Organisation de la chromatine et transcription au cours du développement embryonnaire précoce des mammifères

French. Au cours de mon parcours, mes recherches ont dans un premier temps eu pour objectif de comparer les chromosomes de différentes espèces de mammifères par des techniques de marquages en bandes et d'hybridation en fluorescence. Ces informations m'ont permis de clarifier les relations phylogénétiques entre certains taxons et de discuter de la barrière d'espèces. Ensuite, toujours par des méthodes de cytogénétique classique et moléculaire, j'ai étudié l'impact de la présence d'une anomalie chromosomique équilibrée sur la ségrégation des chromosomes lors de la méiose mâle ou femelle. J'ai notamment pu estimer le pourcentage de spermatozoïdes et d'ovocytes aneuploïdes chez les bovins porteurs de la translocation 1/29. Mes recherches actuelles visent à comprendre les mécanismes épigénétiques et moléculaires intervenant lors de deux étapes essentielles du développement embryonnaire précoce : l'activation du génome embryonnaire et le passage de l'état totipotent à pluripotent, chez les mammifères (souris, lapin, bovin). Dans ce cadre, je m'intéresse aux liens entre l'organisation de la chromatine et la régulation de la transcription de séquences qui la composent. A cette fin, j'ai conduit la mise au point d'approches qui permettent des analyses en cellule unique de l'organisation de la chromatine (ADN et histones) et de la localisation des transcrits (ARN) tout en conservant la structure tridimensionnelle du noyau des embryons (immunomarquage de protéines et hybridation in situ en fluorescence). Les séquences satellites majeures, spécifiques de l'hétérochromatine péricentromérique, ont une organisation spatiale relâchée et transcrivent fortement avant la mise en route du génome puis s'agrègent et transcrivent très peu par la suite. A l'inverse, j'ai pu montré que les séquences des gènes ribosomiques sont dans un état agrégé et ne sont pas transcrites avant la mise en route du génome puis sont de plus en plus dispersées quand elles sont transcrites. Ainsi l'organisation spatiale (relâchée - agrégée) semble corrélée avec l'état de transcription (actif - inactif) de ces séquences entre le stade 1-cellule et le stade morula chez la souris.

English. During my career, my research initially focused on comparing chromosomes from different mammalian species using banding and fluorescence hybridization techniques. This information allowed me to clarify the phylogenetic relationships between these taxa and to discuss the species barrier. Then, still by classical and molecular cytogenetics methods, I studied the impact of the presence of a balanced chromosomal abnormality on the segregation of chromosomes during male or female meiosis. In particular, I was able to assess the percentage of aneuploid spermatozoa and oocytes in cattle carrier of the 1/29 translocation. My current research aims to understand the epigenetic and molecular mechanisms involved in two essential steps of early embryonic development: embryonic genome activation and the transition from totipotency to pluripotency in mammals (mouse, rabbit, cattle). In this context, I am interested in the links between the organization of chromatin and the regulation of the transcription of sequences that compose it. To this end, I directed the development of approaches that allow single-cell analyzes of the organization of chromatin (DNA and histones) and the localization of transcripts (RNA) while maintaining the three-dimensional structure of the nucleus of embryos (protein immunolabeling combined with fluorescence in situ hybridization). The major satellite sequences specific of pericentromeric heterochromatin have a relaxed spatial organization and are strongly transcribed before the embryonic genome activation and then aggregate and are transcribed very little thereafter. Conversely, I was able to show that ribosomal gene sequences are in an aggregated state and are not transcribed before genome activation, and are in more and more dispersed state when they are transcribed. So, the spatial organization (relaxed - aggregated) seems correlated with the (active - inactive) transcription state of these sequences between the 1-cell stage and the morula stage in mice.
