Biomarker responses in oysters Crassostrea hongkongensis in relation to metal contamination patterns in the Pearl River Estuary, southern China النص الكامل

The Pearl River Estuary (PRE) is the third largest estuary in China, where estuarine organisms are under metal stress at various biological levels. Based on the metal concentrations measured in oyster Crassostrea hongkongensis, we documented a change in dominance of metal contamination from Cd, Cr, Cu, Ni and Zn to Ag, Cd, Cu and Zn. In general, metal concentrations were higher in upstream stations and displayed a clear up-downstream gradient. Compared to the historical values, we noted the reductions in Cd, Cr and Ni concentrations, and the changing inputs due to evolving industrial activities were responsible for shaping the metal contamination profile in the PRE region. Along with metal concentrations, a suite of biomarkers was analyzed. Among the metals measured in the oyster tissues, Ag, Cd, Cu, Ni and Zn showed the strongest associations with pro-oxidant and oxidative stress responses (superoxide dismutase, lipid peroxidation and lysosomal membrane destabilization) and detoxification responses (glutathione and metallothionein), suggesting that the present metal contamination still exerts significant amount of stress in biota in the PRE. Metal contamination in estuaries in China is still severe compared to other countries, therefore continuous efforts should be taken to monitor the changing metal profiles with necessary control and remediation measures.

China has been faced with severe haze pollution, which is hazardous to human health. Among the air pollutants, PM2.5 (particles with an aerodynamic diameter ≤ 2.5 μm) is the most dangerous because of its toxicity and impact on human health and ecosystems. However, there has been limited research on PM2.5 particle toxicity. In the present study, we collected daily PM2.5 samples from January 1 to March 31, 2018 and selected samples to extract water-soluble species, including SO42−, NO3−, WSOC, and NH4+. These samples represented clean, good, slight, moderate, and heavy pollution days. After extraction using an ultrasonic method, PM2.5 solutions were obtained. We used Chlorella as the test algae and studied the content of chlorophyll a, as well as the variation in fluorescence when they were placed into the PM2.5 extraction solution, and their submicroscopic structure was analyzed using transmission electron microscopy (TEM). The results showed that when the air quality was relatively clean and good (PM2.5 concentration ≤ 75 μg m−3), the PM2.5 extraction solutions had no inhibiting effects on Chlorella, whereas when the air quality was polluted (PM2.5 concentration > 75 μg m−3) and heavily polluted (PM2.5 concentration > 150 μg m−3), with increasing PM2.5 concentrations and exposure time, the chlorophyll a content in Chlorella decreased. Moreover, the maximum photochemical quantum yield (Fv/Fm) of Chlorella obviously decreased, indicating chlorophyll inhibition during polluted days with increasing PM2.5 concentrations. The effects on the chlorophyll fluorescence parameters were also obvious, leading to an increase of energy dissipated per unit reaction center (DIo/RC), suggesting that Chlorella could survive when exposed to PM2.5 solutions, whereas the physiological activities were significantly inhibited. The TEM analysis showed that there were few effects on Chlorella cell microstructure during clean days, whereas plasmolysis occurred during light- and medium-polluted days. With increasing pollution levels, plasmolysis became more and more apparent, until the organelles inside the cells were thoroughly destroyed and most of the parts could not be recognized.

Feammox, i.e., Fe(III) reduction coupled to anaerobic ammonium oxidation, is a potential alternative to ammonium removal in natural and artificial ecosystems. However, the efficiency of Feammox is quite low to restrain its practical application in wastewater/solid disposal. In this study, three batch experiments, including control (Fe₂O₃/AQDS-free), Fe₂O₃ group (25 mM Fe₂O₃ only) and AQDS-Fe₂O₃ group (25 mM Fe₂O₃ and 0.6 mM AQDS), were conducted in 200 mL serum vials to explore whether AQDS can promote Feammox. Results showed that the nitrogen removal efficiency of the AQDS-Fe₂O₃ group was 82.6%, compared with 64.3% of the Fe₂O₃ group and 46.0% in the control. AH₂QDS, the reduced state of AQDS, was detected in the AQDS-Fe₂O₃ group. Another experiment indicated that AH₂QDS was oxidized back to AQDS by Fe₂O₃. These results suggested that AQDS/AH₂QDS had been serving as electron shuttles between ammonium and Fe₂O₃ to successively forward the oxidation of NH₄⁺. X-ray diffraction analysis showed that new Fe(III) species were found in the systems, implying that a Fe(II)/Fe(III) cycle also occurred. In agreement, both iron-reducing and oxidizing bacteria were detected in the systems.

