Shoot culture requirements of rambutan (Nephelium lappaceum)

Tissue culture requirements of rambutan were investigated. Initial explants and nodal cuttings were derived from aseptic seedlings which were germinated on nutrient solution (Peter's fertilizer, 20-20-20). The board spectrum experiment consisted of 196 treatment combinations of mineral nutrient (1/2 and 1 x MS), sucrose (2 and 4 percent), indole acetic acid at 0; 1.0; 10 and 100 mg L at the power of -1 or 2,4 dichlorophenoxyacetic acid at 0.10; 1.0 and 10 mg L at the power of -1 and cytokinis (BA and Kinetin) at 0; 0.10; 1.0 and 10 mg L at the power of -1 growth of shoots was observed on 1/2 x MS, 2 percent sucrose, 0-10 mg L at the power of -1 IAA and 0-10 mg L at the power of -1 BA or kinetin. In subsequent experiment, shoot proliferation could be increased to 87 percent on 1/4 x MS, 2 percent sucrose, 1 mg L at the power of -1 and 0.1 mg L at the power of -1 IAA.
