Harbor porpoises (Phocoena phocoena) in the North and Baltic Seas are exposed to anthropogenic influences including acoustic stress and environmental contaminants. In order to evaluate immune responses in healthy and diseased harbor porpoise cells, cytokine expression analyses and lymphocyte proliferation assays, together with toxicological analyses were performed in stranded and bycaught animals as well as in animals kept in permanent human care. Severely diseased harbor porpoises showed a reduced proliferative capacity of peripheral blood lymphocytes together with diminished transcription of transforming growth factor-β and tumor necrosis factor-α compared to healthy controls. Toxicological analyses revealed accumulation of polychlorinated biphenyls (PCBs), dichlorodiphenyldichloroethylene (DDE), and dichlorodiphenyltrichloroethane (DDT) in harbor porpoise blood samples. Correlation analyses between blood organochlorine levels and immune parameters revealed no direct effects of xenobiotics upon lymphocyte proliferation or cytokine transcription, respectively. Results reveal an impaired function of peripheral blood leukocytes in severely diseased harbor porpoises, indicating immune exhaustion and increased disease susceptibility.

The biodegradation of polychlorinated-p-dioxins and dibenzofurans (PCDD/Fs) has been recently demonstrated in a single reactor under hypoxic conditions. Maintaining hypoxic conditions through periodic aerations results in a marked fluctuation of reduction–oxidation (redox) potential. To further assess the effects of redox fluctuations, we operated two fed-batch continuously stirred tank reactors (CSTRs) with sophisticated redox controls at different anoxic/oxic fluctuations to reduce PCDD/Fs in contaminated soil. The results of long-term reactor operation showed that the CSTR with redox fluctuations at a narrow range (−63 ± 68 mV) (CSTR_A) revealed a higher substrate hydrolysis level and PCDD/F degradation rate than did the CSTR with a redox potential that fluctuated at a broad range (−13 ± 118 mV) (CSTR_B). In accordance with analyses of bacterial 16S rRNA genes, the designated hypoxic conditions with added compost supported survival of bacterial populations at a density of approximately 10⁹ copies/g slurry. The evolved core microbiome was dominated by anoxic/oxic fluctuation-adapted Bacteroidetes, Alphaproteobacteria, and Actinobacteria, with higher species diversity and functionality, including hydrolysis and degradation of dioxin-like compounds in CSTR_A than in CSTR_B. Taken together, the overall results of this study expand the understanding of redox fluctuations in association with the degradation of recalcitrant substrates in soil and the corresponding microbiome.

Eutrophication of lakes has become one of the world's most serious environmental problems, resulting in an urgent need to monitor and provide safeguards to control water quality. Results from analysis of lake trophic status based on calculated throphic state index (TSI) showed that 69.5% of the surveyed 277 lakes were in a state of eutrophication. Significant logarithmic relationships between light absorption of optically active components (aOACs) and TSI (R2 = 0.78) existed: TSI = 13.64 × ln(aOACs)+43.24, and the regression relationship between aOACs and TSI had a better degree of fit (R2) than the currently used reflectance-TSI relationship. aOACs appeared to be a good predictor of TSI estimation in lake ecosystems. The relationship coefficient (aOACs-TSI) slightly varied with lake type, and relationships in saline lakes and phy-type lakes were shown to be more robust than the relationship with the total lake data. This study highlights the quantification of the trophic status in lakes using aOACs, which realized the monitoring of trophic status in lakes using inherent optical properties on a large-scale. To our knowledge this is the first investigation to assess the variability of trophic status in lakes across China. The assessment trophic state of lakes based on aOACs provides a new way to monitor the trophic status of lakes, and findings may have applications for monitoring large-scale and long-term trophic patterns in lakes using remote sensing techniques.

Various crustaceans are farmed using aquaculture, and food deprivation or fasting can occur due to changing of environmental or management strategies. However, the molecular mechanisms underlying responses to starvation in crustaceans remain unclear. To address this, 12 hepatopancreas transcriptomes were compared for oriental river prawn (Macrobrachium nipponense) from four fasting stages (0, 7, 14 and 21 d). Gene Ontology functional annotation and Kyoto Encyclopaedia of Genes and Genomes pathway enrichment analysis of differentially expressed genes were subsequently performed. During the early stages of starvation (0–7 d), drug metabolism via the cytochrome P450 pathway and metabolism of xenobiotics by the cytochrome P450 pathway were enriched, suggesting that they metabolised compounds generated under starvation stress. As starvation proceeded (7–14 d), the retinol (vitamin A) metabolism pathway was activated, based on three up-regulated genes (CYP3, ADH and UGT), along with the two p450 pathways. Meanwhile, vitamin A was gradually consumed. As acute starvation was reached (14–21 d), vitamin A deficiency decreased the mRNA expression levels of IGF-I that is involved in the mTOR signalling pathway, which ultimately affected the growth and development of M. nipponense. Our results implicate drug/xenobiotic metabolism by cytochrome P450s in adaptation to starvation stress. Furthermore, metabolic cascades (CYP and retinol pathways) and growth (mTOR signalling) pathways are clearly triggered in crustaceans during starvation. The findings expand our understanding of the genes associated with hepatopancreas functioning in M. nipponense, and the underlying molecular mechanisms that govern the responses of crustaceans to starvation stress.