Transformation of organic microcontaminants (OMCs) during wastewater treatments results in the generation of transformation products (TPs), which can be more persistent than parent compounds. Due to reuse of reclaimed wastewater (RWW) for crop irrigation, OMCs and TPs are released in soils being capable to translocate to crops. Furthermore, OMCs are also susceptible to transformation once they reach the soil or crops. The recalcitrant antiepileptic carbamazepine (CBZ) and some of its frequently reported TPs have been found in agricultural systems. However, there is no knowledge about the fate in reuse practices of multiple CBZ TPs that can be formed during wastewater treatment processes. For the first time, this work presents a study of the behavior of CBZ TPs generated after a conventional Ultraviolet-C (UVC) treatment in an agricultural environment. The UVC-treated water was used for the irrigation of lettuces grown under controlled conditions. The latter was compared to the fate of TPs generated in the peat and plant by irrigation with non-treated water containing CBZ. A suspect screening strategy was developed to identify the TPs using liquid chromatography coupled to quadrupole-time-of-flight (LC-QTOF-MS). The results revealed the presence of 24 TPs, 22 in UVC-treated water, 11 in peat and 9 in lettuce leaves. 4 of the TPs identified in peat (iminostilbene, TP 271B, TP 285A-B); and 3 in leaves (10–11 dihydrocarbamazepine, TP 271A-B) were not previously reported in soils or edible parts of crops, respectively. Comparing the TPs found in peat and lettuces derived from both irrigation conditions, no significant differences regarding TPs formation or occurrence were observed. UVC treatment did not contribute to the formation of different TPs than those generated by transformation or metabolism of CBZ in peat or plant material. This research improves the current knowledge on the fate of CBZ TPs in agricultural systems because of reuse practices.

Although amino modified nanopolystyrene could cause toxicity on environmental organisms, the effect of amino modification on nanopolystyrene toxicity is still largely unclear. We here employed Caenorhabditis elegans as an animal model to compare the effects between pristine and amino modified nanopolystyrene particles in inducing reproductive toxicity. Nanopolystyrene (35 nm) could cause the damage on gonad development as indicated by the endpoints of number of total germline cells, length of gonad arm, and relative area of gonad arm. Nanopolystyrene exposure also reduced the reproductive capacity as reflected by the endpoints of brood size and number of fertilized eggs in uterus. Moreover, amino modification enhanced nanopolystyrene toxicity on both the gonad development and the reproductive capacity. Additionally, induction of germline apoptosis and formation of germline DNA damage contributed to the enhancement of nanopolystyrene toxicity in reducing reproductive capacity by amino modification. Our results highlight the potential environmental risk of amino modified nanopolystyrene in inducing reproductive toxicity on gonad development and reproductive capacity of environmental organisms.

Heavy metal data measured by portable X-ray fluorescence (PXRF), especially by in-situ PXRF, are usually affected by multiple soil factors, such as soil moisture (SM), soil organic matter (SOM), and soil particle size (SPZ). Thus, a correction may be needed. However, traditionally-used correction methods, such as non-spatial linear regression (LR), cannot effectively correct the spatially non-stationary influences of the related soil factors on PXRF analysis. Moreover, these correction methods are not robust to outliers. In this study, robust geographically weighted regression (RGWR) was used to correct in-situ and ex-situ PXRF data of soil Pb in a peri-urban agricultural area of Wuhan City, China. The accuracy of the corrected PXRF data by RGWR was compared with those by non-spatial and spatial but non-robust methods (i.e., LR and GWR). In addition, to find an appropriate method of using the corrected PXRF data for more accurate spatial prediction, we compared robust ordinary kriging with the corrected PXRF data as part of hard data (ROK-CPXRF) and robust ordinary cokriging with the corrected PXRF data as auxiliary soft data (RCoK-CPXRF). Results showed that (i) RGWR obtained higher correction accuracy than LR and GWR on both the in-situ and ex-situ PXRF data; (ii) the accuracy of the RGWR-corrected in-situ PXRF data was increased nearly to that of the RGWR-corrected ex-situ PXRF data; (iii) given the same amount of sample data, ROK-CPXRF obtained higher prediction accuracy than RCoK-CPXRF. It is concluded that the methods suggested in this study may largely promote the application of in-situ PXRF technique for rapid and accurate soil heavy metal investigation in large-scale areas.

Cu, Mn, Fe, Zn, Cd and Pb levels were measured in liver and muscle samples of Rainbow Trout Oncorhynchus mykiss collected from three watersheds with different land-uses: native forest, exotic plantation, and agriculture in Chile, during January, April, July, and October 2012. Cd and Pb levels were not detected in the liver and muscle, probably since they are under the detection limits. Higher metal concentrations (liver-muscle tissues) were detected in samples from agriculture and exotic plantation streams, whereas trout from native forest streams had lower metal concentrations. Higher metal concentrations were detected in liver tissue compared to muscle tissue, and both negatively correlated to the length and weight of the fish. This suggest the liver had higher ability to accumulate Cu, Mn, Fe and Zn compared to muscle tissue. The concentration range of Fe and Zn recorded in the muscle are within the range reported by other authors, whereas Mn and Cu concentrations are higher than reported in the literature. However, at all sites the concentration of selected metals were below the limits permitted by current legislation (FAO), and therefore did not put the human population at risk, suggesting that is eating wild rainbow trout safe in Chile.

