Kinetic study on the production of rennet via aerobic fermentation of wheat bran by Rhizopus chinensis Saito Biotech 3273

Established growth models were investigated to describe the kinetics of aerobic batch fermentation of wheat bran by Rhizopus chinensis BIOTECH 3273 for the production of microbial rennet. The maximum number of back-mix fermentors for continuous fermentation was determined. Preliminary material flow analysis was evaluated. The exponential growth of the fungus started 6 hours after the introduction of the microorganism. Biomass yield reached its peak at 15 to 18 hours of fermentation while sugar concentration rapidly declined until after 10 to 12 hours. Biomass production and the rate of sugar depletion were significantly lower int he 2 percent wheat bran than in the 4 and 6 percent substrate. No significant differences were found between the 4 and 6 percent concentration. The growth models of Monod, Tessier, Moser, and Contois were found not applicable to describe the kinetics of fermentation. The Andrews equation which accounts for the effect of substrate inhibition provided the best description. The growth parameters calculated were 4.12/hr, 1.64 mg/mL, and 0.09 mg/mL for Umax, Ks, and K1, respectively. The maximum number of backmix fermentors obtained for continuous fermentation of the 2, 4, and 6 percent substrate concentration were 7, 9 and 13, respectively. No waste product was identified during fermentation.
