Multifaceted toxicity assessment of catalyst composites in transgenic zebrafish embryos النص الكامل

Recent development in the field of nanomaterials has given rise into the inquiries regarding the toxicological characteristics of the nanomaterials. While many individual nanomaterials have been screened for their toxicological effects, composites that accompany nanomaterials are not common subjects to such screening through toxicological assessment. One of the widely used composites that accompany nanomaterials is catalyst composite used to reduce air pollution, which was selected as a target composite with nanomaterials for the multifaceted toxicological assessment. As existing studies did not possess any significant data regarding such catalyst composites, this study focuses on investigating toxicological characteristics of catalyst composites from various angles in both in-vitro and in-vivo settings. Initial toxicological assessment on catalyst composites was conducted using HUVECs for cell viability assays, and subsequent in-vivo assay regarding their direct influence on living organisms was done. The zebrafish embryo and its transgenic lines were used in the in-vivo assays to obtain multifaceted analytic results. Data obtained from the in-vivo assays include blood vessel formation, mutated heart morphology, and heart functionality change. Our multifaceted toxicological assessment pointed out that chemical composites augmented with nanomaterials can too have toxicological threat as much as individual nanomaterials do and alarms us with their danger. This manuscript provides a multifaceted assessment for composites augmented with nanomaterials, of which their toxicological threats have been overlooked.

The Atlantic killifish (Fundulus heteroclitus) is a resilient estuarine species that may be subjected to anthropogenic contamination of its natural habitat, by toxicants such as nickel (Ni). We investigated Ni accumulation and potential modes of Ni toxicity, in killifish, as a function of environmental salinity. Killifish were acclimated to 4 different salinities [0 freshwater (FW), 10, 30 and 100% seawater (SW)] and exposed to 5 mg/L of Ni for 96 h. Tissue Ni accumulation, whole body ions, critical swim speed and oxidative stress parameters were examined. SW was protective against Ni accumulation in the gills and kidney. Addition of Mg and Ca to FW protected against gill Ni accumulation, suggesting competition with Ni for uptake. Concentration-dependent Ni accumulation in the gill exhibited saturable relationships in both FW- and SW-acclimated fish. However SW fish displayed a lower Bmax (i.e. lower number of Ni binding sites) and a lower Km (i.e. higher affinity for Ni binding). No effect of Ni exposure was observed on critical swim speed (Ucrit) or maximum rate of oxygen consumption (MO2max). Markers of oxidative stress showed either no effect (e.g. protein carbonyl formation), or variable effects that appeared to depend more on salinity than on Ni exposure. These data indicate that the killifish is very tolerant to Ni toxicity, a characteristic that may facilitate the use of this species as a site-specific biomonitor of contaminated estuaries.

Although the adverse impact of fine particulate matter (i.e., PM2.5) on human health has been well acknowledged, little is known of the health effects of its specific constituents. Over the past decade, the annual average dust concentrations in Beijing were approximately ∼14 μg m−3, a value that poses a great threat to the city's 20 million residents. In this study, we quantify the potential long-term health damages in Beijing resulting from the dust exposure that occurred from 2000 to 2011. Each year in Beijing, nearly 4000 (95% CI: 1000–7000) premature deaths may be associated with long-term dust exposure, and ∼20% of these deaths are attributed to lung cancer. A decomposition analysis of the inter-annual variability of premature deaths in Beijing indicates that dust concentrations determine the year-to-year tendency, whereas population growth and lung cancer mortality rates drive the increasing tendency of premature death. We suggest that if Beijing takes effective measures towards reducing dust concentrations (e.g., controlling the resuspension of road dust and the fugitive dust from construction sites) to a level comparable to that of New York City's, the associated premature deaths will be significantly reduced. This recommendation offers “low-hanging fruit” suggestions for pollution control that would greatly benefit the public health in Beijing.

In the present study we analyze the effect of seed treatment by a range of nano-TiO2 concentrations on the growth of Arabidopsis thaliana plants, on the vitamin E content and the expression of its biosynthetic genes, as well as activity of antioxidant enzymes and lipid peroxidation. To conduct the mechanistic analysis of nano-TiO2 on plants growth and antioxidant status we applied nanoparticles concentrations that are much higher than those reported in the environment. We find that as the concentration of nano-TiO2 increases, the biomass, and chlorophyll content in 5-week-old Arabidopsis thaliana plants decrease in a concentration dependent manner. In opposite, higher nano-TiO2 concentration enhanced root growth. Our results indicate that a high concentration of nano-TiO2 induces symptoms of toxicity and elevates the antioxidant level. We also find that the expression levels of tocopherol biosynthetic genes were either down- or upregulated in response to nano-TiO2. Thermoluminescence analysis shows that higher nano-TiO2 concentrations cause lipid peroxidation. To the best of our knowledge, this is the first report concerning the effect of nano-TiO2 on vitamin E status in plants. We conclude that nano-TiO2 affects the antioxidant response in Arabidopsis thaliana plants. This could be an effect of a changes in vitamin E gene expression that is diminished under lower tested nano-TiO2 concentrations and elevated under 1000 μg/ml.