Coke oven emissions (COEs) are common particle pollutants in occupational environment and the major constituents of COEs are polycyclic aromatic hydrocarbons (PAHs). Previously, we identified aberrant methylation of the fms related tyrosine kinase 1 (FLT1) gene over the course of benzo(a)pyrene (BaP)-induced cell transformation via genome-wide methylation array. To quantify FLT1 methylation, we established a bisulfite pyrosequencing assay and examined the FLT1 hypermethylation in several human cancers. The results revealed that 70.0% (21/30 pairs) of lung cancers harbored hypermethylated FLT1 and concomitant suppression of gene expression compared to the adjacent tissues. This implies that FLT1 hypermethylation might play a role in malignant cell transformation. In addition, FLT1 hypermethylation and gene suppression appeared in primary human lymphocytes in a dose-response manner following COEs treatment. To explore whether FLT1 methylation is correlated with COEs exposure and DNA damage, we recruited 144 male subjects who had been exposed to high levels of COEs and 84 male control subjects. Notably, the FLT1 methylation in peripheral blood lymphocytes (PBLCs) of the COEs-exposed group (19.8 ± 3.2%) was enhanced by 17.9% compared to that of the control group (16.8 ± 2.8%) (P < 0.001). The FLT1 methylation status was positively correlated with urinary 1-hydroxypyrene (1-OHP) levels, an internal exposure marker of PAHs (β = 0.029, 95% CI = 0.010–0.048, P = 0.003) and positively correlated with DNA damage (βOTM = 0.024, 95% CI = 0.007–0.040, P = 0.005; βTₐᵢₗ DNA = 0.035, 95% CI = 0.0017–0.054, P < 0.001) indicated by comet assay. Taken together, these findings indicate that FLT1 might be a tumor suppressor, and its hypermethylation might contribute to PAHs-induced carcinogenicity.

Civil aircraft emissions during landing and takeoff (LTO) are important air pollutant sources, but have been given insufficient attention in China. Accurate estimation of these emissions is limited by a lack of important parameters, such as detailed flight information and the dynamic time in climb and approach modes during LTO that are dependent on mixing layer height (MLH). We developed a flight-time/flight-height relationship using real-time height information in Aircraft Meteorological Data Relay data, and then calculated the actual time for each flight in those two modes based on the actual MLH from meteorological observation. Hourly emissions of civil aircraft were then estimated based on the database of each flight. Total emissions of NOx, CO, SO2, HC and PM from LTO cycles of domestic flights in China during 2015 were 37.78 Gg, 30.25 Gg, 12.00 Gg, 2.38 Gg and 0.75 Gg, respectively. Substantial monthly, daily and hourly variations of emissions due to the flight schedule as well as MLH were calculated. Large differences were found between the new estimation and emissions calculated based on traditional method. Compared with the emissions estimated based on default parameter obtained from International Civil Aviation Organization, the average difference of annual emission among airports with new estimation for various pollutants was approximately 30.3% in climb mode and 81.4% in approach mode; compared with the emissions estimated based on the method proposed by China National Guide, the average difference of annual emission among airports were 37.4% (NOx), 8.4% (CO), 73.1% (HC) and 58.1% (PM) during LTO process. The monthly airport-specific emissions per LTO were also proposed. These can provide necessary and meaningful support for the revision of the values in National Guide.

Hyperbranched polymers are nanomaterials belonging to the class of dendritic architectures with increasing applications in many diverse fields. We studied the toxicity of two hyperbranched polymers to the freshwater crustacean Daphnia magna. A hyperbranched hydroxyl-terminated polyester and a commercial hyperbranched polyamidoamine, Helux-3316 were tested for the acute immobilization of daphnids, the overproduction of reactive oxygen species and the activity of the antioxidant enzymes catalase and glutathione S-transferase. The effect for D. magna immobilization was higher for the hyperbranched polyamidoamine Helux-3316, which was attributed to the presence of primary amino groups on its surface. Following exposure to both hyperbranched polymers, a clear overproduction of reactive oxygen species took place accompanied by concentration-dependent enzymatic antioxidant response. Our results showed that the overproduction of reactive oxygen species activated antioxidant defence mechanisms and was responsible for the immobilization of daphnids exposed to both hyperbranched polymers. We showed evidence of the uptake of fluorescently labelled Helux-3316 that accumulated into the gastrointestinal tract of D. magna, and its removal via excretion within fecal pellets. This is the first work reporting the internalization of hyperbranched polymers in aquatic organisms.
