Impacts of selenium supplementation on soil mercury speciation, and inorganic mercury and methylmercury uptake in rice (Oryza sativa L.)
2019
Rice grain is known to accumulate methylmercury (MeHg) and has been confirmed to be the major pathway of MeHg exposure to residents in mercury (Hg) mining areas in China. Selenium (Se) supplementation has been proven to be effective in mitigating the toxicity of Hg. To understand how Se supplementation influences soil Hg speciation, a wide range of Se (0–500 mg/kg) was applied to Hg polluted paddy soils in this study, which decreased MeHg concentration in soil from 2.95 ± 0.36 to 0.69 ± 0.16 μg/kg (or 77%). After Se addition, humic acid state Hg (F4) was transformed into strong-complexed state Hg (F5), indicating that Hg bound up to the non-sulfur functional groups of humic acid (non-RSH) was released and reabsorbed by strong binding Se functional group (F5). As a result, inorganic Hg (IHg) was reduced by >48%, 18%, and 80% in root, stem, and grain, respectively, however, the reduction was not apparent in leaf. Substantial reductions were also found for MeHg in grain and root, but not in stem and leaf. Soil is suggested to be the main source of both MeHg and IHg in rice grain. Such a finding may provide an idea for improving Hg-polluted paddies through controlling soil IHg and MeHg. Further research on the molecular structure of the strong-complexed Hg in F5 should be conducted to elucidate the mechanism of Hg-Se antagonism.

Schizophrenia is a devastating neuropsychiatric disorder with increasing concern. Limited studies have been conducted to assess the relationship between short-term exposure to ambient air pollution and schizophrenia attacks. This study aimed to investigate the associations between short-term air pollution exposure and schizophrenia outpatient visits based on a time-series study performed in China. Daily data of schizophrenia outpatient admissions and air pollution from 1 October 2010 to 31 December 2013 were collected in Xi'an, a heavily-polluted city in China. We utilized a time-series Poisson regression model to examine the associations between short-term air pollution and schizophrenia outpatient visits with different lag days. A total of 34,865 outpatient-visits for schizophrenia were identified. A 10 μg/m³ increase of PM₁₀, SO₂, and NO₂ concentrations corresponded to 0.289% (95% Cl: 0.118%, 0.460%), 1.374% (95% Cl: 0.723%, 2.025%), and 1.881% (95% Cl: 0.957%, 2.805%) elevation in outpatient-visits for schizophrenia at lag 0, and the associations appeared to be stronger, although not statistically significantly, in females and in middle and older age adults (40 and over). The most significant associations were observed on the concurrent day in different lag models. In conclusion, short-term exposure to ambient air pollution (PM₁₀, SO₂, and NO₂) can be associated with increased risk of daily outpatient visits for schizophrenia, which may contribute to the further understanding of the potential adverse effects of air pollution in schizophrenia and other neuropsychiatric disorders.

T-2 toxin is an unavoidable contaminant in human food, animal feeds, and agricultural products. T-2 toxin has been found to impair male reproductive function. But, few data is available that reveals the reproductive toxicity mechanism. In the study, male Kunming mice were orally administrated with T-2 toxin at the doses of 0, 0.5, 1 or 2 mg/kg body weight for 28 days. The body and reproductive organs weight, the concentration, malformation rate and ultrastructure of sperm in cauda epididymis were detected. Oxidative stress biomarkers and apoptosis were also measured in testes. Histological change of testes was performed by H&E and TUNEL staining. T-2 toxin down-regulated body and reproductive organs (testis, epididymis and seminal vesicle) weight, sperm concentration, increased sperm malformation rate and damaged the ultrastructure of sperm and structure of testes. T-2 toxin treatment increased the reactive oxygen species (ROS) and malondialdehyde content, while, decreased the total anti-oxidation capacity (T-AOC) and the superoxide dismutase activity in testes. T-2 toxin exposure increased the TUNEL-positive germ cells, the activities and mRNA expressions of caspase-3, caspase-8 and caspase-9, the mRNA expression of Bax, and inhibited the Bcl-2 mRNA expression. Furthermore, the expressions of caspase-3, caspase-8 caspase-9 and Bax were positively correlated with ROS level, but negatively correlated with T-AOC in testis. In summary, T-2 toxin caused spermatogenesis disorder associated with the germ cell apoptosis medicated by oxidative stress, impairing the male reproductive function.