The number of products containing engineered nanomaterials (ENMs) has increased due to their high industrial relevance as well as their use in diverse consumer products. At the end of their life cycle ENMs might be released to the environment and therefore concerns arise regarding their environmental impact. In order to track their fate upon disposal, it is crucial to establish methods to trace ENMs in complex environmental samples and to differentiate them from naturally-occurring nanoparticles. The goal of this study was to distinctively trace ENMs by (non-invasive) detection methods. For this, fluorescent ENMs, namely quantum dots (QDs), were distinctively traced in complex aqueous matrices, and were still detectable after a period of two months using fluorescence spectroscopy. In particular, two water-dispersible QD-species, namely CdTe/CdS QDs with N-acetyl-l-cysteine as capping agent (NAC-QDs) and surfactant-stabilized CdSe/ZnS QDs (Brij®58-QDs), were synthesized to examine their environmental fate during disposal as well as their potential interaction with naturally-occurring substances present in landfill leachates. When QDs were spiked into a leachate from an old landfill site, alteration processes, such as sorption, aggregation, agglomeration, and interactions with dissolved organic carbon (DOC), led to modifications of the optical properties of QDs. The spectral signatures of NAC-QDs deteriorated depending on residence time and storage temperature, while Brij®58-QDs retained their photoluminescence fingerprints, indicating their high colloidal stability. The observed change in photoluminescence intensity was mainly caused by DOC-interaction and association with complexing agents, such as fulvic or humic acids, typically present in mature landfill leachates. For both QD-species, the results also indicated that pH of the leachate had no significant impact on their optical properties. As a result, the unique spectroscopic fingerprints of QDs, specifically surfactant-stabilized QDs, allowed distinctive tracing in complex aqueous waste matrices in order to study their long-term behavior and ultimate fate.

Tetrabromobisphenol A (TBBPA) is the brominated flame retardant with the highest production volume and its bioaccumulation in environment has caused both human health and environmental concerns, however the fate and metabolism of TBBPA in plants is unknown. We studied the fate, metabolites, and transformation of 14C-labeled TBBPA in rice cell suspension culture. During the incubation for 14 days, TBBPA degradation occurred continuously in the culture, accompanied by formation of one anisolic metabolite [2,6-dibromo-4-(2-(2-hydroxy)-propyl)-anisole] (DBHPA) (50% of the degraded TBBPA) and cellular debris-bound residues (46.4%) as well as mineralization (3.6%). The cells continuously accumulated TBBPA in the cytoplasm, while a small amount of DBHPA (2.1% of the initially applied TBBPA) was detectable inside the cells only at the end of incubation. The majority of the accumulated residues in the cells was attributed to the cellular debris-bound residues, accounting for 70–79% of the accumulation after the first incubation day. About 5.4% of the accumulation was associated with cell organelles, which contributed 7.5% to the cellular debris-bound residues. Based on the fate and metabolism of TBBPA in the rice cell suspension culture, a type II ipso-substitution pathway was proposed to describe the initial step for TBBPA degradation in the culture and balance the fate of TBBPA in the cells. To the best of our knowledge, our study provides for the first time the insights into the fate and metabolism of TBBPA in plants and points out the potential role of type II ipso-hydroxylation substitution in degradation of alkylphenols in plants. Further studies are required to reveal the mechanisms for the bound-residue formation (e.g., binding of residues to specific cell wall components), nature of the binding, and toxicological effects of the bound residues and DBHPA.

Halogenated persistent organic pollutants (Hal-POPs) are significant contaminants in the indoor environment that are related to many human diseases. Ingestion of indoor dust is considered the major pathway of Hal-POP exposures, especially for children aged 3–6 years. Alongside a retrospective study on the associations between typical Hal-POP exposure and childhood asthma in Shanghai, indoor dust samples from asthmatic and non-asthmatic children's homes (n = 60, each) were collected. Polybrominated diphenyl ethers (PBDEs), polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) were measured by GC–MS. BDE-209, PCB-8 and p,p′-DDE were the predominant components in each chemical category. The concentrations of most Hal-POPs were significantly higher in the asthmatic families. The associations between Hal-POP exposure and asthma occurrence were examined by calculating the odds ratios (ORs) using a logistic regression model. A positive association was found between p,p′-DDE in indoor dust and childhood asthma (OR = 1.825, 95%CI: 1.004, 3.317; p = 0.048). The average daily doses of Hal-POP intake were calculated using the method provided by the USEPA. Non-carcinogenic health risks were preliminarily assessed. Our study indicated that exposure to p,p’-DDE via indoor dust may contribute to childhood asthma occurrence. Non-carcinogenic health risks were not found with the intake of Hal-POPs via the ingestion of indoor dust.

