Detection of PSTVd [potato spindle tuber viroid] with 32P-cDNA probe by nucleic acid spot hybridization

More than one thousand samples of 32 potato varieties including TPS [True Potato Seed] parents, principal varieties, introduced germplasm, parents used in breeding and selected lines from main potato growing areas: Heilongjiang province, Inner Mongolia, Shandong and Yunnan province were tested by nucleic acid spot hybridization (NASH) using 32P-cDNA probe for infection of potato spindle tuber viroid (PSTVd). Potato cultivars Tiger Head and Lushu No. 1 were severely infected while infection in most other varieties was limited. Potato varieties Zhi-278, Hu-H1, Hu-H2, 7402-17, Dan 82-133,8205-2, 8212, 68-Red, CIP-24 were PSTVd-free; Kuannae, Purple Flower White, 292-20, Zhengshu No. 2, Harvest White and Mira in Yunnan province showed slight infection. Others that were infected by PSTVd to some extent, include Kexin No. 1, 2, 3, 4, 6, 7, Schwalbe, Mira in Wumeng, Tongshu No. 8 and Neishu No. 3.