Terbuthylazine (TBA) has replaced atrazine in many EU countries, becoming one of the most frequently detected pesticides in natural waters. TBA is a compound of emerging concern, due to its persistence, toxicity and proven endocrine disruption activity to wildlife and humans. Techniques applied in water treatment plants remove only partially this herbicide and poor attention is given to the generation and fate of by-products, although some of them have demonstrated an estrogenic activity comparable to atrazine. This paper summarizes the environmental occurrence of TBA and its main metabolite desethylterbuthylazine and reports the performance of an innovative electrochemical cell equipped with a solid polymer electrolyte (SPE) sandwiched between a Ti/RuO₂ cathode and a Boron-Doped Diamond anode, operating at constant current, in the treatment of an aqueous solution of TBA. The herbicide removal in the first 30 min of treatment increases from 42% to 92% as the applied current is increased from 100 to 500 mA. The rate of degradation at 500 mA decreases between 30 and 60 min, with a final abatement of 97%. An 89% removal was reached at 100 mA when the initial TBA concentration was raised from 0.1 to 4 mg L⁻¹ and less than 1% of the herbicide was converted in desethylterbuthylazine and minor metabolites. No chemicals are needed, no sludge is produced. Further research is encouraged, as this technology may be promising for the achievement of a zero-discharge removal of different emerging pollutants as pesticides, pharmaceuticals and personal care products.

We have carried out a comprehensive analysis of six air pollutants (particles with an aerodynamic diameter less than 2.5 μm (PM₂.₅) and less than 10 μm (PM₁₀), carbon monoxide (CO), sulfur dioxide (SO₂), nitrogen dioxide (NO₂), and ozone (O₃)) in western China, including the spatiotemporal characteristics of air pollutants, their relationship with meteorological factors and emission sources, and the efficiency of emission control strategies for the region. Based hourly observations at 23 sites in western China from June 2016 to May 2017, concentrations of most pollutants were higher outside the Tibetan Plateau, lowest in summer and highest in winter, the exception being O₃. This was partially because meteorological conditions in winter were found to the most unfavorable to pollutant dispersion and dilution than other seasons. Pollutant concentrations at most sites were correlated with the residential emissions which were higher in winter, but anti-correlated with the industrial emissions which were lower during the winter holiday period. The Weather Research and Forecasting with Chemistry (WRF-Chem) simulations of four pollution control strategies indicated that reduction of residential emissions is crucial to alleviate PM₂.₅, PM₁₀, and CO pollution in western China, although reduction of industrial and transport emissions can reduce SO₂ and NO₂, respectively. Since PM₂.₅ and PM₁₀ were also found to be the species most and next frequently responsible for extremely serious pollution in western China, respectively, we recommend pollution control regulations that target residential emissions.

There have been an increasing number of automobile vehicles in cities, so that newly developed residential areas are mostly designed with underground parking garages (UPGs). For naturally ventilated UPGs, the ventilation performance may be insufficient to discharge totally vehicle-induced pollutants out of the enclosed underground spaces, which consequently results in threats to residents' health. This study, therefore, aims at examining the patterns of pollutant concentrations in naturally ventilation UPGs as well as their sensitivities to traffic volume. In particular, the naturally ventilated UPGs’ weekday particulate matters (PM₂.₅ and PM₁₀), CO₂ and TVOC concentration as well as their relationships between traffic volume were quantitively evaluated based on field measurements in eight residential areas in Baoding, China. Results indicated that daily average PM₂.₅, PM₁₀, CO₂ and TVOC concentrations in studied UPGs were 105.81 μg/m³, 464.17 μg/m³, 571 ppm and 24 ppb, respectively. The PM₂.₅ concentrations in UPGs were slightly higher than that in ambient environments, while the PM₁₀ concentrations in UPGs were significantly higher. Furthermore, both PM₁₀ and TVOC concentrations in UPGs were in significant relationships with traffic volume at the p < 0.01 level, while the concentration of UPG PM₂.₅ generally exhibited a significant correlation (p < 0.01) with that of the ambient. Nevertheless, a combination of traffic volume, the ambient and accumulative effect was much better to explain the hourly PM₁₀ concentration in UPGs. These findings will be conducive to instruct engineers with fundamental knowledge of UPG ventilation design.

The effect of fluoride as an ongoing topic has attracted much attentions due to the decline in overall human fertility worldwide. However, whether fluorine causes a temporary stimulus or permanent damage to the male reproductive system, as well as the mechanism of fluoride influencing spermatogenesis remained unclear. 48 adult male rats were randomly divided into four groups (twelve each). Control group received the distilled water, while the other three groups were treated with 25, 50, 100 mg/L NaF via drinking water for 8 weeks. Six rats from each group were selected randomly to detect the levels of various indices related to spermatogenesis. The remaining rats were given only distilled water and left for recovery of a period of 2 weeks. Results showed that the levels of serum CK, ALP, CHE, BUN, UA, and Cr, testis morphology and the ultrastructure of sperm acrosome and chromatoid body (CB) were significantly changed by fluoride. Interestingly, the elongated spermatid counts, spermatids elongation ratio, and mRNA expressions of Prm1/2 and MIWI, TDRD1, TDRD 6, TDRD7, PABP, and Hsp72 related to CB decreased markedly in fluoride treatment groups compared to the control. Furthermore, the expression levels of DDX25 and associated regulatory proteins like CRM1, HMG2, H4, TP2, and PGK2 were down-regulated by fluoride. After 2-weeks withdrawal period, out of the 19 altered spermatogenesis indicators, 15 indicators in 100 mg/L group and 3 indicators in 50 mg/L group still exhibited a significant change, while none showed change in 25 mg/L group. These results proved that the reversibility of fluoride toxicity is dose-dependent on the male reproductive system. Meanwhile, fluoride caused unrestored arrest during the haploid period of spermatogenesis, where reduced DDX25 and associated regulatory proteins play a crucial role in this process, which could provide the underlying insights to the toxic mechanism of fluoride induced male reproductive toxicity.

