Excretion of polycyclic aromatic hydrocarbon metabolites (OH-PAHs) in cattle urine in Ghana

Previous studies of polycyclic aromatic hydrocarbons (PAHs) in particulate matter, soils and livers of wild rats indicated that the city centre of Kumasi, Ghana has been severely polluted with high cancer potency. Cattle urine were therefore collected from Kumasi (urban) and Offinso (rural), Ghana: to determine concentrations of urinary PAH metabolites (OH-PAHs); and find their association with sex; and to estimate exposure of cattle to PAHs from the different sites. From the results, geometric mean concentrations (adjusted by specific gravity), GMSG, showed that 2-OHNaphthalene (2-OHNap) was the most abundant OH-PAH in cattle urine from all study sites, and naphthalene-containing-mothballs might have contributed significantly to the levels. There was no significant difference between urinary OH-PAHs concentrations in cattle from urban and rural sites except for 2-OHPhe and 4-OHPhe, and similar to urban areas, rural sites could also be polluted with PAHs. GMSG of 2-OHNap in cattle urine in Kokote (21.9 ± 6.51 ng/mL; a rural area), was significantly higher compared to the other sites followed by Oforikrom (4.15 ± 4.37 ng/mL; urban). The GMSG concentration (ng/mL) of the sum of OH-PAHs decreased in the order, Kokote (44.7) > Oforikrom (7.87) > Saboa (6.98) > Santasi (6.68) > and Twumasen Estate (5.23). The high concentrations of urinary 2-OHNap, 2-3-OHFlu, 2-OHPhe, 3-OHPhe and 4-OHPhe in Kokote indicated high PAHs exposure to cattle in this area or different/specific source of PAHs exposure. GMSG of 2-OHNap was significantly higher in male cattle compared to females while 1-9-OHPhe was significantly higher in females.

Most of the organohalogenated contaminants (OHCs) have high environmental stability and are lipophilic in nature, thus bioaccumulate through the various routes e.g., inhalation, dermal contact and food intake. Human exposure to these OHCs can induce adverse health effects. Studies on the occurrence of OHCs in human samples from Saudi Arabia are scarce. Therefore, this study aimed at providing preliminary insight on the occurrence of polychlorinated biphenyls (PCBs), organochlorine pesticides (OCPs) and polybrominated biphenyl ethers (PBDEs) in diabetic and non-diabetic donors from KSA. Serum samples were collected from type 2 diabetic patients (n = 40) and control donors (n = 20) to study the impact of OHCs on their health. For the first time we studied the difference of ƩOHCs in type 2 diabetic and control participants. The order of obtained results was ƩOCPs (35–650 ng/g lw)> ƩPCBs (15–90 ng/g lw)> ƩPBDEs (1.5–68 ng/g lw). The major contributors were p,p′-DDE (median 44 ng/g lw), PCB 153 (2.3 ng/g lw), PCB 138 (2.1 ng/g lw), BDE 153 (1.2 ng/g lw) and BDE 47 (0.85 ng/g lw). Exposure to different OHCs between male and female donors was not significantly different (p > 0.05). However, ƩPCBs and ƩOHCs were significantly higher (p < 0.05) in diabetic donors than those of control group. We computed significantly positive correlations (p < 0.05) among different OHCs and between OHCs and age factor. The current study highlights the presence of different OHCs in humans from Jeddah, KSA. This is a preliminary study based on small sample size but our results suggested that detailed studies are required to understand the sources of these pollutants and their impact on human health.

We performed toxicological study of mice exposed to lead by quantifying fatty acids in brain of the mice. This study suggests that the introduced analytical method had an extremely high tolerance against impurities such as water and extractives; thus, it led to the enhanced resolution in visualizing the spectrum of fatty acid profiles in animal brain. Furthermore, one of the biggest technical advantages achieved in this study was the quantitation of fatty acid methyl ester profiles of mouse brain using a trace amount of sample (e.g., 100 μL mixture). Methanol was screened as the most effective extraction solvent for mouse brain. The behavioral test of the mice before and after lead exposure was conducted to see the effect of lead exposure on fatty acid composition of the mice’ brain. The lead exposure led to changes in disease-related behavior of the mice. Also, the lead exposure induced significant alterations of fatty acid profile (C16:0, C 18:0, and C 18:1) in brain of the mice, implicated in pathology of psychiatric diseases. The alteration of fatty acid profile of brain of the mice suggests that the derivatizing technique can be applicable to most research fields associated with the environmental neurotoxins with better resolution in a short time, as compared to the current protocols for lipid analysis.

