with lower deleterious effects of metal contamination in populations from highly contaminated environments, although the underlying evolutionary mechanisms remain to be investigated. This study highlights the importance of considering multiple stressors effects and intraspecific variability of sensitivity to refine our ability to predict the effects of environmental contaminants on aquatic wildlife.

Indoor air quality ((IAQ) in classrooms was associated with the daily exposure of school-age children who are particularly vulnerable to air pollutants exposure, while few data exist to evaluate classroom indoor air quality nationwide in China. The subsample of the CIEHS 2018 study was performed in 66 classrooms of 22 primary schools nationwide in China. Temperature, relative humidity, PM₂.₅, PM₁₀, CO₂, CO, formaldehyde concentrations, bacteria and fungi were detected in all classrooms by using the instruments that meet the specified accuracy. The ratios of indoor to outdoor (I/O) of PM₂.₅ were calculated in each classroom to identify whether the indoor environment the pollutants comes from outdoors. The indoor PM₂.₅, PM₁₀, CO, HCHO, bacteria and fungi GM concentration are 47.40 μg/m³, 72.91 μg/m³, 0.37 mg/m³, 0.02 mg/m³, 347.51 CFU/m³ and 362.76 CFU/m³, respectively. We observed that there were 66.5%, 52.6%, 22.4%, 1.8%, and 9.6% of the classrooms that exceeded the guideline values of PM₂.₅, PM₁₀, CO₂, HCHO, and bacteria, respectively. It should be attention that all of the classroom's PM₂.₅ concentrations in Shijiazhuang and Nanning, PM₁₀ concentrations in Nanning, CO₂ concentration in Lanzhou were exceeded the suggested values. Bacteria contamination in Shijiazhuang's classrooms is also serious. All classroom CO concentrations meet the requirement. The results indicated that classroom indoor PM₂.₅ was significantly positively correlated with indoor PM₁₀ and CO₂, while was negative correlated with temperature, CO, and fungi. Our results suggest that indoor air pollution in classrooms was a severe problem in Chinese primary schools. It is necessary to strengthen ventilation in the classroom to improve indoor air quality. What's more, a healthy learning environment should be created for primary school students.

In the present study, the daily dose in terms of particle surface area received by citizens living in different low- and middle-income countries, characterized by different lifestyles, habits, and climates, was evaluated. The level of exposure to submicron particles and the dose received by the populations of Accra (Ghana), Cairo (Egypt), Florianopolis (Brazil), and Nur-Sultan (Kazakhstan) were analyzed. A direct exposure assessment approach was adopted to measure the submicron particle concentration levels of volunteers at a personal scale during their daily activities. Non-smoking adult volunteers performing non-industrial jobs were considered. Exposure data were combined with time-activity pattern data (characteristic of each population) and the inhalation rates to estimate the daily dose in terms of particle surface area. The received dose of the populations under investigation varied from 450 mm² (Florianopolis, Brazil) to 1300 mm² (Cairo, Egypt). This work highlights the different contributions of the microenvironments to the daily dose with respect to high-income western populations. It was evident that the contribution of the Cooking & Eating microenvironment to the total exposure (which was previously proven to be one of the main exposure routes for western populations) was only 8%–14% for low- and middle-income populations. In contrast, significant contributions were estimated for Outdoor day and Transport microenvironments (up to 20% for Cairo, Egypt) and the Sleeping & Resting microenvironment (up to 28% for Accra, Ghana), highlighting the effects of different site-specific lifestyles (e.g. time-activity patterns), habits, socioeconomic conditions, climates, and outdoor air quality.

Seven perfluorinated and polyfluorinated substances (PFASs), namely perfluorononanoic acid (PFNA), perfluorodecanoic acid (PFDA), perfluorooctanoic acid (PFOA), perfluorooctane sulfonic acid (PFOS), perfluoroheptanoic acid (PFHpA), perfluorohexanoic acid (PFHxA), and perfluoro-1,10-decanedicarboxylic acid (PFDDA), were evaluated in urine and hair samples from children (age: 4–6 years, N = 53), airborne particles sampled at 17 kindergartens, and tap water and bottled water samples. All samples were collected in Hong Kong. The analytical results suggested widespread PFAS contamination. All target PFASs were detected in at least 32% of urine samples, with geometric mean (GM) concentrations ranging from 0.18 to 2.97 ng/L, and in 100% of drinking water samples at GM concentrations of 0.18–21.1 ng/L. Although PFOS and PFDDA were not detected in hair or air samples, the other target PFASs were detected in 48–70% of hair samples (GM concentrations: 2.40–233 pg/g) and 100% of air samples (GM concentrations: 14.8–536.7 pg/m³). In summary, the highest PFAS concentrations were detected in airborne particles measured in kindergartens. PFOA was the major PFAS detected in hair, urine, and drinking water samples, while PFOA, PFDA, and PFHpA were dominant in airborne particles. Although a significant difference in PFAS concentrations in hair samples was observed between boys and girls (p < .05), no significant sex-related difference in urinary PFAS or paired PFAS (hair/urine) concentrations was observed.

Complex air pollutant sources and distinct meteorological conditions resulted in unique wintertime haze pollution in the Harbin-Changchun (HC) metropolitan area, China’s only national-level city cluster located in the severe cold climate region. In this study, field observation and air quality modeling were combined to investigate fine particulate matter (PM₂.₅) pollution during a six-month long heating season in HC’s central city (Harbin). The model significantly underpredicted PM₂.₅ and organic carbon (by up to ∼230 μg/m³ and 110 μgC/m³, respectively, in terms of daily average) when levoglucosan concentrations were above 0.5 μg/m³. Based on a synthesis of levoglucosan concentrations and fire counts, the large gaps were attributed to underestimation of open burning emissions by the model. However, the model tended to overpredict elemental carbon (more significantly at higher NO₂), likely pointing to an overestimation of vehicle emissions. With increasing levoglucosan, the difference between observed and simulated nitrate (nitrateₒbₛ ‒ nitrateₘₒd, i.e., Δnitrate) showed a transition from negative to positive values. The positive Δnitrate were attributed to underprediction of the open-burning related nitrate, whereas the negative Δnitrate were likely caused by overprediction of nitrate from other sources (presumably vehicle emissions). The dependence of Δnitrate on levoglucosan indicated that with stronger impact of open burning, the overprediction effect was gradually offset and finally overwhelmed. Influence of open burning on sulfate formation was evident as well, but less apparent compared to nitrate. This study illustrates how the uncertainties in open burning emissions will influence PM₂.₅ simulation, on not only primary components but also secondary species.
