Experimental addition of nitrogen to a whole forest ecosystem at Gårdsjön, Sweden (NITREX): Nitrate leaching during 26 years of treatment

Chronic high deposition of nitrogen (N) to forest ecosystems can lead to increased leaching of inorganic N to surface waters, enhancing acidification and eutrophication. For 26 years nitrogen has been added as ammonium nitrate (NH₄NO₃) at 40 kg N ha⁻¹ yr⁻¹ to a whole forested catchment ecosystem at Gårdsjön, Sweden, to experimentally simulate the transition from a N-limited to N-rich state. Over the first 10 years of treatment there was an increasing amount of nitrate (NO₃⁻) and to a lesser extent ammonium (NH₄⁺) lost in runoff, but then N leaching stabilised, and for the subsequent 16 years the fraction of N added lost in runoff remained at 9%. NO₃⁻ concentrations in runoff were low in the summer during the first years of treatment, but now are high throughout the year. High frequency sampling showed that peaks in NO₃⁻ concentrations generally occurred with high discharge, and were enhanced if high discharge coincided with occasions of N addition. Approximately 50% of the added N has gone to the soil. The added N is equivalent to 140 years of ambient N deposition. At current ambient levels of N deposition there thus appears to be no immediate risk of N saturation at this coniferous forest ecosystem, and by inference to other such N-limited forests in Scandinavia.

Several studies have been carried out to examine nitrous oxide (N₂O) emissions from agricultural soils in the past. However, the emissions of N₂O particularly during amelioration of acidic soils have been rarely studied. We carried out the present study using a rice-rapeseed rotation soil (pH 5.44) that was amended with dolomite (0, 1 and 2 g kg⁻¹ soil) under 60% water filled pore space (WFPS) and flooding. N₂O emissions and several soil properties (pH, NH₄⁺N, NO₃⁻-N, and nosZ gene transcripts) were measured throughout the study. The increase in soil pH with dolomite application triggered soil N transformation and transcripts of nosZ gene controlling N₂O emissions under both water regimes (60% WFPS and flooding). The 60% WFPS produced higher soil N₂O emissions than that of flooding, and dolomite largely reduced N₂O emissions at higher pH under both water regimes through enhanced transcription of nosZ gene. The results suggest that ameliorating soil acidity with dolomite can substantially mitigate N₂O emissions through promoting nosZ gene transcription.

Antibiotics have been widely used in human and veterinary medicine to both treat and prevent disease. Due to their high water solubility and low bioavailability, many antibiotic residues have been found in aquatic environments. Fish are an indispensable link between the environmental pollution and human health. However, the chronic effects of environmental concentrations of antibiotics in fish have not been thoroughly investigated. Sulfamethoxazole (SMX) and oxytetracycline (OTC) are frequently detected in aquatic environments. In this study, zebrafish were exposed to SMX (260 ng/L) and OTC (420 ng/L) for a six-week period. Results indicated that exposure to antibiotics did not influence weight gain of fish but increased the metabolic rate and caused higher mortality when treated fish were challenged with Aeromonas hydrophila. Furthermore, exposure to antibiotics in water resulted in a significant decrease in intestinal goblet cell numbers, alkaline phosphatase (AKP), acid phosphatase (ACP) activities, and the anti-oxidant response while there was a significant increase in expression of inflammatory factors. Antibiotic exposure also disturbed the intestinal microbiota in the OTC-exposed group. Our results indicated that environmental antibiotic concentrations can impair the gut health of zebrafish. The potential health risk of antibiotic residues in water should be evaluated in the future.

The present experiment was conducted to explicate the genotoxic effects of profenofos, an organophosphate insecticide, on the erythrocytes of silver barb (Barbonymus gonionotus). Silver barb were exposed to a solution of 10% and 50% of lethal concentrations (LC₅₀) of profenofos as sub-lethal concentrations at different days (1, 7, 15, and 30 d), along with a control (0% profenofos). Subsequent recovery patterns were assessed allowing the fish exposed to profenofos free water for the same period that they were exposed to profenofos. Our results revealed that with the progression of time and concentration, fish exposed to profenofos showed significantly (p < .05) higher level of erythrocytic nuclear abnormalities (ENA) such as micronuclei, bi-nuclei, degenerated nuclei, notched nuclei, nuclear bridge and nuclear buds, as well as erythrocytic cellular abnormalities (ECA) such as echinocytic, elongated, fusion, spindle, tear-drop and twin shaped cells. After exposure, the silver barb recovered spontaneously, and the abnormal erythrocytic parameters were normalized with a concentration- and duration-dependent fashion. Therefore, these abnormalities and their recovery can be used to assess the toxic levels of pesticides on aquatic organisms. There is great potential to use this technique as in vivo to predict susceptibility of aquatic animals to environmental pollution.