Size-segregated particulate pollutants (PM<0.95, PM0.95–1.5, PM1.5–3.0, PM3.0–7.2 and PM>7.2) were collected over Patiala (30.33°N, 76.40°E; 250 m amsl), a semi-urban city located in northwestern Indo-Gangetic Plain (IGP), during October, 2012 to September, 2013. Mass concentration of total suspended particulates (TSP), derived by summation of particulate (aerosol) mass in different size range, varied from 88 to 387 μg m−3 with highest mass concentration (∼55% of total mass) in submicron size (PM<0.95) during the entire study period, which broadly reflects relative higher contribution of various anthropogenic sources (emissions from biomass and bio-fuel burning, vehicles, thermal power plants, etc) to ambient particles. Concentration of SO42−, NO3−, NH4+, K+ and Ca2+ exhibited large variability ranging from 0.52 to 40, 0.20 to 19, 0.14 to 12, 0.06 to 5.3 and 0.08 to 5.6 μg m−3, respectively, in different size ranges with varying size distribution for most of the species, except NH4+. A strong linear correlation (r = 0.97) between (SO42− + NO3−) and (K+ + NH4+) concentrations has been observed in submicron particles collected in different seasons, suggesting the formation of secondary inorganic salts. However, relatively poor correlation is observed in higher size ranges where significant correlation between (SO42− + NO3−) and (Ca2+ + Mg2+) has been observed. These observations indicate the acid neutralization by dust in coarser modes of particles. Chemical composition of submicron particulates (PM<0.95) in different seasons as well as for whole year was used to identify PM sources through the application of Positive Matrix Factorization (PMF, version 5.0) model. Based on annual data, PMF analyses suggests that six source factors namely biomass burning emission (24%), vehicular emission (22%), secondary organic aerosols (20%), power plant emission (13%), secondary inorganic aerosols (12%) and mineral dust (9%) contribute to PM<0.95 loading over the study region. Such studies are important in dispersion modeling, health impact assessment, and planning of pollution mitigation strategies.

Aquaculture in mangrove wetlands has been developed rapidly, causing various environmental problems (e.g., antibiotic residue). In the present study, the levels and distributions of a well-known class of antibiotics (fluoroquinolones; FQs), including norfloxacin (NOR), ciprofloxacin (CIP), and enrofloxacin (ENR), were examined in sediment and mangrove plant (Aegiceras corniculatum) from a mangrove wetland in the Zhanjiang Mangrove National Nature Reserve, South China. NOR and CIP were detected in all sediment samples, with concentrations ranging from 4.3 to 64.2 ng/g and from 7.62 to 68.5 ng/g on a basis of dry weight (dw), respectively, whereas ENR was found with relatively lower frequency (<78%) and lower concentrations (<19.3 ng/g). Sediments in mangrove rhizosphere area contained considerably higher concentrations of all FQs (except for ENR). FQs were largely varied in mangrove plant tissues; NOR and ENR were not detected in leaf and root samples, respectively. CIP featured an increasing tendency from the root to the upper parts of plants, whereas a decreasing trend was found for NOR. Three bioconcentration factors (BCFs) of FQs, including BCFs for roots (BCFr), branches (BCFb), and leaves (BCFl) were calculated, and most of them exceeded 1. Especially for NOR, its BCFr can reach up to 9.9, indicating that Aegiceras corniculatum has a strong ability to accumulate FQs from sediment and/or surrounding environment. For NOR and CIP, strong positive relationships were observed between BCFr and concentrations in root, but no significant correlations were observed between BCFr and root lipid of mangrove plant. More studies are needed to investigate the uptake mechanism of antibiotics in mangrove plants.

Evaluating potential adverse effects of complex chemical mixtures in the environment is challenging. One way to address that challenge is through more integrated analysis of chemical monitoring and biological effects data. In the present study, water samples from five locations near two municipal wastewater treatment plants in the St. Croix River basin, on the border of MN and WI, USA, were analyzed for 127 organic contaminants. Known chemical-gene interactions were used to develop site-specific knowledge assembly models (KAMs) and formulate hypotheses concerning possible biological effects associated with chemicals detected in water samples from each location. Additionally, hepatic gene expression data were collected for fathead minnows (Pimephales promelas) exposed in situ, for 12 d, at each location. Expression data from oligonucleotide microarrays were analyzed to identify functional annotation terms enriched among the differentially-expressed probes. The general nature of many of the terms made hypothesis formulation on the basis of the transcriptome-level response alone difficult. However, integrated analysis of the transcriptome data in the context of the site-specific KAMs allowed for evaluation of the likelihood of specific chemicals contributing to observed biological responses. Thirteen chemicals (atrazine, carbamazepine, metformin, thiabendazole, diazepam, cholesterol, p-cresol, phenytoin, omeprazole, ethyromycin, 17β-estradiol, cimetidine, and estrone), for which there was statistically significant concordance between occurrence at a site and expected biological response as represented in the KAM, were identified. While not definitive, the approach provides a line of evidence for evaluating potential cause-effect relationships between components of a complex mixture of contaminants and biological effects data, which can inform subsequent monitoring and investigation.

