Influence of pre-equilibration time on the resumption and subsequent completion of first meiosis in cryopreserved bovine oocytes

This study examined the meiotic competence of immature bovine oocytes (n=166) following vitrification 24h post-culture in a 5% incubator at 39 deg C. Oocytes were assigned to 4 treatment groups (T1 - 1 min, T2 - 3 min, T3 - 5 min, and T4 - 10 min) as to the lenght of pre-equilibration in Vitrified Solution A (VSA) and a control group. After thawing and culture, 9 out of 33 oocytes in T3 underwent germinal vesicle breakdown stage (87.88%), in which 9 (30.03%) of these progressed to the M-11 stage of the first meiotic division as compared to 71.43% maturation rate in the control group. Morphological appearance of vitrified oocytes post-thawing was also noted. It was found out that the incidence of good oocytes after thawing was evident in T3 (87.9%) followed by T4 (84.8%). Moreover, all of the treatment groups presented moderate cumulus cell expansion indicating that expansion do not correlate with the completion of nuclear maturation