In order to understand the impacts of regional emission changes and local tourism on sulfur and nitrogen wet deposition in Jiuzhaigou National Nature Reserve of southwestern China, wet deposition was monitored at a background site (Rize) and a tourist-affected site (PE: park entrance) in the reserve during 2015–2016. The observation data were compared between Rize and PE and between 2010–2011 and 2015–2016 monitoring campaigns. Also, the observation data were used in the Positive Matrix Factorization (PMF) model to identify the major sources of sulfur and nitrogen wet deposition. The results show that although local tourism emissions had considerable contributions to NH₄⁺, NO₂⁻, NO₃⁻, and SO₄²⁻ concentrations in wet deposition (p < 0.05), most of the annual Volume Weighted Mean (VWM) concentrations of these four ions were likely from emissions outside Jiuzhaigou. Annual wet deposition fluxes of the four ions were also affected more by precipitation and regional emissions than by local emissions. Although annual precipitation was higher at Rize (818 mm) during 2015–2016 than at another background site near Long Lake (LL: 752 mm) during 2010–2011, the annual concentrations and fluxes of SO₄²⁻ and NO₃⁻ wet deposition decreased by 77% and 74% for SO₄²⁻ and by 12% and 19% for NO₃⁻, respectively, most likely due to regional emission reductions. Similar large reductions in SO₄²⁻ and NO₃⁻ concentrations have been also found in some other sites in southwestern China. In contrast, the annual concentration and flux of NH₄⁺ wet deposition at Rize during 2015–2016 were 1.4 and 1.2 times of that measured at LL during 2010–2011, respectively. The results of source apportionment analysis and tour bus emission estimates suggest that elevated NH₄⁺ wet deposition was possibly related to NH₃ emissions from local tour buses, but additional studies on NH₃ emissions from tour buses in the reserve are needed to confirm this.

The effects of Nereis diversicolor bioturbation on the remobilization and bioavailability of polycyclic aromatic hydrocarbons from estuarine sediment were determined after 60 d in a laboratory experiment. The release fluxes and mass transfer coefficients showed that bioturbation by N. diversicolor can lead to a significant remobilization of polycyclic aromatic hydrocarbons (PAHs) from estuarine sediments. Bioturbation enhanced the release of PAHs from sediment to water by accelerating the transport of sediment particles to the sediment-water interface followed by PAHs desorption to the water. The bioavailability of PAHs was described by SPMD-sediment accumulation factors (SSAF). The SSAF of low molecular weight PAHs with bioturbation was significantly higher than that of PAHs without bioturbation, and there were no significant variations in high-molecular-weight PAHs. Our results revealed that N. diversicolor bioturbation significantly increased PAHs release from sediment to water but only increased the bioavailability of low-molecular-weight PAHs.

Methylisothiazolinone (MIT) has been widely used to control bacterial growth in reverse osmosis (RO) systems. However, MIT's toxicity on microalgae should be determined because residual MIT is concentrated into RO concentrate (ROC) and might have a severe impact on microalgae-based ROC treatment. This study investigated the tolerance of Scenedesmus sp. LX1 to MIT and revealed the mechanism of algal growth inhibition and toxicity resistance. Scenedesmus sp. LX1 was inhibited by MIT with a half-maximal effective concentration at 72 h (72 h-EC50) of 1.00 mg/L, but the strain recovered from the inhibition when its growth was not completely inhibited. It was observed that this inhibition's effect on subsequent growth was weak, and the removal of MIT was the primary reason for the recovery. Properly increasing the initial algal density significantly shortened the adaptation time for accelerated recovery in a MIT-containing culture. Photosynthesis damage by MIT was one of the primary reasons for growth inhibition, but microalgal cell respiration and adenosine triphosphate (ATP) synthesis were not completely inhibited, and the algae were still alive even when growth was completely inhibited, which was notably different from observations made with bacteria and fungi. The algae synthesized more chlorophyll, antioxidant enzymes of superoxide dismutase (SOD) and catalase (CAT), and small molecules, such as reduced glutathione (GSH), to resist MIT poisoning. The microalgae-based process could treat the MIT-containing ROC, since MIT was added for only several hours a week in municipal wastewater reclamation RO processes, and the MIT average concentration was considerably lower than the maximum concentration that algae could tolerate.