Ambient noise has increased in extent, duration and intensity with significant implications for species’ lives. Birds especially, because they heavily rely on vocal communication, are highly sensitive towards noise pollution. Noise can impair the quality of a territory or hamper the transmission of vocal signals such as song. The latter has significant fitness consequences as it may erode partner preferences in the context of mate choice. Additional fitness costs may arise if noise masks communication between soliciting offspring and providing parents during the period of parental care. Here, we experimentally manipulated the acoustic environment of blue tit (Cyanistes caeruleus) families within their nest boxes with playbacks of previously recorded highway noise and investigated the consequences on parent-offspring communication. We hypothesized that noise interferes with the acoustic cues of parental arrival and vocal components of offspring begging. As such we expected an increase in the frequency of missed detections, when nestlings fail to respond to the returning parent, and a decrease in parental provisioning rates. Parents significantly reduced their rate of provisioning in noisy conditions compared to a control treatment. This reduction is likely to be the consequence of a parental misinterpretation of the offspring hunger level, as we found that nestlings fail to respond to the returning parent more frequently in the presence of noise. Noise also potentially masks vocal begging components, again contributing to parental underestimation of offspring requirements. Either way, it appears that noise impaired parent-offspring communication is likely to reduce reproductive success.

The shift from rural lifestyles to urban living has dramatically altered the way humans interact and live across the globe. With over 50% of the world’s populations living within cities, and significant increases expected over the next 50 years, it is critical that changes to social, economic and environmental sustainability of cities globally be implicit. Protecting and enhancing aquatic ecosystems, which provide important ecosystem services, is challenging. A number of factors influence pollutants in urban waterways including changes in land-use, impervious area and stormwater discharges, with sediment-bound pollution a major issue worldwide. This work aimed to investigate the spatial and temporal distribution of trace metals in freshwater sediments from six urbanised catchment over a 30-year period. It provides an estimate of pollution using a geoaccumulation index and examines possible toxicity using a probable effect concentration quotient (mPECq). Results showed significant temporal changes in metal concentrations over time, with lead generally decreasing in all but one of the sites, attributed to significant changes in environmental policies and the active elimination of lead products. Temporal changes in other metals were variable and likely dependent on site-specific factors. While it is likely that diffuse pollution is driving changes in zinc, for metals such as lead, chromium and copper, it is likely that watershed landuse and/or point sources are more important. The results clearly indicated that changes to watershed landuse, environmental policy and pollution abatement programs are all driving changes in sediment quality, highlighting the utility of long-term sediment monitoring for assessment of urban watershed condition. While this study has demonstrated the utility of detecting long-term changes in metal concentrations, this approach could easily be adapted to detect and assess future trends in other hydrophobic contaminants and emerging chemicals of concern, such as synthetic pyrethroids, providing essential information for the protection of catchment.

China's urbanization and the subsequent public vulnerability to degenerated environment is important to global public health. Among the environmental problems, fine particulate (PM2.5) pollution has become a serious hazard in rapidly urbanizing China. However, quantitative information remains inadequate. We thus collected PM2.5 concentrations and population census records, to illustrate the spatial patterns and changes in the PM2.5 hazard levels in China, and to quantify public vulnerability to the hazard during 2000–2010, following the air quality standards of World Health Organization. We found that 28% (2.72 million km2) of China's territory, including 78% of cities (154 cities) with a population of >1 million, was exposed to PM2.5 hazard in 2010; a 15% increase (1.47 million km2) from 2000 to 2010. The hazards potentially impacted the health of 72% of the total population (942 million) in 2010, including 70% of the young (206 million) and 76% of the old (71 million). This was a significant increase from the 42% of total the population (279 million) exposed in 2000. Of the total urban residents, 76% (501 million) were affected in 2010. Along with PM2.5 concentration increase, massive number of rural to urban migration also contributed greatly to China's urban public health vulnerability.
