The cytotoxicity, genotoxicity and oxidative stress of malachite green (MG) was investigated using the fish Channa striata kidney (CSK) and Channa striata gill (CSG) cell lines. Five concentrations ranging from 0.001 to 10 μg mL⁻¹were tested in three independent experiments. Cytotoxicity was assessed by 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, Rhodamine 123 and Alamar Blue. The mitochondrial changes and apoptosis of MG-exposed cells were observed by Rhodamine 123 and acridine orange/ethidium bromide (AO/EB) staining, respectively. In vitro potential DNA damaging effect of MG was tested using comet assay. Mitochondrial damage, apoptosis and DNA fragmentation increased in a concentration-dependent manner. Additionally, DNA electrophoretic mobility experiments were carried out to study the binding effect of MG to double-stranded DNA (dsDNA) of cells. DNA shift mobility experiments showed that MG is capable of strongly binding to linear dsDNA causing its degradation. Biochemical parameters such as lipid peroxidation (MDA), catalase (CAT) activity and reduced glutathione (GSH) levels were evaluated after exposure to MG. In CSK and CSG cell lines exposed to MG for 48 h, a significant increase in lipid peroxidation, which might be associated with decreased levels of reduced glutathione and catalase activity in these cell lines (p < 0.001), was observed.

In this work, a suite of diagnostic biomarkers was applied to seven cetacean species to evaluate the role of the feeding habits and migratory behavior in the toxicological status of these species from the Gulf of California, Mexico. We investigate the interspecific differences in cytochrome P450 1A1 and 2B (CYP1A1 and CYP2B, respectively), aryl hydrocarbon receptor and E2F transcription factor 1 and the contaminants levels [organochlorine compounds, polybrominated diphenyl ethers (PBDEs) and polycyclic aromatic hydrocarbons (PAHs)] in four odontocete species (common bottlenose dolphin, long-beaked common dolphin, sperm whale and killer whale) and three mysticete species (blue whale, fin whale, and Bryde’s whale) using skin biopsy. Differences in contaminant levels and molecular biomarker responses between the odontocete and mysticete species have been pointed out. The canonical discriminant analysis on principal component analysis factors, performed to reveal clustering variables, shows that odontocete are characterised by the highest levels of lipophilic contaminants compared to the mysticete, with the highest levels of polychlorinated biphenyls, dichlorodiphenyltrichloroethanes and PBDEs detected in killer whale and the lowest levels in Bryde’s whale. The biomarker data show interspecific differences amongst the seven species, revealing highest CYP1A and CYP2B protein levels in the mysticete fish-eating species (Bryde’s whale). In conclusion, three main factors seem to regulate the biomarker responses in these species: (a) the inductive ability of persistent organic pollutants and PAHs; (b) the different evolutionary process of the two CYPs related to the different feeding habits of the species; (c) the migratory/resident behaviour of the mysticete species in this area.

Bioindicators can reflect the effects of pollutants on their metabolism, being widely used to assess environmental stress. In this sense, the crab Procambarus clarkii has been previously proposed to monitor the contamination in Doñana National Park (southwest Spain) using conventional biomarkers. In this work, a metabolomic approach based on direct infusion mass spectrometry, which allows an easy and quick study of a large number of metabolites in a single run, was used for pollution assessment of this area, considering the biological response of this organism to contamination. In addition, metal accumulation in crab tissues was determined. Thus, the integrated analysis of metabolomic and metallomic data enabled the study of metabolic response of the organism against pollution. Several metabolites were discovered as potential biomarkers of pollution, such as decreased levels of carnosine, alanine, niacinamide, acetoacetate, pantothenic acid, ascorbate, glucose-6-phosphate, arginine, glucose, lactate, phospholipids, and tryglicerides, as well as elevated levels of acetyl carnitine, phosphocholine, choline, and uric acid. In this way, metal-induced toxicity could be related to metabolic impairments, principally oxidative stress, metabolic dysfunction, and dyslipidemia.

