Recombinant galectins from Hemonchus contortus inhibit goat cytokine mRNA transcription of peripheral blood mononuclear cells in vitro

Chinese. 以体外试验观察雌雄捻转血矛线虫半乳糖凝集素重组蛋白(rHco-gal-m/f)对山羊外周血单核细胞多种细胞因子mRNA转录水平的影响及其量效关系。选用2岁健康山羊5只,分别从颈静脉无菌采血,分离外周血单核细胞(PBMCs),以ConA或LPS刺激的同时分别加入0 μg•ml-1,10 μg•ml-1、20 μg•ml-1和40μg•ml-1的rHco-gal-m/f,进行体外培养。用RT-PCR方法,以肌动蛋白基因(β-actin)作内标,定量分析单核淋巴细胞白介素-1β(IL-1β)、白介素-2(IL-2)、白介素-4(IL-4)、白介素-6(IL-6)、干扰素-γ(IFN-γ)和肿瘤坏死因子-α(TNF-α)的mRNA丰度。重组雄性捻转血矛线虫半乳糖凝集素(rHco-gal-m)可以抑制上述细胞因子mRNA的转录,并呈现出明显的剂量依赖性;重组雌性捻转血矛线虫半乳糖凝集素(rHco-gal-f)可抑制IL-1β、IL-4、IFN-γ and TNF-α的转录,也呈现出明显的剂量依赖性。重组捻转血矛线虫半乳糖凝集素能降低多种细胞因子在体外的转录。

The effects of recombinant galectins of male and female Hemonchus contortus (rHco-gal-m/f) on the mRNA levels of IL-1β, IL-2, IL-4, IL-6, interferon-γ (IFN-γ) and tumor necrosis factor-α (TNF-α) of goat peripheral blood mononuclear cells (PBMCs) were examined in this study. Blood samples were collected from five, randomly selected two year old healthy goats. PBMCs were separated and cultured in vitro with ConA or LPS and varying concentrations of rHco-gal-m/f at 0 μg•ml-1, 10 μg•ml-1, 20 μg•ml-1 and 40 μg•ml-1. Semi-quantitative reverse transcription (RT)-PCR was employed to test the synthesis of mRNA. The mRNA transcriptions of IL-1β, IL-2, IL-4, IL-6, IFN-γ and TNF-α in the PBMCs were inhibited by rHco-gal-m in a dose-dependent manner. The rHco-gal-f inhibited mRNA synthesis of IL-1β, IL-4, IFN-γ and TNF-α of the same cells in similar fashion. The findings suggested that the recombinant galectin proteins of Hemonchus contortus could decrease the transcription of cytokines in vitro.
