Molecular cloning of the movement protein gene of abaca bunchy top nanovirus (ABTV)

Abaca bunchy top disease caused by the virus ABTV is the most devastating viral disease of abaca in the Philippines. Efforts to control ABTV are geared towards crop improvement, production and protection while very little work has been done on the molecular aspects of the disease. Characterization and cloning of the different ABTV viral genes could be very useful in the design and implementation of control strategies. One such viral gene is the movement protein gene. Total DNA was extracted from leaves infected with a local ABTV isolate using a modified CTAB extraction method. Oligonucleotide primers based on published sequence of DNA component 4 of a related virus, banana bunchy top virus (BBTV) were synthesized. The primers target gene fragments encoding for the movement protein of the virus which could be part in the ABTV disease development. The 380 bp fragment generated using polymerase chain reaction (PCR) was cloned and sequenced. Preliminary sequence showed that the ABTV movement protein gene is 96% homologous to BBTV movement protein gene. Further sequence analysis, hybridization and gene expression studies are currently being done for the characterization of the gene. Additional data will help elucidate the biology of ABTV virus-host interactions and relationships between ABTV and BBTV.