Microplastics are plastics that measure less than 5 mm in diameter. They enter the marine environment as primary sources directly from industrial uses, as well as secondary sources resulting from the degradation of large plastic debris. To improve the knowledge of microplastic pollution in China, we investigated samples from 53 estuarine sediment locations collected with a box corer within the Changjiang Estuary. Microplastics (<5 mm) were extracted from sediments by density separation, after which they were observed under a microscope and categorized according to shape, color and size. Identification was carried out using Micro-Fourier-Transform Infrared Spectroscopy (μ-FT-IR).The abundance of microplastics in the Changjiang Estuary was mapped. The mean concentration was 121 ± 9 items per kg of dry weight, varying from 20 to 340 items per kg of dry weight. It was found that the concentration of microplastics was the highest on the southeast coast of Shanghai. The distribution pattern of microplastics may be affected by the Changjiang diluted water in summer. All of the microplastics collected were categorized according to shape, color and size. Among which fiber (93%), transparent (42%) and small microplastics (<1 mm) (58%) were the most abundant types. No clear correlation between microplastics and the finer sediment fraction was found. Rayon, polyester, and acrylic were the most abundant types of microplastics identified, indicating that the main source of microplastics in the Changjiang Estuary was from washing clothes (the primary source). It is possible to compare microplastic abundance in this study with the results of other related studies using the same quantification method. The identification of microplastics raises the awareness of microplastic pollution from drainage systems. The prevalence of microplastic pollution calls for monitoring microplastics at a national scale on a regular basis.

We investigated rhizosphere effects on the distributions and compositions of polybrominated diphenyl ethers (PBDEs), novel brominated flame retardants (NBFRs), and dechlorane plus (DPs) in rhizosphere soils (RS) and non-rhizosphere soils (NRS) in an e-waste recycling area in South China. The concentrations of PBDEs, NBFRs, and DPs ranged from 13.9 to 351, 11.6 to 70.8, and 0.64 to 8.74 ng g⁻¹ in RS and 7.56 to 127, 8.98 to 144, and 0.38 to 8.45 ng g⁻¹ in NRS, respectively. BDE-209 and DBDPE were the dominant congeners of PBDEs and NBFRs, respectively. PBDEs, NBFRs, and DPs were more enriched in RS than NRS in most vegetables species. Further analysis suggested that the differentiation of the rhizosphere effect on halogenated flame retardants (HFRs) was not solely controlled by the octanol-water coefficients. This difference was also reflected by the correlations between total organic carbon (TOC) and PBDEs, NBFRs, or DPs, which indicated that organic carbon was a more pivotal controlling factor for PBDEs and DPs than for NBFRs in soil. We also found significant positive correlations between PBDEs and their replacement products, which indicated a similar emission pattern and environmental behaviour.

With current global changes, the combination of several stressors such as temperature and contaminants may impact species distribution and ecosystem functioning. In this study, we evaluated the combined impact of two metals (Ni and Cr) with a thermal stress (from 12 to 17 °C) on biomarker responses in two bivalves, Dreissena rostriformis bugensis and Dreissena polymorpha. Biomarkers are informative tools to evaluate exposure and effects of stressors on organisms. The set of 14 biomarkers measured here was representative of both physiologic (filtration activity) and cellular antioxidant and detoxification mechanisms. Our aim was to study the response pattern of both species, and its meaning in terms of invasive potential. The implications for the use of these mussels in environmental monitoring are also discussed. Results evidenced that the two species do not respond to multiple stressors in the same way. Indeed, the effects of contamination on biomarker responses were more marked for D. polymorpha, especially under nickel exposure. While we cannot conclude as to the effect of temperature, invasiveness could be influenced by species sensitivity to contaminants. The physiological and cellular differences between D. polymorpha and D. r. bugensis might also be of concern for environmental risk assessment. The two species present differential bioaccumulation patterns, filtration activity and cellular biomarker responses. If D. polymorpha populations decline, their substitution by D. r. bugensis for biomonitoring or laboratory studies will not be possible without a deeper understanding of biomarker responses of the new invasive.