Heavy metal contamination is widespread in Bangladesh. Previous studies have observed lead increases blood pressure over time. However, the role of other metal contaminants and essential micronutrients, which could also adversely affect blood pressure or act as protective factors, is understudied. We therefore evaluated the associations of lead, manganese, and selenium with blood and pulse pressure trajectories. We prospectively followed placebo-assigned participants nested within a randomized trial for the prevention of arsenic-related skin cancer (n = 255). Blood lead, manganese, and selenium were measured at baseline; blood pressure was measured at baseline and at 3 biennial follow-up examinations. Mixed-effect linear regression models were used to estimate associations with average annual changes in systolic, diastolic, and pulse pressure. In models simultaneously adjusted for baseline blood lead, manganese, and selenium concentrations in addition to other potential confounders, lead was linearly associated with increases in systolic blood pressure, but not with diastolic blood pressure or pulse pressure. A non-linear association was observed for manganese, such that mid-range concentrations were associated with decreases in systolic, diastolic, and pulse pressure. Baseline selenium concentrations in the highest quartile were also associated with longitudinal decreases in both systolic and diastolic blood pressure, while null associations were observed with pulse pressure. In exploratory analyses, the combination of mid-range manganese and high selenium concentrations completely offset lead-associated increases in blood and pulse pressure. The results indicate a direct, linear association of lead exposure with systolic blood pressure, and manganese and selenium exposures within certain ranges may have a blood pressure-lowering effect in this population.

Nanoplastics are widely used in modern life, for example, in cosmetics and daily use products, and are attracting concern due to their potential toxic effects on environments. In this study, the uptake of nanopolystyrene particles by Caenorhabditis elegans (C. elegans) and their toxic effects were evaluated. Nanopolystyrene particles with sizes of 50 and 200 nm were prepared, and the L4 stage of C. elegans was exposed to these particles for 24 h. Their uptake was monitored by confocal microscopy, and various phenotypic alterations of the exposed nematode such as locomotion, reproduction and oxidative stress were measured. In addition, a metabolomics study was performed to determine the significantly affected metabolites in the exposed C. elegans group. Exposure to nanopolystyrene particles caused the perturbation of metabolites related to energy metabolism, such as TCA cycle intermediates, glucose and lactic acid. Nanopolystyrene also resulted in toxic effect including induction of oxidative stress and reduction of locomotion and reproduction. Collectively, these findings provide new insights into the toxic effects of nanopolystyrene particles.

2019
Vignier, J. | Rolton, A. | Soudant, P. | Chu, F.L.E. | Robert, R. | Volety, A.K.
The Deepwater Horizon (DWH) disaster released crude oil in the Gulf of Mexico for 87 days, overlapping with the reproductive season and recruitment of the oyster Crassostrea virginica. The pelagic larval life stages of C. virginica are particularly vulnerable to contaminants such as polycyclic aromatic hydrocarbons (PAHs) and oil droplets. Based on their lipophilic properties, PAHs and oil droplets can adsorb onto phytoplankton and filter-feeding C. virginica larvae may be exposed to these contaminants bound to suspended sediment, adsorbed onto algal and other particles, or in solution. This study examined the effects of exposure of C. virginica larvae to algae mixed with DWH oil. In a 14-day laboratory exposure, 5 day-old C. virginica larvae were exposed to Tisochrysis lutea mixed with four concentrations of unfiltered DWH oil (HEWAF) in a static renewal system. Larval growth, feeding capacity, abnormality and mortality were monitored throughout the exposure. Total PAH (n = 50) content of the water medium, in which larvae were grown, were quantified by GC/MS-SIM. Oil droplets were observed bound to algae, resulting in particles in the size-range of food ingested by oyster larvae (1–30 μm). After 14 days of exposure, larval growth and survival were negatively affected at concentrations of tPAH50 as low as 1.6 μg L⁻¹. GC/MS-SIM analysis of the exposure medium confirmed that certain PAHs were also adsorbed by T. lutea and taken up by oyster larvae via ingestion of oil droplets and/or contaminated algae. Long-term exposure to chronic levels of PAH (1.6–78 μg tPAH50 L⁻¹) was shown to negatively affect larval survival. This study demonstrates that dietary exposure of oyster larvae to DWH oil is a realistic route of crude oil toxicity and may have serious implications on the planktonic community and the food chain.