Cakmak, Dragan | Perovic, Veljko | Saljnikov, Elmira | Jaramaz, Darko | Sikiric, Biljana
Elevated arsenic (As) concentrations in soil are often found in the vicinity of certain mineral deposits that have been, or are currently, under exploitation, regardless of the target resource. Detailed study of such areas for safe agriculture requires considerable financial costs and long periods of time. Application of an appropriate spatial model that describes the behavior of arsenic in soil and plants can significantly ease the whole investigation process. This paper presents a model of ecological security of an area that, in the past, was an antimony mine and has a naturally high content of arsenic. For simulation and modeling the geographic information science (GIS) technology with the inserted predictors influencing the accessibility of As and its content in plants was used. The results obtained were the following: (1) a categorization of contaminated soils according to soil properties was developed; (2) the proposed methodology allows focusing on particular suspect area saving an energy and human resource input; and (3) new safe areas for growing crops in contaminated area were modeled. The application of the proposed model of As solubility to various crops grown around a former antimony mine near the village of Lisa, southwest Serbia showed that significant expansion of the areas suitable for growing potato, raspberry, and pasture was possible.

Alier, M. | Osto, M Dall | Lin, Y.-H. | Surratt, J. D. | Tauler, R. | Grimalt, J. O. | van Drooge, B. L.
Water-soluble organic compounds (WSOCs), represented by anhydro-saccharides, dicarboxylic acids, and polyols, were analyzed by gas chromatography interfaced to mass spectrometry in extracts from 103 PM₁ and 22 PM₂.₅ filter samples collected in an urban background and road site in Barcelona (Spain) and an urban background site in Los Angeles (USA), respectively, during 1-month intensive sampling campaigns in 2010. Both locations have similar Mediterranean climates, with relatively high solar radiation and frequent anti-cyclonic conditions, and are influenced by a complex mixture of emission sources. Multivariate curve resolution-alternating least squares analyses were applied on the database in order to resolve differences and similarities in WSOC compositions in the studied sites. Five consistent clusters for the analyzed compounds were obtained, representing primary regional biomass burning organic carbon, three secondary organic components (aged SOC, isoprene SOC, and α-pinene SOC), and a less clear component, called urban oxygenated organic carbon. This last component is probably influenced by in situ urban activities, such as food cooking and traffic emissions and oxidation processes.
اظهر المزيد [+] اقل [-]The ability of biologically based wastewater treatment systems to remove emerging organic contaminants—a review
2014
Garcia-Rodríguez, Aida | Matamoros, Víctor | Fontàs, Clàudia | Salvadó, Victòria
Biologically based wastewater treatment systems are considered a sustainable, cost-effective alternative to conventional wastewater treatment systems. These systems have been used and studied for the treatment of urban sewage from small communities, and recently, it has been reported that they can also effectively remove emerging organic contaminants (EOCs). EOCs are a new group of unregulated contaminants which include pharmaceutical and personal care products, some pesticides, veterinary products, and industrial compounds among others that are thought to have long-term adverse effects on human health and ecosystems. This review is focused on reporting the ability of biologically based wastewater treatment systems to remove EOCs and the main elimination mechanisms and degradation processes (i.e., biodegradation, photodegradation, phytoremediation, and sorption) taking place in constructed wetlands, ponds, and Daphnia and fungal reactors.

Kim, M. P. (Maksim Pavlovich) | Guerra, P. | Alaee, M. | Smyth, S. A.
Four novel brominated flame retardants (NBFRs), i.e., decabromodiphenylethane (DBDPE), 1,2-bis(2,4,6-tribromophenoxy) ethane (BTBPE), pentabromoethylbenzene (PBEB), and hexabromobenzene (HBB) were studied in 377 liquid samples and 288 solid samples collected from 20 wastewater treatment plants. Lagoon, primary, secondary, and advanced treatment processes were included, in order to investigate NBFR occurrence and the effects of WWTP operational conditions on NBFR removal. Median influent and effluent levels were 14 to 3,700 and 1.0 to 180 pg/L respectively, with DBDPE being the highest in both. Overall median removal efficiencies for DBDPE, BTBPE, HBB, and PBEB across all process types were 81 to 93, 76 to 98, 61 to 97, and 54 to 97 %, respectively with advanced treatment processes obtaining the best removals. NBFRs removal was related to retention time, surface loading rate, and biomass concentration. Median NBFR levels in treated biosolids were 80 to 32,000 pg/g, influenced by solids treatment processes.

