Application of a luminescence-based biosensor for assessing naphthalene biodegradation in soils from a manufactured gas plant النص الكامل

Despite numerous reviews suggesting that microbial biosensors could be used in many environmental applications, in reality they have failed to be used for which they were designed. In part this is because most of these sensors perform in an aqueous phase and a buffered medium, which is in contrast to the nature of genuine environmental systems. In this study, a range of non-exhaustive extraction techniques (NEETs) were assessed for (i) compatibility with a naphthalene responsive biosensor and (ii) correlation with naphthalene biodegradation. The NEETs removed a portion of the total soil naphthalene in the order of methanol > HPCD > βCD > water. To place the biosensor performance to NEETs in context, a biodegradation experiment was carried out using historically contaminated soils. By coupling the HPCD extraction with the biosensor, it was possible to assess the fraction of the naphthalene capable of undergoing microbial degradation in soil. Exposure of microbial biosensors to cyclodextrin solutions allows the assessment of the degradable fraction of contaminants in soil.

Among air pollutants, ozone is the most important stressor to vegetation, which undergoes damage and biomass reduction after penetration of ozone molecules into the leaf tissues through the stomata. Stomatal ozone fluxes are considered the governing factor needed to assess risk to plant health due to ozone. Although this parameter may be calculated by modeling, direct measurements are scarce. Moreover, southern European situations, especially regarding Italy, require special attention due to the decoupling between ozone concentrations and fluxes. This work reviews ozone flux measurements made during the last 15 years through Italy. The dependence of ozone fluxes on environmental factors such as water supply is stronger than the dependence on the surface covering species.

Between 1992 and 2004, air contamination with lead (Pb), cadmium (Cd), and zinc (Zn) in Warsaw, Poland, was monitored annually with moss (Sphagnum fallax) bags on a network of 230 sites covering the entire city. During the study the highest contamination was near the Warszawa Steel Mill, northwestern Warsaw. Lead concentrations in moss bags decreased in time, while those of Cd and Zn did not show clear trends. Between 1994 and 2004, Pb, Cd, and Zn were also monitored in the Crimean linden (Tilia Euchlora) foliage along the main city avenue and in a northwestern warsaw park. Lead concentrations decreased more near the avenue than in the park, indicating that the phasing-out of leaded gasoline had a major effect on reduced Pb contamination in Warsaw. At the same time, foliar concentrations of Cd and Zn in both areas decreased much less. Between 1992 and 2004, concentrations of Pb decreased, while those of Cd and Zn remained little changed in moss bags and linden foliage exposed to ambient air in Warsaw, Poland.

Soil organic matter (SOM) is generally believed not to influence the sorption of glyphosate in soil. To get a closer look on the dynamics between glyphosate and SOM, we used three approaches: I. Sorption studies with seven purified soil humic fractions showed that these could sorb glyphosate and that the aromatic content, possibly phenolic groups, seems to aid the sorption. II. Sorption studies with six whole soils and with SOM removed showed that several soil parameters including SOM are responsible for the strong sorption of glyphosate in soils. III. After an 80 day fate experiment, ~40% of the added glyphosate was associated with the humic and fulvic acid fractions in the sandy soils, while this was the case for only ~10% of the added glyphosate in the clayey soils. Glyphosate sorbed to humic substances in the natural soils seemed to be easier desorbed than glyphosate sorbed to amorphous Fe/Al-oxides.

This study investigated the dissipation kinetics of oxytetracycline in soils under aerobic and anoxic conditions. Laboratory experiments showed that the dissipation of oxytetracycline in soil followed firstorder reaction kinetics and its dissipation rates decreased with increasing concentration. Oxytetracycline dissipated faster in soil under aerobic conditions than under anoxic conditions. The half-lives for oxytetracycline in soil under aerobic conditions ranged between 29 and 56 days for non-sterile treatments and 99-120 days for sterile treatments, while under anoxic conditions the half-lives of oxytetracycline ranged between 43 and 62 days in the non-sterile soil and between 69 and 104 days in the sterile soil. This suggests microbes can degrade oxytetracycline in agricultural soil. Abiotic factors such as strong sorption onto soil components also played a role in the dissipation of oxytetracycline in soil.

