Molecular mapping of a thermosensitive genetic male sterility gene in rice

Bulked segregant analysis was employed in conjunction with RAPD (Randomly Amplified Polymorphic DNA) technique using a population developed from a cross IR32364 TGMS x IR68. Survey of 389 Operon random primers revealed a 5 RAPD fragments linked to the gene, of which 4 were sterility linked (OPF 18-2600, OPAA7-550, OPAC 1-300, OPB19-750) and 1 sterility linked (OPAC3-640). Futher analysis with the homozygous sterile and homozygous fertile individuals showed cosegregation of these markers with the TGMS trait. Use of OPF18-2600 and OPAC3-640 RAPD fragments as probe revealed polymorphism in southern analysis. Southern and RAPD analysis of progenies indicated that the TGMS gene is flanked by these markers on both sides. The OPAC3-640 fragment was mapped on the chromosome 6 using a doubled haploid mapping population. Therefore the TGMS gene is believed to be present in the chromosome. Cloning and sequencing of some of these fragments are being done in order to convert it in PCR marker for use in marker-assisted selection