The dissipation of famoxadone as well as the behaviour of its metabolites in environmental samples such as water and soil is a major concern. In this study, the dissipation of the target compound in both matrices was carried out applying an analytical method based on ultra-high performance liquid chromatography coupled to Orbitrap mass spectrometry (UHPLC-Orbitrap-MS). The dissipation of famoxadone was monitored over a period of 100 days after the plant protection product, Equation Pro®, was administered to the target matrices. This study was performed at two doses, normal and double in the case of soils and fivefold instead of double dose in water. The concentration of famoxadone steadily decreased during the monitoring period in both matrices. Half-life (DT₅₀) values were lower than 30 days in most cases except for loam soils, for which it was 35 days. Therefore, persistence of this pesticide in both matrices was low. Famoxadone metabolites such as IN-KF015 ((5RS)-5-methyl-5-(4-phenoxyphenyl)-1,3- oxazolidine-2,4-dione) and IN-JS940 ((2RS)-2-hydroxy-2-(4- phenoxyphenyl)propanoic acid) were detected in both matrices and their concentration increased while the concentration of the parent compound decreased. Metabolite IN-JS940 was the compound detected at highest concentration for both matrices. In water the maximum concentration was 20% of the initial famoxadone content and in soils it was 50% of initial famoxadone content. In addition, another metabolite, IN-MN467 ((5RS)-5-methyl-3-[(2-nitrophenyl)amino]- 5-(4-phenoxyphenyl)-1,3-oxazolidine-2,4-dione), was detected in soils, following the same behaviour as the other metabolites. These results provided ample information about the behaviour of metabolites and the necessity of knowing their toxicity in both matrices in order to detect possible risks for living beings.

Rapid development of Bohai and Yellow Sea Economic Rim has led to the concern of emerging contamination of marine environments. This study investigated the spatial distribution of antibiotic resistance genes (ARGs) in Bohai and Yellow Sea areas. A large scale sampling from Bohai Sea, Yellow Sea and the major cities along the coastline from the mouth of Yalu River to the Yangtze River was performed. The spatial distribution of target ARGs based on the absolute abundances was in the trend of river water ≈ coastal water > the Bohai Sea > the Yellow Sea, inshore > offshore and inner bay > bay mouth. The total absolute abundances of selected ARGs in the coastal waters (1.23 × 10⁴–3.94 × 10⁵ copies/mL) were about 1–4 orders of magnitude higher than those in the sea (21.1–8.00 × 10³ copies/mL). The abundances of ARGs fluctuated greatly in the Yellow Sea and the coastal areas. Sulfonamide resistance genes hold the highest abundances in the Bohai and Yellow Sea (up to 2.13 × 10³ copies/mL of sul1 and 6.23 × 10³ copies/mL of sul2), followed by tetracycline and quinolone resistance genes, while qnrA hold the highest abundances in coastal areas (up to 3.66 × 10⁵ copies/mL). The distribution coefficients of target genes between sediments and corresponding water samples were more than 1.0 in the majority of different aquatic systems. According to the principle component analysis and redundancy analysis, water samples collected from the sea clustered together while those from the coastal zone and rivers were separated. Ammonium and nitrate played important roles in the distribution and variation of ARGs. Co-occurrence network analysis revealed that the potential multi-antibiotics resistant bacteria were detected with higher abundances in the Yellow Sea than in the Bohai Sea. These observations provided a comprehensive new insight into the pollution status of ARGs in the Bohai and Yellow Sea areas.

2019
González-Alcaraz, M Nazaret | Malheiro, Catarina | Cardoso, Diogo N. | Loureiro, Susana
Water availability is paramount in the response of soil invertebrates towards stress situations. This study aimed to evaluate the effects of forecasted soil moisture scenarios on the avoidance behavior of two invertebrate species (the arthropod Folsomia candida and the soft-bodied oligochaete Enchytraeus crypticus) in soils degraded by different types of anthropogenic metal(loid) contamination (mining soil and agricultural soil affected by industrial chemical wastes). Different soil moisture contents (expressed as % of the soil water holding capacity, WHC) were evaluated: 50% (standard soil moisture conditions for soil invertebrates' tests); 75% (to simulate increasing soil water availability after intense rainfalls and/or floods); 40%, 30%, 25% and 20% (to simulate decreasing soil water availability during droughts). Invertebrates’ avoidance behavior and changes in soil porewater major ions and metal(loid)s were assessed after 48 h exposure. Soil incubations induced a general solubilization/mobilization of porewater major ions, while higher soil acidity favored the solubilization/mobilization of porewater metal(loid)s, especially at 75% WHC. Folsomia candida preferred soils moistened at 50% WHC, regardless the soils were contaminated or not and the changing soil porewater characteristics. Enchytraeus crypticus avoided metal(loid) contamination, but this depended on the soil moisture conditions and the corresponding changes in porewater characteristics: enchytraeids lost their capacity to avoid contaminated soils under water stress situations (75% and 20–25% WHC), but also when contaminated soils had greater water availability than control soils. Therefore, forecasted soil moisture scenarios induced by global warming changed soil porewater composition and invertebrates capacity to avoid metal(loid)-contaminated soils.