The effects on the growth of tomato seedlings and cadmium accumulation of earthworm mucus and a solution of amino acids matching those in earthworm mucus was studied through a hydroponic experiment. The experiment included four treatments: 5 mg Cd L⁻¹ (CC), 5 mg Cd L⁻¹ þ 100 mL L⁻¹ earthworm mucus (CE), 5 mg Cd L⁻¹ þ 100 mL L⁻¹ amino acids solution (CA) and the control (CK). Results showed that, compared with CC treatment, either earthworm mucus or amino acids significantly increased tomato seedling growth and Cd accumulation but the increase was much higher in the CE treatment compared with the CA treatment. This may be due to earthworm mucus and amino acids significantly increasing the chlorophyll content, antioxidative enzyme activities, and essential microelement uptake and transport in the tomato seedlings. The much greater increase in the effect of earthworm mucus compared with amino acid treatments may be due to IAA-like substances in earthworm mucus.

Agricultural landscapes, although often highly altered in nature, provide habitat for many species of amphibian. However, the persistence and health of amphibian populations are likely to be compromised by the escalating use of pesticides and other agricultural chemicals. This review examines some of the issues relating to exposure of amphibian populations to these chemicals and places emphasis on mechanisms of toxicity. Several mechanisms are highlighted, including those that may disrupt thyroid activity, retinoid pathways, and sexual differentiation. Special emphasis is also placed on the various interactions that may occur between different agro-chemicals and between chemicals and other environmental factors. We also examine the indirect effects on amphibian populations that occur when their surrounding pond communities are altered by chemicals. The literature on the various mechanisms by which amphibians may be affected by agricultural chemicals is reviewed.

The study was prompted to characterize the B-type esterase activities in the terrestrial snail Xeropicta derbentina and to evaluate its sensitivity to organophosphorus and carbamate pesticides. Specific cholinesterase and carboxylesterase activities were mainly obtained with acetylthiocholine (Km = 77.2 mM; Vmax = 38.2 mU/mg protein) and 1-naphthyl acetate (Km = 222 mM, Vmax = 1095 mU/mg protein) substrates, respectively. Acetylcholinesterase activity was concentration-dependently inhibited by chlorpyrifos-oxon, dichlorvos, carbaryl and carbofuran (IC50 = 1.35 x 10-5-3.80 x 10-8 M). The organophosphate-inhibited acetylcholinesterase activity was reactivated in the presence of pyridine-2-aldoxime methochloride. Carboxylesterase activity was inhibited by organophosphorus insecticides (IC50 = 1.20 x 10-5-2.98 x 10-8 M) but not by carbamates. B-esterase-specific differences in the inhibition by organophosphates and carbamates are discussed with respect to the buffering capacity of the carboxylesterase to reduce pesticide toxicity. These results suggest that B-type esterases in X. derbentina are suitable biomarkers of pesticide exposure and that this snail could be used as sentinel species in field monitoring of Mediterranean climate regions. The level of production of nanoparticles will inevitably lead to their appearance in air, water, soils, and organisms. A theoretical framework that relates properties of nanoparticles to their biological effects is needed to identify possible risks to human health and the environment. This paper considers the properties of dispersed metallic nanoparticles and highlights the relationship between the chemical stability of these nanoparticles and their in vitro toxicity. Analysis of published data suggests that chemically stable metallic nanoparticles have no significant cellular toxicity, whereas nanoparticles able to be oxidized, reduced or dissolved are cytotoxic and even genotoxic for cellular organisms. The ability of metallic nanoparticles to be oxidized, reduced or dissolved in biological media can be used to predict their toxicity in vitro.

In the frame of a French monitoring program, tri- to deca- polybromodiphenylethers (PBDE) have been measured in maternal and cord serum, adipose tissue, and breast milk samples, collected from 93 volunteer women during caesarean deliveries. The seven major tri- to heptaBDE (BDE-28, 47, 99, 100, 153, 154, and 183) were detected in adipose tissue and breast milk with cumulated median values of 2.59 and 2.51 ng g-1 l w. Nine highly brominated octa- to decaBDE (BDE-196, 197, 201, 202, 203, 206, 207, 208 and 209) was performed in the same samples, with cumulated median values of 2.73 and 3.39 ng g-1 l w in adipose tissue and breast milk, respectively. At this opposite, median levels of octa- to decaBDE in maternal and cord serum appeared significantly higher than the levels of tri- to heptaBDE in the same matrices, i.e. 8.85 and 12.34 versus 0.98 and 0.69 ng g-1 l w, respectively. This study provides the first data at the French level regarding the occurrence of PBDE in human, and demonstrates the exposure of both human foetus at late stage of pregnancy and newborn at early stages of life through breastfeeding with special emphasis on the presence of highly brominated compounds.