2014
Akhmat, Ghulam | Zaman, Khalid | Shukui, Tan | Irfan, Danish | Khan, Muhammad Mushtaq
The objective of the study is to examine the causal relationship between energy consumption and environmental pollutants in selected South Asian Association for Regional Cooperation (SAARC) countries, namely, Bangladesh, India, Nepal, Pakistan, and Srilanka, over the period of 1975–2011. The results indicate that energy consumption acts as an important driver to increase environmental pollutants in SAARC countries. Granger causality runs from energy consumption to environmental pollutants, but not vice versa, except carbon dioxide (CO₂) emissions in Nepal where there exists a bidirectional causality between CO₂ and energy consumption. Methane emissions in Bangladesh, Pakistan, and Srilanka and extreme temperature in India and Srilanka do not Granger cause energy consumption via both routes, which holds neutrality hypothesis. Variance decomposition analysis shows that among all the environmental indicators, CO₂ in Bangladesh and Nepal exerts the largest contribution to changes in electric power consumption. Average precipitation in India, methane emissions in Pakistan, and extreme temperature in Srilanka exert the largest contribution.

2014
Lancellotti, I. | Toschi, T. | Passaglia, E. | Barbieri, L.
The principal plant nutrients are phosphorous, nitrogen and potassium. Among these compounds, phosphorous is the most critical: it reacts rapidly, becoming an insoluble compound. The combination of zeolitites with phosphate materials (zeoponic substrate) agrees to a gradual and controlled phosphorous release in soils: phosphorous for plant uptake is released by the combination of dissolution and ion-exchange reactions. Animal bone ashes, rich in phosphorous and leached alone, release little amounts of soluble phosphorous and a great deal of alkaline sodium and potassium. Concerning chabazitic-zeolitite, it encourages a both gradual and growing soluble phosphorous release from animal bone ashes, in accordance with clinoptilolitic- and phillipsitic-zeolitite abilities; in particular, that release increases, thanks to both a higher zeolitite/bone ash ratio and ammonium enrichment of zeolitite. The use of zeolitite is environmentally sustainable in Italy because large amounts of deposits of zeolitite were present in Italy.

Chronic exposure to heavy metals has long been recognized as being capable of increasing head and neck cancer (HNC) incidence, such as laryngeal (LC) and nasopharyngeal (NPC), among exposed human populations. The aim of the present study was to evaluate the concentrations of arsenic (As) and cadmium (Cd) in the blood of 145 patients (LC and NPC) and 351 controls in order to establish a potential relationship between these factors and the occurrence of LC and NPC. Mean blood levels of As and Cd in patients (5.67 and 3.51 μg/L, respectively) were significantly higher than those of controls (1.57 and 0.74 μg/L, respectively). The blood levels of As and Cd were mostly significantly higher than those of controls (p < 0.05) after controlling the other risk factors of HNC including tobacco smoking and chewing, and alcohol drinking. Cd levels in blood increase significantly with the number of occupational exposure years for patients (p < 0.05). However, seafood was not found to be contributing as an exposure source. Among these risk factors, smoking (>30 pack years) and occupational exposure (>20 years) presented the most significant association with HNC (OR = 10.22 and 10.38, respectively, p < 0.001). Cd level in blood sample of cases that are occupationally exposed/tobacco users (smokers and chewers) were higher than that of non-occupationally exposed/nontobacco users (p < 0.001). The logistic regression model illustrated that HNC (LC + NPC) was significantly associated with blood levels of As (OR = 2.41, p < 0.001) and Cd (OR = 4.95, p < 0.001).