Timber harvest has many effects on aquatic ecosystems, including changes in hydrological, biogeochemical, and ecological processes that can influence mercury (Hg) cycling. Although timber harvest's influence on aqueous Hg transformation and transport are well studied, the effects on Hg bioaccumulation are not. We evaluated Hg bioaccumulation, biomagnification, and food web structure in 10 paired catchments that were either clear-cut in their entirety, clear-cut except for an 8-m wide riparian buffer, or left unharvested. Average mercury concentrations in aquatic biota from clear-cut catchments were 50% higher than in reference catchments and 165% higher than in catchments with a riparian buffer. Mercury concentrations in aquatic invertebrates and salamanders were not correlated with aqueous THg or MeHg concentrations, but rather treatment effects appeared to correspond with differences in the utilization of terrestrial and aquatic basal resources in the stream food webs. Carbon and nitrogen isotope data suggest that a diminished shredder niche in the clear-cut catchments contributed to lower basal resource diversity compared with the reference of buffered treatments, and that elevated Hg concentrations in the clear-cut catchments reflect an increased reliance on aquatic resources in clear-cut catchments. In contrast, catchments with riparian buffers had higher basal resource diversity than the reference catchments, indicative of more balanced utilization of terrestrial and aquatic resources. Further, following timber harvest THg concentrations in riparian songbirds were elevated, suggesting an influence of timber harvest on Hg export to riparian food webs. These data, coupled with comparisons of individual feeding guilds, indicate that changes in organic matter sources and associated effects on stream food web structure are important mechanisms by which timber harvest modifies Hg bioaccumulation in headwater streams and riparian consumers.

Fluoroquinolone antibiotics (FQs) are considered to be emerging environmental contaminants that have been detected extensively in aquatic environment. It is of quite importance to explore FQs interacting with dissolved organic matter (DOM). The interactions of FQs with DOM were examined by nuclear magnetic resonance (NMR) spectroscopy, fluorescence quenching, UV–vis, Fourier transform infrared (FT-IR) spectroscopic techniques. The bindings of FQs to DOM had one single binding site and their quenching mechanisms were static, which were evaluated by the Stern-Volmer and Site-binding equations. Addition of DOM could result in micro-environmental changes of fluorophores groups in FQs. The location adjacent oxygen right of Ofloxacin (OFL) and the aromatic ring (the adjacency replaced by two nitrogen-containing groups) of Ciprofloxacin (CIP), Enrofloxacin (ENR), Norfloxacin (NOR) might be highly affected by DOM molecule. The negative enthalpy change (ΔH⁰), negative entropy change (ΔS⁰) and the positive Gibbs' energy change (ΔG⁰) figured out that the binding processes were exothermic but not thermodynamic favorable, the formation of HA-FQs complexes would be powered chiefly by the ΔS⁰. H-bonding, electrostatic effect, van der Waals force were the acting force in the binding reactions and the π-π stacking effect was the major binding force under alkaline conditions. Moreover, the protonated, deprotonated, or partially protonated state of FQs were found to have different binding capacity to DOM, and the binding reactions for FQs-HA system were suppressed as the ionic strength increased. Meanwhile, alterations of FQs conformation in the presence of DOM were evaluated by FT-IR and UV–vis spectra.

Metal pollution has been associated with anthropogenic activities, such as effluents and emissions from mines. Soil could be exposure route of wild rats to metals, especially in mining areas. The aim of this study was to verify whether soil exposure under environmentally relevant circumstances results in metal accumulation and epigenetic modifications. Wistar rats were divided to three groups: 1) control without soil exposure, 2) low-metal exposure group exposed to soil containing low metal levels (Pb: 75 mg/kg; Cd: 0.4), and 3) high-metal exposure group exposed to soil (Pb: 3750; Cd: 6). After 1 year of exposure, the metal levels, Pb isotopic values, and molecular indicators were measured. Rats in the high-group showed significantly greater concentrations of Pb and Cd in tissues. Higher accumulation factors (tissue/soil) of Cd than Pb were observed in the liver, kidney, brain, and lung, while the factor of Pb was higher in the tibia. The obtained results of metal accumulation ratios (lung/liver) and stable Pb isotope ratios in the tissues indicated that the respiratory exposure would account for an important share of metal absorption into the body. Genome-wide methylation status and DNA methyltransferase (Dnmt 3a/3b) mRNA expressions in testis were higher in the high-group, suggesting that exposure to soil caused metal accumulation and epigenetic alterations in rats.