Persistent organic pollutants (POPs) such as dichlorodiphenyltrichloroethanes (DDTs), hexachlorobenzene (HCB), hexachlorocyclohexanes (HCHs), and polychlorinated biphenyls (PCBs) have been spreading to Antarctica for over half a century. Penguins are effective indicators of pelagic concentrations of POPs. We synthesized the literature on penguins to assess temporal trends of pelagic contamination in Antarctica, using fat and eggs to monitor changes from 1964 to 2011. DDT/DDE ratios suggest long-range atmospheric transport. Average DDT in fat (ww) increased from 44 ng g−1 in the 1960s, peaked at 171 ng g−1 in the mid-1980s, and then declined slowly to the present level of 101 ng g−1. Temporal trends in HCB contamination rose into the 1990s before declining. ∑HCHs in fat was ∼5 ng g−1 from 1960 to 1979, peaking at 33 ng g−1 during the period 1980–1989 before declining to ∼5 ng g−1 from 1990 to present. PCBs rose substantially from 1970 to 2009 in fat, varying more than DDTs and HCB in both fat and eggs. Antarctic penguins are good biological indicators of global DDT and HCB emissions, but the existing data are insufficient regarding HCHs and PCBs.

Difenoconazole, a typical triazole fungicide, inhibits lanosterol-14R-demethylase (CYP51) to prevent fungal sterol synthesis and its residues are frequently detected in the environment due to its wide application. Previous studies have demonstrated that difenoconazole altered the triglyceride levels, and gene expression relevant to cholesterol biosynthesis in zebrafish. However, endocrine-disruption in the hypothalamus-pituitary-gonadal-liver (HPGL) axis, the effects of transferring to offspring, and the underlying mechanisms of difenoconazole in aquatic organisms are still unknown. In this study, we defined the effects of difenoconazole at environmental concentrations on endocrine disturbance using zebrafish as an experimental model. The results indicated that difenoconazole induced a significant change in the somatic index, and pathological variations in tissues, and steroid hormone levels. RT-PCR experiments further confirmed that difenoconazole significantly induced expression alteration of lhr, hsd3β, hsd11β, cyp19a in the ovary and star, cyp19a, cyp3c1 in the testis, and erα genes in livers. In addition, difenoconazole exposure in parental zebrafish affected the hatchability and length of its offspring. Moreover, the burdens of difenoconazole and difenoconazole alcohol in females were higher than in males. These findings highlighted that difenoconazole exposure at environmentally relevant concentrations elicited estrogenic endocrine-disruption effects via altering homeostasis of sex steroid hormones in the HPGL axis and the adverse effects can be transferred to the offspring.

Studies are underway to gather information about the mode of action (MOA) of emerging pollutants that could guide practical environmental decision making. Previously, we showed that propranolol, an active pharmaceutical ingredient, had adverse effects on Daphnia magna that were similar to its pharmaceutical action. In order to characterize the mode of action of propranolol in D. magna, which is suspected to be organ-specific pharmaceutical action or baseline toxicity, we performed time-series monitoring of behavior along with heart rate measurements and nuclear magnetic resonance (NMR) based metabolite profiling. Principle component analysis (PCA) and hierarchical clustering were used to categorize the mode of action of propranolol among 5 chemicals with different modes of action. The findings showed that the mode of action of propranolol in D. magna is organ-specific and vastly different from those of narcotics, even though metabolite regulation is similar between narcotic and non-narcotic candidates. The method applied in this study seems applicable to rapid characterization of the MOA of other cardiovascular pharmaceutical ingredients.
